On the basis of a search of the literature and extensive inquiries to clinicians, this paper catalogues those cases that contain any information concerning possible neuropathologic changes in cases of childhood autism or psychosis. In all, 33 cases were identified. Twenty-nine cases are from the literature; six of these are sibling pairs. Four cases were folnd by inquiry and have not been reported heretofore. Because of limitations in both the clinical and neuropathologic data in most of the case reports, the paper serves primarily as a source of reference to the 33 cases identified. The infor

In attempting to control the pain of disseminated carcinoma, a variety of agents has been injected intrathecally. This report deals with the neuropathologic changes in spinal roots and spinal cord of four patients into whose lumbar sacs 100 to 285 ml of distilled water was infused. Survival following the procedure ranged from 9 to 50 days. In most severely affected case (285 ml instilled 50 days prior to death) the abnormalities were: (1) severe breakdown of axons and myelin sheaths of the lower cauda equina and to a lesser extent of other roots at levels as high as the eighth cervical segment

Chemotherapy is frequently associated with long-term cognitive impairments in cancer survivors that negatively impact their quality of life. Effective mitigation strategies for cancer therapy-related cognitive impairments (CRCI) are still underdeveloped. Our clinical studies on breast cancer patients treated with doxorubicin (Adriamycin®, ADR) and cyclophosphamide (CYP) found significant CRCI associated with neurodegenerative and neuroinflammatory signatures. Current preclinical and clinical studies highlight the potential of cannabidiol (CBD) for alleviating cognitive deficits in neurodegener

INTRODUCTION: Primary Progressive Aphasia (PPA) clinical syndromes do not align consistently with underlying pathology. This study aimed to identify language markers for specific neuropathologies using both standard clinical tests and narrative speech analysis.
METHODS: We analyzed data from 82 autopsy-confirmed PPA cases, including Alzheimer's disease (AD), transactive DNA-binding protein 43 (TDP-43) type C (TDP-C), Pick's disease, and 4R-tauopathies (progressive supranuclear palsy/ cortico-basal degeneration (PSP/CBD). Linear mixed-effects regression was used to analyze performance on stand

Naegleria fowleri, the causative agent of primary amoebic meningoencephalitis, produces an acute infection of the central nervous system. Although human cases are rare, the rapid progression and more than 95% mortality rate have made the disease an important subject of neuropathological research. Here, we discuss an integrated comparative review of the pathology of N. fowleri infection in humans and animals, highlighting conserved routes of neuroinvasion, shared patterns of necrotising and haemorrhagic meningoencephalitis, and cross-species similarities in inflammatory and vascular injury. The

Studies of centenarians and supercentenarians provide insight into aging and substrates of cognitive impairment at the extreme limits of age. In this study, we examined pathologic findings in 15 centenarians (99-109 years) and 7 supercentenarians (111-115 years) who underwent autopsy at our institution during a 25-year period between 2000 and 2025 and compared them with a cohort of 24 older adults (70-90 years) who underwent autopsy during 2024-2025. Of the 22 centenarians and supercentenarians, 15 underwent full autopsies and 7 had brain only examinations. The average age at death of the cent

Alzheimer's disease (AD) is characterized by the hallmark pathologies of β-amyloid plaques and neurofibrillary tangles (NFTs), which vary across brain regions and between affected individuals. As a rapid research method to quantify hallmark pathologies and their relationships with the surrounding cytoarchitecture across the brain, we trialed tissue microarrays (TMAs) in combination with multiplex immunofluorescence and semi-automated image analysis. We assayed the hallmark pathologies across grey and white matter sites of 11 different brain regions and quantified astrocytes, microglia, and neu

Life-threatening thrombotic events and neurological symptoms are prevalent in COVID-19 and are persistent in patients with long COVID experiencing post-acute sequelae of SARS-CoV-2 infection1-4. Despite the clinical evidence1,5-7, the underlying mechanisms of coagulopathy in COVID-19 and its consequences in inflammation and neuropathology remain poorly understood and treatment options are insufficient. Fibrinogen, the central structural component of blood clots, is abundantly deposited in the lungs and brains of patients with COVID-19, correlates with disease severity and is a predictive bioma

