Determining the effects of paternal obesity on sperm chromatin at histone H3 lysine 4 tri-methylation in relation to the placental transcriptome and cellular composition.

Paternal obesity has been implicated in adult-onset metabolic disease in offspring. However, the molecular mechanisms driving these paternal effects and the developmental processes involved remain poorly understood. One underexplored possibility is the role of paternally induced effects on placenta development and function. To address this, we investigated paternal high-fat diet-induced obesity in relation to sperm histone H3 lysine 4 tri-methylation signatures, the placenta transcriptome, and cellular composition. C57BL6/J male mice were fed either a control or high-fat diet for 10 weeks begi

The role of paternal obesity in fetal development, pregnancy complications, and childhood outcomes: a systematic review.

The Developmental Origins of Health and Disease theory proposes that environmental influences during early life have long-term effects on health outcomes. Although maternal factors have been extensively studied, the impact of paternal obesity on fetal development and pregnancy outcomes has received limited attention. This systematic review aimed to evaluate the effects of paternal obesity on fetal development and pregnancy complications within the DOHaD framework. English-language studies published between 2020 and 2025 were searched in PubMed, Scopus, Web of Science, and the Cochrane Library

Investigating the significance of the transgenerational impact of high and repeated doses of ivermectin: Effects on paternal testis histopathology, pups' development, and sexual behavior.

Paternal exposure to environmental challenges is critical for the offspring's future health, and the transmission of acquired traits through generations increases the risk of offspring developing diseases. Ivermectin (IVM) is widely used in veterinary and human medicine to treat parasitosis. Our previous studies showed that IVM acute administration induced disorganization of the germinal epithelium and could cause damage to sperm production. Thus, this study investigated the effects of paternal exposure to repeated high ivermectin doses on paternal testis histology. After mating, their pups' d

The impact of maternal and paternal birth weights on infant birth weights: the Japan environment and children's study.

This study aimed to evaluate the association between parental and infant birth weights in Japan. In total, 37,504 pregnant Japanese women and their partners were included in this birth cohort study. A multinomial logistic regression model was used to evaluate the associations of parental birth weights with small-for-gestational-age (SGA) or large-for-gestational-age (LGA) infants. Associations between parental birth weight and low birth weight (LBW) infants or macrosomia were also examined, and linear associations between parental birth weight and SGA or LGA were found. The adjusted odds ratio

Associations of maternal weight status with the risk of offspring atopic dermatitis and wheezing by 1 year of age.

BACKGROUND: Maternal pre-pregnancy body mass index (BMI) and gestational weight gain (GWG) have been linked to offspring allergic disorders. However, associations observed in previous studies were inconsistent and might be confounded by unmeasured familial factors. We aimed to examine the associations of maternal weight with offspring allergic disorders by using paternal BMI as a negative control exposure. METHODS: We included the data of 10,522 children from the Born in Guangzhou Cohort Study, 2012-2017. Data on maternal weight were obtained from questionnaires and obstetric records, and pat

Does early paternal involvement predict offspring developmental diagnoses?

BACKGROUND: A long line of research has illustrated that fathers play an important role in the development of their children. Few studies, however, have examined the impact of paternal involvement at the earliest stages of life on developmental diagnoses in childhood. AIMS: The present study extends this line of research by exploring the possibility that paternal involvement prenatally, postnatally, and at the time of birth may influence offspring risk for various diagnoses in childhood. STUDY DESIGN: A quasi-experimental, propensity score matching design was used to create treatment and con

Parental psychological distress during pregnancy and childhood cardiovascular development. The Generation R Study.

BACKGROUND: Maternal psychological distress during pregnancy might lead to higher fetal cortisol exposure, which subsequently leads to fetal cardiovascular developmental adaptations and cardiovascular dysfunction in later life. AIMS: We examined whether maternal and paternal psychological distress was associated with the cardiovascular outcome measurements in school age children. STUDY DESIGN AND SUBJECTS: In a population-based prospective cohort study among 4831 children, we assessed maternal and paternal psychological distress during pregnancy by questionnaire, using the Brief Symptom Inve

Maternal and paternal exercise regulate offspring metabolic health and beta cell phenotype.

