Oxidative stress and female infertility: the role of follicular fluid soluble receptor of advanced glycation end-products (sRAGE) in women with endometriosis.

Oxidative Stress (OS) relates to the pathophysiology of endometriosis by activation of the inflammation process in the ovary, abdomen, peritoneum and endometrium. Advanced Glycation end-products (AGEs) cause oxidative damage to the follicles of the ovary. This study aims to investigate the correlation of follicular fluid soluble receptor of AGEs (FF sRAGE) with fertility-related parameters in infertile women with endometriosis. From January 2012 to July 2015 twenty-four women diagnosed with mild to moderate endometriosis aged 28-38 years underwent assisted reproduction. sRAGE levels measured i

Dietary Exposure to Acrylamide Has Negative Effects on the Gastrointestinal Tract: A Review.

Changing eating habits and an increase in consumption of thermally processed products have increased the risk of the harmful impact of chemical substances in food on consumer health. A 2002 report by the Swedish National Food Administration and scientists at Stockholm University on the formation of acrylamide in food products during frying, baking and grilling contributed to an increase in scientific interest in the subject. Acrylamide is a product of Maillard's reaction, which is a non-enzymatic chemical reaction between reducing sugars and amino acids that takes place during thermal processi

Glycation of fibronectin impairs angiopoietin-1/Tie-2 signaling through uncoupling Tie-2-α5β1 integrin crosstalk.

The dysfunction of angiopoietin-1 (Ang-1)/Tie-2 signaling pathways has been implicated in diabetic complications. However, the underlying molecular mechanisms remain unclear. Fibronectin (FN) is thought to have an important role in regulating Ang-1/Tie-2 signaling activation. But no previous study has investigated the effects of FN glycation on Ang-1/Tie-2 signaling. In the present study, FN was glycated by methylglyoxal (MGO) to investigate whether the glycation of FN contributes to diabetes-induced Ang-1/Tie-2 signaling impairment and to understand the molecular mechanisms involved. The resu

In vitro glycation of a tissue-engineered wound healing model to mimic diabetic ulcers.

Diabetic foot ulcers are a major complication of diabetes that occurs following minor trauma. Diabetes-induced hyperglycemia is a leading factor inducing ulcer formation and manifests notably through the accumulation of advanced glycation end-products (AGEs) such as N-carboxymethyl-lysin. AGEs have a negative impact on angiogenesis, innervation, and reepithelialization causing minor wounds to evolve into chronic ulcers which increases the risks of lower limb amputation. However, the impact of AGEs on wound healing is difficult to model (both in vitro on cells, and in vivo in animals) because i

Inhibition of advance glycation end products formation, gastrointestinal digestion, absorption and toxicity: A comprehensive review.

Advanced glycation end-products (AGEs) are the final products of the non-enzymatic interaction between reducing sugars and amino groups in proteins, lipids and nucleic acids. In numerous diseases, such as diabetes, neuropathy, atherosclerosis, aging, nephropathy, retinopathy, and chronic renal illness, accumulation of AGEs has been proposed as a pathogenic mechanism of inflammation, oxidative stress, and structural tissue damage leading to chronic vascular issues. Current studies on the inhibition of AGEs mainly focused on food processing. However, there are few studies on the inhibition of AG

Divergent sensory-metabolic fingerprints in Longjing peach tea beverage: Decoding quality evolution under HPP versus UHT sterilization.

Thermal sterilization often compromises beverage quality, making non-thermal sterilization a potential alternative. This study systematicall … d by UHT was mainly attributed to the loss of heat-sensitive flavor substances and the formation of Maillard reaction and thermal oxidation products. The impact of HPP, however, primarily resulted from the release and complexation of compounds under high pressure. This research elucidated the differential effects of the two sterilization techniques at both sensory …

Tea Polyphenols as Inhibitors of Furan Formed in the Maillard Model System and Canned Coffee Model.

In this article, the effects of sugars and amino acids on furan formation via the Maillard reaction in low-moisture model systems were investigated. Glucose and alanine are important furan precursors, and the effects of the heating temperature, heating time, and molar ratio of glucose to alanine on furan formation were studied in glucose/alanine model system by response surface methodology. The heating temperature greatly affected furan formation. The maximum furan concentration was obtained with a glucose-to-alanine molar ratio of 0.83:1.00, by heating at 151 °C for 41 min. Tea polyphenols ef…

A Novel Umami Peptide Selector (UPS) for Identifying Key Peptides from Maillard-Reacted Chicken Hydrolyzates: From Taste Evaluation to Antioxidant Properties.

The Maillard reaction (MR) enhances the flavor of chicken enzymatic hydrolyzates, yet pinpointing key umami peptides remains challenging due to complex matrices. Sensory evaluation showed increased umami intensity despite decreased equivalent umami concentration (EUC: 0.29-0.41 g/100  g dry weight), suggesting that peptides may contribute substantially to the observed umami perception beyond the contribution captured by EUC. We developed a novel Umami Peptide Selector (UPS) integrating quantitative peptidomics with predicted taste thresholds to identify potent peptides. Guided by UPS, five key…

Effect of rice flour extrusion on α-dicarbonyl compounds, protein glycation products, and the formation of acrylamide and 5-hydroxymethylfurfural in bread crust-like system.

Extrusion conditions and particle size significantly influenced the formation of Maillard reaction products (MRP) in rice flour and bread crust-like model system. In extruded rice flour (<132 μm and > 132-200 μm; barrel temperatures 80 °C and 120 °C; feed moisture 15% and 30%), 3-deoxyglucosone and methylglyoxal were the predominant α-dicarbonyl compounds. In rice flour samples, higher feed moisture (30%) combined with elevated temperature increased 3-deoxyglucosone and furosine concentrations, while N-ε-carboxymethyllysine and N-ε-carboxyethyllysine levels declined. In crust-like model system…

Antibody-based detection of advanced glycation end-products: promises vs. limitations.

Advanced glycation end-products (AGEs) of the Maillard reaction were originally measured according to their fluorescent and browning properties. A subsequent study with instrumental analyses such as high-performance liquid chromatography and gas chromatography mass spectrometry more clearly demonstrated the involvement of each AGE structure in pathological conditions. Furthermore, immunochemical methods have also been developed to clarify the localization of AGEs in tissues and measurement of AGEs in multiple clinical samples. Although the involvement of AGEs in age-related diseases has progre…

Divergent specificity of PatA, GabT, and IlvE defines the branched transamination of Nε-carboxymethyllysine and its metabolite N-carboxymethylcadaverine in Escherichia coli.

Thermal food processing generates Nε-carboxymethyllysine (CML), a key advanced glycation end product (AGE) and marker of the Maillard reaction in food. Escherichia coli utilizes CML as a nitrogen source. While SpeC initiates degradation by decarboxylating CML to N-carboxymethylcadaverine (CM-Cad), the enzymes liberating the nitrogen remained unknown. Here, we identify PatA, GabT, and IlvE as glutamate-dependent transaminases responsible for CML and CM-Cad transamination. Our results reveal a branched metabolic network, not a linear pathway: PatA shows specificity toward both substrates, while …