BACKGROUND: This review provides an overview of obsessive-compulsive disorder (OCD) symptoms, including the four partially distinct subtypes of the disorder, current diagnostic criteria, and common comorbidities. Critically, it focuses on the etiology of OCD, including its underlying neuropathology, and examines cognitive dysfunction in OCD.
METHODS: This review study was conducted by library method.
RESULTS: We show how dysfunction in cortico-striato-thalamo-cortical (CSTC) circuits may underpin symptoms; and shed light on the putative neurochemistry within these loops such as the role of s

BACKGROUND: Parkinson's disease (PD) is a progressive neurodegenerative disorder associated with a loss of dopaminergic (DA) neurons. Despite symptomatic therapies, there is currently no disease-modifying treatment to halt neuronal loss in PD. A major hurdle for developing and testing such curative therapies results from the fact that most DA neurons are already lost at the time of the clinical diagnosis, rendering them inaccessible to therapy. Understanding the early pathological changes that precede Lewy body pathology (LBP) and cell loss in PD will likely support the identification of novel

Pediatric brain tumors are an incredibly diverse group of neoplasms and neuropathological tumor classification is an essential part of patient care. Classification of pediatric brain tumors has changed considerably in recent years as molecular diagnostics have become incorporated with routine histopathology in the diagnostic process. This article will focus on the fundamental major histologic, immunohistochemical, and molecular features that neuropathologists use to make an integrated diagnosis of pediatric brain tumors. This concise review will focus on tumors that are integral to the central

BACKGROUND AND PURPOSE: Neuropathology plays a major role in deciphering disease mechanisms in multiple sclerosis (MS). This review article describes recent advances in neuropathological research related to inflammatory demyelinating diseases.
METHODS: A retrospective review of neuropathological studies published during the last two decades was conducted.
RESULTS: The importance of neuropathology is generally seen in its contribution to the diagnosis of diseases of the nervous system and, in particular, in neuro-oncology. However, when it also includes analysis of the global three-dimensiona

Tauopathies display a spectrum of phenotypes from cognitive to affective behavioral impairments; however, mechanisms promoting tau pathology and how tau elicits behavioral impairment remain unclear. We report a unique interaction between polyamine metabolism, behavioral impairment, and tau fate. Polyamines are ubiquitous aliphatic molecules that support neuronal function, axonal integrity, and cognitive processing. Transient increases in polyamine metabolism hallmark the cell's response to various insults, known as the polyamine stress response (PSR). Dysregulation of gene transcripts associat

Sudden Unexplained Death in Childhood (SUDC) is the unexpected death of a child over age 12 months that remains unexplained after a thorough case investigation, including review of the child's medical history, circumstances of death, a complete autopsy and ancillary testing (1). First defined in 2005, SUDC cases are more often male, with death occurring during a sleep period, being found prone, peak winter incidence, associated with febrile seizure history in ~28% of cases and mild pathologic changes insufficient to explain the death (1, 2). There has been little progress in understanding the

Brain temperature is a physiologic parameter that depends on metabolism-related intracerebral heat production and heat loss by cerebral circulation to the rest of the body and then to the external environment. Despite the importance of temperature as a metabolism-related parameter and a factor affecting neural activity and function, it is generally believed that brain temperature is a tightly regulated and highly stable homeostatic parameter. To challenge this view, we present data on fluctuations in brain temperature occurring in rats following exposure to various arousing stimuli and during

Embryo/fetus is much more radiosensitive than neonatal and adult human being. The main potential effects of pre-natal radiation exposure on the human brain include growth retardation, small head/brain size, mental retardation, neocortical ectopias, callosal agenesis and brain tumor which may result in a lifetime poor quality of life. The patterns of prenatal radiation-induced effects are dependent not only on the stages of fetal development, the sensitivity of tissues and organs, but also on radiation sources, doses, dose rates. With the increased use of low dose radiation for diagnostic or ra
