Oncogenic osteomalacia induced by schwannoma in a patient with neurofibromatosis.

A 62-year-old woman known to be suffering from Von Recklinghausen disease and osteomalacia was operated on for a pelvic mass. Pathological examination of the excised tumor showed it to be a schwannoma and this tumor was considered to be responsible for the osteomalacia. The return of previously abnormal laboratory data to normal values and the impressive clinical and subjective improvement further support the assumption that the excised schwannoma was responsible for the pathology observed in this patient.

Expression of c-myc oncogene in coeliac disease.

A monoclonal antibody, produced by peptide immunisation was used to detect the distribution of p62c/myc by immunohistology in normal and coeliac small intestinal mucosa. The effect of gluten in four treated coeliac patients was investigated by taking serial jejunal biopsy specimens for six hours after a 10 g oral gluten challenge. There was a progressive increase in p62c/myc staining intensity in the villus enterocytes extending to the crypts, which accompanied the classical morphological changes occurring in the mucosa.

In vitro assessment of the oncogenic potential of nitroimidazole radiosensitizers.

Two hypoxic cell radiosensitizers, RSU-1069 and Ro-03-8799 were investigated for their in vitro cytotoxicity and ability to induce oncogenic transformation and sister chromatid exchanges in the C3H 10T1/2 cell system. Their effects were then compared to those of the clinically used sensitizer misonidazole. Equitoxic doses of Ro-03-8799 and RSU-1069 were approximately 3-fold and 150-fold less than misonidazole, respectively, with both agents exhibiting dose and contact time dependence for cell killing. Both sensitizers appeared no more oncogenic than misonidazole when administered at equitoxic

Vitamin D metabolite levels in oncogenic osteomalacia.

Measurements of 1,25 (OH)2D3 and other metabolites of vitamin D in a patient with oncogenic osteomalacia confirm the selective, reversible deficiency of 1,25 (OH)2D3 in this syndrome, and indicate the rapidity of normalization (within days) of the hormone level and associated hypophosphatemia after resection of the tumor. In this patient, the tumor was an intranasal hemangiopericytoma.

Targeting Oncogenic Gene Fusions in Lung Cancer.

Oncogenic gene fusions define a clinically important subset of non-small cell lung carcinoma (NSCLC) for which targeted therapies have transformed outcomes. Over the past 15 years, successive generations of tyrosine kinase inhibitors (and more recently, monoclonal antibodies) have demonstrated substantial improvements in response rates, progression-free survival, and central nervous system control across multiple fusion-defined populations. This review consolidates current evidence for FDA-approved therapies targeting ALK, ROS1, RET, NTRK, and NRG1 fusions in lung cancer, highlighting mechanis

LIN28B: LINeage-dependent oncogene or tumor suppressor?

INTRODUCTION: Dry eye disease (DED) is a multifactorial disorder characterized by tear film instability, ocular surface inflammation, and epithelial compromise, often predisposing patients to complications such as acute keratitis and persistent corneal epithelial defects. Amniotic membrane therapy has emerged as a valuable treatment modality for DED due to its ability to protect against mechanical trauma, modulate inflammation and fibrosis, promote corneal epithelial migration, and help restore ocular surface homeostasis in refractory cases. METHODS: This retrospective clinical series evaluat

The Expression and Oncogenic Role of NTSR2 in Human Glioblastoma.

BACKGROUND/AIMS: Neurotensin receptor 2 (NTSR2) plays critical pro-tumor roles in multiple human solid tumors. However, the expression characteristics, biological function and molecular mechanism of NTSR2 in glioblastoma (GBM) remain poorly elucidated. METHODS: Bioinformatics analysis was used to analyze NTSR2 expression and its clinical prognostic significance in GBM. NTSR2 expression in glioma tissues was validated by immunohistochemistry, Western blot (WB) and RT-qPCR, while immunofluorescence staining detected its subcellular localization. In vitro functional assays were performed to eval

A Novel Mechanism of STAT3 Activation by Oncogenic Signaling.

CARP-1, a perinuclear phospho-protein, is a biphasic regulator of cell survival and apoptosis signaling. We previously found that UV cross-linking of proteins from HeLa cervical cancer cells resulted in STAT3 interacting with the CARP-1 (614-638) peptide. Mutagenesis and co-IP-WB experiments revealed that CARP-1 interacts with a 40-amino-acid epitope from positions 441-480 (CE Epitope) located in the STAT3 DNA-binding domain. Overexpression of mutant STAT3 with in-frame deletion of the CE epitope (Gst-STAT3 (ΔCE) mutant), but not Gst-STAT3 (WT), failed to translocate to the nucleus in IL-6-tre

Oncogenic role of PHF23 in gastric cancer progression.

