Evaluation of NHS carbamates as a potent and selective class of endocannabinoid hydrolase inhibitors.

Monoacylglycerol lipase (MAGL) is a principal metabolic enzyme responsible for hydrolyzing the endogenous cannabinoid (endocannabinoid) 2-ar … xplore NHS carbamates more broadly and demonstrate their potential as inhibitors of endocannabinoid hydrolases and additional enzymes from the serine hydrolase class. We extensively characterize an NHS carbamate 1a (MJN110) as a potent, selective, and in-vivo-active MAGL inhibitor. Finally, we demonstrate that MJN110 alleviates mechanical allodynia in a rat …

Metabolism and neurotoxicity of homocysteine thiolactone in mice: protective role of bleomycin hydrolase.

Genetic or nutritional disorders in homocysteine (Hcy) metabolism elevate Hcy-thiolactone and cause heart and brain diseases. Hcy-thiolactone has been implicated in these diseases because it has the ability to modify protein lysine residues and generate toxic N-Hcy-proteins with auto-immunogenic, pro-thrombotic, and amyloidogenic properties. Bleomycin hydrolase (Blmh) has the ability to hydrolyze L-Hcy-thiolactone (but not D-Hcy-thiolactone) to Hcy in vitro, but whether this reflects a physiological function has been unknown. Here, we show that Blmh (-/-) mice excreted in urine 1.8-fold more H

Inhibition of soluble epoxide hydrolase limits niacin-induced vasodilation in mice.

BACKGROUND: The use of niacin in the treatment of dyslipidemias is limited by the common side effect of cutaneous vasodilation, commonly termed flushing. Flushing is thought to be due to release of the vasodilatory prostanoids prostaglandin D2 (PGD2) and prostaglandin E2 from arachidonic acid metabolism through the cyclooxygenase pathway. Arachidonic acid is also metabolized by the cytochrome P450 system, which is regulated, in part, by the enzyme soluble epoxide hydrolase (sEH). METHODS: These experiments used an established murine model in which ear tissue perfusion was measured by laser Do

Prevention of hypertension in DOCA-salt rats by an inhibitor of soluble epoxide hydrolase.

Cyclooxygenase and lipoxygenase metabolism of arachidonic acid produces compounds important in cardiovascular control. Further, arachidonic acid can be metabolised by cytochrome p450 to produce epoxyeicosatrienoic acids (EETs). These derivatives are inactivated by soluble epoxide hydrolase (sEH). The potential role of these EETs in hypertension and cardiac remodelling has been determined using the selective sEH inhibitor, N-adamantyl-N'-dodecylurea (ADU), in deoxycorticosterone acetate (DOCA)-salt hypertensive rats. Experiments were performed on male Wistar rats following uninephrectomy alone

S-adenosylhomocysteine hydrolase deficiency in a human: a genetic disorder of methionine metabolism.

We report studies of a Croatian boy, a proven case of human S-adenosylhomocysteine (AdoHcy) hydrolase deficiency. Psychomotor development was slow until his fifth month; thereafter, virtually absent until treatment was started. He had marked hypotonia with elevated serum creatine kinase and transaminases, prolonged prothrombin time and low albumin. Electron microscopy of muscle showed numerous abnormal myelin figures; liver biopsy showed mild hepatitis with sparse rough endoplasmic reticulum. Brain MRI at 12.7 months revealed white matter atrophy and abnormally slow myelination. Hypermethionin

Comparative development of Brugia pahangi and variation in acid hydrolase enzyme titers in.

The development of the filarial nematode Brugia pahangi was monitored and compared in susceptible (BLACK EYE) and refractory (ROCK) strains of Aedes aegypti. Simultaneously, the activities of acid phosphatase, beta-glucuronidase, alpha-glucosidase, and N-acetyl-beta-glucosaminidase were measured. Three- to five-day-old females of both strains were fed on infected and uninfected clawed jirds (Meriones unguiculatus) then dissected or homogenized at 2 h, at 24-h intervals for 5 days, and at 8 and 10 days after treatment. Enzyme activities were assayed by a fluorometric procedure. The susceptible

Hydrolase activity of 150 Malassezia furfur isolates of different clinical origin.

Apart from pityriasis versicolor, Malassezia furfur is thought to play a significant role in the pathogenesis of seborrhoic eczema and Malassezia folliculitis. However, it has not been clarified whether in addition to host factors (e.g. immune status, greasy skin), yeast-dependent activities are responsible for manifestation of the disease. In this context interstrain variability of hydrolase activity of Malassezia isolates might be significant. For determination of hydrolase activity, washed yeast suspension was applied to selective agar for pathogenic fungi containing 8% (v/v) Tween 80 or Tw

Dividing the α-amylase family GH57 of starch hydrolases and related enzymes into subfamilies using evolutionary, clustering and functional criteria.

