Constructing High-Efficiency Polyoxometalate-Based Antibacterial Hydrogels for Wearable Sensors.

Conductive flexible hydrogel are widely used in wearable electronics owing to its desired conductivity, flexibility, adhesion, and mechanical properties similar to human tissue. Nevertheless, conductivity and bacterial infections are always critical issues for the long-term use of hydrogel wearable sensors. Herein, a multifunctional polyoxometalate-based hydrogel with both antibacterial and sensing performances are prepared by integrating polydopamine-functionalized polyoxometalates (POMs) particles into polyacrylamide matrix. To obtain rapid gelation times (to seconds), a dual autocatalytic s

Micelle-to-Gel: Thermosensitive intra-articular hydrogels for osteoarthritis management.

Osteoarthritis (OA) is a chronic and degenerative joint disease with a rising incidence worldwide. Current therapeutic approaches primarily focus on symptom relief through systemic administration, which raises safety concerns related to side effects and long-term use. In this context, the local administration of natural compounds with anti-inflammatory and anti-arthritic properties, such as β-Lapachone constitutes an interesting alternative. In this work, we prepared and characterized injectable thermosensitive hybrid hydrogels loaded with β-Lapachone. A comprehensive characterization of the h…

Thermoresponsive Hydrogels with Tunable Viscoelasticity for Extended Ocular Drug Delivery.

The conformation of charged polymers is heavily dependent on interactions with the surrounding salt ions. By adding calcium ions to hyaluronan-Pluronic F-127 complexes, we prepared colloidal coacervates with decreased viscosity at cold temperatures (G' of 10 Pa) and enhanced mechanical properties at physiological temperatures (G' of 19 kPa) that improved the stability of the gel and prolonged drug release. The release of ophthalmic drugs, brinzolamide, dorzolamide, and dexamethasone, was tuned from 2 weeks to four months by changing drug lipophilicity. The low-viscosity hydrogel enabled topica

Smart Polymer-Derived Injectable Hydrogels: Current Status and Future Perspectives.

Because of their tissue-conforming qualities, in situ gelation, and less invasive distribution, injectable hydrogels (IHs) have become a revolutionary class of soft materials with enormous potential in biomedical applications. The ability of stimuli-responsive polymer-derived smart injectable hydrogels (SIHs) to react dynamically to external stimuli like temperature, redox potential, pH, or enzyme activity has drawn more attention than any other. This responsiveness enables precise spatiotemporal control over therapeutic delivery, tissue regeneration, and self-healing capabilities. Recent adva…

Granular polyrotaxane microgels as injectable hydrogels for corneal tissue regeneration.

Corneal diseases, a leading cause of global vision impairment, present challenges in treatment due to corneal tissue donor scarcity and transplant rejection. Hydrogel biomaterials in the form of corneal implants for tissue regeneration, while promising, have faced obstacles related to cellular and tissue integration. This study develops and investigates the potential of granular polyrotaxane (GPR) hydrogels as a scaffold for corneal keratocyte growth and transparent tissue generation. Employing host-guest driven supramolecular interactions, we developed injectable, cytocompatible hydrogels. By…

Modeling Cystic Fibrosis Chronic Infection Using Engineered Mucus-like Hydrogels.

The airway mucus of patients with cystic fibrosis has altered properties, which create a microenvironment primed for chronic infections that are difficult to treat. These complex polymicrobial airway infections and corresponding mammalian-microbe interactions are challenging to model in vitro. Here, we report the development of mucus-like hydrogels with varied compositions and viscoelastic properties reflecting differences between healthy and cystic fibrosis airway mucus. Models of cystic fibrosis and healthy airway microenvironments were created by combining the hydrogels with relevant pathog…

Synergistic Dual Antibacterial Activity of Magnetite Hydrogels Doped with Silver.

In this work, we utilized poly-N-isopropylacrylamide (NIPAM), magnetic nanoparticles (MNPs), and silver nitrate to prepare magnetic hydrogel microparticles doped with silver, which exhibited a dual antimicrobial effect. The antibacterial effect of these composites was mediated by the antimicrobial activity of silver and the magnetic hyperthermic induction, which we believe increased biofilm disruption and silver release into the surrounding bacterial biofilms. The prepared particles were characterized by using several analytical techniques. The particles exhibited a porous morphology impregnat

Permselectivity of Silk Fibroin Hydrogels for Advanced Drug Delivery Neurotherapies.

A promising trend in tissue engineering is using biomaterials to improve the control of drug concentration in targeted tissue. These vehicular systems are of specific interest when the required treatment time window is higher than the stability of therapeutic molecules in the body. Herein, the capacity of silk fibroin hydrogels to release different molecules and drugs in a sustained manner was evaluated. We found that a biomaterial format, obtained by an entirely aqueous-based process, could release molecules of variable molecular weight and charge with a preferential delivery of negatively ch…

Soft Polyethylene Glycol Hydrogels Support Human PSC Pluripotency and Morphogenesis.

