The evolutionary genomics of novel endosymbiosis in wild rhizobia bacteria.

The advent of endosymbiosis underlies evolutionary innovation and ecosystem function. However, whether free-living partners tend to benefit … novel endosymbionts derived from more closely related SI donor and recipient strains showed greater nitrogen fixation. However, consistent with selection on the SI for broad horizontal transfer, we did not detect phylogenetic limits to SI transmission, and the SI was able to displace other genomic elements residing at its characteristic tRNA gene insertion site. We …

Ocean warming interacts with elevated nitrogen to modulate carbon storage-excretion balance in Gracilariopsis lemaneiformis.

Macroalgae farming represents a proposed approach for carbon dioxide removal (CDR), as macroalgae exhibit distinct advantages over other blue carbon taxa in cultivation scalability and cost effectiveness. Yet how future warming and coastal nitrogen enrichment will jointly affect macroalgal carbon fixation and sequestration remains poorly understood. Here, we investigated physiological and biochemical responses (growth, photosynthesis, pigment content, and carbon accumulation and excretion) of the commercially important seaweed Gracilariopsis lemaneiformis under a cross-factorial design of two

Metagenomic Insights into Microbial Functional Potential Associated with Soil Carbon, Nitrogen, and Phosphorus Cycling Along an Elevational Gradient in a Warm-Temperate Forest.

Soil microbial functional potential is crucial to maintaining forest productivity and ecosystem functions. However, how microbially mediated soil nutrient cycling responds to environmental changes, particularly those caused by variations in elevation, remains poorly understood. Using the natural temperature gradient in a temperate mountain forest, this study investigated the differences in functional microbial groups and functional genes involved in soil carbon, nitrogen and phosphorus cycling along the elevation gradient, and analyzed the associations between environmental factors and these d

Plasma-assisted ammonia synthesis utilizing water as a hydrogen source: progress, challenges, and prospects.

Ammonia, a vital chemical precursor primarily used in nitrogen fertilizer production, is also a promising hydrogen carrier. However, conventional ammonia synthesis methods, notably the Haber-Bosch process, are energy-intensive and environmentally unsustainable. This review explores the emerging field of plasma-based ammonia synthesis using water as a hydrogen source, offering a greener and potentially sustainable alternative. Non-thermal plasma technology, operating under mild conditions, facilitates nitrogen fixation by activating inert nitrogen molecules through electron impact, collisions, …

GmNIGT2a/2b-mediated transcriptional cascade buffers high-nitrogen-triggered nodule senescence in soybean.

Soil inorganic nitrogen (N) availability is a critical determinant of symbiotic nitrogen fixation efficiency, making it essential for legumes to respond appropriately and effectively to N fluctuations. Here, we identify a pair of high N response factors GmNIGT2a/2b (NITRATE-INDUCIBLE GARP-TYPE TRANSCRIPTIONAL REPRESSOR). GmNIGT2a/2b are activated by GmNLP4a/b (NIN-like proteins) under high N. The nodules of gmnigt2a/2b double mutants exhibit exacerbated reduction in nitrogenase activity and accelerated senescence in response to high N. Integration of RNA-seq and DAP-seq analyses reveals that G…

Insights into Fe0, Fe2O3, and Fe3O4-Mediated humification and nitrogen transformation during composting.

This study evaluated Fe0, Fe2O3, and Fe3O4 as regulators of humification during composting of chicken manure (CM) and wine grape pomace (WGP). Results showed that Fe2O3 and Fe3O4 substantially enhanced humification, raising H2O2 (15-47 %) and ·OH (3-16 %) and thereby driving aromatic CC formation and hastening HA production and aromatization. Specifically, Fe2O3 upregulated hydroxylamine dehydrogenase and the nitrogenase molybdenum-iron protein. Correspondingly, nitrification and nitrogen fixation increased by 4.9 % and 6.7 %, respectively, with enrichment of key taxa (e.g., Oceanobacillus, Th…

'Candidatus Viridifilum namsaraevi' gen. nov., sp. nov., a new mesophilic filamentous anoxygenic phototrophic bacterium from the mesothermal lake Umkhey (Buryatia).

The evolution of phototrophy and carbon fixation in the phylum Chloroflexota remains incompletely understood, largely due to the scarcity of cultured mesophilic representatives. Here, we report the isolation and genomic characterization of a novel filamentous anoxygenic phototrophic bacterium from a microbial mat in the mesothermal Lake Umkhey (Southern Siberia). Although the original culture was lost, its genome was recovered via metagenome-assembled genome reconstruction from environmental samples, enabling a detailed phylogenomic and metabolic analysis. The genome of umkhey_bin13 (proposed

Impact of biochar amendment on the abundance and structure of diazotrophic community in an alkaline soil.

