M6A-modified circArhgap26 attenuates cardiac ischemia‒reperfusion injury by suppressing plakophilin-1 palmitoylation.

Cardiac ischemia‒reperfusion (I/R) injury is a leading cause of disability and mortality worldwide, but the underlying mechanism remains largely unknown. Despite the emerging recognition of circular RNAs (circRNAs) as pivotal regulators of cardiac development and disease, their roles in cardiac I/R injury have yet to be thoroughly investigated. In this study, we identified a circRNA named circArhgap26, which is regulated by m6A modification. The expression of circArhgap26 was significantly decreased in the I/R myocardium. Cardiac-specific overexpression of circArhgap26 ameliorated cardiac dysf

Lipid Disorder and PIP2-Regulated Clustering of Syntaxin-1 JMD-TMD Regions Govern Membrane Fusion Competence.

Syntaxin-1 (Stx1), a core neuronal SNARE protein, regulates membrane docking and fusion during neurotransmitter release. Although Stx1 clustering is widely observed at presynaptic active zones and in neuroendocrine cells, how Stx1 isoforms and lipid interactions organize the plasma membrane prior to fusion remains unclear. Here, we performed MARTINI coarse-grained molecular dynamics (cgMD) simulations of 1-10 copies of the juxtamembrane and transmembrane domains (JMD-TMDs) of Stx1A and Stx1B embedded in plasma membrane models to investigate membrane lipid disorder, Stx1 JMD-TMD clustering dyna

Identification and validation of palmitoylation-related biomarkers in gestational diabetes mellitus.

Palmitoylation plays a crucial role in the pathophysiology of diabetes, and an increase in palmitoylation may inhibit the function of insulin receptors, thereby affecting the progression of gestational diabetes mellitus (GDM). However, its involvement in gestational diabetes mellitus (GDM) remains underexplored. This study analyzed GDM-related datasets and 30 palmitoylation-related genes (PRGs), identifying MNDA, FCGR3B, and AQP9 as significantly upregulated biomarkers in GDM samples. Consistent with the dataset analysis, reverse transcription-polymerase chain reaction (RT-qPCR) confirmed elev

Palmitoylation at a conserved cysteine residue facilitates gasdermin D-mediated pyroptosis and cytokine release.

Gasdermin D (GSDMD)-mediated pyroptotic cell death drives inflammatory cytokine release and downstream immune responses upon inflammasome activation, which play important roles in host defense and inflammatory disorders. Upon activation by proteases, the GSDMD N-terminal domain (NTD) undergoes oligomerization and membrane translocation in the presence of lipids to assemble pores. Despite intensive studies, the molecular events underlying the transition of GSDMD from an autoinhibited soluble form to an oligomeric pore form inserted into the membrane remain incompletely understood. Palmitoylation at Residue C221 of Japanese Encephalitis Virus NS2A Protein Contributes to Viral Replication Efficiency and Virulence.

Palmitoylation of viral proteins is crucial for host-virus interactions. In this study, we examined the palmitoylation of Japanese encephalitis virus (JEV) nonstructural protein 2A (NS2A) and observed that NS2A was palmitoylated at the C221 residue of NS2A. Blocking NS2A palmitoylation by introducing a cysteine-to-serine mutation at C221 (NS2A/C221S) impaired JEV replication in vitro and attenuated the virulence of JEV in mice. NS2A/C221S mutation had no effect on NS2A oligomerization and membrane-associated activities, but reduced protein stability and accelerated its degradation through the

All about that fat: Lipid modification of proteins in Cryptococcus neoformans.

Lipid modification of proteins is a widespread, essential process whereby fatty acids, cholesterol, isoprenoids, phospholipids, or glycosylphospholipids are attached to polypeptides. These hydrophobic groups may affect protein structure, function, localization, and/or stability; as a consequence such modifications play critical regulatory roles in cellular systems. Recent advances in chemical biology and proteomics have allowed the profiling of modified proteins, enabling dissection of the functional consequences of lipid addition. The enzymes that mediate lipid modification are specific for b

EFR3s are palmitoylated plasma membrane proteins that control responsiveness to G-protein-coupled receptors.

