Plasmacytomas are neoplasms of plasma cells that can involve the bone marrow, the bone itself, or soft tissue without bone marrow involvement. Extramedullary plasmacytomas are most commonly found in the upper respiratory tract in a total of 80% of all cases. Extramedullary plasmacytomas have also been documented in locations such as the central nervous system. This form of plasmacytoma may be seen as a solitary entity or in patients with multiple myeloma. We present a case of a 66-year-old female with a history of multiple myeloma on maintenance therapy with lenalidomide who has been experienc

Most thyroid lesions are of epithelial origin characterized by a papillary and/or follicular pattern. However, lesions involving plasma cells or plasmacytoid cytomorphology that mimic plasma cells can rarely be encountered in the thyroid gland. Thyroid lesions involving true plasma cells include benign entities such as chronic lymphocytic thyroiditis and neoplasms including plasmacytoma. Thyroid lesions that present with plasmacytoid features include medullary thyroid carcinoma. Rendering the correct diagnosis may be difficult due to the rarity of these lesions, overlapping cytomorphologic fea

Epithelioid vascular neoplasms of the bone are classified by the World Health Organization (WHO) into only two tiers: low-grade epithelioid hemangioma (EH) and a more malignant category including both epithelioid hemangioendothelioma and epithelioid angiosarcoma. The World Health Organization defines bone EH as a locally aggressive neoplasm with no connotation of benign or intermediate malignancy. We reviewed three cases of EH in our lab archives with the perspective of appraising their histomorphological approach toward diagnosis. Patients were in the age range of 15-25 years. The site of the

Neoplasms and pseudotumors of the temporomandibular joint (TMJ) are very uncommon. Early recognition of such will prevent therapeutic delay and may have a dramatic impact on patient morbidity and mortality. Included in rare TMJ lesions are the following: 1) synovial chondromatosis, 2) osteochondroma, 3) osteoma, 4) osteoblastoma, 5) pigmented villonodular synovitis, 6) ganglion, 7) synovial cyst, 8) simple bone cyst, 9) aneurysmal bone cyst, 10) epidermal inclusion cyst, 11) hemangioma, 12) nonossifying fibroma, 13) Langerhans cell histiocytosis, 14) plasma cell myeloma, 15) sarcoma.

BACKGROUND: Plasma cell neoplasm and/or plasma cell myeloma (PCM) is a mature B-cell lymphoproliferative neoplasm of plasma cells that secrete a single homogeneous immunoglobulin called paraprotein or M-protein. Plasma cells accumulate in the bone marrow (BM) leading to bone destruction and BM failure. Diagnosis of PCM is based on clinical, radiologic, and pathological characteristics. The percent of plasma cells by manual differential (bone marrow morphology), the white blood cell (WBC) count, cytogenetics, fluorescence in situ hybridization (FISH), microarray, and next-generation sequencing

UNLABELLED: Whether multiple myeloma (MM) and light-chain amyloidosis (AL) stem from terminally differentiated plasma cells (PC) or earlier clonotypic B cells remains under debate. Addressing this issue would improve accurate diagnosis and treatment monitoring. We performed single-cell and exome sequencing on highly purified MM and AL samples to define in which B-cell development stage the driver genetic alterations are present. Clonotypic B-cell receptors (BCR) were detected in ≤0.4% of B-cell precursors and mature B cells from patients with MM and AL. Paired single-cell transcriptomes confir

Clonotypic B cells have been proposed as a potential disease reservoir in multiple myeloma and light-chain amyloidosis, but Gonzalez and colleagues demonstrate that they do not necessarily share the malignant potential of disease clones. Their study shows that the key genetic hallmarks of disease are largely restricted to the plasma-cell compartment and raises caution that assays detecting minimal residual disease may yield false-positive results by detecting rare clonotypic B cells. See related article by Gonzalez et al., p. 558.

Primary solid neoplasms of the extratesticular tissues are rare. The reported prevalence rate of paratesticular neoplasms is between 3% and 16% of all patients referred for scrotal ultrasonography. A plasmacytoma is a discrete, solitary mass of malignant monoclonal plasma cells that can arise in any part of the body. In this report, we present a case of a paratesticular solid mass detected in an 80-year-old patient that proved to be primary extraosseous plasmacytoma on surgery, and discuss its sonographic features.

