[Studies on the evaluation of exposure to industrial chemicals].

Among the biological exposure indices of lead, lead in plasma was the most direct indicator of current exposure. Lead mobilized into plasma as well as in urine could be used as an indicator of the internal dose of lead. The ratio of non-treated to restored activity of delta-aminolevulinic acid dehydratase (ALA-D) was a more specific index than ALA-D activity itself at low levels of lead exposure, excluding the familial or genetic variation in the activity. The methods using HPLC for determining heme intermediate improved the evaluation of the lead effect: delta-aminolevulinic acid in plasma, b

Sulfate homeostasis. I. Effect of salicylic acid and its metabolites on inorganic sulfate in rats.

Homeostasis of inorganic sulfate, a physiologic anion necessary for both detoxification and biosynthetic reactions, involves predominantly capacity-limited renal clearance mechanisms. The objective of this investigation was to examine the effect of salicylic acid (SA) and its major metabolites, salicyluric acid and salicyl phenolic glucuronide, on the serum concentrations and renal clearance of inorganic sulfate in rats. Animals were studied using a crossover design in which they received a bolus i.v. injection (75 mg/kg) and infusion (approximately 0.26 mg/min/kg) of SA or the same volume of

Rapid and sensitive determination of acetylsalicylic acid and its metabolites using reversed-phase high-performance liquid chromatography.

A rapid and sensitive high-performance liquid chromatographic technique was developed for the simultaneous determination of gentisic acid, salicyluric acid, acetylsalicylic acid and salicylic acid in plasma and serum. The method involved a single deproteinization step and separation using a reversed-phase column eluted with a buffered methanol (35%) mobile phase. Detection was achieved with a variable-wavelength ultraviolet detector set at 235 nm and a given chromatographic analysis could be completed in less than 10 min. The method was tested in both human and animal (rat) models given a sing

Comparative aspirin absorption kinetics after administration of sodium- and potassium-containing buffered solutions.

Twelve fasting normal volunteers received three aspirin dosage forms in a single 325-mg dose in a complete crossover study; the plasma aspirin and salicylic acid levels and the urine salicylic acid and salicyluric acid levels were measured over 10 h. The three dosage forms included an unbuffered tablet and two effervescent solutions, one with sodium bicarbonate-citrate buffer and the other with potassium bicarbonate-citrate buffer. A significantly faster absorption rate was observed with the sodium bicarbonate-citrate buffer, when compared with the potassium bicarbonate-citrate buffer and the

Evaluation of metabolite profiles as biomarkers for the pharmacological effects of thiazolidinediones in Type 2 diabetes mellitus patients and healthy volunteers.

WHAT IS ALREADY KNOWN ABOUT THIS SUBJECT: * Many studies have investigated the effects of thiazolidinediones on isolated biochemical markers (biomarkers) or sets of markers in Type 2 diabetes mellitus (T2DM) patients and healthy volunteers. * However, a limited number of parameters is not capable of capturing the broad response to pharmacological intervention with these types of (pleiotropic) drugs, which are known to activate the nuclear transcription factor peroxisome proliferator activated receptor gamma (PPARgamma). * Our study tested the new hypothesis (primary objective) that nuclear mag

Sex differences in salicylic acid metabolism in streptozotocin induced diabetes in rats.

The metabolism of salicylic acid was studied in male and female streptozotocin-induced diabetic Wistar rats. Results obtained showed that in both sexes there was a significant increase in urinary excretion of salicyluric acid in diabetic rats when compared to controls (P less than or equal to 0.001). Within the diabetic groups, there was a significant increase in the urinary excretion of salicyluric acid in the female in comparison to the male rats (P less than or equal to 0.01). A statistically significant increase was observed in urinary excretion of salicyl-glucuronic acid in diabetic femal

A Narrative Review of Candidate Biomarkers of Total Fruit and Vegetable Intake.

While an abundance of cohort studies has suggested that greater consumption of fruits and vegetables (FVs) is associated with reduced risk of chronic diseases, current self-report based dietary assessment tools are susceptible to biases that can reduce their accuracy in estimating dietary intakes, and consequently, the risk estimates obtained from this research. As a result, considerable attention has been directed towards the identification and validation of dietary biomarkers, which represent a more objective method for estimating FV intake and have the potential to improve the validity of f

Effect of End-Tethered Methoxy-PEO Chain Density on Uremic Toxin Adsorption.