OBJECTIVE: Poor maternal and paternal environments increase the risk for obesity and diabetes in offspring, whereas maternal and paternal exercise in mice can improve offspring metabolic health. We determined the effects of combined maternal and paternal exercise on offspring health and the effects of parental exercise on offspring pancreas phenotype, a major tissue regulating glucose homeostasis. RESEARCH DESIGN AND METHODS: Breeders were high fat fed and housed±running wheels before breeding (males) and before and during gestation (females). Offspring groups were: both parents sedentary (Se

Adrenocortical Tumors in Children With Constitutive Chromosome 11p15 Paternal Uniparental Disomy: Implications for Diagnosis and Treatment.

Pediatric adrenocortical tumors (ACTs) are rare and heterogeneous. Approximately 50% of children with ACT carry a germline TP53 variant; however, the genetic underpinning of remaining cases has not been elucidated. In patients having germline TP53 variants, loss of maternal chromosome 11 and duplication of the paternal copy [paternal uniparental disomy, (UPD)] occurs early in tumorigenesis and explains the overexpression of IGF2, the hallmark of pediatric ACT. Beckwith-Wiedemann syndrome (BWS) is also associated with overexpression of IGF2 due to disruption of the 11p15 loci, including segment

Childhood experiences of parenting and age at menarche, age at menopause and duration of reproductive lifespan: Evidence from the English Longitudinal Study of Ageing.

OBJECTIVES: The parent-child relationship is critical for human development, yet little is known about its association with offsprings' reproductive health outside the context of abuse and neglect. We investigated whether childhood experiences of poor-quality parenting (characterized as decreased parental care and increased parental overprotection) are associated with women's reproductive timing and lifespan. STUDY DESIGN: Observational study of 2383 women aged 55-89 years in 2007 from the English Longitudinal Study of Ageing (ELSA). Multinomial logistic regression models were estimated. MAI

Fathers' Sensitivity in Infancy and Externalizing Problems in Middle Childhood: The Role of Coparenting.

The present study examined the role of father sensitivity and couple coparenting quality in the first 2 years of life in relation to the development of externalizing behavior problems in middle childhood, focusing on the unique role of fathers. In this study, 125 mothers, fathers, and their first-born children were followed from 8 months to age 7 years. Paternal sensitivity was rated when infants were 8 and 24 months old. Fathers were videotaped at home playing, feeding, and changing their 8-month-old infants' clothes. They also were videotaped in a lab playing with their 24-month-olds and sol

Parental origin of the supernumerary chromosome in trisomy 18.

The parental origin of an extra chromosome in Edwards syndrome has been investigated in 23 families by the combination of the VNTR probe pERT25, two microsatellite polymorphisms for D18S34 and D18S40, and several two-allele polymorphisms. Of the 23 cases, 22 were informative, with 17 (77%) being maternal and 5 (23%) paternal in origin. These results support the previous investigations, suggesting that trisomy 18 is predominantly of maternal origin, although a higher rate of paternally derived cases was observed than previously reported. A significant increase in maternal age was found to be as

Mothers' and fathers' social support trajectories and children's prosocial development: A dyadic perspective.

The current study examined mothers' and fathers' dyadic trajectories of perceived social support and their associations with children's prosociality. Data were drawn from 4,329 children (52% male, 48% female; 44% Black, 22% Hispanic, 17% White) in a prospective birth cohort study of low-income families. Repeated-measures latent class analysis identified three trajectories of social support at the level of the mother-father dyad from birth to age five: "High and Concordant" (64%), "Paternal-Advantaged, Declining Maternal" (20%), and "Maternal-Advantaged" (16%) social support. Child prosocial be

Parental smoking before childbirth and multiple sclerosis onset in their child: a case-control study.

BACKGROUND: Multiple sclerosis (MS) pathogenesis involves both environmental and genetic factors. Any stage of life, including the prenatal and perinatal years, may be impacted by environmental factors linked to the onset and progression of MS. It is essential to assess these environmental elements in order to avoid the start of illness. However, there is a lack of research on the effects of parental smoking before childbirth. OBJECTIVE: This study aimed to investigate the association between MS and parental cigarette consumption before childbirth. DESIGN AND METHODS: We conducted a case-con

Association of Cumulative Paternal Early Life Stress With White Matter Maturation in Newborns.

IMPORTANCE: Early life stress (ELS) has been shown to affect brain development and health outcomes. Recent animal studies have linked paternal early stress exposures with next-generation outcomes. Epigenetic inheritance through the male germline has been suggested to be one of the mechanisms. OBJECTIVES: To test whether paternal ELS, as measured using the Trauma and Distress Scale, is associated with neonate brain development. DESIGN, SETTING, AND PARTICIPANTS: This cohort study included data from participants from the prospective 2-generation FinnBrain Birth Cohort, which was collected from
