Effect of castration and cryptorchidism on fatty acid content of ovine adipose tissue.

Fatty acid analyses of perinephric and subcutaneous adipose tissue were made on 50 of Mehraban and Bakhriari breeds lambs to determine the effect of castration and cryptorchidism on ovine lipid composition. The level of stearic acid was significantly higher (p < 0·05) in cryptorchids than in wethers. Rams were also higher (p < 0·05) in the percentage of linoleic acid of the internal adipose tissue than wethers. The percentage of palmitic acid of subcutaneous tissue was higher (p < 0·05) in cryptorchid than rams. There was a tendency toward less saturated (palmitic and stearic) acids, but more

A human hepatocellular in vitro model to investigate steatosis.

The present study was designed to define an experimental model of hepatocellular steatosis with a fat overaccumulation profile in which the metabolic and cytotoxic/apoptotic effects could be separated. This was accomplished by defining the experimental conditions of lipid exposure that lead to significant intracellular fat accumulation in the absence of overt cytotoxicity, therefore allowing to differentiate between cytotoxic and apoptotic effects. Palmitic (C16:0) and oleic (C18:1) acids are the most abundant fatty acids (FFAs) in liver triglycerides in both normal subjects and patients with

PRAP1 aggravates palmitate-induced insulin resistance in skeletal muscle cells through a CD36-dependent mechanism.

Proline-rich acidic protein 1 (PRAP1) is involved in intestinal lipid handling, but its role in skeletal muscle insulin resistance is unknown. We examined circulating PRAP1 in normal diet- and high-fat diet-fed mice and evaluated PRAP1 expression and secretion in palmitate-treated intestinal epithelial cells, together with its functional effects in differentiated C2C12 myotubes. PRAP1 expression and secretion were assessed following palmitate exposure, while C2C12 myotubes were treated with recombinant PRAP1 and palmitate followed by insulin stimulation. CD36 expression, fatty acid uptake, int

Presynaptically acting snake venom phospholipase A2 enzymes attack unique substrates.

Synaptosomes were incubated with bovine serum albumin (BSA) to examine whether the presynaptic action of snake venom phospholipase A2 (PLA2) toxins is due either to the release of fatty acids resistant to extraction by BSA or to the liberation of a specific fatty acid type. In the presence of BSA (0.5% or 1.0%) two PLA2 enzymes from Naja naja atra and Naja naja kaouthia snake venoms that do not have a predominant presynaptic action at the neuromuscular junction (PS-) did not stimulate acetylcholine (ACh) release from synaptosomes. In contrast, two PLA2 enzymes (beta-bungarotoxin, scutoxin) tha

Abnormal lung surfactant related to essential fatty acid deficiency in a neonate.

A low-birth-weight infant, suffering from chronic bronchopulmonary dysplasia following hyaline membrane disease and recurrent episodes of necrotizing enterocolitis, developed biochemical evidence of essential fatty acid (EFA) deficiency in the plasma. Fatty acid composition of phosphatidylcholine and phosphatidylglycerol in the lung lavage fluid was abnormal. Plasma changes included a decrease in the level of linoleic acid and an increased level of palmitic, palmitoleic, oleic, and 5,8,11-eicosatrienoic acid to arachidonic acid being greater than 0.4:1. A lower than normal level of palmitic ac

HIF-1α expression as a protective strategy of HepG2 cells against fatty acid-induced toxicity.

Free fatty acid-induced lipotoxicity via increased endoplasmic reticulum (ER) stress and hepatocyte apoptosis is a key pathological mechanism of non-alcoholic steatohepatitis. A role of hypoxia-inducible factor 1α (HIF-1α) in this process has been suggested, but direct evidence is lacking. Here, we used HepG2 cells as a model to study whether HIF-1α can reduce palmitic acid-induced lipotoxicity and ER stress. In HepG2 cells treated with 500 µM palmitic acid, HIF-1α expression increased transiently, the decline was associated with increased cleaved caspase-3 expression. Overexpression and knock

Differential effects of saturated and unsaturated fatty acids on vascular reactivity in isolated mesenteric and femoral arteries of rats.

