Spg20-/- mice reveal multimodal functions for Troyer syndrome protein spartin in lipid droplet maintenance, cytokinesis and BMP signaling.

Hereditary spastic paraplegias (HSPs; SPG1-48) are inherited neurological disorders characterized by lower extremity spasticity and weakness. Loss-of-function mutations in the SPG20 gene encoding spartin cause autosomal recessive Troyer syndrome (SPG20), which has additional features of short stature, cognitive deficits and distal amyotrophy. To identify cellular impairments underlying Troyer syndrome, we generated Spg20-/- mice, which exhibit progressive gait defects. Although gross central nervous system pathology appeared largely normal, cerebral cortical neurons cultured from neonatal Spg2

Plin2 Coordinates Immune and Metabolic Reprogramming in Lacrimal Gland Aging.

PURPOSE: Aging impairs lacrimal gland function, contributing to dry eye syndrome and reduced quality of life. This study aimed to identify core molecular regulators of lacrimal gland aging and delineate their roles in immune imbalance and metabolic dysfunction. METHODS: Bulk transcriptomic profiling and single-cell RNA sequencing (scRNA-seq) were performed on lacrimal glands from young and aged C57BL/6 mice. Differentially expressed genes (DEGs) were screened, followed by machine learning-based feature selection using least absolute shrinkage and selection operator regression and support vect

Apoptosis inhibitor of macrophage (AIM) diminishes lipid droplet-coating proteins leading to lipolysis in adipocytes.

Under fasting conditions, triacylglycerol in adipose tissue undergoes lipolysis to supply fatty acids as energy substrates. Such lipolysis is regulated by hormones, which activate lipases via stimulation of specific signalling cascades. We previously showed that macrophage-derived soluble protein, AIM induces obesity-associated lipolysis, triggering chronic inflammation in fat tissue which causes insulin resistance. However, the mechanism of how AIM mediates lipolysis remains unknown. Here we show that AIM induces lipolysis in a manner distinct from that of hormone-dependent lipolysis, without

An immunohistochemical approach to differentiate hepatic lipidosis from hepatic phospholipidosis in rats.

Hepatocellular vacuolation can be a diagnostic challenge since cytoplasmic accumulations of various substances (lipid, water, phospholipids, glycogen, and plasma) can have a similar morphology. Cytoplasmic accumulation of phospholipids following administration of cationic amphiphilic drugs (CAD) can be particularly difficult to differentiate from nonphosphorylated lipid accumulations at the light microscopic level. Histochemical methods (Sudan Black, Oil Red-O, Nile Blue, etc.) can be used to identify both nonphosphorylated and/or phosphorylated lipid accumulations, but these techniques requir

Regulation of ADRP expression by long-chain polyunsaturated fatty acids in BeWo cells, a human placental choriocarcinoma cell line.

Transplacental transfer of maternal fatty acids is critical for fetal growth and development. In the placenta, a preferential uptake of fatty acids toward long-chain polyunsaturated fatty acids (LCPUFAs) has been demonstrated. Adipose differentiation-related protein (ADRP) is a lipid droplet-associated protein that has been ascribed a role in cellular fatty acid uptake and storage. However, its role in placenta is not known. We demonstrate that ADRP mRNA and protein are regulated by fatty acids in a human placental choriocarcinoma cell line (BeWo) and in primary human trophoblasts. LCPUFAs of

Energy metabolism alteration and gene expression reprogramming in a cell model of high fat load non-alcoholic fatty liver disease.

BACKGROUND: Metabolic rewiring plays a crucial role in the energy metabolism of hepatocytes during steatosis. However, the precise alterations in energy metabolism remain unclear. The aim of this study was to investigate the effects of lipid exposure on cellular energy metabolism and gene expression in a cellular model of non-alcoholic fatty liver disease (NAFLD). METHODS AND RESULTS: We induced hepatocyte steatosis in the hepatocyte cell line Huh7 via treatment with high levels of palmitate and oleate for 24 h. We then analysed transcriptomics, proteomics, mitochondrial phenotypes, and cellul

TBC1D15 protects alcohol-induced liver injury in female mice through PLIN5-mediated mitochondrial and lipid droplet contacting.

OBJECTIVE: Alcohol-induced hepatic steatosis and mitochondrial dysfunction are progressive conditions contributing to the development of alcoholic liver disease (ALD), often leading to cirrhosis and hepatocellular carcinoma. TBC1D15, a Rab7 GTPase-activating protein (GAP), has been implicated in mitochondrial homeostasis, however, its role in ALD remains elusive. This study aimed to investigate the functional role of TBC1D15 in ALD and elucidate the underlying mechanisms. METHODS: Female TBC1D15flox/flox mice and hepatocyte-specific overexpression of TBC1D15 mice were fed a Lieber-DeCarli eth

Delayed liver regeneration after partial hepatectomy in adipose differentiation related protein-null mice.

