Novel insights into an old disease: recent developments in scabies mite biology.

PURPOSE OF REVIEW: Scabies is a serious disease of both humans and other animals caused by infestation of the skin with the ectoparasitic mi … disease control programmes. Other studies have begun to decipher the molecular basis of the complex host-parasite interactions underlying scabies infestations. Scabies mites have developed complex mechanisms to interfere with the host defence processes that may also enhance the survival of the associated skin microbiome, consistent with the epidemiological evidence. Recentl …

Non-genetic red blood cell variability can modulate hemoglobin consumption by intracellular Plasmodium falciparum.

Natural, non-genetic variation among host cells can shape infection outcomes, yet its role in intracellular parasite dynamics remains poorly … arasite feeding rates. This perspective reframes parasite Hb consumption as an emergent property of host-parasite interactions. More broadly, it highlights how natural variability among host cells can influence the course of intracellular infections, with potential implications for predicting pathogen growth and optimizing drug treatment strategies.

Decoding Leukocyte Dynamics: Functional Biomarkers and Precision Diagnostics in Parasitic Infections.

BACKGROUND: Parasitic infections affect over 1.5 billion people globally, with leukocytes serving as critical mediators of both protective immunity and immunopathology. Traditional diagnostic approaches relying on static cell counts lack specificity and fail to capture the functional complexity of host-parasite interactions. OBJECTIVE: This review synthesizes cutting-edge evidence on how leukocyte functional dynamics, encompassing transcriptional, metabolic, and spatial profiles, revolutionize diagnostic precision, prognostic stratification, and therapeutic development in parasitic diseases. …

How Apicomplexa Parasites Secrete and Build Their Invasion Machinery.

Apicomplexa are obligatory intracellular parasites that sense and actively invade host cells. Invasion is a conserved process that relies on the timely and spatially controlled exocytosis of unique specialized secretory organelles termed micronemes and rhoptries. Microneme exocytosis starts first and likely controls the intricate mechanism of rhoptry secretion. To assemble the invasion machinery, micronemal proteins-associated with the surface of the parasite-interact and form complexes with rhoptry proteins, which in turn are targeted into the host cell. This review covers the molecular advan

Parasite co-infection: an ecological, molecular and experimental perspective.

Laboratory studies of pathogens aim to limit complexity in order to disentangle the important parameters contributing to an infection. However, pathogens rarely exist in isolation, and hosts may sustain co-infections with multiple disease agents. These interact with each other and with the host immune system dynamically, with disease outcomes affected by the composition of the community of infecting pathogens, their order of colonization, competition for niches and nutrients, and immune modulation. While pathogen-immune interactions have been detailed elsewhere, here we examine the use of ecol

Virulent Disease Epidemics Can Increase Host Density by Depressing Foraging of Hosts.

AbstractAll else equal, parasites that harm host fitness should depress densities of their hosts. However, parasites that alter host traits may increase host density via indirect ecological interactions. Here, we show how depression of foraging rate of infected hosts can produce such a hydra effect. Using a foraging assay, we quantified reduced foraging rates of a zooplankton host infected with a virulent fungal parasite. We then parameterized a dynamical model of hosts, parasites, and resources with this foraging function, showing how foraging depression can create a hydra effect. Mathematica

Molecular Mechanisms of Clonorchis sinensis-Host Interactions and Implications for Vaccine Development.

Infections caused by Clonorchis sinensis remain a significant public health challenge for both humans and animals, causing pyogenic cholangitis, cholelithiasis, cholecystitis, biliary fibrosis, and even cholangiocarcinoma. However, the strategies used by the parasite and the immunological mechanisms used by the host have not yet been fully understood. With the advances in technologies and the accumulated knowledge of host-parasite interactions, many vaccine candidates against liver flukes have been investigated using different strategies. In this review, we explore and analyze in-depth the imm…

Host specificity of gastrointestinal parasites in free-ranging sloths from Costa Rica.

The diversity and host specificity of gastrointestinal parasites infecting free-ranging sloths is poorly known. We compared gastrointestinal parasites of two sloth species from Costa Rica-three-fingered sloths (Bradypus variegatus) and two-fingered sloths (Choloepus hoffmanni)-for the first time in both a primary forest and an urban habitat. We asked whether host-parasite interactions were predominantly structured by host identity, the habitats in which hosts occurred, or both. Coproparasitology revealed protozoa and nematode eggs from both host species, but cestode eggs were recorded only in …

Human Acanthocephaliasis: a Thorn in the Side of Parasite Diagnostics.

