Vitamin E did not prevent platelet activation, but prevented increase of lipid peroxides and renal dysfunction in DOCA-salt hypertensive rats.

Changes in platelet aggregability patterns and lipid peroxide levels and their relationship were examined in DOCA-salt hypertensive rats dur … ot prevent in vivo platelet activation due to hypertension, but greatly prevented the elevations of lipid peroxides in serum, the heart and the kidney, and serum urea nitrogen. These results suggested that in vivo platelet activation, an increase of lipid peroxides, and renal dysfunction occur in this order due to hypertension, and that the latter two are signifi …

AA-Induced Lipid Peroxidation at the Center of Tumor Cell Apoptosis and Ferroptosis.

Polyunsaturated fatty acids (PUFAs), especially gamma-linolenic acid (GLA), arachidonic acid (AA), eicosapentaenoic acid (EPA), and docosahexaenoic acid (DHA), can block HMG-CoA reductase activity and thus decrease cholesterol synthesis/formation. Cholesterol has antioxidant actions and thus reduces the formation of ROS (reactive oxygen species). In contrast, PUFAs augment the generation of ROS. PUFAs undergo peroxidation to form lipid peroxides that are toxic to tumor cells but not normal cells. GPX4 (glutathione peroxidase) reduces the formation of lipid peroxides and thus enhances tumor cel…

[Practical aspects of lipid emulsion stability and peroxidation processes in parenteral nutrition].

The stability of lipid emulsions used in parenteral nutrition (PN) is a key determinant of both the safety and effectiveness of intravenous nutritional therapy. This review summarizes the main physicochemical factors influencing the stability of ternary PN admixtures, emphasizing the role of electrolytes, pH, osmolarity, temperature, light exposure, and handling during storage and administration. Mechanisms leading to emulsion destabilization-such as lipid globule coalescence or phase separation-are described, highlighting the importance of maintaining a mean particle diameter below 5 µm to pr

Oroxylin A attenuates sepsis-associated coagulopathy by targeting the ALOX12-lipid peroxidation.

Sepsis is a life-threatening syndrome driven by dysregulated thromboinflammation. Effective therapies are currently lacking due to the clinical limitations and bleeding risks associated with conventional anticoagulants. In this study, we demonstrate that Oroxylin A, a natural flavonoid, mitigates sepsis-associated coagulopathy by targeting arachidonate 12-lipoxygenase (ALOX12)-mediated lipid peroxidation. Using murine models of cecal ligation and puncture (CLP) and bacterial sepsis, we found that Oroxylin A significantly improved survival, restored platelet counts, and normalized coagulation p

Volatile Biomarkers in Exhaled Breath for In Vivo Lipid Peroxidation and Ferroptosis Detection.

Human breath volatile metabolites have attracted increasing attention as a source of non-invasive biomarkers. However, the biosynthetic pathways of most breath-borne compounds remain unclear, and their relationships to physiological or pathological states are still poorly defined, which has greatly hindered the clini cal application of breath biomarkers.Ferroptosis is an iron-dependent process driven by lipid peroxidation that exacerbates tissue inflammation and has been implicated in a wide range of age-associated disorders. Importantly, some oxidized lipids generated during ferroptosis are v

N -aldehyde modified phosphatidylethanolamines formed by lipid peroxidation inhibit efferocytosis.

BACKGROUND: Impairments in efferocytosis, the phagocytic clearance of apoptotic cells, contribute to the development of chronic inflammation and promote the formation of large necrotic cores within atherosclerotic plaques. A better understanding of the factors that impair efferocytosis could lead to improved interventions. Many chronic inflammatory conditions feature excessive lipid peroxidation, which generates reactive lipid aldehydes that can potentially form N -aldehyde-modified phosphatidylethanolamines (NAMPs). We therefore investigated the extent to which levels of NAMPs are elevated un

Lipid peroxidation in cancer: a double-edged sword shaping progression and therapy.

Lipid Peroxidation (LPO) is a process driven by oxidative stress that is notoriously implicated for triggering various diseases. In cancer, however, LPO acts as a double-edged sword, fueling tumor growth while showing promise as a treatment target. This review performs a deep dive into this duality and highlights its twofold involvement across various malignancies, including colorectal, breast, and lung cancers. LPO is closely linked with oxidative stress, which contributes to DNA damage, chronic inflammation, and other processes that cause cancer initiation. Other LPO-derived mechanisms, such

P53 Regulation upon Lipid Peroxidation and Ferroptosis for Intervention against Atherogenesis.

