Risk of infection following exposures to human parvovirus B19.

In response to concern about the effect of human parvovirus B19 infection of the fetus, we have developed estimates of the risk of adults becoming infected following B19 exposures at home, in schools or day-care centers, and in hospitals. These estimates can then be used with other data to estimate the risk to the fetus of a B19 exposure during pregnancy. The risk to the fetus equals the rate of maternal susceptibility to infection times the rate of maternal infection following the specific type of exposure times the rate of fetal death following maternal infection. Data from studies of outbre…

[Molecular biological demonstration of parvovirus infection in fetal tissue].

We report on the morphological findings in 16 fetuses with serologically confirmed maternal parvovirus B19 infection. Typical routine morphological findings of the hydropic fetuses were the presence of abnormal erythroblasts with typical nuclear inclusions. These infected cells positively stained immunohistochemically with antibodies against a recombinant virus protein, as well as they ultrastructurally contained virus particles. Using in-situ hybridization techniques, we were able to demonstrate the presence of parvovirus B19 genome in the infected cells, while no other cell type was shown to…

[Prevalence of anti-human parvovirus antibody in pregnant women].

Human parvovirus B19 (B19) has been shown to be associated with erythema infectiosum. Recently, it was reported that when a pregnant woman is infected with B19, the fetus in her uterus sometimes becomes hydropic and results in a stillbirth. But no epidemiologic study of pregnant women in Japan has been performed yet. We have established an in vitro propagation system of B19 virions and an assay system for detecting anti-B19 antibody by indirect immunofluorescence (IF) staining. We examined the positive rate of anti-B19 antibody among 337 normal blood donors, 329 normal pregnant women, 23 early…

Pathogenesis of disease caused by Aleutian mink disease parvovirus.

A review of the pathogenesis of Aleutian mink disease parvovirus (ADV) infection based on recent knowledge gained by the author and collaborators is given. The review focuses mainly on the following topics. 1) Development of an easy, sensitive and fast assay for detection of ADV antigens and antibodies directed against these antigens. A highly sensitive rocket line immunoelectrophoretic assay (RLIE) was developed. This assay turned out to be 32 times more sensitive than the counter current electrophoresis assay routinely used to detect anti-ADV antibodies in ADV eradication programs, and moreo…

Transmission of human parvovirus B19 by coagulation factor concentrates.

The prevalence of antibody to human parvovirus B19 was determined in 86 children with congenital bleeding disorders. Forty-seven of 53 boys (89%) receiving non-heat-treated factor VIII or prothrombin complex concentrates were anti-B19 IgG positive compared with 38% of their age-matched controls and 48% of children treated with cryoprecipitate. Acute B19 virus infection occurred in 2 boys 3-4 weeks after they had received the same batch of commercial factor VIII concentrate. Of 11 susceptible children who had only received heat-treated National Health Service factor VIII concentrate (8Y), 1 acq…

Parvovirus associated aplastic crisis in homozygous sickle cell disease.

Aplastic crises in homozygous sickle cell disease in Jamaica predominantly affect children and occur in epidemics. Of 67 cases in a cohort study of 314 children with homozygous sickle cell disease, 62 were attributable to human parvovirus infection. Affected children were aged 0.5-12.5 years, and the incidence rose to 28% by 10 years. No recurrences were seen. Symptoms and signs on presentation were attributable to the viraemia and acute anaemia. Asymptomatic thrombocytopenia was common. Blood transfusion was given in 54 cases (87%). Thirty eight children (61%) were admitted to hospital, 16 of…

Cellular determinants of parvovirus B19 susceptibility in the human placenta.

Parvovirus B19 (B19V) is a prevalent human pathogen that can cross the placenta by a mechanism that remains unknown, posing a risk of severe fetal complications, particularly during the first trimester of pregnancy. We investigated the expression of B19V-specific receptors in the three trophoblast cell types, cytotrophoblasts (CTBs), syncytiotrophoblasts (STBs), and extravillous trophoblasts (EVTs), and assessed their susceptibility to infection. VP1uR, the receptor that mediates viral uptake and infection in erythroid progenitor cells, is expressed in CTBs and STBs, but not in EVTs. Globoside…

Novel Parvovirus in Pigs Associated with Exophthalmos and Erythema, the Netherlands.

