The constitutive desmosomal plaque protein desmoplakin plays a vital part in keratinocyte adhesion in linking the transmembranous desmosomal cadherins to the cytoplasmic keratin filament network. Recently, mutations in desmoplakin have been shown to underlie some cases of the autosomal dominant disorder, striate palmoplantar keratoderma, as well as an autosomal recessive condition characterized by dilated cardiomyopathy, woolly hair, and keratoderma. Here, we describe two unrelated individuals with a new autosomal recessive genodermatosis characterized by focal and diffuse palmoplantar keratod

Skin fragility, palmoplantar keratoderma, and characteristic woolly hair are defining features of skin fragility-woolly hair syndrome (SFWHS), an extremely rare inherited disorder. This condition is classified within the group of desmosomal disorders, which are associated with compromised structural integrity of the skin and hair. Clinically, affected individuals often present with hair fragility, fragile skin, recurrent erosions, nail abnormalities, and varying degrees of palmoplantar thickening beginning in early childhood. In this case report, we describe a patient with this extremely rare

A morphological and biochemical analysis was made of cornified envelopes isolated from patients with different congenital disorders. Nomarski contrast microscopy of the envelopes showed that their morphology was not greatly altered in several types of keratoderma and parapsoriasis, but it was grossly modified in ichthyotic disorders. The various types of ichthyoses, keratoderma palmoplantare, KID syndrome and parapsoriasis showed, after cyanogen-bromide cleavage, peptide patterns similar to those obtained from healthy subjects. In contrast, envelopes from patients with Darier's disease, congen

Six cases of a dermatosis characterized by "music box spine" keratotic papules limited to the palms and soles have been reported. The names that have been used for this type of dermatosis most commonly incorporate the term porokeratosis; for example, "porokeratosis palmaris et plantaris," or "punctuate porokeratotic keratoderma." We suggest the name "spiny keratoderma of the palms and soles," because the clinical, histologic, and electron microscopic features of this entity are distinct from porokeratosis. In addition, unlike porokeratosis, this entity has not shown malignant potential. We rep

Pachyonychia congenita (PC) is a rare genodermatosis caused by mutations in any of the four genes KRT6A, KRT6B, KRT16, or KRT17, which can lead to dystrophic, thickened nails and focal palmoplantar keratoderma, among other manifestations. Although classically subdivided into two major variants, PC-1 (Jadassohn-Lewandowski syndrome) and PC-2 (Jackson-Lawler syndrome), according to the localization of the mutations in the KRT6A/KRT16 or KRT6B/KRT17 genes, respectively, a classification system based on the mutant gene (PC-6a, PC-6b, PC-16 and PC-17) has been recently proposed. We report a 2-year-

In 1901, Vörner described a diffuse keratoderma of palms and soles with autosomal dominant inheritance. Histopathologically, this disease has the typical features of epidermolytic hyperkeratosis. Clinical examination does not allow differentiation between keratoderma of the Vörner type and the keratoderma described by Thost in 1880 and Unna in 1883. Reexamination of the family originally seen by Thost revealed histopathological signs of epidermolytic hyperkeratosis, confirming that keratoderma of the Vörner type is present in this family. The clinical features and variability of this palmoplan

Tegafur is a prodrug of 5-fluorouracil (5-FU) with a similar spectrum of antitumor activity. It is used in the treatment of advanced gastrointestinal neoplasms. Over 5-FU, tegafur has the advantage of oral administration and less hematologic toxicity. Gastrointestinal toxicity is its main dose-limiting factor. The cutaneous adverse effects of tegafur include mucositis, photosensitivity, diffuse or nail-restricted hyperpigmentation, palmoplantar erythrodysesthesia syndrome, palmoplantar keratoderma, sclerodactyly and Raynaud phenomenon. We report here the case of a patient who developed acral h

Synaptosomal-associated protein 29 (SNAP29) is a t-SNARE protein that is implicated in intracellular vesicle fusion. Mutations in the SNAP29 gene have been associated with cerebral dysgenesis, neuropathy, ichthyosis, and keratoderma syndrome (CEDNIK). In patients with 22q11.2 deletion syndrome, mutations in SNAP29 on the nondeleted chromosome are linked to similar ichthyotic and neurological phenotypes. Here, the authors report a patient with cerebral dysgenesis, neuropathy, ichthyosis, and keratoderma syndrome who presented with global developmental delay, polymicrogyria, dysgenesis of the co

Palmoplantar keratoderma in humans is a condition defined by an abnormally thickened cornified skin layer on the hands and feet. In animals, the corresponding disease is commonly termed paw pad hyperkeratosis. It can be acquired due to repeated trauma, infections, cancer, or inflammatory dermatoses, or inherited due to pathogenic variants in genes involved in skin development. More than 60 different genes involved in the development of palmoplantar keratoderma have been described. Here, we investigated a female Labrador Retriever showing hyperkeratosis on all four paw pads and most digital pad

BACKGROUND: Pathogenic variants in SERPINA12 have recently been identified in patients with palmoplantar keratoderma (PPK). Clinically, SERPINA12-associated PPK shares substantial overlap with Nagashima-type palmoplantar keratoderma (NPPK) caused by SERPINB7 deficiency, although inflammatory manifestations appear to be more prominent in SERPINA12-associated disease. The molecular basis underlying these similarities and differences remains unclear.
METHODS: Virtual knockout analyses of SERPINA12 and SERPINB7 were performed using the scTenifoldKnk framework. Functional enrichment analyses, incl

We describe the case of a 32-year-old male patient who had presented from birth with generalized ichthyosiform dermatosis, palmoplantar keratoderma with constrictive bands around the fingers and keratotic plaques in a linear arrangement, located in the large skin folds. The dermatopathological examination showed orthokeratotic hyperkeratosis and epidermal hyperplasia with hypergranulosis. No other members of the patient's family were affected. With these clinical and histological findings, the diagnosis was keratosis linearis with ichthyosis congenita and sclerosing keratoderma (KLICK syndrome
