Parietal dysfunction during number processing in children with fetal alcohol spectrum disorders.

Number processing deficits are frequently seen in children prenatally exposed to alcohol. Although the parietal lobe, which is known to mediate several key aspects of number processing, has been shown to be structurally impaired in fetal alcohol spectrum disorders (FASD), effects on functional activity in this region during number processing have not previously been investigated. This fMRI study of 49 children examined differences in activation associated with prenatal alcohol exposure in five key parietal regions involved in number processing, using tasks involving simple addition and magnitu

Impaired acuity of the approximate number system underlies mathematical learning disability (dyscalculia).

Many children have significant mathematical learning disabilities (MLD, or dyscalculia) despite adequate schooling. The current study hypothesizes that MLD partly results from a deficiency in the Approximate Number System (ANS) that supports nonverbal numerical representations across species and throughout development. In this study of 71 ninth graders, it is shown that students with MLD have significantly poorer ANS precision than students in all other mathematics achievement groups (low, typically, and high achieving), as measured by psychophysical assessments of ANS acuity (w) and of the ma

The effects of a secondary task on forward and backward walking in Parkinson's disease.

BACKGROUND: People with Parkinson's disease (PD) often fall while multitasking or walking backward, unavoidable activities in daily living. Dual tasks involving cognitive demand during gait and unfamiliar motor skills, such as backward walking, could identify those with fall risk, but dual tasking while walking backward has not been examined in those with PD, those who experience freezing of gait (FOG), or healthy older controls. METHODS: A total of 78 people with PD (mean age = 65.1 +/-9.5 years; female, 28%) and 74 age-matched and sex-matched controls (mean age = 65.0 +/-10.0 years; female,

Incorporating Lesion-Level Heterogeneity in a Branching-Process Model of Ovarian Cancer Metastasis.

Danesh et al. (J. Theor. Biol., 2012) estimated the ovarian cancer screening window at roughly 2.9 years, using a multi-type branching process and the hitting time of an aggregate mass threshold. We extend the model of Danesh et al. to incorporate the lesion-level heterogeneity characteristic of intraperitoneal metastasis, replacing the homogeneous secondary compartment by a vector of distinct lesions whose formation follows a Chinese Restaurant Process with concentration parameter α . We prove three results. Because established metastases divide faster than the peritoneal source compartment,

Predicting biological age using an accumulated neurotoxicity biomarker for amyloid beta oligomers.

This study proposes using accumulated neurotoxicity, defined as the time integral of Aβ oligomer concentration, as a biomarker for neuronal aging. A relationship between biological age and accumulated neurotoxicity is proposed. Numerical analysis guided the development of a new analytical solution linking the biological and calendar ages of neurons. The effects of Aβ monomer and oligomer half-lives, key indicators of proteolytic efficiency, on biological age are examined. Both constant and age-dependent (exponentially increasing) half-life scenarios are considered. The findings indicate that i

A partially shared magnitude system: Common and specialized neural substrates underlying spatial, numerical, and temporal arithmetic.

Magnitude, such as time, space and number, constitutes a foundational construction of perceptual and cognitive experience. A Theory of Magnitude proposes that these dimensions are processed by a shared magnitude system. However, it remains unclear how this system supports the manipulation of multiple continuous magnitude information, such as arithmetic operation across domains and whether it operates as a fully shared or partially specialized subsystem. Using functional magnetic resonance imaging, we investigated magnitude arithmetic in numerical, spatial and temporal domains. Conjunction anal

Numerical Analysis of BIMs for Stochastic SIR and SIS Models with Variable Contact Diffusion Rates.

Qualitative properties of adequate numerical methods for stochastic differential equations to simulate the stochastic counterparts of some known models with variable contact diffusion rates in mathematical epidemiology are investigated, especially SIS and SIR models with non-constant total population sizes on bounded domains. We show the convergence of numerical approximations based on Balanced Implicit Methods (BIMs) by proving several properties; including positivity, invariance, stability, mean and mean square consistency, local uniform boundedness, mean square Hölder continuity and mean sq

Hierarchical mathematical modelling of patients with myeloproliferative neoplasms captures interferon-α treatment responses and allows for personalised and population predictions.

The Philadelphia chromosome-negative myeloproliferative neoplasms (MPNs) are a group of haematological malignancies triggered by a driver mutation, most commonly the JAK2 V617F mutation, which is acquired in a haematopoietic stem cell. The diseases are characterised by an overproduction of myeloid cells and may result in severe complications such as thrombosis, myelofibrotic transition, and potentially progression to acute myeloid leukaemia. Treatment with interferon-α (IFN-α) can potentially deplete the disease-driving malignant stem cell population, thereby leading to long-term remission. We

Mathematical modeling of the effects of potassium ions on the tumor immune system and virotherapy.

To understand how extracellular potassium ions influence both anti-tumor immunity and virotherapy in cancer progression, we construct two mathematical models informed by our experimental results. For the tumor immune system, our analysis shows a high concentration of extracellular potassium ions diminishes the killing rate of tumor cells by immune cells. Our model confirms that the stimulation coefficient of immune cells by tumor cells remains a crucial parameter in the presence of extracellular potassium ions, which largely controls the overall tumor growth. For virotherapy, we obtain a formu

Optimal control of monomers and oligomers degradation in an Alzheimer's disease model.

