BACKGROUND: High heterogeneity among individuals with internet gaming disorder (IGDs) hampers the discovery of neuroimaging findings. To address this heterogeneity, this study adopted the newly proposed approach based on individualized gray matter morphological differences to objectively characterize neuroanatomical subtypes of IGDs.
METHODS: 92 IGDs and 56 demographically matched healthy controls (HCs) were recruited for structural imaging scans and clinical tests. Heterogeneity through discriminative analysis identifies IGD subtypes through individual local GMV aberrances. Voxel-wise volume

Signaling through nicotinic acetylcholine receptors (nAChRs) plays a role in cocaine reward and reinforcement, suggesting that the cholinergic system could be manipulated with therapeutics to modulate aspects of cocaine use disorder (CUD). We examined the interaction between nAChRs and cocaine reinforcement by expressing a hypersensitive β2 nAChR subunit (β2Leu9'Ser) in the ventral tegmental area of male Sprague Dawley rats. Compared to control rats, β2Leu9'Ser rats acquired (fixed ratio) intravenous cocaine self-administration faster and with greater likelihood. By contrast, β2Leu9'Ser rats w

Drugs of abuse regulate the activity of the mesolimbic dopamine (DA) system, and drug-induced changes in ventral tegmental area (VTA) cellular activity and gene regulation are linked to behavioral outputs associated with addiction. Previous work from our lab determined that VTA serum- and glucocorticoid-inducible kinase 1 (SGK1) transcription and catalytic activity were increased by repeated cocaine administration; however, it was unknown if these biochemical changes contributed to cocaine-elicited behaviors. Using transgenic and viral-mediated manipulations, we investigated the role of VTA SG

Ten male opiate addicts, who were current heroin injectors, underwent positron emission tomographic (PET) scanning during exposure to a sequence of six alternating drug related and neutral video cues, on two occasions. After the second scan, each subject received heroin or placebo using a randomised single-blind procedure. This design allowed the investigation of patterns of brain activity during a range of self-reported cue evoked emotional states, both in the presence and absence of heroin. Self-reports of 'urge to use' correlated strongly with increased regional blood flow (rCBF) in the inf

Alterations in blink rate have been reported in several neuropsychiatric disorders presumed to result from abnormal central dopaminergic functions. Increased blink rate in schizophrenia, Tardive dyskinesia, Tourette's syndrome and Meige's disease are associated with enhanced dopaminergic functions. Parkinson's disease is associated with reduced dopaminergic functions and decreased blink rate. Thus, blink rate may reflect striatal and mesolimbic dopaminergic activity. Since acute light exposure suppresses melatonin production and darkness stimulates melatonin secretion, blinking may serve to re

The effects of a new TRH analogue, YM-14673 (N alpha-[[(S)-4-oxo-2-azetidinyl]carbonyl]-L-histidyl-L-prolinamide dihydrate) on spontaneous motor activity (SMA) were compared with those of TRH in rats. Intraperitoneal, oral and intracerebral administrations of YM-14673 caused a significant and dose-dependent increase in SMA. TRH also showed similar actions as YM-14673 did, except after oral dosing. These actions of YM-14673 were about 20-100 times more potent than those of TRH. Pretreatment with haloperidol, alpha-methyl-p-tyrosine (alpha-MT) and amino-oxyacetic acid (AOAA) but not atropine, in

Substance addiction is a chronic psychiatric disorder characterized by compulsive substance use, impaired behavioral control, and high relapse rates. Although mesolimbic dopaminergic reward system dysfunction is a key neurobiological substrate of substance addiction, it cannot fully explain persistent drug craving, addiction-related memory, or compulsive drug-seeking and drug-taking behaviors after abstinence. Repeated exposure to addictive substances can induce oxidative stress, activation of pattern-recognition receptors, increased blood-brain barrier permeability, and gut-brain axis dysregu

GPR143, originally identified as the gene product of ocular albinism 1 (OA1), a G protein-coupled receptor (GPCR), can function as a receptor for l-3,4-dihydroxyphenylalanine (DOPA), a precursor of dopamine (DA). To examine the physiological and pathophysiological roles of GPR143, we analyzed the behavior of Gpr143 gene-deficient (Gpr143-/y) mice. We performed comprehensive behavioral analyses including the prepulse inhibition test, sucrose preference test, light-dark exploration test, etc. and microarray analysis. Gpr143-/y mice displayed a mixed psychosis-like phenotype: impaired prepulse in

The mesolimbic dopamine (DA) system has been implicated in pair bond formation. However, the involvements of DA release, real-time activities, and electrophysiological activities of D1/D2 medium spiny neurons (MSNs) in the nucleus accumbens (NAc) shell in pair bonding remain unclear. This work verified that male mandarin voles after pair bonding released higher levels of DA in the NAc shell and displayed higher levels of D1 MSNs activity and lower levels of D2 MSNs activity upon sniffing their partners compared to upon sniffing an unknown female. Moreover, pair bonding induced differential alt

Alcohol use disorder is a severe mental health condition causing medical consequences and preterm death. Alcohol activates the mesolimbic dopamine system leading to an increase of extracellular dopamine (DA) in the nucleus accumbens, an event that is associated with the reinforcing effects of alcohol. Cholinergic interneurons (CIN) are important modulators of accumbal DA signalling, and depletion of accumbal CIN attenuates the alcohol-induced increase in extracellular DA. The aim of this study was to explore the functional role of accumbal CIN in alcohol-related behaviour. To this end, ablatio