The yeast Efr3p protein is a main regulator of the Stt4p phosphatidylinositol 4-kinase at contact sites between the endoplasmic reticulum and the plasma membrane. A mutation in its fly homologue Rbo, leads to diminished light responses in the eye attributed to progressively impaired PLC signaling. Here, we find that Efr3s plays a role in maintaining responsiveness to the type-I angiotensin II (AngII) receptors. siRNA-mediated depletion of EFR3A and EFR3B impaired the sustained phase of cytosolic Ca(2+) response to high concentration of AngII in HEK293 cells that express wild type but not trunc

Context-dependent roles of palmitoylation in acute respiratory distress syndrome: integrating inflammation, cell death and repair.

Acute respiratory distress syndrome (ARDS) is a life-threatening condition characterised by dysregulated inflammation, immune imbalance and impaired alveolar repair. Despite advances in supportive care, effective targeted therapies remain limited. Palmitoylation, a reversible lipid-based post-translational modification, has recently emerged as a regulatory mechanism in ARDS pathogenesis. Acting in a context-dependent manner, palmitoylation affects key processes, including immune activation, programmed cell death and epithelial remodelling. Accumulating evidence suggests that palmitoylation may

Columbianadin targets TRIM7 to maintain P2X7 palmitoylation, inhibiting cuproptosis in synovial M2 macrophages.

BACKGROUND: Traumatic synovitis often arises from sports injuries, and early intervention is crucial for preserving joint health. Columbianadin (CBN), a natural phytochemical, has demonstrated efficacy in inhibiting inflammatory progression through Chinese herbal injection therapy; however, its underlying mechanism remains unclear. PURPOSE: This study aims to investigate the molecular mechanism by which CBN maintains P2X7 palmitoylation, characterize TRIM7 as a novel cuproptosis-alert protein, and provide a theoretical foundation for the bench-to-bedside translation of CBN in treating traumat

Complementary crosstalk between palmitoylation and phosphorylation events in MTIP regulates its role during Plasmodium falciparum invasion.

Post-translational modifications (PTMs) including phosphorylation and palmitoylation have emerged as crucial biomolecular events that govern many cellular processes including functioning of motility- and invasion-associated proteins during Plasmodium falciparum invasion. However, no study has ever focused on understanding the possibility of a crosstalk between these two molecular events and its direct impact on preinvasion- and invasion-associated protein-protein interaction (PPI) network-based molecular machinery. Here, we used an integrated in silico analysis to enrich two different catalogu

Legionella pneumophila Effector LpdA Is a Palmitoylated Phospholipase D Virulence Factor.

Legionella pneumophila is a bacterial pathogen that thrives in alveolar macrophages, causing a severe pneumonia. The virulence of L. pneumophila depends on its Dot/Icm type IV secretion system (T4SS), which delivers more than 300 effector proteins into the host, where they rewire cellular signaling to establish a replication-permissive niche, the Legionella-containing vacuole (LCV). Biogenesis of the LCV requires substantial redirection of vesicle trafficking and remodeling of intracellular membranes. In order to achieve this, several T4SS effectors target regulators of membrane trafficking, w

Palmitoylation of gephyrin controls receptor clustering and plasticity of GABAergic synapses.

Postsynaptic scaffolding proteins regulate coordinated neurotransmission by anchoring and clustering receptors and adhesion molecules. Gephyrin is the major instructive molecule at inhibitory synapses, where it clusters glycine as well as major subsets of GABA type A receptors (GABAARs). Here, we identified palmitoylation of gephyrin as an important mechanism of strengthening GABAergic synaptic transmission, which is regulated by GABAAR activity. We mapped palmitoylation to Cys212 and Cys284, which are critical for both association of gephyrin with the postsynaptic membrane and gephyrin cluste

Global analysis of protein palmitoylation in African trypanosomes.

Many eukaryotic proteins are posttranslationally modified by the esterification of cysteine thiols to long-chain fatty acids. This modification, protein palmitoylation, is catalyzed by a large family of palmitoyl acyltransferases that share an Asp-His-His-Cys Cys-rich domain but differ in their subcellular localizations and substrate specificities. In Trypanosoma brucei, the flagellated protozoan parasite that causes African sleeping sickness, protein palmitoylation has been observed for a few proteins, but the extent and consequences of this modification are largely unknown. We undertook the

Targeting the ZDHHC9-mediated STAT1 palmitoylation-phosphorylation conversion inhibits gastric cancer progression.

