Hijacking anaerobic metabolism to restore antibiotic efficacy in Pseudomonas aeruginosa.

Bacterial pathogens regularly encounter oxygen (O2) limitation at sites of infection in the human body. Many pathogens exhibit antibiotic recalcitrance under low O2 conditions, spotlighting the need for new therapeutics that are effective in O2-limited environments. We demonstrate that several classes of antibiotics display limited toxicity against hypoxic cultures of the opportunistic pathogen, Pseudomonas aeruginosa, where the pathogen exhibits antibiotic tolerance. In O2-limited host environments, many pathogens employ nitrate respiration, marking this anaerobic bacterial metabolism as an e

An N-acetyltransferase influences AreA nuclear localization and nitrogen utilization in the basidiomycete Ganoderma lucidum.

Nitrogen metabolism is fundamental to fungal growth, development, and the biosynthesis of bioactive metabolites. The GATA transcription factor AreA plays a pivotal regulatory role in nitrogen metabolite repression. While the molecular mechanisms by which AreA responds to nitrogen sources have been relatively well elucidated in ascomycetes, they remain poorly understood in basidiomycetes. Here, using the macrofungus Ganoderma lucidum as a model, we elucidated a molecular mechanism by which an N-acetyltransferase (Nat) influences AreA nuclear entry under non-preferred nitrogen sources, thereby p

The Noncanonical Pathway for In Vivo Nitric Oxide Generation: The Nitrate-Nitrite-Nitric Oxide Pathway.

In contrast to nitric oxide, which has well established and important roles in the regulation of blood flow and thrombosis, neurotransmission, the normal functioning of the genitourinary system, and the inflammation response and host defense, its oxidized metabolites nitrite and nitrate have, until recently, been considered to be relatively inactive. However, this view has been radically revised over the past decade and more. Much evidence has now accumulated demonstrating that nitrite serves as a storage form of nitric oxide, releasing nitric oxide preferentially under acidic and/or hypoxic c

Trehalose-6-Phosphate Phosphatase I (TPPI) Regulates Floral Transition, Nitrogen Responses, and Photosynthetic Performance in Arabidopsis.

Trehalose-6-phosphate (T6P) is a key signalling metabolite that integrates carbon availability with development and stress responses in plants. T6P levels are controlled by trehalose phosphate synthase (TPS) and trehalose-6-phosphate phosphatase (TPP) enzymes; however, while TPS enzymes have been studied extensively, the physiological functions of individual TPPs remain incompletely understood. Here, we investigated the role of TPPI in Arabidopsis using loss-of-function tppi mutants, a complemented line (tppi+35S::TPPI), and TPPI-overexpressing (TPPI-OEX) plants. The tppi mutant exhibited dela

Nitric Oxide-Dependent Regulation of Oxygen-Related Processes in a Rat Model of Lead Neurotoxicity: Influence of the Hypoxia Resistance Factor.

BACKGROUND/AIMS: Lead exposure is known to induce oxidative stress and neurotoxicity. Nitric oxide (NO) plays an important role in modulating oxidative stress, with L-arginine as a precursor of NO and Nω-nitro-L-arginine (L-NNA) as an inhibitor of NO synthase, an enzyme that catalyses the production of nitric oxide (NO) from L-arginine. METHODS: This study investigated the differential effects of L-arginine and L-NNA on markers of oxidative stress and biochemical changes in brain tissue from rats with different levels of resistance to hypoxia exposed to lead nitrate. Rats with either low or h

Microbiota responses to mutations affecting NO homeostasis in Arabidopsis thaliana.

Interactions between plants and microorganisms are pivotal for plant growth and productivity. Several plant molecular mechanisms that shape these microbial communities have been identified. However, the importance of nitric oxide (NO) produced by plants for the associated microbiota remains elusive. Using Arabidopsis thaliana isogenic mutants overproducing NO (nox1, NO overexpression) or down-producing NO (i.e. nia1nia2 impaired in the expression of both nitrate reductases NR1/NIA1 and NR2/NIA2; the 35s::GSNOR1 line overexpressing nitrosoglutathione reductase (GSNOR) and 35s::AHB1 line overexp

Evidence for post-translational regulation of NrtA, the Aspergillus nidulans high-affinity nitrate transporter.

Here, influx and efflux of (13)NO(3)(-), and net fluxes of (14)NO(3)(-) and (14)NO(2)(-), were measured in Aspergillus nidulans mutants niaD171 and niiA5, devoid of nitrate reductase (NR) and nitrite reductase (NiR) activities, respectively. Transcript and protein abundances of NrtA, the A. nidulans principal high-affinity NO(3)(-) transporter, were determined using semiquantitative reverse transcription-polymerase chain reaction and western blots, respectively. (13)NO(3)(-) influx in niaD171 was negligible relative to wild-type values, whereas efflux to influx ratios increased nine-fold. Neve

Adaptations in electron transport chain complexes of clinical Rhodococcus equi revealed through comparative genomics.

Rhodococcus equi is an important veterinary and zoonotic pathogen that survives inside macrophage phagolysosomes. Nitrate reductase NarGHI is a known virulence factor for this pathogen. This enzyme complex serves as the terminal electron acceptor in anaerobic respiration in other pathogens, such as the phylogenetically related Mycobacterium tuberculosis. A recent study demonstrated that R. equi survives under anaerobic conditions. In this work, we endeavored to uncover the electron transport chain complexes of R. equi. By in silico comparison to known complexes in the Transporter Classificatio

Isolation of anaerobic respiratory mutants of Shewannella putrefaciens and genetic analysis of mutants deficient in anaerobic growth on Fe3+.

