D2 dopamine and CB2 cannabinoid receptor cooperation restores striatal LTP in an epilepsy model.

Increasing evidence suggests that subcortical structures contribute to seizure susceptibility and cognitive impairment in several epileptic … clamp recordings to assess neuronal excitability, synaptic transmission, and plasticity of striatal medium spiny neurons. Our results reveal progressive alterations in corticostriatal transmission and long-term synaptic plasticity, including long-term potentiation (LTP). Electrophysiological and biochemical assessments showed a reduced NMDA/AMPA ratio and reduced expr …

Cell-type-specific CAG repeat expansions and toxicity of mutant Huntingtin in human striatum and cerebellum.

Brain region-specific degeneration and somatic expansions of the mutant Huntingtin (mHTT) CAG tract are key features of Huntington's disease (HD). However, the relationships among CAG expansions, death of specific cell types and molecular events associated with these processes are not established. Here, we used fluorescence-activated nuclear sorting (FANS) and deep molecular profiling to gain insight into the properties of cell types of the human striatum and cerebellum in HD and control donors. CAG expansions arise at mHTT in striatal medium spiny neurons (MSNs), cholinergic interneurons and …

SWI/SNF chromatin remodeler complex within the reward pathway is required for behavioral adaptations to stress.

Enduring behavioral changes upon stress exposure involve changes in gene expression sustained by epigenetic modifications in brain circuits, … havioral responses to cocaine without impacting midbrain dopamine neurons activity. Within striatal medium spiny neurons, Brg1 gene inactivation reduces the expression of stress- and cocaine-induced immediate early genes, increases levels of heterochromatin and at a global scale decreases chromatin accessibility. Altogether these data demonstrate the pivotal function …

Signaling in striatal neurons: the phosphoproteins of reward, addiction, and dyskinesia.

The striatum is a deep region of the forebrain involved in action selection, control of movement, and motivation. It receives a convergent excitatory glutamate input from the cerebral cortex and the thalamus, controlled by dopamine (DA) released in response to unexpected rewards and other salient stimuli. Striatal function and its dysfunction in drug addiction or Parkinson's disease depend on the interplay between these neurotransmitters. Signaling cascades in striatal medium-sized spiny neurons (MSNs) involve multiple kinases, phosphatases, and phosphoproteins, some of which are highly enrich

Aerobic Exercise Improves Motor Performance by Reducing Total ERK Expression and Hyperexcitability of Striatal D2-MSNs in MPTP-Induced Parkinsonian Mice.

Aerobic exercise improves motor dysfunction in Parkinson's disease (PD), but the striatal cell-type-specific mechanisms underlying this effect remain incompletely understood. Here, we investigated whether altered total extracellular signal-regulated kinase (ERK) expression and Mapk1/3-associated functional changes in dorsolateral striatal dopamine D2 receptor-expressing medium spiny neurons (D2-MSNs) are associated with exercise-related functional recovery in 1-methyl-4-phenyl-1,2,3,6-tetrahydropyridine (MPTP)-induced parkinsonian mice. Mice underwent four weeks of moderate-intensity treadmill…

Dopamine and serotonin cotransmission filters striatonigral synaptic activity via 5-HT1B receptor activation.

The substantia nigra pars reticulata (SNr), a key basal ganglia output nucleus, is modulated by dopamine (DA) believed to be released locally from midbrain DA neurons. Although DA has been proposed to regulate γ-aminobutyric acid (GABA) release from medium spiny neuron (MSN) terminals via presynaptic D1 receptors, the precise mechanisms remain unclear. Using presynaptic optical recordings of synaptic vesicle fusion, calcium influx in D1-MSN synapses together with postsynaptic patch-clamp recordings from SNr neurons, we found that DA inhibits D1-MSN GABA release in a frequency-dependent manner.

Direct and indirect pathway neurons in ventrolateral striatum differentially regulate licking movement and nigral responses.

Drinking behavior in rodents is characterized by stereotyped, rhythmic licking movement, which is regulated by the basal ganglia. It is unclear how direct and indirect pathways control the lick bout and individual spout contact. We find that inactivating D1 and D2 receptor-expressing medium spiny neurons (MSNs) in the ventrolateral striatum (VLS) oppositely alters the number of licks in a bout. D1- and D2-MSNs exhibit different patterns of lick-sequence-related activity and different phases of oscillation time-locked to the lick cycle. On the timescale of a lick cycle, transient inactivation o…

New insights into the mechanism of drug-induced dyskinesia.

