Subcellular distribution, molecular dynamics and catabolism of plasmalogens in myocardium.

Recent studies have implicated accelerated sarcolemmal phospholipid catabolism as a mediator of the lethal sequelae of atherosclerotic heart disease. We have demonstrated that plasmalogens are the predominant phospholipid constituents of canine myocardium and that plasmalogens are hydrolyzed by a novel calcium independent plasmalogen selective phospholipase A2. Since the activities of phospholipases are modulated by the molecular dynamics and interfacial characteristics of their phospholipid substrates, we compared the molecular dynamics of plasmenylcholine and phosphatidylcholine vesicles by …

[Effect of chronic alcoholism on the state of the microcirculatory bed of the myocardium].

Ultrastructure of myocardial capillaries of rats was studied in cases of chronic alcohol intoxication, experimental alcoholic cardiomyopathy (ACM) and its correction with antioxidants (vitamin E, dibunol). Alterations in the microcirculatory bed were similar in all groups of animals irrespective of disturbances in cardiomyocytes. Cardiomyocyte ultrastructure was improved after treatment with antioxidants, but capillary bed was the same as in untreated animals. High dibunol doses caused the onset of perivascular sclerosis. Disturbances in the microcirculatory bed are, probably, the first step i

[Mechanical noise of the myocardium as an index of sarcoplasmic reticulum activity].

The effects of caffeine (0.5-12 mM) and cooling on the mechanical noise (MN) and relaxation rate of contractile response (RR) were studied in experiments on rat papillary muscles. Mean frequency of MN and RR were found to have similar temperature dependences from 12 to 32 degrees C. MN amplitude was more sensitive to the action of sarcoplasmic reticulum activity blocker, caffeine, than RR one. MN as an index of sarcoplasmic reticulum activity made it possible to demonstrate a competition between caffeine and local anaesthetics described earlier for skeletal muscles.

Responses of the canine myocardium to stimulation of thoracic cardiac nerves.

The thoracic cardiac nerves were stimulated in each of 21 dogs anesthetized with alpha chloralose. Recordings were made of heart rate, blood pressure, and contractile force from all four cardiac chambers. Walton-Brodie strain-gauge arches were sutured to both atria, and to three locations of each ventricle, representing both anterior and posterior surfaces. The functional autonomic components of each nerve were summarized and classified into four basic types. Types I and II were both located medial to the thoracic vagi. These were shown to contain both sympathetic and parasympathetic component

Protection of ischemic myocardium by nitroglycerin: experimental and clinical results.

Nitroglycerin (NTG) traditionally has bben avoided in the treatment of pain caused by acute myocardial infarction because of the belief that NTG-induced decrease in arterial pressure and concomitant reflex increase in heart rate might extend the ischemic process. However, recent experimental and clinical investigations cast doubt on this concept. For example, when the left anterior descending coronary artery is acutely occluded in normal dogs or in dogs when chronic coronary occlusions and extensive collaterals, NTG reduces ST-segment evevation (and presumably myocardial ischemia). This saluta

Pathology of the myocardium and the conduction system in sudden coronary death.

This review summarizes previously unpublished and recently published autopsy findings of prehospital sudden coronary death (SCD) in four different counties (Olmsted, Minnesota; Albany, New York; Dade, Florida; and San Francisco Bay Area, California), totaling 868 patients. The prevalence of cardiomegaly and significant coronary atherosclerosis, and the relative infrequency of acute coronary thrombosis in prehospital SCD, well documented in the past, have been reaffirmed in current studies. Differences in the patient populations and laboratory techniques notwithstanding, these independent autop

Electrolyte and morphologic alterations of myocardium in adriamycin-treated rabbits.

Rabbits receiving adriamycin (ADR) on a chronic schedule developed significant histopathologic, ultrastructural and tissue electrolyte alterations of the ventricular myocardium. Rabbits that developed clinicopathologic evidence of cardiomyopathy with ADR had histologic lesions of the myocardium, including perivascular fibrosis, interstitial fibrosis and edema and myocytolysis. Ultrastructurally, large vacuoles resembling distended sarcoplasmic reticulum displaced the contractile elements and mitochondria, which were diminished in number within affected myocytes. Frequently, mitochondria appear…

Triiodothyronine protects infarcted myocardium by reducing apoptosis and preserving mitochondria.

Myocardial infarction (MI) is a leading cause of heart failure, with thyroid hormone (TH) signaling playing a key role in heart function and postinfarct recovery. Despite evidence of TH administration's safety in cardiac patients, inconsistent therapeutic outcomes and limited understanding of its mechanisms hinder clinical translation. This study aims to investigate the long-term effect of acute triiodothyronine (T3) administration following MI and to elucidate the mechanisms of its cardioprotective actions. To this end, two doses (40 μg/kg) of T3 were administered immediately after injury and