BACKGROUND & AIMS: Adult hepatocytes undergo cell cycle progression and proliferation in response to partial hepatectomy (PH). Transient lipid accumulation within hepatocytes preceding the peak proliferative phase is a characteristic feature of regenerating livers. However, the molecular mediators and mechanisms responsible for lipid accumulation in regenerating livers are not well understood. Adipose differentiation related protein (ADRP; Plin2) regulates hepatic triglyceride storage and Plin2-deficient (Plin2(-/-)) mice have significantly reduced triglyceride (TG) content in the liver. We so

Is placental tissue protein 17b/TIP47 a new factor in cervical cancer genesis?

We identified novel members of the placental tissue protein 17 (PP17) protein family which consists of different-size variants of PP17; cDNAs of PP17a and PP17b variants have also been cloned and sequence analyzed. By Western-blot analysis in cervical carcinoma tissues we found overexpression of PP17b. Compared to healthy controls a mean five-fold increase in the amount of PP17b was also detected in the sera of untreated cervical carcinoma patients, which declined after radical operations. In our recent findings, in sera of inoperable cervical carcinoma patients, we also found elevated PP17b l

METTL3 Promotes Lipid Deposition and Pulmonary Fibrosis by Destabilizing PLIN2 in a m6A-Dependent Manner.

Idiopathic pulmonary fibrosis (IPF) is a lethal lung disorder that is associated with aberrant lipid metabolism. N6-methyladenosine (m6A) methylation is involved in IPF progression. However, whether METTL3, a m6A methyltransferase, modulates lipid metabolism in IPF remains unknown. In this study, we aimed to investigate the effects of METTL3 on fibrosis and lipid accumulation and identify the molecular mechanism. PF mouse model was generated by bleomycin. WI-38 cells were stimulated with TGF-β1 to mimic the IPF-caused damage. Lipids were observed using Nile Red staining and marker detection. F

Downregulation of Perilipin1 by the Immune Deficiency Pathway Leads to Lipid Droplet Reconfiguration and Adaptation to Bacterial Infection in Drosophila.

Lipid droplets (LDs), the highly dynamic intracellular organelles, are critical for lipid metabolism. Dynamic alterations in the configurations and functions of LDs during innate immune responses to bacterial infections and the underlying mechanisms, however, remain largely unknown. In this study, we trace the time-course morphology of LDs in fat bodies of Drosophila after transient bacterial infection. Detailed analysis shows that perilipin1 (plin1), a core gene involved in the regulation of LDs, is suppressed by the immune deficiency signaling, one major innate immune pathway in Drosophila D

An ethanolic extract of Artemisia scoparia inhibits lipolysis in vivo and has antilipolytic effects on murine adipocytes in vitro.

An ethanolic extract of Artemisia scoparia (SCO) has metabolically favorable effects on adipocyte development and function in vitro and in vivo. In diet-induced obese mice, SCO supplementation significantly reduced fasting glucose and insulin levels. Given the importance of adipocyte lipolysis in metabolic health, we hypothesized that SCO modulates lipolysis in vitro and in vivo. Free fatty acids and glycerol were measured in the sera of mice fed a high-fat diet with or without SCO supplementation. In cultured 3T3-L1 adipocytes, the effects of SCO on lipolysis were assessed by measuring glycer

Modulation of lipid droplets by Mycobacterium leprae in Schwann cells: a putative mechanism for host lipid acquisition and bacterial survival in phagosomes.

The predilection of Mycobacterium leprae (ML) for Schwann cells (SCs) leads to peripheral neuropathy, a major concern in leprosy. Highly infected SCs in lepromatous leprosy nerves show a foamy, lipid-laden appearance; but the origin and nature of these lipids, as well as their role in leprosy, have remained unclear. The data presented show that ML has a pronounced effect on host-cell lipid homeostasis through regulation of lipid droplet (lipid bodies, LD) biogenesis and intracellular distribution. Electron microscopy and immunohistochemical analysis of lepromatous leprosy nerves for adipose di

Astrocyte-Associated ABCA1/APOE Responses Accompany Hippocampal Lipid Remodeling and Ectopic Neuronal Accumulation During Epileptogenesis.

