Mannose-binding lectin and the risk of HIV transmission and disease progression in children: a systematic review.

BACKGROUND: Mannose-binding lectin (MBL) can activate the complement system by binding to carbohydrates, such as those presented on the HIV virion surface. It is unclear whether genetically determined MBL deficiency is related to vertical HIV transmission and disease progression in HIV-infected children. METHODS: A literature search of Medline, Embase and Cochrane Central Register identified all relevant studies on MBL and HIV infection in children. We extracted information on the characteristics of the study group, method of MBL analysis, outcome definitions, follow-up and the risk estimates…

Elevated levels of vitamin D and deficiency of mannose binding lectin in dengue hemorrhagic fever.

BACKGROUND: Altered plasma concentrations of vitamin D and mannose binding lectin (MBL), components of innate immunity, have been shown to be associated with the pathogenesis of viral infections. The objective of the present study was to find out whether plasma concentrations of MBL and vitamin D are different in patients with dengue fever (DF) and dengue hemorrhagic fever (DHF). THE RESULTS: The plasma concentrations of vitamin D and MBL were assessed in 48 DF cases, 45 DHF cases and 20 apparently healthy controls using ELISA based methods. Vitamin D concentrations were found to be higher am…

The mannose receptor binds Trichuris muris excretory/secretory proteins but is not essential for protective immunity.

Trichuris muris is a natural mouse model of the human gastrointestinal nematode parasite Trichuris trichiura and it is well established that a T helper type 2-dominated immune response is required for worm expulsion. Macrophages accumulate in the large intestine of mice during infection and these cells are known to express the mannose receptor (MR), which may act as a pattern recognition receptor. The data presented here show for the first time that T. muris excretory/secretory products (E/S) induce bone-marrow-derived macrophages (BMDM) to produce several cytokines and have MR-binding activit…

Mannose-binding lectin as a predictor of microalbuminuria in type 1 diabetes: an inception cohort study.

Inflammation and complement activation via the mannose-binding lectin (MBL) pathway have been suggested to play a role in the pathogenesis of diabetic microvascular complications. The association between the complement-activating protein MBL and the development of persistent microalbuminuria was evaluated in an inception cohort of 286 newly diagnosed type 1 diabetic patients consecutively admitted to the Steno Diabetes Center between 1 September 1979 and 31 August 1984. Serum MBL was measured with an immunofluorometric assay in 270 of the patients (159 men) after 3 years of diabetes duration. …

[Effect of local anesthetics on the transport of mannose in the microsomal membranes of Aspergillus niger van Tieghem].

The influence of various local anaesthetics has been studied on the mannose transfer enzymic system which is localized in Aspergillus niger van Tieghem microsomal membranes. The n-alkanes and tertiary amines do not seem to react on the first step of the reaction: i.e. the polyprenylphosphate-mannose biosynthesis. However two amines, the dibucaine and to a lesser extent the tetracaine, inhibit the mannose transfer from the polyprenylphosphate-mannose to the endogenous proteins.

Mannose attenuates CCl4-induced liver fibrosis by reducing oxidative stress through AMPK-dependent NRF2 signaling.

Mannose, a naturally occurring hexose, has been implicated in the regulation of metabolism and inflammation. Although emerging evidence suggests its therapeutic potential in liver diseases, the mechanisms by which mannose modulates liver fibrosis, particularly in relation to oxidative stress, remain poorly defined. This study aimed to investigate the protective effects of mannose in liver fibrosis and elucidate the underlying mechanisms. A CCl4-induced mouse model of liver fibrosis was established with mannose administered in drinking water. Transcriptomic analysis was performed to explore pot…

Mannose-engineered Eu-MOF for ratiometric fluorescent/colorimetric dual-detection and inhibition of E. coli.

Reliable detection and fast inhibition of Escherichia coli (E. coli) are critical for public health and food safety, so the detection-inhibition-integrated strategies become critical. Herein, a mannose-engineered 3,5-dicarboxyphenylboronic acid/curcumin dual-ligand Eu-metal-organic framework (Eu-MOF) was designed as dcMOF@man. Mannose realized the specific identification to E. coli; Eu-MOF exhibited dual-emission at 350 and 633 nm for robust ratiometric fluorescence sensing to E. coli as boronic acid-surface glycan interaction enhanced the emission at 350 nm, while mannose-FimH-mediated intera…

Augmenting hepatitis C E1E2 glycoprotein antibody detection via an alternative lectin-assisted mannose-binding ELISA.