The aggregation and accumulation of oligomers of misfolded Aβ-amyloids in the human brain is one of the possible causes for the onset of the Alzheimer’s disease in the early stage. We introduce and study a new ODE model for the evolution of Alzheimer’s disease based on the interaction between monomers, proto-oligomers, and oligomers of Aβ amyloid protein in a small portion of the human brain, based upon biochemical processes such as polymerization, depolymerization, fragmentation and concatenation. We further introduce the possibility of controlling the evolution of the system via a treatment

A 3D-1D-0D multiscale model of the neuro-glial-vascular unit for synaptic and vascular dynamics in the dorsal vagal complex.

Cerebral blood flow regulation is critical for brain function, and its disruption is implicated in various neurological disorders. Many existing models do not fully capture the complex, multiscale interactions among neuronal activity, astrocytic signaling, and vascular dynamics, especially in key brainstem regions. In this work, we present a 3D-1D-0D multiscale computational framework for modeling the neuro-glial-vascular unit (NGVU) in the dorsal vagal complex (DVC). Our approach integrates a quadripartite synapse model, which captures the dynamic interactions among excitatory and inhibitory

The roles of continuous and discontinuous proliferations on hepatitis B virus infection.

The proliferation of both uninfected and infected hepatocytes, as well as the recycling effects of rcDNA-containing capsids are two key mechanisms playing significant roles in the persistence and clearance of hepatitis B virus (HBV) infection. In this study, the temporal dynamics of this viral infection is investigated through two intercellular mathematical models considering proliferation of both types of hepatocytes (uninfected and infected) and recycling effects of capsids. Both models are formulated on the basis of a key finding in the existing literature: mitosis of an infected hepatocyte

Modelling Mucus Clearance in Sinuses: Thin-Film Flow Inside a Fluid-Producing Cavity Lined with an Active Surface.

The paranasal sinuses are a group of hollow spaces within the human skull, surrounding the nose. They are lined with an epithelium that contains mucus-producing cells and tiny hairlike active appendages called cilia. The cilia beat constantly to sweep mucus out of the sinus into the nasal cavity, thus maintaining a clean mucus layer within the sinuses. This process, called mucociliary clearance, is essential for a healthy nasal environment and disruption in mucus clearance leads to diseases such as chronic rhinosinusitis, specifically in the maxillary sinuses, which are the largest of the para

Information-Theoretic Analysis of a Model of CAR-4-1BB-Mediated NFκB Activation.

Systems biology utilizes computational approaches to examine an array of biological processes, such as cell signaling, metabolomics and pharmacology. This includes mathematical modeling of CAR T cells, a modality of cancer therapy by which genetically engineered immune cells recognize and combat a cancerous target. While successful against hematologic malignancies, CAR T cells have shown limited success against other cancer types. Thus, more research is needed to understand their mechanisms of action and leverage their full potential. In our work, we set out to apply information theory on a ma

Modeling Intracellular Delay in Within-Host HIV Dynamics Under Conditioning of Drugs of Abuse.

Drugs of abuse, such as opiates, have been widely associated with the enhancement of HIV replication, the acceleration of disease progression, and severe neuropathogenesis. Specifically, the presence of drugs of abuse (morphine) switches target cells (CD4[Formula: see text] T cells) from lower-to-higher susceptibility to HIV infection. The effect of such switching behaviors on viral dynamics may be altered due to the intracellular delay (the replication time between viral entry into a target cell and the production of new viruses by the infected cell). In this study, we develop, for the first

A Two-phase mixture model of platelet aggregation.

We present a two-phase model of platelet aggregation in coronary-artery-sized blood vessels. The model tracks the number densities of three platelet populations as well as the concentration of a platelet activating chemical. Through the formation of elastic bonds, activated platelets can cohere with one another to form a platelet thrombus. Bound platelets in a thrombus move in a velocity field different from that of the bulk fluid. Stresses produced by the elastic bonds act on the bound platelet material. Movement of the bound platelet material and that of the background fluid are coupled thro

Typical numerosity adaptation despite selectively impaired number acuity in dyscalculia.

It has been suggested that a core deficit of the "number sense" may underlie dyscalculia. We test this idea by measuring perceptual adaptation and discrimination thresholds for numerosity and object size in a group of dyscalculic and typical preadolescents (N = 71, mean age 12). We confirmed that numerosity discrimination thresholds are higher in developmental dyscalculia, while size thresholds are not affected. However, dyscalculics adapted to numerosity in a similar way to typicals. This suggests that although numerosity thresholds are selectively higher in dyscalculia, the mechanisms for pe

Neurostructural correlate of math anxiety in the brain of children.

Adequate mathematical competencies are currently indispensable in professional and social life. However, mathematics is often associated with stress and frustration and the confrontation with tasks that require mathematical knowledge triggers anxiety in many children. We examined if there is a relationship between math anxiety and changes in brain structure in children with and without developmental dyscalculia. Our findings showed that math anxiety is related to altered brain structure. In particular, the right amygdala volume was reduced in individuals with higher math anxiety. In conclusion

Estimating the Size of the HCV Infection Prevalence: A Modeling Approach Using the Incidence of Cases Reported to an Official Notification System.

In this paper we propose two methods to give a first rough estimate of the actual number of hepatitis C virus (HCV)-infected individuals (prevalence) taking into account the notification rate of newly diagnosed infections (incidence of notification) and the size of the liver transplantation waiting list (LTWL) of patients with liver failure due to chronic HCV infection. Both approaches, when applied to the Brazilian HCV situation converge to the same results, that is, the methods proposed reproduce both the prevalence of reported cases and the LTWL with reasonable accuracy. We use two methods
