Regulation of CNS Lipids by Protease Activated Receptor 1.

Disruptions in the metabolism of cholesterol and other lipids are strongly implicated in the pathogenesis of neurological disease. The CNS is highly enriched in cholesterol, which is primarily synthesized de novo. Cholesterol synthesis is also rate limiting for myelin regeneration. Given that knockout of the thrombin receptor (Protease Activated Receptor 1 (PAR1)) accelerates myelin regeneration, here we sought to determine the potential regulatory actions of PAR1 in CNS cholesterol and lipid metabolism in the intact adult CNS and during myelin regeneration. We present quantitative PCR and RNA…

Lipids as pharmacological targets in age-related lens disease.

Cataract and presbyopia are the leading causes of age-related vision impairment worldwide, yet non-surgical management options remain limited. Age-related changes in the lipid composition of the crystalline lens have been implicated in their pathophysiology, highlighting the lens lipidome as a potential therapeutic target. In this review we summarize how recent advances in high-resolution and spatial lipidomics have clarified age- and region-specific changes in the lens lipidome, and we evaluate recent research efforts to utilize topical lipid-replenishing formulations and lipid-modifying smal

Zinc-Coordinated Lipids: Facilitators for Enhanced mRNA Delivery Efficacy.

The advancement of mRNA therapeutics necessitates more efficient delivery systems. Current clinically advanced lipid nanoparticles (LNPs) primarily bind mRNA through electrostatic interactions, with limited exploration of other intermolecular forces that can benefit multiple delivery processes. Here, we design zinc-coordinated lipids (Zn-CL) to formulate LNPs, enabling high mRNA delivery efficacy in vivo. The zinc-coordinated moieties in ionizable lipids show strong affinity toward phosphate groups, yielding tight yet reversible mRNA encapsulation. In addition, due to the abundance of phosphat…

Role of Lipids in the Pathogenesis of Parkinson's Disease.

Aggregation of α-synuclein (αSyn) and its accumulation as Lewy bodies play a central role in the pathogenesis of Parkinson's disease (PD). However, the mechanism by which αSyn aggregates in the brain remains unclear. Biochemical studies have demonstrated that αSyn interacts with lipids, and these interactions affect the aggregation process of αSyn. Furthermore, genetic studies have identified mutations in lipid metabolism-associated genes such as glucocerebrosidase 1 (GBA1) and synaptojanin 1 (SYNJ1) in sporadic and familial forms of PD, respectively. In this review, we focus on the role of li…

Affinity of Malassezia and Other Yeasts for Pulmonary Lipids.

Pulmonary surfactant, the primary substance lining the epithelium of the human Lower Respiratory Tract (LRT), is rich in lipids, with dipalmitoyl-phosphatidylcholine (DPPC) being the most abundant. Although surfactants are known to have antifungal activity against some yeast species, the significant presence of species like Malassezia restricta in the lung mycobiome suggests that these yeasts may exhibit some level of lipo-tolerance or even lipo-affinity for pulmonary lipids. This study explored the affinity and tolerance of yeasts, identified as significant members of the lung microbiome, to …

The role of lipids in regulating macrophage antifungal immunity.

Macrophages are critical components of the antifungal immune response. Disturbance in the number or function of these innate immune cells can significantly increase susceptibility to invasive fungal infections. Pathogenic fungi cause billions of infections every year and have an unmet clinical need, with many infections associated with unacceptably high mortality rates that primarily affect vulnerable patients with underlying immune defects. Lipid metabolism has been increasingly appreciated to significantly influence macrophage function, particularly of macrophages residing in lipid-rich orga

Optimization of ionizable lipids for aerosolizable mRNA lipid nanoparticles.

Although mRNA lipid nanoparticles (LNPs) are highly effective as vaccines, their efficacy for pulmonary delivery has not yet fully been established. A major barrier to this therapeutic goal is their instability during aerosolization for local delivery. This imparts a shear force that degrades the mRNA cargo and therefore reduces cell transfection. In addition to remaining stable upon aerosolization, mRNA LNPs must also possess the aerodynamic properties to achieve deposition in clinically relevant areas of the lungs. We addressed these challenges by formulating mRNA LNPs with SM-102, the clini

Bioactive signalling lipids as drivers of chronic liver diseases.

Lipids are important in multiple cellular functions, with most having structural or energy storage roles. However, a small fraction of lipids exert bioactive roles through binding to G protein-coupled receptors and induce a plethora of processes including cell proliferation, differentiation, growth, migration, apoptosis, senescence and survival. Bioactive signalling lipids are potent modulators of metabolism and energy homeostasis, inflammation, tissue repair and malignant transformation. All these events are involved in the initiation and progression of chronic liver diseases. In this review,…

Increased unsaturated lipids underlie lipid peroxidation in synucleinopathy brain.

