Novel intragraft regulatory lymphoid structures in kidney allograft tolerance.

Intragraft events thought to be relevant to the development of tolerance are here subjected to a comprehensive mechanistic study during long-term spontaneous tolerance that occurs in C57BL/6 mice that receive life sustaining DBA/2 kidneys. These allografts rapidly develop periarterial Treg-rich organized lymphoid structures (TOLS) that form in response to class II but not to class I MHC disparity and form independently of lymphotoxin α and lymphotoxin β receptor pathways. TOLS form in situ in the absence of lymph nodes, spleen, and thymus. Distinctive transcript patterns are maintained over ti

Targeting age-specific changes in CD4+ T cell metabolism ameliorates alloimmune responses and prolongs graft survival.

Age impacts alloimmunity. Effects of aging on T-cell metabolism and the potential to interfere with immunosuppressants have not been explored yet. Here, we dissected metabolic pathways of CD4+ and CD8+ T cells in aging and offer novel immunosuppressive targets. Upon activation, CD4+ T cells from old mice failed to exhibit adequate metabolic reprogramming resulting into compromised metabolic pathways, including oxidative phosphorylation (OXPHOS) and glycolysis. Comparable results were also observed in elderly human patients. Although glutaminolysis remained the dominant and age-independent sour

Morphometric analysis of the placenta in the New World mouse Necromys lasiurus (Rodentia, Cricetidae): a comparison of placental development in cricetids and murids.

BACKGROUND: Stereology is an established method to extrapolate three-dimensional quantities from two-dimensional images. It was applied to placentation in the mouse, but not yet for other rodents. Herein, we provide the first study on quantitative placental development in a sigmodontine rodent species with relatively similar gestational time. Placental structure was also compared to the mouse, in order to evaluate similarities and differences in developmental patterns at the end of gestation. METHODS: Fetal and placental tissues of Necromys lasiurus were collected and weighed at 3 different s

Protective role of complement C5a in an experimental model of Staphylococcus aureus bacteremia.

The complement system is a key component of the innate immune response. Here, we have examined the role of complement anaphylatoxin C5a in an experimental model of Staphylococcus aureus bacteremia. Our data provide compelling evidence for a protective role of C5a during staphylococcal bloodstream infection.

A mouse model of the recrudescence of latent tuberculosis in the elderly.

The present study was designed to attempt to construct an animal model of the recrudescence of pulmonary tuberculosis by following the course of infection in mice exposed at 3 months of age to a low dose aerosol of Mycobacterium tuberculosis Erdman. The results show that such mice died of overwhelming lung disease at a median age of 24 months; in contrast, uninfected mice lived 4 to 6 months longer. Examination of infected mice over the period of 20 to 24 months of age provided evidence to suggest that recrudescence over this period of time was a general phenomenon within the infected mice, an

Repeated generalized seizures can produce calcified cardiac lesions in DBA/1 mice.

Studies suggest that cardiorespiratory dysfunction likely contributes to sudden unexpected death in epilepsy (SUDEP). Seizures result in autonomic and respiratory dysfunction, leading to sympathetic hyperactivity and respiratory distress, including apnea. While the heart is vulnerable to catecholamine surges and hypoxia, it remains unknown if repetitive generalized seizures lead to cardiac damage. DBA/1 mice exhibit seizure-induced respiratory arrest (S-IRA) following generalized audiogenic seizures (AGS), which can be resuscitated using a rodent ventilator. In the current study, we induced di

A study of the development of thoracic vertebrae in the mouse assisted by autoradiography.

Evidence is accumulating that the current concept of resegmentation during vertebral formation is no longer valid. In order to shed some light on this controversial issue, in the present investigation the development of the vertebral column was studied in a graded series of mouse embryos by conventionally stained serial sections and methyl thymidine 3H autoradiography. It was found that the vertebral bodies do not originate from the upper and lower halves of sclerotomes but from a continuous central tissue mass surrounding the notochord, the perichordal tube. The caudal sclerotome half gives r

The ameliorative effects of choline on ethanol-induced cell death in the neural tube of susceptible BXD strains of mice.