With the incidence and prevalence of lymphoid neoplasms increasing, a comparison of survival patterns by subtype may provide critical clues for improving the disease burden. We conducted a comprehensive survival analysis for 254,702 lymphoid neoplasm cases diagnosed during 1973-2003 at 17 Surveillance, Epidemiology and End Results (SEER) registries according to the World Health Organization (WHO) classification introduced in 2001. The best survival was observed for Hodgkin lymphoma among young patients, and the worst survival was observed among cases with plasma cell neoplasms, particularly pl

BACKGROUND: Greater adiposity and height have been associated with increased risk of haematological malignancies. Associations for disease subtypes are uncertain.
METHODS: A cohort of 1.3 million middle-aged U.K. women was recruited in 1996-2001 and followed for 10 years on average. Potential risk factors were assessed by questionnaire. Death, emigration, and incident cancer were ascertained by linkage to national registers. Adjusted relative risks were estimated by Cox regression.
RESULTS: During follow-up, 9162 participants were diagnosed with lymphatic or haematopoietic cancers. Each 10 k

With the advent of precision genomics, hereditary predisposition to hematopoietic neoplasms- collectively known as hereditary predisposition syndromes (HPS)-are being increasingly recognized in clinical practice. Familial clustering was first observed in patients with leukemia, which led to the identification of several germline variants, such as RUNX1, CEBPA, GATA2, ANKRD26, DDX41, and ETV6, among others, now established as HPS, with tendency to develop myeloid neoplasms. However, evidence for hereditary predisposition is also apparent in lymphoid and plasma--cell neoplasms, with recent disco

INTRODUCTION: Amyloidosis is characterized by extracellular protein fibril deposition. Light-chain (AL) amyloidosis may be associated with plasma cell dyscrasias. Isolated mediastinal involvement is exceptionally rare and may mimic malignant tumors.
CASE PRESENTATION: A 69-year-old woman presented with a progressively enlarging anterior chest wall mass caused by an invasive mediastinal lesion. Biopsy revealed amyloid deposition. She underwent bilateral thoracotomy, sternectomy, en-bloc chest wall resection, and reconstruction. Final pathology confirmed AL amyloidosis associated with a kappa-r

Multiple myeloma is a tumour of antibody-secreting plasma cells characterized by clonal expansion and accumulation of monotypic plasma cells in the bone marrow. It is an incurable malignant neoplasm accounting for 10% all hematological malignancies. Globally, the annual percentage of new cancer cases and deaths attributed to multiple myeloma is estimated at about 0.8% and 1%, respectively. Furthermore, its global incidence ranges from 0.5 - 12/100,000 population. It causes hypercalcemia, renal insufficiency, anemia, thrombocytopenia, leucopenia, bone lesions, bone fractures, spinal stenosis, a

Neoplastic proliferation of plasma cells is called plasma cell dyscrasias, and these neoplasms can present as a solitary neoplasm or multiple myeloma. Extramedullary plasmacytoma, in particular pancreatic plasmacytoma, is a rare manifestation of multiple myeloma. Although computerized tomography is useful for the diagnosis of extramedullary plasmacytoma, there are no specific radiologic markers that distinguish it from adenocarcinoma. Histological confirmation by biopsy is necessary for accurate diagnosis and management of the tumor. Endosonography is the most sensitive method for the diagnosi

Plasmacytoma is a rare malignant tumor originating from plasma cells either located in the bone marrow (known as solitary bone plasmacytoma or SBP) or from outside the bone, usually arising from the mucosa (known as extramedullary plasmacytoma). Extramedullary plasmacytoma (EMP) in the head and neck region is extremely uncommon, and therefore this case involving the paranasal sinuses is reported for its rarity and to demonstrate the importance of imaging in the diagnosis and follow-up of the disease. Both pathologies can progress to multiple myeloma, a much more common manifestation of the con

Solitary plasmacytomas are localized single tumors of monoclonal plasma cells that occur in two variants: solitary plasmacytoma of bone and extraosseous plasmacytoma. Solitary plasmacytoma of bone accounts for only 1%-2% of plasma cell lesions, and extraosseous plasmacytoma is also approximately 1%. These are both very uncommon at the skull base. We report two cases of plasmacytic neoplasms at the skull base where the differential diagnosis included pituitary adenoma when the tumor was sellar/suprasellar, or other bone-related tumors such as chordoma/chondrosarcoma when clivus/sphenoid bones w

An Extramedullary Plasmacytoma (EMP) is characterized by a neoplastic proliferation of clonal plasma cells outside the medullary cavity. EMPs are a rare occurrence compared to other malignant plasma cell disorders and account for approximately 3-5% of plasma-cell neoplasms. Although most cases of EMP are not immediately life threatening at diagnosis, EMPs can progress to Multiple Myeloma (MM) and thus, warrant monitoring. Currently, there are no standard guidelines for when and how to monitor patients who are diagnosed with or treated for a solitary plasmacytoma. We present a case of solitary

Multiple myeloma, solitary plasmacytoma, and extramedullary plasmacytoma constitute a continuous disease spectrum of plasma cell neoplasms. In the larynx, although extramedullary plasmacytoma in the supraglottic region has been sometimes reported, plasma cell neoplasm with involvement of the thyroid cartilage is extremely rare. We report a case of multiple myeloma involving the thyroid cartilage. A 72-year-old male patient presented with a pathological fracture of the cervical vertebrae. CT scan revealed low-density areas within the thyroid cartilage, but the laryngeal mucosa and submucosal so