Dyskinesia is an extrapyramidal movement disorder characterized by involuntary, repetitive, irregular motions that affect the mouth and face … em. However, recent evidence has suggested that we should focus on the possible damage of GABAergic medium spiny neurons (MSNs). MSNs are the first station in the corticostriato-thalamo-cortical circuit that regulates the amplitude and velocity of movements. Two pathways can be distinguished in this circuit: a direct pathway, which increases movements (hyperkinesia), …

Genetic deletion of A2A adenosine receptors in the striatum selectively impairs habit formation.

A(2A) receptors are a major class of G-protein-coupled receptors for adenosine. Highly expressed in the striatum, on the projection neurons giving rise to the striatopallidal or "indirect" pathway, they have been implicated in sleep, addiction, and other processes, yet their role in the control of striatal circuits and behavior remains unclear. Using established assays from the instrumental learning paradigm, we showed that mice with striatum-specific deletion of A(2A) receptors were selectively impaired in habit formation. After training that generated habitual lever pressing in wild-type con

Longitudinal modeling of human neuronal aging reveals the contribution of the RCAN1-TFEB pathway to Huntington's disease neurodegeneration.

Aging is a common risk factor in neurodegenerative disorders. Investigating neuronal aging in an isogenic background stands to facilitate analysis of the interplay between neuronal aging and neurodegeneration. Here we perform direct neuronal reprogramming of longitudinally collected human fibroblasts to reveal genetic pathways altered at different ages. Comparative transcriptome analysis of longitudinally aged striatal medium spiny neurons (MSNs) in Huntington's disease identified pathways involving RCAN1, a negative regulator of calcineurin. Notably, RCAN1 protein increased with age in reprog…

Somatostatin receptor 1 and 5 double knockout mice mimic neurochemical changes of Huntington's disease transgenic mice.

BACKGROUND: Selective degeneration of medium spiny neurons and preservation of medium sized aspiny interneurons in striatum has been implicated in excitotoxicity and pathophysiology of Huntington's disease (HD). However, the molecular mechanism for the selective sparing of medium sized aspiny neurons and vulnerability of projection neurons is still elusive. The pathological characteristic of HD is an extensive reduction of the striatal mass, affecting caudate putamen. Somatostatin (SST) positive neurons are selectively spared in HD and Quinolinic acid/N-methyl-D-aspartic acid induced excitotox…

Exercise promotes glutamate transporter-mediated glutamate uptake in the striatum to regulate MSN plasticity and alleviate behavioral and functional impairments in PD rats.

This study investigated whether exercise ameliorates behavioral deficits in a rat model of Parkinson's disease (PD) by regulating medium spiny neuron (MSN) plasticity via striatal astrocytic excitatory amino acid transporters (EAATs). The PD model was induced by 6-hydroxydopamine (6-OHDA) and confirmed by apomorphine rotation. Exercised rats underwent 4 weeks of treadmill training. We assessed motor function using beam walking and cylinder tests. Striatal pathology was evaluated via tyrosine hydroxylase (TH) immunohistochemistry, glutamate (Glu) enzyme-linked immunosorbent assay, c-Fos tracing

Assessment of Cell-Type-Specific Excitatory Synaptic Strength in the Dorsolateral Striatum of Goal-Directed and Habitual Cocaine-Seeking Behavior.

With repeated exposure to addictive drugs, there is a shift from drug abuse to drug addiction that is mediated by the transition from goal-directed to habitual control. It is well known that the development of habitual control over behavior relies upon cell-type-specific synaptic changes in both D1 and D2 medium spiny neurons (MSNs) in dorsal striatum. Specifically, habitual behavior is mediated by increased synaptic strength in D1 and D2 MSNs in dorsolateral striatum (DLS), suggesting similar cell-type-specific synaptic changes may underlie the development of habitual cocaine-seeking behavior…

A Golgi study of neuronal architecture in a genetic mouse model for Lesch-Nyhan disease.

Lesch-Nyhan disease (LND) is an inherited disorder associated with deficiency of hypoxanthine-guanine phosphoribosyltransferase (HPRT), an enzyme essential for purine recycling. The clinical manifestations of the disorder and several neurochemical studies have pointed towards a defect in the striatum, but histological studies of autopsied brain specimens have not revealed any consistent abnormalities. An HPRT-deficient (HPRT-) mouse that has been produced as a model for the disease also exhibits neurochemical abnormalities of the striatum without obvious histological correlates. In the current