Tumor protein 53 (P53), as an intracellular regulator of antioxidant responses, participates in the expression of antioxidant defense and lipid metabolism as well as the synthesis of genes in cells. The balance of oxidation and reduction can be disrupted by many pathological conditions, and the role of the antioxidant system in protecting the equilibrium state from pathological effects, such as reactive lipids, is crucial. In particular, the excessive accumulation of lipid peroxidation products is a key factor driving the occurrence and development of various diseases. Ferroptosis is an iron-d

Lipid Peroxidation Levels in Saliva and Plasma of Patients Suffering from Periodontitis.

Lipid peroxidation (LPO) participates in the development of various diseases, including periodontitis, and malondialdehyde (MDA) is its terminal product. Therefore, in the present study, salivary and plasma MDA levels in 30 periodontitis patients were compared to those in 20 healthy controls, as well as in relation to periodontal therapy in order to assess its effectiveness. Periodontal status was assessed via plaque index, gingival index, papilla bleeding index, probing depth and clinical attachment level, while salivary and plasma MDA levels were determined by the ELISA method. The periodont

Ferroptosis and cardiovascular disease: role of free radical-induced lipid peroxidation.

Cardiovascular disease (CVD), including heart attack, stroke, heart failure, arrhythmia, and other congenital heart diseases remain the leading cause of morbidity and mortality worldwide. The leading cause of deaths in CVD is attributed to myocardial infarction due to the rupture of atherosclerotic plaque. Atherosclerosis refers a condition when restricted or even blockage of blood flow occurs due to the narrowing of blood vessels as a result of the buildup of plaques composed of oxidized lipids. It is well-established that free radical oxidation of polyunsaturated fatty acids (PUFAs) in lipop

Lipid peroxidation products as predictors of oxidant-mediated disease in preterm infants.

BACKGROUND: Preterm infants are susceptible to "oxygen radical diseases" (ORD). 8-isoprostane (8-IP) is a bioactive eicosanoid generated by reactive oxygen species-catalyzed peroxidation of arachidonic acid. Malondialdehyde (MDA) is generated by the decomposition of oxidant-induced lipid hydroperoxides. We hypothesize that the development of ORD is associated with elevated plasma 8-IP on day 0-1, and increasing urine levels of MDA in the first month. METHODS: Preterm (<32 weeks, n = 39) and term (n = 39) infants were recruited at birth. Plasma 8-IP was quantified by ELISA on day 0-1, and urin

Effect of combined G6PD deficiency and diabetes on protein oxidation and lipid peroxidation.

BACKGROUND: Oxidative Stress, an imbalance in the pro-oxidant/antioxidant homeostasis, occurs in many physiological and non-physiological processes and several human diseases, including diabetes mellitus (DM) and glucose-6-phosphate dehydrogenase (G6PD) deficiency. Since the incidence of G6PD deficiency in Jordan and many parts of the world is high, this study aimed to measure the effect of G6PD deficiency on the oxidative markers and the antioxidant glutathione (GSH) in diabetic and non-diabetic individuals. METHODS: Whole blood G6PD deficiency was screened by the fluorescent spot method, an

Arachidonic Acid Kills Staphylococcus aureus through a Lipid Peroxidation Mechanism.

Staphylococcus aureus infects every niche of the human host. In response to microbial infection, vertebrates have an arsenal of antimicrobial compounds that inhibit bacterial growth or kill bacterial cells. One class of antimicrobial compounds consists of polyunsaturated fatty acids, which are highly abundant in eukaryotes and encountered by S. aureus at the host-pathogen interface. Arachidonic acid (AA) is one of the most abundant polyunsaturated fatty acids in vertebrates and is released in large amounts during the oxidative burst. Most of the released AA is converted to bioactive signaling

A high throughput biochemical fluorometric method for measuring lipid peroxidation in HDL.

Current cell-based assays for determining the functional properties of high-density lipoproteins (HDL) have limitations. We report here the development of a new, robust fluorometric cell-free biochemical assay that measures HDL lipid peroxidation (HDLox) based on the oxidation of the fluorochrome Amplex Red. HDLox correlated with previously validated cell-based (r = 0.47, p<0.001) and cell-free assays (r = 0.46, p<0.001). HDLox distinguished dysfunctional HDL in established animal models of atherosclerosis and Human Immunodeficiency Virus (HIV) patients. Using an immunoaffinity method for capt