A genetic approach was used to study (dissimilatory) ferric iron (Fe3+) reduction in Shewanella putrefaciens 200. Chemical mutagenesis procedures and two rapid plate assays were developed to facilitate the screening of Fe3+ reduction-deficient mutants. Sixty-two putative Fe3+ reduction-deficient mutants were identified, and each was subsequently tested for its ability to grow anaerobically on various compounds as sole terminal electron acceptors, including Fe3+, nitrate (NO3-), nitrite (NO2-), manganese oxide (Mn4+), sulfite (SO3(2-)), thiosulfate (S2O3(2-)), trimethylamine N-oxide, and fumara

Preliminary functional analysis of the subgingival microbiota of cats with periodontitis and feline chronic gingivostomatitis.

The subgingival microbial communities of domestic cats remain incompletely characterized and it is unknown whether their functional profiles are associated with disease. In this study, we used a shotgun metagenomic approach to explore the functional potential of subgingival microbial communities in client-owned cats, comparing findings between periodontally healthy cats and cats with naturally occurring chronic periodontitis, aggressive periodontitis, and feline chronic gingivostomatitis. Subgingival samples were subjected to shotgun sequencing and the metagenomic datasets were analyzed using

Microbial-Mediated Coevolution of Vanadium and Ferric Iron Minerals in Sedimentary Environments.

Rising vanadium(V) contamination severely threatens environmental safety and human health. Microbial-mediated pentavalent V [V(V)] reduction, an effective route to V detoxification, can be influenced by coexisting redox-sensitive matters. However, the interaction between V(V) and widely distributed ferric iron [Fe(III)] minerals still remains largely unknown. This study investigated the coupled transformations of V(V) and Fe(III) minerals in biosystems with Bacillus subtilis. Fe(III) minerals, particularly low-crystallinity ferrihydrite, markedly enhanced V(V) reduction. Concurrently, B. subti

[Effects of Trichoderma on nitrogen absorption and use efficiency in Lycium chinense roots under saline stress].

To clarify the mechanisms underlying the improvement of Trichoderma on Chinese wolfberry (Lycium chinense) growth under saline stress, we analyzed the effects of application of organic fertilizer, Trichoderma agent and fertilizer on nitrogen uptake, assimilation, accumulation and use efficiency in Chinese wolfberry, based on a pot experiment with coastal saline soil. The organic fertilizer was the sterilization substance of Trichoderma fertilizer without viable Trichoderma, without any difference in the content of nutrients (such as nitrogen, phosphorus and potassium) between them. The results

Response of inoculated foliar fed pea plants (Pisum sativum L.) to reduced Mo supply.

The application of nutrients to the roots and leaves of inoculated pea plants grown under conditions of reduced Mo supply was studied. Pea plants (Pisum sativum L.) were grown on liquid nutrient solution excluding Mo from the media until the 35th day under glasshouse conditions. Plants were inoculated with the bacterial suspension of Rhizobium leguminosarum Bv. Vicae, strain D293 at approximately 10(8) cells per cm3. The foliar fertilizer Agroleaf was applied at 0.3% concentration. Changes in the root nodulation and the activities of the enzymes connected with nitrogen assimilation pathway (ni

From cultivation to cancer: formation of N-nitrosamines and other carcinogens in smokeless tobacco and their mutagenic implications.

Tobacco use is a major cause of preventable morbidity and mortality globally. Tobacco products, including smokeless tobacco (ST), generally contain tobacco-specific N-nitrosamines (TSNAs), such as N'-nitrosonornicotine (NNN) and 4-(methylnitrosamino)-1-(3-pyridyl)-butanone (NNK), which are potent carcinogens that cause mutations in critical genes in human DNA. This review covers the series of biochemical and chemical transformations, related to TSNAs, leading from tobacco cultivation to cancer initiation. A key aim of this review is to provide a greater understanding of TSNAs: their precursors

Transcriptome and metabolome analyses reveal the mechanisms by which H2S improves energy and nitrogen metabolism in tall fescue under low-light stress.

Hydrogen sulfide (H2S) functions as a signaling molecule affecting plant growth, development, and stress adaptation. Tall fescue (Festuca arundinacea Schreb.), a bioenergy crop, encounters significant challenges in agricultural production owing to low light by shading. However, the influence of H2S on tall fescue under low light stress (LLS) remains unclear. To examine the role of H2S in acclimation of tall fescue to low light, we conducted combined analyses of physiological traits, metabolomics, and transcriptomics. These results showed that H2S mitigated LLS-induced inhibition of photosynthe

Rice husk-intensified cathode driving bioelectrochemical reactor for remediating nitrate-contaminated groundwater.

To achieve economical and eco-friendly denitrification, rice husk-intensified cathode driving bioelectrochemical reactor (RCBER) was constructed with rice husk as solid-phase carbon source and microbial carrier. Results demonstrated that the application of current improved the utilization of rice husk and enhanced the denitrification, and the quenching of anodic hydroxyl radicals by rice husk also improved the microbial resistance to current. The highest nitrate removal rate as 0.34 mg-N/(L∙d), higher economic benefits, i.e., current efficiency as 31.6% and energy consumption as 2.43 kWh/g NO3
