Neuritic sprouting with aberrant expression of the nitric oxide synthase III gene in neurodegenerative diseases.

Neuronal loss, synaptic disconnection and neuritic sprouting correlate with dementia in Alzheimer's disease (AD). Nitric oxide (NO) is an important synaptic plasticity molecule generated by nitric oxide synthase (NOS) oxidation of a guanidino nitrogen of L-arginine. Experimentally, the NOS III gene is modulated with neuritic sprouting. In a previous study, NOS III expression was found to be abnormal in cortical neurons, white matter glial cells, and dystrophic neurites in AD and Down syndrome brains. The present study demonstrates the same abnormalities in neuronal and glial NOS III expression…

Direct neurite-mast cell communication in vitro occurs via the neuropeptide substance P.

Communication between nerves and mast cells is a prototypic demonstration of neuroimmune interaction. However, whether mast cell activation … lization. Neither bradykinin nor scorpion venom had any direct effect on the RBLs in the absence of neurites. Addition of a neutralizing substance P Ab or a neurokinin (NK)-1 receptor antagonist, but not an NK-2 receptor antagonist, dose-dependently prevented the RBL activation that resulted as a consequence of neural activation by either bradykinin or sco …

Comparison of neurite outgrowth induced by intact and injured sciatic nerves: a confocal and functional analysis.

Mechanisms regulating axon growth in the peripheral nervous system have been studied by means of an in vitro bioassay, the tissue section culture, in which regenerating neurons are grown on substrata made up of tissue sections. Sections from intact and degenerated sciatic nerves proved to be different in their ability to support neurite outgrowth of embryonic chick sensory neurons from both qualitative and quantitative points of view. On denervated nerve sections, the total length of neurites elaborated per neuron was almost twice that found on intact nerve sections. In addition, confocal micr…

Enhancement of neurite outgrowth by the soluble form of human L1 (neural cell adhesion molecule).

L1, a neural cell adhesion molecule, is involved in neurite outgrowth, migration and fasciculation. Although L1 is a membrane glycoprotein e … ed with polyethylenimine (PEI) alone, on substrate-bound L1 or in medium containing soluble L1. The neurites induced by L1, both the substrate-bound form and the soluble form, were 2-3 times longer than those cultured on PEI. The ability of the soluble L1 to induce neurite formation was slightly greater than that of the substrate L1. The present results de …

GTP and guanosine synergistically enhance NGF-induced neurite outgrowth from PC12 cells.

Six per cent of rat pheochromocytoma (PC12) cells extended neurites (processes greater than one cell diameter in length) in the presence of 300 microM extracellular GTP or 300 microM guanosine for 48 hr, compared to only 2.5% of cells in control cultures. In the presence of 40 ng/ml of 2.5S NGF, about 20-35% of PC12 cells had neurites after 48 hr, and the addition of 300 microM guanosine or GTP together with NGF synergistically increased the proportion of cells with neurites to 40-65%. GTP and guanosine also increased the average number of branches per neurite, from 0.6 in NGF-treated cultures…

N-acetyl-lactosamine in the rat olfactory system: expression and potential role in neurite growth.

Primary sensory olfactory neurons exhibit a mosaic topographical projection from the olfactory neuroepithelium in the nasal cavity to the olfactory bulb formation of the telencephalon. Axons from primary neurons that are widely scattered in the epithelium terminate in discrete regions of the olfactory bulb. It has been hypothesised that carbohydrates present on the surface of primary olfactory axons mediate selective fasciculation and the formation of the topographical pathway. We examined the expression of the disaccharide N-acetyl-lactosamine in both the developing and the adult rat olfactor

Myelin-associated glycoprotein inhibits neurite/axon growth and causes growth cone collapse.

We have previously shown that myelin-associated glycoprotein (MAG) inhibits neurite growth from a neuronal cell line. In this study we show that 60% of axonal growth cones of postnatal day 1 hippocampal neurons collapsed when they encountered polystyrene beads coated with recombinant MAG (rMAG). Such collapse was not observed with denatured rMAG. Neurite growth from rat embryonic hippocampal and neonatal cerebellar neurons was also inhibited about 80% on tissue culture substrates coated with rMAG. To investigate further the inhibitory activity of MAG in myelin, we purified myelin from MAG-defi

Rapidly progressing dementia with filopodia-like cytoskeletal neuritic anomalies, but without Alzheimer changes.

An unusual case of dementia is reported. The patient was a woman who died at the age of 69 years and 6 months after a two years history of organic dementia. Possibly the disease was familial. Examination of the brain at autopsy revealed no atrophy. In routine histology the brain seemed to be normal. However, when the sections were stained with highly sensitive techniques such as an antibody to phosphorylated Tau (PHF-1), widespread neuritic pathology was discovered. Probably both, axons and dendrites were involved. Only few perikarya were reactive with the antibody. In some of them, morphologi

Substrate-bound GP130/F11 will promote neurite outgrowth: evidence for a cell surface receptor.

GP130/F11 is a neuronal cell adhesion molecule belonging to the Ig superfamily. Preliminary evidence suggests that it may be important for the fasciculation of axons and cell-cell interactions during synaptogenesis. Previous work has shown that when F3 (the mouse homologue) is transfected into Chinese hamster ovary (CHO) cells it will act as a substrate for neuronal growth and adhesion. In this study we have shown that isolated GP130/F11 will also act as a substrate for the growth of chick dorsal root ganglia cell neurons. The concentration of GP130/F11 on the surface of sympathetic neurons, g

Characteristics of neurite outgrowth from rat spinal ganglia: effects of serum and segmental level.