In the CAZy classification, four glycoside hydrolase families - GH13, GH57, GH119 and GH126 - have been designated as α-amylase families. The present study applied to a dataset of over 5000 family GH57 sequences, delivers a functionally balanced subdivision of the family GH57 into ten subfamilies. Eight of these subfamilies bear a specific enzyme activity: GH57_1, 4-α-glucanotransferases; GH57_2, amylopullulanases; GH57_3, α-amylases; GH57_4, α-galactosidases; GH57_5, α-glucan-branching enzymes; GH57_6, non-specified amylases; GH57_7, amylopullulanases-cyclomaltodextrinases; and GH57_8, maltog

Effects of multiple deletions of murein hydrolases on viability, septum cleavage, and sensitivity to large toxic molecules in Escherichia coli.

The multiplicity of murein hydrolases found in most bacteria presents an obstacle to demonstrating the necessity of these potentially autolytic enzymes. Therefore, Escherichia coli mutants with deletions in multiple murein hydrolases, including lytic transglycosylases, amidases, and DD-endopeptidases, were constructed. Even a mutant from which seven different hydrolases were deleted was viable and grew at a normal rate. However, penicillin-induced lysis was retarded. Most of the mutants were affected in septum cleavage, which resulted in the formation of chains of cells. All three enzymes were…

Lysosomal acid hydrolases in established lymphoblastoid cell lines, transformed by Epstein-Barr virus, from patients with genetic lysosomal storage diseases.

Lysosomal acid hydrolases were determined in established lymphoblastoid cell lines, transformed in vitro by Epstein-Barr virus (EBV) from lymphocyte-rich cell populations isolated from the peripheral blood of patients with genetic lysosomal storage diseases--Hurler syndrome, Scheie syndrome, GM1-gangliosidosis type 1 and type 2, Tay-Sachs disease, and I-cell disease--and from obligate heterozygotes for these diseases. The respective enzyme activity was undectectable in lymphoblastoid cells from the patients, but not from controls. Obligate heterozygotes could not always be distinguished from c…

The influence of heavy metal ions, spermine and sodium nitroprusside on ATP-hydrolases of cell membranes of rat colon smooth muscle.

The specific features of functional lability of the rat colon smooth muscle (CSM) АТР-hydrolases were studied. Na+,K+-AТРase activity is effectively inhibited by divalent ions of both transition (≥ 0,1 µM) and nontransition (≥ 1 µM) heavy metals in succession by efficiency: Cu2+ > Fe2+ ≥ Cd2+ (10 µM). Polyamine spermine (0,5-1,0 mM) is a weak Na+,K+-AТРase inhibitor at saturation concentrations of ions and substrate. Sodium nitroprusside (1 mM) as nitric oxide-generating compound exhibits weak Na+,K+-AТРase inhibition only after prolonged preincubation with membranes. Mg2+-АТР-hydrolase activi…

A ubiquitin carboxyl-terminal hydrolase facilitates H2A.Z removal in response to phosphate starvation.

The histone variant H2A.Z plays a critical role in the modulation of environmental responses, but the mechanisms regulating H2A.Z dynamics in response to stresses remain poorly understood in plants. Here we identify the ubiquitin carboxyl-terminal hydrolase UCH2 as a Pi-responsive chromatin regulator that promotes Pi starvation-induced (PSI) gene activation. We show that PHR1 recruits UCH2 to PSI gene promoters under Pi deficiency, where UCH2 deubiquitinates H2A.Z and promotes reduction in H2A.Z occupancy at PSI gene promoters. Loss of UCH2 impairs this reduction in H2A.Z occupancy, represses

Gut microbial bile salt hydrolase as a metabolic gatekeeper in digestive homeostasis and disease.

The gut microbiota exerts broad control over host physiology through tightly coordinated metabolic networks. Among these, bile salt hydrolase (BSH), a microbial enzyme, serves as a key mediator of microbiota-host crosstalk. As a key upstream gateway reaction in bile acid metabolism, BSH hydrolyzes conjugated bile acids and reshapes the composition and distribution of the intestinal bile acid pool. This remodeling alters bile acid signaling quality and influences the host energy metabolism, immune homeostasis and intestinal barrier integrity. In this review, we summarize the core biological fun