In 2023, around 850 million people globally were affected by chronic kidney disease, which leads to the retention of uremic toxins and excess fluid in the blood. This study examines the adsorption of these toxins to poly(ethylene oxide) (PEO) films, known for their low-fouling properties. The gold surfaces were treated with 5 mM end-thiolated methoxy-terminated PEO (m-PEO) and analyzed using dynamic contact angle measurements, X-ray photoelectron spectroscopy, and spectroscopic ellipsometry to confirm the PEO film's presence and determine chain density. The adsorption of 25 different uremic to

Systemic responses of BALB/c mice to Salmonella typhimurium infection.

Salmonella typhimurium is a bacterial pathogen that poses a great threat to humans and animals. In order to discover hosts' responses to S. typhimurium infection, we collected and analyzed biofluids and organ tissues from mice which had ingested S. typhimurium. We employed (1)H NMR spectroscopy coupled with multivariate data analysis and immunological techniques. The results indicate that infection leads to a severe impact on mice spleen and ileum, which are characterized by splenomegaly and edematous villi, respectively. We found that increased levels of itaconic acid were correlated with the

Impact of coexposure on toluene biomarkers in rats.

1. Toluene (TOL) is widely used in industry. Occupational exposure to TOL is commonly assessed using TOL in blood, hippuric acid and ortho-cresol. Levels of these biomarkers may depend on factors potentially interfering with TOL biotransformation, such as the presence of other solvents in the workplace. Mercapturic acids (MAs) could be an alternative to the "traditional" TOL biomarkers. 2. This study aims (1) to investigate in rat the effects of an exposure to vapours mixtures on the TOL metabolism, and (2) to assess how well MAs performed in these contexts compared to the traditional TOL biom

Development of a thin-layer chromatography-angiotensin-converting enzyme bioassay coupled with digital image analysis for effect-directed screening of enzyme inhibitors.

A novel thin-layer chromatography (TLC)-based bioassay coupled with digital image analysis (DIA) was developed for rapid and non-targeted screening of angiotensin-converting enzyme (ACE) inhibitors in complex natural matrices. Conventional ACE inhibitory assays lack spatial resolution and rely on indirect measurements, limiting their applicability for effect-directed analysis (EDA). In this study, an on-surface enzymatic assay was implemented using hippuryl-histidyl-leucine (HHL) as substrate, followed by in situ derivatization of the generated hippuric acid with p-dimethylaminobenzaldehyde (D

Lymphocyte Phenotypes and Protein-Bound Uremic Toxins as Determinants of Clinical Outcomes in Hemodialysis Patients.

The impact of protein bound uremic toxins (PBUTs) and lymphocyte alterations in morbidity and mortality in patients on hemodialysis (HD) is of great concern. The aim of this study was the assessment of association between PBUTs, immunosenescent lymphocytes' phenotype and clinical events [cardiovascular, severe infections (hospitalization due to infection, respiratory infection), all-cause mortality] during 2-year follow-up. In this prospective observational study, lymphocytes' phenotype of 54 patients on HD and 31 age-matched controls was analyzed by flow cytometry, and simultaneously, PBUT se

Evaluation of synergistic factors on BTEX urinary biomarkers in motorcycle riders exposed to heavy traffic in a megacity.

Fuel combustion is a major source of volatile organic compounds such as benzene, a known human carcinogen. Benzene is metabolized into intermediate biomarkers that can be used to assess the extent of exposure. This study aimed to investigate the association between urinary concentrations of creatinine (Cr), muconic acid (MA), trans, trans-muconic acid (tt-MA), and hippuric acid (HA) with demographic, lifestyle, and occupational factors among motorcyclists exposed to heavy traffic in the Shiraz metropolis, both before and after shift work. Data on demographic characteristics, lifestyle habits,

Plug-and-play metabolic transducers expand the chemical detection space of cell-free biosensors.

Cell-free transcription-translation systems have great potential for biosensing, yet the range of detectable chemicals is limited. Here we provide a workflow to expand the range of molecules detectable by cell-free biosensors through combining synthetic metabolic cascades with transcription factor-based networks. These hybrid cell-free biosensors have a fast response time, strong signal response, and a high dynamic range. In addition, they are capable of functioning in a variety of complex media, including commercial beverages and human urine, in which they can be used to detect clinically rel