Acanthocephala is a phylum of parasitic pseudocoelomates that infect a wide range of vertebrate and invertebrate hosts and can cause zoonotic infections in humans. The zoologic literature is quite rich and diverse; however, the human-centric literature is sparse, with sporadic reports over the past 70 years. Causal agents of acanthocephaliasis in humans are reviewed as well as their biology and life cycle. This review provides the first consolidated and summarized report of human cases of acanthocephaliasis based on English language publications, including epidemiology, clinical presentation,

The influence of selected gastrointestinal parasites on apoptosis in intestinal epithelial cells.

Studies on the parasite-host interaction may provide valuable information concerning the modulation of molecular mechanisms as well as of the host immune system during infection. To date, it has been demonstrated that intestinal parasites may affect, among others, the processes of digestion in the gastrointestinal system of the host, thus limiting the elimination of the parasite, the immune response as well as inflammation. However, the most recent studies suggest that intestinal parasites may also affect modulation of the apoptosis pathway of the host. The present paper presents the latest sc

Is SELDI-TOF a valid tool for diagnostic biomarkers?

The genome revolution is providing fresh insights into host and parasite genomes, and new tools are becoming available for examining host-parasite interactions at the proteome level. Technologies such as surface-enhanced laser desorption/ionization time-of-flight (SELDI-TOF) mass spectrometry (MS) can be applied to discover biomarkers (alterations in both host and parasite proteomes) associated with parasitic diseases. Such biomarkers can represent host proteins, fragments of host proteins or parasite proteins that appear in body fluids or tissues following infection. Individual biomarkers or …

The role of autophagy in protozoan infections.

Autophagy is a conserved, lysosome-dependent degradation system that is used by a wide variety of eukaryotes; during autophagy, intracellular substances are transported to lysosomes for degradation, and thus, autophagy plays a crucial role in cell survival under stress conditions such as starvation and hypoxia. Additionally, cells can eliminate foreign pathogens, such as parasites, bacteria, and viruses, via autophagic clearance. Protozoa are categorized as either intracellular parasitic protozoa or extracellular parasites, which are mostly zoonotic pathogens that pose significant threats to p

Phenotyping Toxoplasma Invasive Skills by Fast Live Cell Imaging.

Host cell invasion by Toxoplasma gondii/T. gondii tachyzoites is an obligate but complex multistep process occurring in second-scale. To capture the dynamic nature of the whole entry process requires fast and high-resolution live cell imaging. Recent advances in T. gondii/host cell genome editing and in quantitative live cell imaging-image acquisition and processing included-provide a systematic way to accurately phenotype T. gondii tachyzoite invasive behaviour and to highlight any variation or default from a standard scenario. Therefore, applying these combined strategies allows gaining deep

Exploring Structural and Physical Properties of Schistosome Eggs: Potential Pathways for Novel Diagnostics?

In this era of increasing demand for sensitive techniques to diagnose schistosomiasis, there is a need for an increased focus on the properties of the parasite eggs. The eggs are not only directly linked to the morbidity of chronic infection but are also potential key targets for accurate diagnostics. Eggs were the primary target of diagnostic tools in the past and we argue they could be the target of highly sensitive tools in the future if we focus on characteristics of their structure and shell surface that could be exploited for enhanced detection. In this review, we discuss the current sta

Porpostoma guamensis n. sp., a philasterine scuticociliate associated with brown-band disease of corals.

Brown band disease of coral is caused by a ciliate that consumes the tissue of the corals in the genus Acropora. We describe the ciliate associated with this disease on Guam, based on: general morphology, division stages, and ciliature observed on live and protargol-stained specimens; modification of the oral structures between divisional stages, observed on protargol-stained specimens; and some aspects of behavior in field and laboratory studies. Porpostoma guamensis n. sp. is elongate and has ciliature typical for the genus; live cells are 70-500 × 20-75 μm; the macronucleus is sausage-like,

Constraints on host use by a parasitic plant.

Consumers do not always utilize all suitable hosts. Understanding why parasitic plants do not always parasitize potentially suitable hosts requires a better understanding of the constraints that limit host use by parasitic plants. In Texas salt marshes, the parasitic plant Cuscuta indecora rarely parasitizes three hosts that support vigorous growth in the greenhouse. We identified three constraints on host use by C. indecora. First, a mismatch between the phenology of C. indecora and some suitable hosts meant that these hosts were not abundant when C. indecora was growing most vigorously, and

Host-Pathogen Interactions in Malaria: Invasion, Neutralization, and Evasion.

Malaria remains a major global health burden despite decades of effort toward its prevention and treatment. Much of this difficulty stems from the complexity of the Plasmodium parasite's life cycle and its interactions with the human host. Here, we review receptor-ligand interactions between the Plasmodium parasite and human host, with a focus on clinically relevant antigens. The parasite uses a variety of proteins at each stage of its life cycle to achieve requisite functions such as motility, cell traversal, and invasion. In turn, the human immune response targets these key parasite antigens