We report an outbreak of infections with a novel protoparvovirus on several commercial farms in the Netherlands and associations with a clinical syndrome in pigs characterized by exophthalmos and erythema. Evidence of involvement of this virus, highly similar to a vulpine parvovirus, was substantiated by long-read sequencing and in situ hybridization techniques.

Persistent Anemia in an HIV Patient: The Role of Parvovirus B19.

Parvovirus B19 (B19V) is a common single-stranded DNA virus that infects humans and is its only host. Although infection is often asymptomatic, B19V can cause significant hematologic disease due to its tropism for erythroid progenitor cells. Immunocompromised individuals, including those with human immunodeficiency virus (HIV), are susceptible to persistent infection and may develop pure red cell aplasia (PRCA) and chronic anemia. Since B19V infection is not routinely monitored in the United States, and its symptoms can overlap with other causes of anemia in HIV, diagnosis may be overlooked. E…

Parvovirus B19 Infection as a Potential Trigger for Systemic Diseases.

It is crucial to perform direct methods like polymerase chain reaction (PCR) beyond serology to diagnose an active viral infection that can trigger a cutaneous/systemic disease, such as fibromyalgia. Indeed, measuring the viral load in tissues, plasma, and serum are diagnostic tools that permit the follow-up of an active infection.

Parvovirus B19 Infection: A Vasculitis Masquerade in an Elderly Patient.

BACKGROUND The profound ability of viral infections to convincingly mimic vasculitis, thereby pathologically influencing vessels of any caliber, is undeniably significant. Notably, adult patients with B19V infection frequently experience joint pain and cutaneous eruptions, which are ostensibly immune responses to the infection and necessitate careful differentiation from autoimmunity. Conversely, vasculitis syndromes represent an amalgamation of diseases characterized by vascular inflammation, predominantly classified based on the impacted vessels' size and location. Although the expedited dia

The seroprevalence of parvovirus B19 infection in pregnant women in Sudan.

Parvovirus B19 (B19V) infection during pregnancy may have serious consequences like fetal anaemia, hydrops fetalis, and fetal loss. Since epidemiological data on B19V infection are generally lacking in Sudan, the current study aimed to determine the seroprevalence of B19V in Sudanese pregnant women. Five hundred women, attending antenatal clinics in Khartoum state between November 2008 and March 2009, were enrolled and screened for B19V IgG and IgM antibodies by enzyme immunoassays. The study revealed a B19V IgG seroprevalence of 61·4%, with one subject positive for IgM. B19V DNA was not detec…

Occurrence of human bocaviruses and parvovirus 4 in solid tissues.

Human bocaviruses 1-4 (HBoV1-4) and parvovirus 4 (PARV4) are recently discovered human parvoviruses. HBoV1 is associated with respiratory infections of young children, while HBoV2-4 are enteric viruses. The clinical manifestations of PARV4 remain unknown. The objective of this study was to determine whether the DNAs of HBoV1-4 and PARV4 persist in human tissues long after primary infection. Biopsies of tonsillar tissue, skin, and synovia were examined for HBoV1-4 DNA and PARV4 DNA by PCR. Serum samples from the tissue donors were assayed for HBoV1 and PARV4 IgG and IgM antibodies. To obtain sp…

[An exceptional cause of acute hepatitis in an adult: parvovirus B19].

There are multiple causes of hepatitis. The most frequent etiologies are viral, usually hepatitis A, B and C viruses. However, other, non-hepatotropic viruses can cause this disease, including parvovirus B19. We present a case of acute hepatitis due to parvovirus B19, as well as a review of the epidemiological, clinical, diagnostic and therapeutic features of this entity.