INTRODUCTION: Fetal alcohol spectrum disorders (FASD) are the leading preventable cause of intellectual disability, providing the impetus for evaluating various potential treatments to ameliorate ethanol's teratogenic effects, particularly in the nervous system. One treatment is the dietary supplement choline which has been shown to mitigate at least some of ethanol's teratogenic effects. The present study was designed to investigate the effects of genetics on choline's efficacy in ameliorating cell death in the developing neural tube. Previously, we examined BXD recombinant inbred mice, and t

A precision surgical approach for complete or partial callosotomy in the mouse.

The corpus callosum (CC) of mice was completely transsected with a thin tungsten knife using a three-cut approach through the dorsal cerebrum just lateral to midline. This method results in almost total transsection of the CC throughout its entire rostrocaudal extent. Advantages of this approach include minimal bleeding and extracallosal damage as well as the possibility of selective transsection of only anterior, middle, or posterior parts of the CC. The technique can be readily adapted to any other rodent species.

[Characteristics of the audiogenic convulsive crisis in mice made sensitive by magnesium deficiency].

Mice of the OF1, C57BL/6, AKR, C3H/He, DBA/2, BALB/C, B6D2F1 and CBA strains are susceptible to audiogenic seizures after 40 or 20 days of acute magnesium deficiency. The duration of the various phases of the audiogenic seizure response (at 100 dBA) (wild running latency period, convulsions latency period and the clonic and tonic convulsions) among these mice was measured. Using the Kruskall-Wallis test, no difference was recorded using these measurements and those obtained for genetically audio-susceptible animals. This shows that acute magnesium deficiency-induced audiogenic seizures develop

Genetic dissection of age-related changes of immune function in mice.

Understanding of the genetic basis of normal and abnormal development of the immune response is an enormous undertaking. The immune response, at the most minimal level, involves interactions of antigen presenting cells (APCs), T and B cells. Each of these cells produce cell surface and soluble factors (cytokines) that affect both autocrine and paracrine functions. A second level of complexity needs to consider the development of the macrophage/monocyte lineage as well as the production of the common lymphoid precursor which undergoes distinct maturation steps in the thymus and periphery to for

Stenotrophomonas maltophilia strains replicate and persist in the murine lung, but to significantly different degrees.

The environmental bacterium Stenotrophomonas maltophilia is increasingly described as a multidrug-resistant pathogen of humans, being associated with pneumonia, among other diseases. But the degree to which S. maltophilia is capable of replicating in a mammalian host has been an issue of controversy. Using a model of intranasal inoculation into adult A/J mice, we now document that S. maltophilia strain K279a, the clinical isolate of S. maltophilia whose complete genome sequence was recently determined, is in fact capable of replicating in lungs, displaying as much as a 10-fold increase in c.f.

Evolution and host-dependent lesions in autoimmune thyroiditis: experimental study in inbred mice.

The evolution of thyroid pathology in experimental autoimmune thyroiditis was studied in several inbred strains of mice. The disease was induced by immunization with mouse thyroglobulin emulsified in Freund's complete adjuvant. In addition to the common lymphoid/mononuclear cell infiltration seen in other animal species with experimental autoimmune thyroiditis, several distinct thyroid lesions occurred only in certain strains of mice, suggesting a strain-dependency of these lesions. These lesions appeared often at the peak of the common lesion of autoimmune thyroiditis. These findings suggest

The use of host strain variation to assess the significance of mucosal mast cells in the spontaneous cure response of mice to the nematode Trichuris muris.