In this review, we aim to draw a connection between drug addiction and overconsumption of highly palatable food (OHPF) by discussing common behaviors and neurochemical pathways shared by these two states. OHPF can stimulate reward pathways in the brain that parallel those triggered by drug use, increasing the risk of dependency. Behavioral similarities between food and drug addiction can be addressed by tracking their stages: loss of control when eating (bingeing), withdrawal, craving, sensitization, and cross-sensitization. The brain adapts to addiction by way of the mesolimbic dopamine syste

Food deprivation (FD) or restriction augments the locomotor activating and reinforcing effects of drugs of abuse. It has been proposed that these effects might be mediated by FD-induced increase in plasma levels of ghrelin, a 28-amino acid orexigenic peptide demonstrated to functionally interact with the mesolimbic dopaminergic system. However, a role for ghrelin has been demonstrated only with psychostimulant drugs and alcohol associated behaviors. We therefore examined the role of ghrelin in ongoing heroin self-administration and FD-induced reinstatement of extinguished heroin seeking. As ex

Several recent reports have demonstrated that opiate action in both the ventral tegmental area (VTA) and the nucleus accumbens (N.Acc.) produces an increase in locomotor activity. In the present experiments, the effect of repeated bilateral injections into these sites of either morphine or the mu opioid receptor agonist Tyr-D-Ala-Gly-NMe-Phe-Gly-ol (DAGO) was investigated. As previously reported with morphine and other opioids, repeated injections of either morphine or DAGO into the VTA produced a progressive enhancement or sensitization of their locomotor activating effects. On the other hand

Mesocorticolimbic circuits are central to theories of methamphetamine and other addictions. White-matter fibres are a necessary component of those circuits. However, the extent to which mesocorticolimbic white-matter tracts are altered in methamphetamine addiction is unknown. The current study addressed this gap as well as sought to determine relationships with neurofunctional and behavioural deficits concerning nondrug rewards. Multimodal MRI was acquired from 92 people with problematic methamphetamine-use experience (MA+) and 98 methamphetamine-naïve controls (MA-). Diffusion tractography ma

BACKGROUND: Symptoms of psychosis are often observed in patients with post-traumatic stress disorder (PTSD) and are driven by aberrant regulation of the mesolimbic dopamine system. We have previously shown that targeting upstream brain regions that regulate dopamine neuron activity, the ventral hippocampus (vHipp), and paraventricular nucleus of the thalamus (PVT) maybe a novel approach to restore dopamine system function. The vHipp and PVT work in concert to regulate ventral tegmental area (VTA) dopamine neuron activity through a multisynaptic circuit that begins with inputs to the nucleus ac

The precise cortical and subcortical mechanisms of Tourette syndrome (TS) are still not fully understood. In the present retrospective analysis, adolescent and adult medication-naïve patients showed increased DA transporter (DAT) binding in nucleus caudate (CAUD), putamen (PUT) and/or whole neostriatum (NSTR). D2 receptor (R) binding and DA release were not different from controls throughout the nigrostriatal and mesolimbocortical system. When patients were medication-free (either medication-naïve or under withdrawal), DAT was still increased in PUT, but not different from controls in CAUD, NS

REM sleep behaviour disorder (RBD) is common in narcolepsy type 1 (NT1). Abnormalities in the reward system have been observed in NT1, possibly related to impaired orexin projections towards the mesolimbic reward system, but also in RBD when associated with Parkinson's disease. Our study aimed to explore the psychobehavioural profile of NT1 patients with and without RBD compared with healthy controls (HC). Forty patients with NT1 were compared with 20 sex- and age-matched HC. All patients with NT1 underwent a video-polysomnography including a measure of REM sleep without atonia (RSWA). The fol

Chronic alcohol intoxication is known to produce neuronal degeneration in the central and peripheral nervous system of experimental animals and of humans. It is suggested that various components of the cytoskeleton undergo profound changes following chronic alcohol use and misuse. Here we studied the expression of the neuronal cytoskeletal microtubule-associated protein 2 (MAP2) following long-term alcohol consumption and subsequent alcohol withdrawal. Alcohol-preferring AA (Alko Alkohol) rats with a high voluntary alcohol consumption for a period of 16 months were compared with age-matched co

Obesity is one of the most serious public health challenges worldwide and has reached the scale of a global epidemic. Its etiology is multifactorial and includes genetic, environmental, hormonal, and neurobiological factors. In recent years, increasing attention has been paid to the role of the gut microbiota in the regulation of energy metabolism, inflammatory processes, and the functioning of the gut-brain axis. An increasing body of evidence suggests that the gut microbiota may influence the dopaminergic system and eating behaviors through bacterial metabolites, immune pathways, and the vag

Nicotine addiction constitutes a significant global health burden, primarily driven by the substance's capacity to dysregulate the brain's reward and stress systems. This chronic relapsing disorder is characterized by robust dependence and high rates of relapse, underscoring the limitations of current therapeutic strategies. Neuropeptide Y (NPY), a 36-amino acid neuromodulator abundantly expressed in the central nervous system, has emerged as a critical regulator of emotional behavior, stress responses, and reward pathways. Its role in the pathophysiology of nicotine addiction is of increasing