Free fatty acid (FFA) intake regulates blood pressure and vascular reactivity but its direct effect on contractility of systemic arteries is not well understood. We investigated the effects of saturated fatty acid (SFA, palmitic acid), polyunsaturated fatty acid (PUFA, linoleic acid), and monounsaturated fatty acid (MUFA, oleic acid) on the contractility of isolated mesenteric (MA) and deep femoral arteries (DFA) of Sprague-Dawley rats. Isolated MA and DFA were mounted on a dual wire myograph and phenylephrine (PhE, 1-10 µM) concentration-dependent contraction was obtained with or without FFAs

(+)-Lipoic acid reduces mitochondrial unfolded protein response and attenuates oxidative stress and aging in an in vitro model of non-alcoholic fatty liver disease.

BACKGROUND: Non-alcoholic fatty liver disease (NAFLD) is a liver disorder characterized by the ac-cumulation of fat in hepatocytes without alcohol consumption. Mitochondrial dysfunction and endoplasmic reticulum (ER) stress play significant roles in NAFLD pathogenesis. The unfolded protein response in mitochondria (UPRmt) is an adaptive mechanism that aims to restore mitochondrial protein homeostasis and mitigate cellular stress. This study aimed to investigate the effects of ( +)-Lipoic acid (ALA) on UPRmt, inflammation, and oxidative stress in an in vitro model of NAFLD using HepG2 cells tre

Linking endoplasmic reticulum stress to cell death in hepatocytes: roles of C/EBP homologous protein and chemical chaperones in palmitate-mediated cell death.

Prolonged endoplasmic reticulum (ER) stress and activation of the unfolded protein response (UPR) have been linked to apoptosis via several mechanisms, including increased expression of C/EBP homologous protein (Chop). Increased long-chain fatty acids, in particular saturated fatty acids, induce ER stress, Chop expression, and apoptosis in liver cells. The first aim of the present study was to determine the role of Chop in lipid-induced hepatocyte cell death and liver injury induced by a methionine-choline-deficient diet. Albumin-bound palmitate increased Chop gene and protein expression in a

Effects of Sex and Obesity on Lipid Metabolism during Low-Intensity Exercise and Postexercise Period.

BACKGROUND: Whether there are sex and obesity differences in fat oxidation and free fatty acid (FFA) mobilization during low-intensity exercise is unclear. METHODS: We studied 21 (nine females) participants without and 24 (16 females) with obesity during 5 h of treadmill walking (~2 mph). We measured energy expenditure (EE) and substrate oxidation (indirect calorimetry) and FFA-palmitate kinetics and oxidation with isotopic palmitate infusions ([U- 13 C]palmitate and [1- 14 C]palmitate or [9,10- 3 H]palmitate). Protective effect of ascorbic acid against renal injury induced by 3-chloropropane-1,2-diol-dipalmitate in rats.

3-monochloropropane-1,2-diol esters (3-MCPDE) are a group of contaminants which are mainly formed during heat processing of edible oil and fat-based foods. The kidney is the primary target organ for the toxic effects of 3-MCPDE. 3-MCPD-di-palmitate exists in a variety of oils and fats, and is the most common and relatively high proportion of 3-MCPDE. In this study, we investigated the protective effect of ascorbic acid on 3-MCPD-di-palmitate-induced renal injury in rats. Thirty 8-week-old male Sprague-Dawley rats were randomly divided into 5 groups, namely control, 3-MCPD-di-palmitate (240 mg/

Inhibitory effect of FO-1561 (S-adenosyl-L-methionine sulfate tosylate) on phospholipase A2.

The mechanism of the protective effect of FO-1561 (S-adenosyl-L-methionine sulfate tosylate) on mitochondria was investigated using rat heart muscle. FO-1561 stimulated CO2 production from palmitic acid in the mitochondria, indicating an increase in energy production. FO-1561 inhibited mitochondrial swelling induced by Ca2+ added exogenously or by addition of snake venom phospholipase A2. Moreover, FO-1561 inhibited phospholipase A2 activity directly. These results suggested that FO-1561 blocked mitochondrial swelling by inhibiting endogenous phospholipase A2 activity and so preventing decreas

Clinical evaluation of the 14C fat absorption test.