Lumenogenesis within the epiblast represents a critical step in early human development, priming the embryo for future specification and patterning events. However, little is known about the specific mechanisms that drive this process due to the inability to study the early embryo in vivo. While human pluripotent stem cell (hPSC)-based models recapitulate many aspects of the human epiblast, most approaches for generating these 3D structures rely on ill-defined, reconstituted basement membrane matrices. Here, we designed synthetic, nonadhesive polyethylene glycol (PEG) hydrogel matrices to bett

Facile photopatterning of perfusable microchannels in hydrogels for microphysiological systems.

Perfusable hydrogels have garnered substantial attention in recent years for the fabrication of microphysiological systems. However, current methodologies to fabricate microchannels in hydrogel platforms involve sophisticated equipment and techniques, which hinder progress of the field. In this protocol, we present a cost-effective, simple, versatile and ultrafast method to create perfusable microchannels of complex shapes in photopolymerizable hydrogels. Our method uses one-step UV photocross-linking and a photomask printed on inexpensive transparent films, to photopattern both synthetic (PEG…

Electro-responsive hyaluronic acid-based click-hydrogels for wound healing.

With the aim of healing challenging skin wounds, electro-responsive click-hydrogels made of hyaluronic acid (clickHA) crosslinked with a modified polyethylene glycol precursor (PEG) were prepared by semi-interpenetrating a conducting polymer, poly(hydroxymethyl-3,4-ethylenedioxythiophene) (PEDOT-MeOH) by oxidative polymerization. The porosity and pore size of the mixed hydrogel, clickHA/PEDOT-MeOH, were both higher than those determined for the hydrogel without PEDOT-MeOH, while a honeycomb-like morphology with PEDOT-MeOH covering the pore walls was observed. Although such PEDOT-MeOH-induced c…

Predicting Fatigue Damage in Hydrogels Through Force-Induced Luminescence Enhancement.

Fatigue damage of polymers occurs under long-term load cycling, resulting in irreversible fracture failure, which is difficult to predict. The real-time monitoring of material fatigue damage is of great significance. Here, tough hydrogels are prepared with force-induced confined luminescence enhancement of carbonated polymer quantum dot (CPD) clusters to realize the visualization of fracture process and the monitoring of fatigue damage. The enhanced interactions induced by force between the clusters and the polymer in the confined space inhibit the non-radiative leaps and promote the radiative…

Programmable adhesion and morphing of protein hydrogels for underwater robots.

Soft robots capable of efficiently implementing tasks in fluid-immersed environments hold great promise for diverse applications. However, it remains challenging to achieve robotization that relies on dynamic underwater adhesion and morphing capability. Here we propose the construction of such robots with designer protein materials. Firstly, a resilin-like protein is complexed with polyoxometalate anions to form hydrogels that can rapidly switch between soft adhesive and stiff non-adhesive states in aqueous environments in response to small temperature variation. To realize remote control over…

Monophasic hyaluronic acid-silica hybrid hydrogels for articular cartilage applications.

Hyaluronic acid (HA), an FDA-approved natural polymer and important component of the extracellular matrix (ECM), has been widely used to develop hydrogels for cartilage regeneration. However, HA hydrogels often exhibit poor mechanical properties and unsuitable degradability, limiting their capability to support cell growth in cartilage. To overcome these challenges, this study modifies HA with a silica precursor and the coupling agent (3-Glycidyloxypropyl) trimethoxysilane (GPTMS) to develop a monophasic organic-inorganic hybrid HA-silica hydrogel. In this system, the inorganic silicate and or…

Rapid in situ forming PEG hydrogels for mucosal drug delivery.

In situ gelling polymeric biomaterials have proven useful as drug delivery vehicles to enable sustained release at sites of disease or injur … inistered. We identified a set of 6 lead formulations, which rapidly form into disulfide-linked PEG hydrogels in 30 seconds or less. These rapidly forming PEG hydrogels were also able to conform and adhere to mucosal tissues via PEG-mucin entanglements and hydrogen bonding. Controlled release of protein-based cargoes from the PEG gels was achieved over seve …

Signaling Pathways Triggering Therapeutic Hydrogels in Promoting Chronic Wound Healing.

In recent years, there has been a significant increase in the prevalence of chronic wounds, such as pressure ulcers, diabetic foot ulcers, and venous ulcers of the lower extremities. The main contributors to chronic wound formation are bacterial infection, prolonged inflammation, and peripheral vascular disease. However, effectively treating these chronic wounds remains a global challenge. Hydrogels have extensively explored as wound healing dressing because of their excellent biocompatibility and structural similarity to extracellular matrix (ECM). Nonetheless, much is still unknown how the h…

Injectable Conductive Hydrogels with Tunable Degradability as Novel Implantable Bioelectrodes.

Bioelectrodes have been developed to efficiently mediate electrical signals of biological systems as stimulators and recording devices. Recently, conductive hydrogels have garnered great attention as emerging materials for bioelectrode applications because they can permit intimate/conformal contact with living tissues and tissue-like softness. However, administration and control over the in vivo lifetime of bioelectrodes remain challenges. Here, injectable conductive hydrogels (ICHs) with tunable degradability as implantable bioelectrodes are developed. ICHs were constructed via thiol-ene reac…
