Soy isoflavones modulate adipokines and myokines to regulate lipid metabolism in adipose tissue, skeletal muscle and liver of male Huanjiang mini-pigs.

Although a growing body of evidence suggests that soy isoflavones help regulate lipid metabolism, the underlying mechanism has not yet been thoroughly clarified. The present study was undertaken to determine the effects of soy isoflavones on the expression of genes involved in lipid metabolism in different adipose tissue depots, skeletal muscle and liver of male Huanjiang mini-pigs, as well as the expression of adipokines and myokines. A total of 36 male Huanjiang mini-pigs were fed basal diet (control, Con), low-dose soy isoflavones (LSI) and high-dose soy isoflavones (HSI). The results showe…

A galactose-free diet enriched in soy isoflavones and antioxidants results in delayed onset of symptoms of Krabbe disease in twitcher mice.

Krabbe disease or globoid cell leukodystrophy is an autosomal recessively inherited disorder caused by the deficiency of galactocerebrosidase, the lysosomal enzyme that catalyzes the hydrolysis of galactose from galactosylceramide and galactosylsphingosine (psychosine). Psychosine accumulation results in the loss of myelin and oligodendrocytes in the brain of Krabbe patients as well as twitcher mice (natural model of human Krabbe disease). The aim of the present research was to investigate in twitcher mice the potential role of a diet deficient in galactose enriched in soy isoflavones and a po…

Enrichment of milk antioxidant activity by dietary supplementation of red clover isoflavone in cows and its improvement on mice intestinal health.

Red clover (Trifolium pratense) isoflavone was supplemented to dairy cows, and antioxidant capacity of milk was assessed. Treated cows increased the activities of antioxidant enzymes, reduced production of oxidation products, and enhanced the concentrations of vitamin E and vitamin C. Moreover, milk fatty acid profile was positive influenced by 8 g/kg red clover isoflavone, with changes in the lower saturated and higher unsaturated fatty acids. We further demonstrated the efficacy of antioxidant capacity of milk in mice, found that milk from cows feeding red clover isoflavone increased the exp

The effect of soybean isoflavone on the dysregulation of NMDA receptor signaling pathway induced by β-amyloid peptides 1-42 in rats.

Synaptic damage is the key factor of cognitive impairment. The purpose of this study was to understand the effect of soybean isoflavone (SIF) on synaptic damage induced by β-amyloid peptide 1-42 (Aβ1-42) in rats. Adult male Wistar rats were randomly divided into control, Aβ1-42, SIF, and SIF + Aβ1-42 (SIF pretreatment) groups according to body weight. SIF was treated orally by gavage in SIF and SIF + Aβ1-42 groups. After 14 days pretreatment with SIF or vehicle, Aβ1-42 was injected into the lateral cerebral ventricle of rats in Aβ1-42 and SIF + Aβ1-42 groups using miniosmotic pump. The level o

Irisolidone, an isoflavone metabolite, represses JC virus gene expression via inhibition of Sp1 binding in human glial cells.

Progressive multifocal leukoencephalopathy (PML) is a fatal demyelinating disease that results from an oligodendrocyte infection caused by the JC virus. Therefore, inhibiting the expression of JC virus is important for preventing and/or treating PML. This study found that irisolidone, an isoflavone metabolite, significantly inhibited the JC virus expression in primary cultured human astrocytes and glial cell lines. Studies examining the underlying mechanism revealed that a mutation of the Sp1 binding site downstream of the TATA box (Sp1-II) dramatically diminished the inhibitory activity of ir

Higher dietary isoflavone intake is associated with lower risk of new-onset hypertension: An 18-year national cohort study.

Prior experimental studies suggest that isoflavones may reduce blood pressure, but epidemiological evidence on their association with hypertension risk remains limited and inconsistent. We hypothesized that dietary isoflavone intake is inversely associated with the risk of new-onset hypertension. This cohort study included 11,402 adults from the China Health and Nutrition Survey who were free of hypertension at baseline. Hazard ratios (HRs) and 95% confidence intervals (CIs) were estimated from Cox proportional-hazards models, adjusting for potential confounders. Over 93,058 person-years, 3,99…

An isoflavone-enriched diet alleviates Parkinson's disease in mice by inhibiting ferroptosis through gut microbiota-mediated serotonin production.

OBJECTIVE: Parkinson's disease (PD) is a severe neurodegenerative disorder whose pathogenesis is closely linked to gut microbiota dysregulation. However, whether and how modulation of gut homeostasis can ameliorate PD remains unclear. Dietary isoflavones have been associated with neuroprotective effects and show strong potential in shaping the composition of the gut microbiota, yet their underlying mechanisms in PD are poorly understood. METHODS: Microbiomics and non-targeted metabolomics were utilized to characterize microbial community composition and metabolic alterations in the MPTP (1-me…

Multi-omics integration identifies GmISO5 as a key regulator of seed isoflavone biosynthesis and implicates it in the response to soybean mosaic virus.

INTRODUCTION: Isoflavones are specialized metabolites that accumulate abundantly in soybean seeds, contributing to nutritional quality, human health benefits, and plant defense. Although the core isoflavone biosynthetic pathway has been well characterized, its upstream regulatory network and potential relationship with plant stress responses remain incompletely understood. OBJECTIVES: This study aimed to identify key regulators of seed isoflavone accumulation and to assess whether the identified regulator is associated with the soybean response to soybean mosaic virus (SMV) infection. METHOD…

TMI, an isoflavone from the fruits of Ficus altissima Blume, overcomes K562/ADR leukemia cells resistance by targeting Nrf2.

Chronic myeloid leukemia (CML) is a myeloproliferative tumor, which is prone to drug resistance during treatment. Previous studies have indicated that Ficus altissima Blume exhibits potential antitumor properties, with 5,7,2'-trihydroxy-4'-methoxy-6-(3-hydroxy-3-methylbutyl)-isoflavone (TMI) as its main active component. However, the antitumour activity of TMI and its underlying mechanisms remain unexplored. This study investigated the anti-proliferative effects of TMI on adriamycin (ADR)-resistant leukemia K562 cells (K562/ADR cells) and the mechanism of overcoming multidrug resistance (MDR).

Potential Protective Effects of Equol (Soy Isoflavone Metabolite) on Coronary Heart Diseases-From Molecular Mechanisms to Studies in Humans.

Equol, a soy isoflavone-derived metabolite of the gut microbiome, may be the key cardioprotective component of soy isoflavones. Systematic reviews have reported that soy isoflavones have no to very small effects on traditional cardiovascular disease risk factors. However, the potential mechanistic mode of action of equol on non-traditional cardiovascular risk factors has not been systematically reviewed. We searched the PubMed through to July 2021 by using terms for equol and each of the following markers: inflammation, oxidation, endothelial function, vasodilation, atherosclerosis, arterial s…