Biological nitrogen (N) fixation contributes to the pool of plant-available N in soil and helps to minimize the use of inorganic N fertilizer in agricultural ecosystems. Although diazotrophs play an important role in the biological fixation of atmospheric N2 in a range of soil types, the knowledge of their response to biochar amendment is still limited. Here, using the nifH gene as a molecular marker, we investigated the short-term effect of biochar application on the abundance, community composition and activity of diazotroph in an alkaline soil. A field trial was established before soybean s

Iron is required to relieve inhibitory effects on NifL on transcriptional activation by NifA in Klebsiella pneumoniae.

In Klebsiella pneumoniae, products of the nitrogen fixation nifLA operon regulate transcription of the other nif operons. NifA activates transcription by sigma54-holoenzyme. In vivo, NifL antagonizes the action of NifA under aerobic conditions or in the presence of combined nitrogen. In contrast to a previous report, we show that depletion of iron (Fe) from the growth medium with the chelating agent o-phenanthroline (20 microM) mimics aerobiosis or combined nitrogen in giving rise to inhibition of NifA activity even under anaerobic, nitrogen-limiting conditions. Adding back Fe in only twofold …

Microalgae carbon fixation integrated with organic matters recycling from soybean wastewater: Effect of pH on the performance of hybrid system.

Microalgae have been considered as promising alternative for CO2 fixation and wastewater purification. In our previous work, a hybrid microalgae CO2 fixation concept has been put forward, which initially used carbonate solution absorb CO2, and then provided obtained bicarbonate as nutrition for microalgae growth to avoid the challenge of low CO2 solubility and carbon fixation efficiency in the conventional process. In this work, the proposed hybrid system was further intensified via integrating soybean wastewater nutrition removal with bicarbonate-carbon (NH4HCO3 and KHCO3) conversion. The inv

Waste iron scraps promote anammox bacteria to resist inorganic carbon limitation.

The anaerobic ammonium oxidation (anammox) process is adversely affected by the limitation of inorganic carbon (IC). In this research, a new technique was introduced to assist anammox biomass in counteracting the adverse effects of IC limitation by incorporating waste iron scraps (WIS), a cheap and easily accessible byproduct of lathe cutting. Results demonstrated that reducing the influent IC/TN ratio from 0.08-0.09 to 0.04 resulted in a 20 % decrease in the nitrogen removal rate (NRR) for the control reactor, with an average specific anammox activity (SAA) of 0.65 g N/g VSS/day. Nevertheless

In situ transcriptomic analysis of the globally important keystone N2-fixing taxon Crocosphaera watsonii.

The diazotrophic cyanobacterium Crocosphaera watsonii supplies fixed nitrogen (N) to N-depleted surface waters of the tropical oceans, but the factors that determine its distribution and contribution to global N(2) fixation are not well constrained for natural populations. Despite the heterogeneity of the marine environment, the genome of C. watsonii is highly conserved in nucleotide sequence in contrast to sympatric planktonic cyanobacteria. We applied a whole assemblage shotgun transcript sequencing approach to samples collected from a bloom of C. watsonii observed in the South Pacific to un

Long-term PFOA and cadmium Co-contamination alters soil carbon, nitrogen, and phosphorus cycling: Insights from metagenomics and metabolomics.

The co-existence of perfluorooctanoic acid (PFOA) and cadmium (Cd) in soil poses a combined threat to microbial communities. However, the ecological effects and underlying mechanisms of their long-term combined exposure remain poorly understood. This study conducted a 90-day soil microcosm experiment to systematically investigate the effects of individual and combined effects of PFOA and Cd on microbial communities. Our results demonstrated that combined pollution of PFOA and Cd significantly affected four soil enzyme activities associated with carbon, nitrogen, and phosphorus cycling. It also

Size Effects of Copper Oxide Nanoparticles on Boosting Soybean Growth via Differentially Modulating Nitrogen Assimilation.

Nanoscale agrochemicals have been widely used in sustainable agriculture and may potentially affect the nitrogen fixation process in legume crops. The present study investigated the size-effects of copper oxide nanoparticles (CuO NPs) on nitrogen assimilation in soybean (G. max (L.) Merrill) plants, which were treated with different sizes (20 and 50 nm) of CuO NPs at low use doses (1 and 10 mg/kg) for 21 days under greenhouse conditions. The results showed that 50 nm CuO NPs significantly increased the fresh biomass more than 20 nm CuO NPs achieved at 10 mg/kg. The activities of N assimilation…

Heterotrophic inorganic carbon fixation varies with temperature and resource availability in marine and soil bacterial isolates.

Inorganic carbon use in cellular metabolism is ubiquitous in all living organisms. The assimilation of inorganic carbon into biomass via heterotrophic inorganic carbon fixation (HICF) is estimated to contribute significantly to global carbon fixation, yet the controls on this process in strictly heterotrophic microorganisms remain poorly understood. To identify potential drivers of HICF, we cultivated well-characterized heterotrophic Gammaproteobacteria (Pseudomonas putida and Pseudoalteromonas carrageenovora) at two temperatures (optimal vs colder) and resource treatments (high vs low organic
