[Correction of immune response using purified staphylococcal toxoid and likopid in the secondary immunodeficiency induced by Coxsackie virus B3].

The action of immunomodulators, purified staphylococcal toxoid (PST) and lycopid, on secondary immunodeficiency state developing during infection caused by Coxsackie virus B3 was studied. This defect was manifested by delayed hypersensitivity to sheep red blood cells (SRBC) and the suppression of neutralizing antibodies to poliomyelitis virus. Depending on the scheme of the experiment, PST normalized the defects of immune response to SRBC or poliovirus, increased suppression or showed no activity. Lycopid corrected the defects of humoral response to SRBC. The combination of PST and lycopid was

Using heat shock proteins as indicators of the immune function in wistar rats during a secondary Trichinella spiralis infection.

The muscle, liver, brain and spleen tissues from Wistar rats with either a primary Trichinella spiralis infection alone, or reinfected 45 days after primary infection, were collected at Days 1, 7, 14, 20 and 27 post reinfection. They were then assayed for levels of four heat shock proteins (HSPs), i.e. hsp90, hsp70, hsp60 and hsp25. Detection and quantitation of the separate HSPs in tissue specimens were achieved using Western blot and image analysis technique, respectively. The results show that the elements affecting altered expression of rat organs' HSP were 'neutralized' by resistance-rela

Immune response to phi X 174 in man. 5. Primary and secondary antibody production in primary biliary cirrhosis.

In primary biliary cirrhosis, the primary immune response to the bacteriophage phi X 174 is normal but the secondary response is significantly reduced. The reduction is primarily of IgG antibody, while IgM is proportionately less affected. These changes may be the result of a reduction in helper T lymphocyte function and may contribute to the increase in the ratio of serum concentration of IgM to IgG.

Hypercalcaemia as an immune-related adverse event secondary to ipilimumab and nivolumab therapy in a patient with metastatic renal cell carcinoma.

We report a case of severe hypercalcaemia after a single cycle of a combination therapy of ipilimumab and nivolumab. Initial evaluation ruled out common causes, including bone metastases, parathyroid hormone/parathyroid hormone-related protein elevation and paraproteinaemia. Her serum 1,25-dihydroxyvitamin D (calcitriol) levels were significantly elevated following the first immunotherapy cycle. Hypercalcaemia responded only to glucocorticoid therapy, with normalisation of serum calcium and calcitriol levels. Recurrence of hypercalcaemia after glucocorticoid discontinuation and subsequent reso

The recall of antigen-specific antibody-secreting cells in Solea senegalensis gives clues on the secondary immune response in flatfish.

In this study, monoclonal antibodies (mAbs) specific for sole immunoglobulin M (IgM) were generated, which allowed us to evaluate the kinetics of IgM+ B cell responses, assess the recruitment of these cells to the peritoneum following vaccination, and compare the dynamics of antigen-specific antibody-secreting cells in the spleen and kidney of primed and boosted fish. Solea senegalensis is a commercially important species in aquaculture, and the development of vaccines is of great interest. Currently, autogenous vaccines provide partial protection against furunculosis, but their efficacy remai

Characterization of Immune Cell Infiltration and Collagen Type III Disorganization in Human Secondary Lymphedema: A Case-control Study.

BACKGROUND: Secondary lymphedema (SL) affects 120 million people globally, posing a lifelong burden for up to 37% of cancer survivors. Chronic inflammation and progressive fibrosis are key drivers of SL, yet detailed characterization of immune cell subpopulations across lymphedema stages is lacking. This study aimed to investigate the immunologic profile of lymphedematous skin and its association with extracellular matrix changes, which could serve as clinical biomarkers or therapeutic targets. METHODS: This case-control study analyzed the skin from 36 patients with and without SL, using immu

Induction and potential molecular mechanism of the enhanced immune response in Procambarus clarkii after secondary encountered with Aeromonas veronii.

For a long time, it was believed that invertebrates do not possess acquired immunity and mainly rely on innate immunity for protection against pathogens infection. However, an increasing number of studies have suggested that some form of "immune memory" can be initiated in invertebrates after primary exposure to the pathogen, which was defined as "specific immune priming". In the present study, two experiments were carried out to determine whether specific immune priming can be induced in crayfish (Procambarus clarkii) by Aeromonas veronii, if so, to identify the underlying mechanism. Once bei

Resident memory CD4+ T lymphocytes mobilize from bone marrow to contribute to a systemic secondary immune reaction.

Resident memory T lymphocytes (TRM ) of epithelial tissues and the Bm protect their host tissue. To what extent these cells are mobilized and contribute to systemic immune reactions is less clear. Here, we show that in secondary immune reactions to the measles-mumps-rubella (MMR) vaccine, CD4+ TRM are mobilized into the blood within 16 to 48 h after immunization in humans. This mobilization of TRM is cognate: TRM recognizing other antigens are not mobilized, unless they cross-react with the vaccine. We also demonstrate through methylome analyses that TRM are mobilized from the Bm. These mobili

Structural modeling and mRNA expression of epididymal β-defensins in GnRH immunized boars: A model for secondary hypogonadism in man.

Human secondary hypogonadism is associated with impaired testicular function, however, little is known about its impact on sperm epididymal maturation. Endocrine disruption in the epididymis could impair the secretion of key proteins, such as β-defensins, responsible for spermatozoa maturation during epididymal transit. This study evaluated the sequence and structural similarities between porcine epididymal β-defensins porcine β-defensins (pBD3), pBD4, pBD125, and pEP2C and their human homologs using bioinformatics integrated with information derived from protein databanks. We then verified wh

Leflunomide: an immune modulating drug that may have a role in controlling secondary infections with review of its mechanisms of action.