The absorption of (14)C-labelled glyceryl tripalmitate was tested in 62 patients by determining the maximum specific activity of CO(2) in the breath after an oral dose. A significant correlation with faecal fat excretion was found. The absorption of (14)C labelled palmitic acid was tested subsequently in 54 patients. In cases of pancreatic steatorrhoea, normal absorption of palmitic acid with impaired absorption of glyceryl tripalmitate was found, whereas in most patients with coeliac disease the absorption of both substances was depressed to the same extent.

Ultrahigh magnetic field selectively promotes starch-fatty acid complexation depending on crystal type and fatty acid unsaturation.

Ultrahigh magnetic field (UMF) processing can modulate the properties of starch and alter starch crystallization. However, the effect of UMF in the formation of starch-fatty acid complexes remains limited, particularly regarding systematic comparisons across different crystalline starch and varied fatty acids. Using A-type wheat starch, B-type potato starch, saturated palmitic acid, and unsaturated palmitoleic acid as model systems, this study examined the effects of UMF treatment at two distinct stages (pretreatment and post-treatment) on starch-fatty acid complexation. UMF treatment at diffe

Influence of reducing compounds on the formation of DNA-protein cross-links in HL-60 cells induced by hexavalent chromium.

The influence of reducing compounds on the formation of DNA--protein cross-links induced by hexavalent chromium was studied in the human cell line HL-60. Analysis of cytoplasmic concentration of ascorbic acid and glutathione by HPLC in these cells showed that ascorbic acid was not detectable (detection limit: 0.1 nmol). The cellular content of glutathione was low (6 nmol/million cells). It could easily be depleted with diethylmaleate. The effect of glutathione, ascorbic acid and ascorbyl palmitate alone, or glutathione in combination with ascorbyl palmitate was investigated. It could be shown

Augmented CPT1A Expression Is Associated with Proliferation and Colony Formation during Barrett's Tumorigenesis.

Obesity is a known risk factor for the development of gastroesophageal reflux disease (GERD), Barrett's Esophagus (BE) and the progression to esophageal adenocarcinoma. The mechanisms by which obesity contributes to GERD, BE and its progression are currently not well understood. Recently, changes in lipid metabolism especially in the context of a high fat diet have been linked to GERD and BE leading us to explore whether fatty acid oxidation plays a role in the disease progression from GERD to esophageal adenocarcinoma. To that end, we analyzed the expression of the rate-limiting enzyme, carni

Cellular fatty acid composition of Kingella species, Cardiobacterium hominis, and Eikenella corrodens.

We determined the cellular fatty acid composition of reference strains and clinical isolates of each of the three Kingella species, Cardiobacterium hominis, and Eikenella corrodens by using capillary gas chromatography. Kingella denitrificans and Kingella kingae contained myristic (14:0) and palmitic (16:0) acids as major acids, whereas cis-vaccenic (18:1 omega 7c) and palmitic acids were the major acids in Kingella indologenes, C. hominis, and E. corrodens. C. hominis differed from the other four species by the absence of 3-hydroxylauric (3-OH-12:0) acid, from K. indologenes by the presence o

Differential effects of oleate on vascular endothelial and liver sinusoidal endothelial cells reveal its toxic features in vitro.

Several fatty acids, in particular saturated fatty acids like palmitic acid, cause lipotoxicity in the context of non-alcoholic fatty liver disease . Unsaturated fatty acids (e.g. oleic acid) protect against lipotoxicity in hepatocytes. However, the effect of oleic acid on other liver cell types, in particular liver sinusoidal endothelial cells (LSECs), is unknown. Human umbilical vein endothelial cells (HUVECs) are often used as a substitute for LSECs, however, because of the unique phenotype of LSECs, HUVECs cannot represent the same biological features as LSECs. In this study, we investigat

Palmitate and group B Streptococcus synergistically and differentially induce IL-1β from human gestational membranes.

INTRODUCTION: Rupture of the gestational membranes often precedes major pregnancy complications, including preterm labor and preterm birth. One major cause of inflammation in the gestational membranes, chorioamnionitis (CAM) is often a result of bacterial infection. The commensal bacterium Streptococcus agalactiae, or Group B Streptococcus (GBS) is a leading infectious cause of CAM. Obesity is on the rise worldwide and roughly 1 in 4 pregnancy complications is related to obesity, and individuals with obesity are also more likely to be colonized by GBS. The gestational membranes are comprised o