Temporal lobe epilepsy (TLE) is characterized by progressive hippocampal remodeling, including reactive astrogliosis and aberrant neuronal positioning. Although lipid disturbances are implicated in epileptogenesis, how astrocyte-associated lipid responses relate to hippocampal remodeling remains poorly understood. Here, human TLE and kainic acid (KA)-treated mouse transcriptomic datasets identified ATP-binding cassette transporter A1 (ABCA1)/Abca1 as a nominal astrocyte-associated candidate, while KA- and pilocarpine-induced status epilepticus (SE) elicited astrocyte-associated ABCA1/apolipopr

Mature Adipocyte-Derived Extracellular Vesicles Activate Hepatic Stellate Cells and Promote Liver Fibrosis via Inflammation.

BACKGROUND AND AIMS: Extracellular vesicles (EVs) play a crucial role in intercellular communication, particularly in metabolic regulation and liver disease. This study aimed to investigate the differential effects of EVs derived from pre-adipocytes (PA) and mature adipocytes (MA) on hepatic stellate cell (HSC) activation in vitro and their role in promoting liver fibrosis in high-fat diet (HFD)-fed mice. METHODS: EVs were isolated from PA and MA following transdifferentiation, which was confirmed by assessing adipogenic markers (PPAR-γ, perilipin, adiponectin, leptin, and Glut4). EVs were ch

Inhibition of adipocyte-macrophage interaction via CSF1-CSF1R by curcumin.

Adipocytes (Ad), macrophages (Mac), and lipopolysaccharide (LPS) in the adipose tissue (AT) of individuals with obesity may jointly contribute to obesity-induced pathogenesis. This study aims to examine the roles of adipocyte-macrophage interaction (AMI) and LPS in obesity-induced pathogenesis, as well as the effects of curcumin (Cur) on these interactions. An indirect co-culture model using conditioned media (CM) from 3T3-L1 Ad and J774A.1 Mac, with LPS, was used. AMI in AT and its association with systemic insulin resistance (IR) were further evaluated in a high-fat diet (HFD)-induced obese

PLIN5 Protects Against Ang II-Induced Podocyte Lipotoxicity by Interacting With FKBP8 and Preserving Lipid Droplet-Mitochondria Contact.

Chronic kidney disease (CKD) remains a major global health challenge. Angiotensin II (Ang II)-induced lipotoxicity is an important contributor to podocyte injury. Perilipin 5 (PLIN5) is a lipid droplet-associated protein that helps maintain cellular metabolic homeostasis. However, how PLIN5 protects podocytes from lipotoxic stress remains incompletely understood. In this study, we generated podocyte-specific PLIN5 knockout mice using the Cre-loxP system and induced PLIN5 overexpression in vivo and in vitro. We found that Ang II markedly downregulated PLIN5 expression in podocytes both in vivo

The fungal biocontrol agent Beauveria bassiana expresses a HsbA domain-containing protein to postpone the rapid cuticle penetration outwards from the insect cadavers.

The insect biocontrol fungi (IBF) 'hidden' within the dead insect bodies come out of the insect cadavers for dissemination until the suitable environment conditions arising. However, details of the IBF balancing fungal cells 'hidden' within the cadavers and penetration of the cuticle outwards for growth remain limited. Here, a HsbA domain-containing protein, BbHsbA, was found to express during the later stages of insect colonization in an IBF, Beauveria bassiana, which distributed in cytoplasm and cell wall. BbHsbA disruption did not significantly affect fungal virulence, while the mutant pene

SLC25A1-targeted gene therapy attenuates liver fibrosis through NEDD4-mediated ubiquitination and degradation of PPARγ.

Restoring lipid droplet (LD) content has been reported to reverse hepatic stellate cell (HSC) activation during liver fibrosis. Although the mitochondrial citrate carrier SLC25A1 is known to drive metabolic reprogramming in cancer and steatohepatitis, its specific role in regulating LDs remains unclear. This study aimed to elucidate the role of SLC25A1 in controlling LD homeostasis during HSC activation and to determine whether therapeutic inhibition of SLC25A1 could ameliorate liver fibrosis by restoring peroxisome proliferator-activated receptor-γ (PPARγ)-dependent lipid storage. Fibro-adipogenic progenitor cells from murine SMA muscles are intrinsically adipogenic.

Spinal muscular atrophy (SMA) is a neurodegenerative disorder caused by mutations in the SMN1 gene. Although classically viewed as a neurogenic disease, SMA patients exhibit poor skeletal muscle regeneration and increased fatty-fibrotic infiltration. Fibro-adipogenic progenitors (FAPs) are mesenchymal precursor cells that contribute to muscle remodeling and underlie fat and fibrosis formation. Because FAPs transiently express Smn1 during regeneration, FAPs were examined in muscles from adult C/C SMA and control mice to determine if reduced Smn activity altered their properties. We performed a