Lipid peroxidation is a process of oxidative degradation of cellular lipids that is increasingly recognized as an important factor in the pathogenesis of neurodegenerative diseases. We were therefore interested in the manifestation of lipid peroxidation in synucleinopathies, a group of neurodegenerative diseases characterized by the central pathology of α-synuclein aggregates, including Parkinson's disease, multiple system atrophy, dementia with Lewy bodies and Alzheimer's disease with Lewy bodies. We assessed lipid peroxidation products, lipid aldehydes, in the amygdala, a common disease-affe…

Endogenous ocular lipids as potential modulators of intraocular pressure.

Elevated intraocular pressure (IOP) is a risk factor in glaucoma, a group of irreversible blinding diseases. Endogenous lipids may be involved in regulation of IOP homeostasis. We present comparative fold analysis of phospholipids and sphingolipids of aqueous humour and trabecular meshwork from human control vs primary open-angle glaucoma and mouse control (normotensive) vs ocular hypertensive state. The fold analysis in control vs disease state was based on ratiometric mass spectrometric data for above classes of lipids. We standardized in vitro assays for rapid characterization of lipids und…

Identifying lipids tightly bound to an integral membrane protein.

Anabaena Sensory Rhodopsin (ASR) is a microbial photosensor from the cyanobacterium Anabaena sp. PCC 7120. It was found in previous studies that ASR co-purifies with several small molecules, although their identities and structural or functional roles remained unclear. Here, we use solid-state nuclear magnetic resonance (SSNMR) spectroscopy and mass spectrometry to characterize these molecules. Numerous correlations atypical for protein amino acids were found and assigned in the SSNMR spectra. The chemical shift patterns correspond to N-acetyl-d-glucosamine, N-acetyl-d-mannosaminuronic acid, a

One-Year Lifestyle Intervention, Muscle Lipids, and Cardiometabolic Risk.

PURPOSE: Low-attenuation muscle (LAM) area at mid-thigh, a computed tomography (CT)-derived index of intramuscular lipids, is associated with insulin resistance, obesity, and type 2 diabetes. The present study aimed at testing the hypothesis that changes in LAM area in trunk muscles from a single abdominal scan could provide relevant information to evaluate the effects of a lifestyle intervention without the use of a mid-thigh CT scan. METHODS: Cardiometabolic risk variables, including waist circumference, lipoprotein-lipid profile, glucose tolerance, and cardiorespiratory fitness, were asses…

Analyzing T-Cell Plasma Membrane Lipids by Flow Cytometry.

Plasma membrane lipid rafts are highly ordered membrane microdomains enriched for glycosphingolipids and cholesterol, which play an important role during T-cell antigen receptor (TCR) signaling. Our previous work has demonstrated that plasma membrane lipid composition is an important determinant of human CD4+ T-cell function and that defects in lipid raft expression contribute to CD4+ dysfunction in patients with autoimmunity. In this chapter we share three flow cytometry-based methods to quantitatively analyze plasma membrane lipid composition in primary human CD4+ T cells. We describe the qu…

Effects of oxidized lipids and lipoproteins on cardiac function.

Oxidative modifications of lipids and lipoproteins have long been linked to the pathogenesis of cardiovascular diseases including atherosclerosis and coronary disease. Furthermore, overwhelming evidence indicate that oxidized lipids are also associated with myocardial dysfunction and cardiomyopathy. Oxidized lipid derivatives are generated by enzymatic and non-enzymatic reactions with unsaturated lipids in the cell and foods. In addition, blood LDL particles are prone to oxidation leading to the formation of oxidized LDL (oxLDL), which is often associated with obesity, diabetes and metabolic d…

Maternal Lipids and Fetal Overgrowth: Making Fat from Fat.

There is increasing recognition that maternal glucose concentrations lower than those previously used for diagnosis of gestational diabetes mellitus (GDM) and targeted for treatment can result in excess fetal growth. Yet, mothers with GDM who appear to have optimal glycemic control and mothers with obesity and normal glucose tolerance still have a significantly increased risk for delivering infants who are large for gestational age, or even more importantly, who have increased adiposity at birth. What is less appreciated is that in addition to glucose, maternal lipids are also substrates for f…

Lipids characterization of ultrasound and microwave processed germinated sorghum.

BACKGROUND: Cereal crops and oilseeds provide diverse pool of fatty acids with characteristic properties. Sorghum (Sorghum bicolor (L.) Moench) provides the staple food with serving as main source of energy and protein. Germination of sorghum generally increases the nutritive value of seeds and the effects of germination on lipids composition of seeds vary greatly with processing conditions. Therefore, the current study was conducted to compare the effect of emerging processing techniques such as ultrasound (US) and microwave (MW) on fatty acids composition and oil yield of sorghum seeds befor…

Variations of cocoon external lipids during wolf spiderlings' development.

This study presents the first characterisation of the silk lipids of the cocoons of a wolf spider (Pardosa saltans). Wolf spiders' maternal behaviour is complex and involves guarding a cocoon for several weeks, and so cocoons must emit cues to ensure their care and the development of juveniles. We investigated cues associated with the cocoon silk. We assessed qualitative changes of the lipid contents and the composition of cocoon silk in relation to the development of P. saltans wolf spider juveniles. Gas chromatography-mass spectrometry of P. saltans' cocoon silk revealed 62 different lipid c…