The outgrowth of neurites from spinal ganglia in tissue culture may be affected by many variables including medium constituents and the age of ganglia. If E15 rats were used, the rate of outgrowth from ganglion explants in response to nerve growth factor was independent of the cranial-caudal position from which the ganglia were derived. This contrasts with the cranial-to-caudal gradient present during embryonic development. The effect of serum on this outgrowth was also studied and ten of 17 lots of human placental serum, two of three lots of pooled human placental serum, and one of two lots o…

Baseline Adjustment and Normalisation in Neurite Outgrowth Assays: A Simulation-Based Comparison of Analytical Methods.

BACKGROUND: When baseline neurite lengths differ between experimental groups, analytical methods may detect, miss or overstate treatment effects. We assessed how commonly used methods recover a baseline-adjusted treatment effect. NEW METHOD: We developed a simulation framework comparing endpoint comparison, change score, percentage change, percent-of-control normalisation and ANCOVA. A known 8 μm treatment effect was simulated under varying baseline imbalances and baseline-growth relationships, with additional analyses of sample size and baseline measurement error. RESULTS: In the worked exa

Neurite orientation dispersion and density imaging in stroke: current applications, limitations, and future directions.

Stroke is currently one of the leading causes of death and disability worldwide among neurological diseases. Although traditional imaging techniques can clearly delineate lesion extent and hemorrhagic patterns, they are limited in quantifying microscopic structural changes and repair processes in early-stage brain tissue damage. Neurite Orientation Dispersion and Density Imaging (NODDI), an advanced diffusion-weighted MRI technique, enables accurate, non-invasive quantification of post-stroke microstructural changes in brain tissue. NODDI objectively quantifies neurite damage and remodeling, p

Discovery of VU6077564: A Selective M1 Antagonist That Promotes Neurite Outgrowth in Adult Sensory Neurons.

Previously, we reported the potent and highly selective M1 antagonists VU0415248 and VU0452865; however, these compounds were limited by high predicted hepatic clearance in rat. This work reports the development of cyclobutylsulfonamide-based analogs of VU0415248 and VU0452865. From this exercise, we identified VU6077564, a peripherally restricted and selective M1 antagonist. Furthermore, we demonstrate the ability of VU6077564 to promote neurite outgrowth in adult rat dorsal root ganglia sensory neurons, highlighting the potential of selective M1 antagonists for the treatment of peripheral ne

Neurite Degradation Mediates the Effect of Amyloid Deposition on Global Cognition in Asymptomatic Older Adults.

This study investigates whether amyloid deposition is associated with neurite degradation in cognitively unimpaired (asymptomatic) older adults, with a focus on brain regions critical for memory and cognition. Using data from the ADNI3 cohort (n = 65; mean age 69.5±5.5), we examined the relationship between amyloid levels (from PET imaging), neurite density (derived from diffusion MRI-based Neurite Orientation Dispersion and Density Imaging, NODDI), and global cognition (MoCA scores). NODDI is a biophysical diffusion MRI model that quantifies microstructural features of brain tissue through me

Association between diffusion MRI-based measures of neurite microstructure and risk of Alzheimer's disease.

Early detection of Alzheimer's disease (AD) is crucial for intervention, but traditional MRI and cognitive assessments may miss pre-symptomatic changes. Advanced diffusion MRI (dMRI) methods, such as Neurite Orientation Dispersion and Density Imaging (NODDI), show promise in identifying early brain changes. We analyzed 65 cognitively unimpaired older adults (25 APOE-e4 carriers, 40 non-carriers) from the ADNI3 dataset. NODDI's neurite density index (NDI) and orientation dispersion index (ODI), volumetric MRI and cognition (MoCA) were analyzed in key brain regions like the hippocampus, fusiform

Soma and neurite density imaging detects brain microstructural impairments in amyotrophic lateral sclerosis.

OBJECTIVE: To investigate whole-brain microstructural changes in amyotrophic lateral sclerosis (ALS) using soma and neurite density imaging (SANDI), a novel multicompartment model of diffusion-weighted imaging that estimates apparent soma and neurite density. METHODS: This study consists of 41 healthy controls and 43 patients with ALS, whose diffusion-weighted data were acquired. The SANDI-derived (including signal fractions of soma (fsoma), neurite (fneurite), and extra-cellular space (fextra)) and diffusion tensor imaging (DTI)-derived metrics were obtained. Voxel-based analyses were perfor

Magnetically guided neurite outgrowth modulated by PIEZO1 in hiPSC-derived retinal ganglion cells.

Retinal ganglion cell (RGC) axon degeneration is one of the major causes of vision loss in glaucoma, where regeneration remains limited by t … te elongation and alignment. The mechanosensitive ion channel PIEZO1 was expressed in both soma and neurites. Specifically, PIEZO1 inhibition by Dooku1 enhanced neurite elongation, and activation by Yoda1 elevated somatic calcium transients without affecting neurite outgrowth. Furthermore, two-photon calcium imaging revealed that somatic calcium activity w …

Insulin and epidermal signals independently shape sexually dimorphic neurite branching in C. elegans.

Sexual dimorphism in neural wiring and behavior arises from both intrinsic genetic programs and environmental cues, yet how these factors interact to shape neuronal morphogenesis remains unclear. Here, we investigate sexually dimorphic collateral branching in PVP cholinergic interneurons of Caenorhabditis elegans. In hermaphrodites, PVP branches form near the vulva and exhibit dynamic morphologies enriched with synaptic proteins for dense core vesicles but not synaptic vesicles, suggesting a role in selective neuropeptide transmission. We find that sex identity is necessary but not sufficient