Gastric cancer (GC) poses a persistent global health burden, largely attributed to its asymptomatic early stages and limited efficacy of existing therapies. PHF23, which has been primarily studied in hematologic malignancies and degenerative joint diseases, has not been explored in gastric cancer. In this study, PHF23 expression was assessed in GC tissues through immunohistochemistry and public dataset analysis. Functional assays, including proliferation, migration, invasion, and apoptosis, were performed following PHF23 knockdown in GC cell lines. Tumorigenic effects were evaluated in nude mo

Analysis and identification of PTBP2 as an oncogene in hepatocellular carcinoma.

PURPOSE: Liver cancer, also known as hepatocellular carcinoma (HCC), is one of the most prevalent and lethal malignancies in the world. The management of HCC depends on the underlying hepatic function and histological stage of the neoplasm, potentially encompassing locoregional therapies, surgical resection, hepatic transplantation, targeted drug therapies, and immunotherapy. However, the outlook for advanced HCC remains poor, with global 5-year survival rates below 15%. Consequently, it is imperative to identify novel tumor biomarkers for HCC prediction. METHODS: Multiple public databases we

Oncogene-Driven Induction of Orthotopic Cholangiocarcinoma in Mice.

Cholangiocarcinoma (CCA) is a malignancy affecting the epithelial cells that line the bile ducts. This cancer shows a poor prognosis and current treatments remain inefficient. Orthotopic CCA mouse models are useful for the development of innovative therapeutic strategies. Here, we describe an orthotopic model of intrahepatic CCA that can be easily induced in mice within 5 weeks at a high incidence. It is achieved by expressing two oncogenes, namely, (i) the intracellular domain of the Notch1 receptor (NICD) and (ii) AKT, in hepatocytes by means of the sleeping beauty transposon system. These p…

FOXP2 confers oncogenic effects in prostate cancer.

Identification oncogenes is fundamental to revealing the molecular basis of cancer. Here, we found that FOXP2 is overexpressed in human prostate cancer cells and prostate tumors, but its expression is absent in normal prostate epithelial cells and low in benign prostatic hyperplasia. FOXP2 is a FOX transcription factor family member and tightly associated with vocal development. To date, little is known regarding the link of FOXP2 to prostate cancer. We observed that high FOXP2 expression and frequent amplification are significantly associated with high Gleason score. Ectopic expression of FOX…

T-ALL can evolve to oncogene independence.

The majority of cases of T-cell acute lymphoblastic leukemia (T-ALL) contain chromosomal abnormalities that drive overexpression of oncogenic transcription factors. However, whether these initiating oncogenes are required for leukemia maintenance is poorly understood. To address this, we developed a tetracycline-regulated mouse model of T-ALL driven by the oncogenic transcription factor Lmo2. This revealed that whilst thymus-resident pre-Leukemic Stem Cells (pre-LSCs) required continuous Lmo2 expression, the majority of leukemias relapsed despite Lmo2 withdrawal. Relapse was associated with a …

Recurrent Superenhancer of the Oncogene POU5F1B in Colorectal Cancers.

Superenhancer usages in single cancer form such as colorectal cancer (CRC) may provide novel efficient targeting candidates. It is unclear whether CRC contains recurrent superenhancers that confer a predisposition to malignancy. We investigated the superenhancer profile of CRC cell line HCT116 and compared it to that of a healthy sigmoid colon. We found that HCT116 had lost most of the normal colon superenhancer activities but gained a new set of tumor-favoring superenhancers that facilitate tumor proliferation, growth signalling, and hypoxia resistance. Inhibiting the superenhancers by JQ-1 t

Alternative polyadenylation is a determinant of oncogenic Ras function.

Alternative polyadenylation of mRNA has important but poorly understood roles in development and cancer. Activating mutations in the Ras oncogene are common drivers of many human cancers. From a screen for enhancers of activated Ras (let-60) in Caenorhabditis elegans, we identified cfim-1, a subunit of the alternative polyadenylation machinery. Ablation of cfim-1 increased penetrance of the multivulva phenotype in let-60/Ras gain-of-function (gf) mutants. Depletion of the human cfim-1 ortholog CFIm25/NUDT21 in cancer cells with KRAS mutations increased their migration and stimulated an epithel

Roles of the RET Proto-oncogene in Cancer and Development.

RET (REarranged during Transfection)is activated by DNA rearrangement of the 3' fragment of the receptor tyrosine kinase gene, namely, RET proto-oncogene, with the 5' fragment of various genes with putative dimerization domains, such as a coiled coil domain, that are necessary for constitutive activation. RET rearrangements have been detected in a variety of human cancers, including thyroid, lung, colorectal, breast, and salivary gland cancers. Moreover, point mutations in RET are responsible for multiple endocrine neoplasia types 2A and 2B, which can develop into medullary thyroid cancer and