In this study we compared several plant lectins for use in a lectin capture ELISAs to enrich HCV glycoproteins from cellular extracts for subsequent detection of HCV glycoprotein-specific antibodies. We selected various high-mannose plant lectins and assessed for their properties to capture HCV glycoproteins. Three lectins, Galanthus nivalis lectin (GNL), Narcissus Pseudonarcissus Lectin (NPL) and Hippeastrum hybrid lectin (HHL) demonstrated effective capturing properties. These were subsequently tested for specificity and sensitivity against viral isolates spanning diverse HCV genotypes. Furt…

Anticancer activity of Pseudomonas fluorescens lectin (PFL) targeting high-mannose glycans on breast cancer cells.

Aberrant glycosylation is one of the key characteristics of cancer cells. High-mannose-type N-glycans are highly expressed in various types of cancer, making them potentially important therapeutic targets. As a basis for developing new therapeutic drugs targeting high-mannose glycans, we employed Pseudomonas fluorescens lectin (PFL) that specifically binds to high-mannose glycans as a model compound, and analyzed the cellular responses of MDA-MB-231 and T47D breast cancer cell lines. In both cell types, PFL induced the internalization and degradation of cancer-associated molecules such as epid…

Targeted delivery of kaempferol via mannose-modified PLGA nanoparticles reprograms macrophages and ameliorates rheumatoid arthritis.

BACKGROUND: While the natural flavonoid Kaempferol (Kae) possesses promising anti-inflammatory and immunomodulatory properties for treating rheumatoid arthritis (RA), it is challenged by poor aqueous solubility and low bioavailability, which impede its targeting of key effector cells like macrophages and limit its clinical utility. PURPOSE: To overcome Kae's pharmaceutical limitations in RA therapy, We developed mannose-modified PLGA nanoparticles (Kae-NPs) for macrophage-targeted delivery and investigated their therapeutic efficacy and underlying mechanisms. METHODS: Integrated bioinformati…

A mannose-rich exopolysaccharide from Weissella cibaria SK01: Purification, characterization, antibiofilm, anticancer, and physicochemical properties.

Among lactic acid bacteria (LAB) strains isolated from minced beef, an effective exopolysaccharide (EPS)-producing bacterium was identified using 16S rRNA gene sequencing as Weissella cibaria SK01. A distinctive EPS, designated EPS-SK01, was produced in relatively high yield (2.79 g/L) and purified to apparent homogeneity by consecutive chromatographic procedures. Structural characterization using monosaccharide composition analysis, FTIR, GPC, and one-dimensional NMR revealed that EPS-SK01 is mainly composed of α-linked mannose residues, necessitating further validation through advanced struc…

Mannose metabolism inhibition sensitizes acute myeloid leukaemia cells to therapy by driving ferroptotic cell death.

Resistance to standard and novel therapies remains the main obstacle to cure in acute myeloid leukaemia (AML) and is often driven by metabolic adaptations which are therapeutically actionable. Here we identify inhibition of mannose-6-phosphate isomerase (MPI), the first enzyme in the mannose metabolism pathway, as a sensitizer to both cytarabine and FLT3 inhibitors across multiple AML models. Mechanistically, we identify a connection between mannose metabolism and fatty acid metabolism, that is mediated via preferential activation of the ATF6 arm of the unfolded protein response (UPR). This in…

Toward understanding tissue-specific symptoms in dolichol-phosphate-mannose synthesis disorders; insight from DPM3-CDG.

The congenital disorders of glycosylation (CDG) are inborn errors of metabolism with a great genetic heterogeneity. Most CDG are caused by defects in the N-glycan biosynthesis, leading to multisystem phenotypes. However, the occurrence of tissue-restricted clinical symptoms in the various defects in dolichol-phosphate-mannose (DPM) synthesis remains unexplained. To deepen our understanding of the tissue-specific characteristics of defects in the DPM synthesis pathway, we investigated N-glycosylation and O-mannosylation in skeletal muscle of three DPM3-CDG patients presenting with muscle dystro…