BACKGROUND: Palmitoyl transferases ZDHHCs and its mediated protein palmitoylation in several malignant tumors have been reported. However, the role in gastric cancer (GC) has not yet been fully elucidated. METHODS: Various experimental techniques were employed to investigate the role of ZDHHCs and related protein palmitoylation in GC, mainly including acyl-biotinyl exchange assay, co-immunoprecipitation, immunohistochemistry, western blot, immunofluorescence, and confocal imaging. RESULTS: We identified widespread protein palmitoylation in GC, and the palmitoylation inhibitor 2-bromopalmitat

Acetyl-CoA acyltransferase 2 palmitoylation drives liver fibrosis by inducing hepatic stellate cell ferroptosis.

Hepatic fibrosis is a major driver of mortality in metabolic dysfunction-associated steatotic liver disease (MASLD)-previously known as non-alcoholic fatty liver disease (NAFLD). While hepatic stellate cell (HSC) activation and myofibroblast accumulation are central to fibrogenesis, the regulatory mechanisms remain incompletely understood. Acetyl-CoA acyltransferase 2 (ACAA2), a pivotal enzyme in fatty acid oxidation, has been implicated in lipid metabolism but has not been investigated as a therapeutic target in MASLD. Here, we show that ACAA2 upregulation in HSCs exacerbates hepatic fibrosis

Palmitoylation of CLDN12 regulated by ZDHHC7 and APT1/2 promotes hepatitis C virus entry.

UNLABELLED: Hepatitis C virus (HCV) entry is a critical step in establishing infection and is highly dependent on host cell co-receptors. We previously identified that the tight junction protein CLDN12 facilitates HCV entry. However, the regulatory mechanisms governing the dynamic post-translational modifications of CLDN12 remain unclear. This study employs targeted mutagenesis, palmitoylation level analysis, plasma membrane localization, manipulation of key enzymes, and functional virology assays to reveal whether and how reversible palmitoylation regulates the function of CLDN12 in HCV entry

Integrated analysis identifies a palmitoylation-associated prognostic model (ACSM5/SKA3) for lung adenocarcinoma across multiple cohorts.

BACKGROUND: Lung adenocarcinoma (LUAD) is the most common subtype of non-small cell lung cancer and remains associated with poor prognosis despite therapeutic advances. Protein palmitoylation, particularly reversible S-palmitoylation, modulates oncogenic signaling, membrane localization, and immune checkpoint stability, but the prognostic landscape of palmitoylation-related genes (PRGs) in LUAD and their relationship with the tumor microenvironment are not well defined. METHODS: PRGs were curated from GeneCards and integrated with transcriptomic and clinical data from The Cancer Genome Atlas

Palmitoylation regulates norepinephrine transporter uptake, surface localization, and total expression with pathogenic implications in postural orthostatic tachycardia syndrome.

Postural orthostatic tachycardia syndrome (POTS) is an adrenergic signaling disorder characterized by excessive plasma norepinephrine, postural tachycardia, and syncope. The norepinephrine transporter (NET) modulates adrenergic homeostasis via the reuptake of extracellular catecholamines and is implicated in the pathogenesis of adrenergic and neurological disorders. In this study, we reveal NET is palmitoylated in male Sprague-Dawley rats and Lilly Laboratory Cell Porcine Kidney (LLC-PK1) cells. S-palmitoylation, or the addition of a 16-carbon saturated fatty acid, is a reversible post-transla

Inhibition of Fatty Acid Translocase (FAT/CD36) Palmitoylation Enhances Hepatic Fatty Acid β-Oxidation by Increasing Its Localization to Mitochondria and Interaction with Long-Chain Acyl-CoA Synthetase 1.

Aims: Impaired fatty acid oxidation (FAO) in mitochondria of hepatocytes causes lipid accumulation and excessive production of reactive oxygen species (ROS) and oxidative damage, leading to nonalcoholic fatty liver disease (NAFLD). Fatty acid translocase (FAT/cluster of differentiation 36 [CD36]), a transmembrane protein that facilitates the uptake of long-chain fatty acids (LCFAs), is recently found to be involved in FAO. The function of FAT/CD36 is associated with its subcellular localization. Palmitoylation, one of the most common lipid modifications, is generally thought to regulate FAT/CD