BACKGROUND: Leflunomide (LEF) is an immune modulator used most commonly for rheumatoid arthritis (RA). The mechanisms of action of LEF also include anti-microbial effects, particularly anti-viral effects. OBSERVATIONS: We present three patients with atopic dermatitis on azathioprine therapy who had multiple verrucae and in two molluscum contagiosum (MC) that were resistant to repeated conventional therapies. These patients were switched to LEF, and all the patients showed complete resolution of their verrucae and MC within 2 months of starting therapy. In addition, all three patients showed e

Visualization of granzyme B-expressing CD8 T cells during primary and secondary immune responses to Listeria monocytogenes.

CD8 T cells contribute to long-term protection against Listeria monocytogenes infection by differentiating into memory T cells. These rapidly respond to antigen or inflammation upon secondary infection. In this study we used CD8 T cells from OT1 mice and CD4 T cells from OT2 mice expressing a fluorescent chimeric granzyme (GZMB-Tom) protein to monitor the primary response to infection with ovalbumin-expressing L. monocytogenes (Lm-OVA). We show that, unlike poorly responding CD4 T cells, CD8 T cells readily proliferated and expressed high levels of GZMB-Tom as early as 2 days after infection.

Drug-induced immune-mediated thrombocytopenia secondary to sunitinib in a patient with metastatic renal cell carcinoma: a case report.

INTRODUCTION: Sunitinib is an oral multi-targeted tyrosine kinase inhibitor approved for first line treatment for metastatic renal cell carcinoma and imatinib-resistant metastatic gastrointestinal stromal tumors. Sunitinib administration can cause myelosuppression resulting in neutropenia and thrombocytopenia. Here we present the case of a patient with metastatic renal cell carcinoma who developed sunitinib-induced immune-mediated thrombocytopenia and who was treated with withdrawal of sunitinib and administration of intravenous immunoglobulin and steroids. CASE PRESENTATION: This case report

Immune responses to vaccinia and influenza elicited during primary versus recent or distant secondary smallpox vaccination of adults.

The kinetics and frequency of the primary response to human smallpox vaccine was compared to that among subjects re-vaccinated within the last 2 years, or after several decades. Vaccination induced local and systemic reactions that were mildest in the distant cohort and more severe in the primary vaccinees. The timing of IFN-gamma responses was similar in all three groups, but of higher frequency in the primary vaccinees. Responses to vaccinia re-immunization between those immunized <2 years ago and those immunized >10 years ago were not significantly different. Neutralizing antibodies were bo

Induction and expression of intestinal humoral immunity in HIV-infected individuals: prospects for vaccination against secondary enteric infections.

In view of the high incidence and severity of AIDS-related mucosal complications and secondary mucosal infections, we have evaluated the impact of HIV infection on the mucosal humoral immune status. By measuring the number of total and specific antibody-secreting cells in duodenal biopsies, we could show that HIV infection induces a pronounced polyclonal B-cell activation already in the early stages of the disease. Furthermore, we could document normal mucosal B-cell responses following oral vaccination, also in the late stages of the disease, and despite a lack of concomitant systemic respons

Identification of key genes associated with persistent immune changes and secondary immune activation responses induced by influenza vaccination after COVID-19 recovery by machine learning methods.

COVID-19 is hypothesized to exert enduring effects on the immune systems of patients, leading to alterations in immune-related gene expression. This study aimed to scrutinize the persistent implications of SARS-CoV-2 infection on gene expression and its influence on subsequent immune activation responses. We designed a machine learning-based approach to analyze transcriptomic data from both healthy individuals and patients who had recovered from COVID-19. Patients were categorized based on their influenza vaccination status and then compared with healthy controls. The initial sample set encomp

Lymphocytic 2',5'-oligoadenylate synthetase activity increases prior to the appearance of neutralizing antibodies and immunoglobulin M and immunoglobulin G antibodies after primary and secondary immunization with yellow fever vaccine.

Primary and secondary immunizations with live, attenuated yellow fever virus vaccine (17D strain) were performed in order to study the course of appearance of virus-neutralizing antibodies and immunoglobulin M (IgM) and IgG antibodies directed against the virus and the interferon-dependent enzyme 2',5'-oligoadenylate synthetase (2',5'AS) activity, determined in homogenates of peripheral B and T lymphocytes. From cellular ATP, this enzyme generates 2',5'-oligoadenylates which mediate degradation of viral mRNA by stimulation of a latent RNase. By day 4 after the first immunization, the earliest

Single-Cell Sequencing Reveals the Immune Characteristics of Secondary Liver Injury Induced Indirectly by CHIKV Infection in Rhesus Macaques.

Chikungunya virus (CHIKV) is now prevalent in multiple regions worldwide, posing a serious threat to human health. In this study, we have successfully established a rhesus macaque model of Chikungunya virus infection. Notably, while no viral load was detected in liver tissue on day 7 post-infection, significant pathological damage was observed. Single-cell RNA sequencing of liver tissue revealed a reduction in B cells following infection. Among T cells, CD8+ T and NKT cells mediated major cytotoxic effects, whereas CD4+ T and memory T cells primarily exerted regulatory functions that further e
