The karyotypes of 103 cases of B cell chronic lymphocytic leukemia were studied following stimulation or establishment of continuous cell lines using the Epstein-Barr virus. In 52 patients metaphases suitable for cytogenetic analysis were obtained; 30 revealed normal karyotypes and 22 abnormal karyotypes. The most frequently encountered abnormalities were 6 cases of extra chromosome 12, 4 cases of structured aberrations concerning chromosome 14, including 2 t(11;14), 4 translocations concerning chromosome 11, and 3 cases of extra chromosome 3. No relationship appears to exist between the abnor

Peripheral lymphocytes of 16 patients with chronic lymphatic leukemia were studied for B and T cell markers with peroxidase-labelled anti-immunogloulin antisera and complement and spontaneous rosetting techniques. Increased numbers of B cells and normal numbers of T cells were noted. A significant observation was the presence of a large proportion of circulating lymphocytes without the conventional markers for T and B cells. This finding is correlated with clinical features.

B-cell acute lymphoblastic leukemia (B-ALL) is an aggressive hematologic malignancy characterized by rapid proliferation of immature lymphoid cells. Despite treatment advancements, relapse remains a significant challenge. Understanding the molecular mechanisms that drive B-ALL progression is essential for developing more effective treatments. Here, we show that extracellular cyclophilin A (eCypA) is elevated in patients with B-ALL via both autocrine and paracrine mechanisms. Moreover, eCypA administration exacerbated B-ALL progression in a mouse model. Mechanistically, eCypA binds to the melan

BACKGROUND AND STUDY AIMS: Historically, adults with B-cell acute lymphoblastic leukemia (ALL) treated with standard intensive chemotherapy in first salvage had a complete response rate of 40% and a 3-year survival rate <10%. The outcome of B-cell ALL on second-line therapy (salvage 1) is changing rapidly with the introduction of novel strategies into both frontline and salvage therapy. The study aim is to define the outcome of adult B-cell ALL on second-line therapy and its predictors in the novel era of ALL therapy.
PATIENTS AND METHODS: A total of 372 adults with B-cell ALL in salvage 1 tr

We report the case of an 8-year-old girl with B-cell precursor acute lymphoblastic leukemia (BCP-ALL), who presented with a symptomatic epidural mass at the initial diagnosis. She had persistent back pain and mild neurological symptoms. Magnetic resonance imaging (MRI) revealed an epidural mass extending from L2 to the sacrum without spinal cord compression. A bone marrow examination confirmed BCP-ALL. A biopsy was deferred because of procedural risks, and systemic chemotherapy combined with corticosteroids was initiated. Her symptoms resolved rapidly, and follow-up MRI showed a marked regress

B-cell acute lymphoblastic leukemia (ALL) with KMT2A rearrangements (KMT2Ar B-ALL) represents a distinct, high-risk subtype in adults, characterized by aggressive disease biology, high relapse rates, and inferior long-term survival. Conventional intensive chemotherapy, even when consolidated with allogeneic hematopoietic stem cell transplantation (allo-HSCT), yields suboptimal outcomes, underscoring the need for novel therapeutic approaches. In recent years, substantial progress has been made with the introduction of antibody-based immunotherapies, cellular immunotherapies, and small molecular

B-cell acute lymphoblastic leukemia (B-ALL) is an aggressive hematological malignancy that primarily affects children but can also occur in adults, progressing rapidly and requiring urgent clinical intervention. Late-stage diagnosis often results in reduced survival rates and typically depends on costly, time-intensive diagnostic procedures. Peripheral blood smear (PBS) imaging plays a central role in the preliminary screening of B-ALL and provides an accessible foundation for computer-assisted diagnosis. To support early and efficient classification, this study proposes a lightweight convolut

Before November 2023, CD19 chimeric antigen receptor (CAR) T-cell therapies had not been approved in China for patients with relapsed or refractory B-cell acute lymphoblastic leukemia (R/R B-ALL), leaving a significant unmet need. In response, inaticabtagene autoleucel (Inati-cel), a novel CD19 CAR T-cell therapy with a distinct single-chain variable fragment (HI19α), was developed and showed promising efficacy in preliminary clinical research. We conducted a phase 2, single-arm, multicenter study of Inati-cel in adult CD19+ R/R B-ALL in China. The primary end point was the overall remission r

Resistance to glucocorticoids (GC), the common agents for remission induction in pediatric B-cell precursor acute lymphoblastic leukemia (BCP-ALL), poses a significant therapeutic hurdle. Therefore, dissecting the mechanisms shaping GC resistance could lead to new treatment modalities. Here, we showed that CD9- BCP-ALL cells were preferentially resistant to prednisone and dexamethasone over other standard cytotoxic agents. Concordantly, we identified significantly more poor responders to the prednisone prephase among BCP-ALL patients with a CD9- phenotype, especially for those with adverse pre

BACKGROUND: Aberrant DNA methylation is an important mechanism by which the normal patterns of microRNA expression are disrupted in human cancers including B-cell precursor acute lymphoblastic leukemia (BCP ALL), the most common pediatric malignancy.
OBJECTIVES: To characterize the methylation profile landscape of microRNA genes in BCP ALL patients.
MATERIAL AND METHODS: We employed Infinium® MethylationEPIC BeadChip Arrays to measure the methylation of microRNA genes from bone marrow samples of children with BCP ALL (n = 38) and controls without neoplasms (n = 4).
RESULTS: This analysis re

Chloroma, is an rare malignant tumor characterized by the extramedullary blast proliferation of myeloid lineages that subsequently change the normal architecture of surrounding tissues. Because this is very rare disease, primary central nervous system myeloid sarcoma has not been widely reported. Myeloid sarcoma, as a group of heterogenetic diseases, presents with differential clinical and histological pictures depending upon the primary affected site. We are presenting a 77-year-old gentleman, with past medical history of chronic lymphoblastic leukemia that was not on any treatment and who pr

Inotuzumab ozogamicin (InO) is a recently US Food and Drug Administration-approved antibody-drug conjugate for the treatment of relapsed/refractory B-cell acute lymphoblastic leukemia (ALL). InO consists of a CD22-targeting immunoglobulin G4 humanized monoclonal antibody conjugated to calicheamicin. Although initially developed for the treatment of non-Hodgkin lymphoma (NHL) because of activity in preclinical models and high response rates in indolent lymphomas, a phase 3 trial was negative and further development focused on CD22+ ALL. Although results in NHL were disappointing, parallel testi

CASE DESCRIPTION: A 17-year-old Friesian gelding was examined at a referral hospital because of a 1-month history of mild exercise intolerance and marked lymphocytosis.
CLINICAL FINDINGS: Physical examination revealed no peripheral lymphadenopathy or other abnormalities. Results of an abdominal palpation examination per rectum and thoracic and abdominal ultrasonographic examinations were unremarkable. B-cell chronic lymphocytic leukemia (CLL) was diagnosed on the basis of severe lymphocytosis and positive expression of the B-cell marker CD20 by lymphocytes in the bone marrow and peripheral bl