A proportion of DBA/2 mice do not reject the large intestinal nematode parasite Trichuris muris. In this strain and the early rejecting NIH and the late rejecting CBA/Ca strains the kinetics of the mast cell accumulation in a primary infection were similar with peak mast cell numbers being recorded on day 20 post-infection. Comparisons between rejector and non-rejector DBA/2 mice showed no differences in the mast cell accumulation. There was no rise in mast cell numbers in response to a secondary infection, in either the NIH or CBA/Ca strains, for at least 3 days after the infection had been e

Protection from Severe Influenza Virus Infections in Mice Carrying the Mx1 Influenza Virus Resistance Gene Strongly Depends on Genetic Background.

UNLABELLED: Influenza virus infections represent a serious threat to human health. Both extrinsic and intrinsic factors determine the severity of influenza. The MX dynamin-like GTPase 1 (Mx1) gene has been shown to confer strong resistance to influenza A virus infections in mice. Most laboratory mouse strains, including C57BL/6J, carry nonsense or deletion mutations in Mx1 and thus a nonfunctional allele, whereas wild-derived mouse strains carry a wild-type Mx1 allele. Congenic C57BL/6J (B6-Mx1(r/r)) mice expressing a wild-type allele from the A2G mouse strain are highly resistant to influenza

Mechanism of NK cell activation: relationship between Qa5+ NK cells and lymphocytes.

Interferon (IF) induces within 20 hr of incubation Qa5+Thy-1- NK cells but Qa5-Thy-1+ cytotoxic cells in cultures of murine splenocytes. The mechanism of Qa5+ NK cell activation has been studied by using tumor necrosis serum (TNS), concanavalin A (Con A), and lipolysaccharide (LPS) as additional NK cell-inducing reagents. It was found that IF and TNS activate precursor NK cells directly without the involvement of accessory cells. The T cell mitogen Con A and the B cell mitogen LPS, on the other hand, induced Qa5+ NK cells only in the presence of lymphocytes. Con A-stimulated T cells and LPS-st

Characterization of auxotrophic mutants of Mycobacterium tuberculosis and their potential as vaccine candidates.

Auxotrophic mutants of Mycobacterium tuberculosis have been proposed as new vaccine candidates. We have analyzed the virulence and vaccine potential of M. tuberculosis strains containing defined mutations in genes involved in methionine (metB), proline (proC), or tryptophan (trpD) amino acid biosynthesis. The metB mutant was a prototrophic strain, whereas the proC and trpD mutants were auxotrophic for proline and tryptophan, respectively. Following infection of murine bone marrow-derived macrophages, H37Rv and the metB mutant strain survived intracellularly for over 10 days, whereas over 90% o

RNA-seq analysis highlights DNA replication and DNA repair associated with early-onset hearing loss in the cochlea of DBA/2J mice.

AIMS: Age-related hearing loss (ARHL) is a significant health concern, and DBA/2J (D2) and C57BL/6 (B6) mouse strains serve as valuable models for its study. B6 mice, harboring a homozygous ahl allele in Cdh23, manifest high-frequency hearing loss at 3 months. In contrast, D2 mice, carrying the R109H variant of the Fascin-2 gene (Fscn2), experience early-onset hearing loss by 3 weeks. Yet, the underlying molecular mechanisms driving early-onset hearing loss in D2 mice remain elusive. This study aimed to identify novel genes and regulatory pathways as therapeutic targets for early deafness. MA

TNFα Blockade Inhibits Both Initial and Continued Control of Pulmonary Coccidioides.

Tumor necrosis factor alpha (TNFα) is a pluripotent cytokine that is important in many infections, though its role in Coccidioides infection remains poorly understood. The need to understand TNFα in Coccidioides infection has increased recently with the widespread use of TNFα inhibitors for a wide variety of autoimmune conditions. Here, we couple the newly developed Coccidioides infection model using strain Cp1038 and C57BL/6 × DBA/2J F1 (B6D2F1) mice. B6D2F1 mice develop long-lasting control of Cp1038. Treatment of B6D2F1 mice with anti-TNFα antibodies permits significant fungal proliferation
