Intracellular activity of fosfomycin against two distinct enteropathogenic bacteria, Salmonella enterica and Listeria monocytogenes, alive inside host cells.

We studied the effect of fosfomycin (FOM) on the intracellular growth of two different facultative intracellular bacteria, Salmonella enterica serovar Typhimurium and Listeria monocytogenes, in an enterocyte-like cell line, Caco-(2). These bacteria replicate in different compartments within the host cells; Salmonella serovar Typhimurium grow inside phagosomes, whereas L. monocytogenes escape the phagosomal environment and multiply in the cytosol of the host cells. At concentrations equal to 0.25 of the MIC and 4 times the MIC, respectively, FOM effectively decreased the number of intracellular…

Deciphering the relationship between sarcopenia and aging: A combined text mining and bioinformatics approach.

AIM: Sarcopenia is recognized as an age-related muscle disease, but there has been no comprehensive analysis of what is different between normal aging and sarcopenia, with an awareness of the worldwide research to date. Therefore, in this study, we used text mining of PubMed articles on sarcopenia and focused our bioinformatics analysis on the items that have been identified. METHODS: This study compared gene-level changes in sarcopenia and normal aging to identify sarcopenia-specific gene changes using high-throughput sequencing data. In particular, text mining analysis was used to identify

PknG Protein of Mycobacterium tuberculosis Targets RGDI-1 to Regulate Rab7l1 GTPase Activity.

The protein kinase G (PknG) protein of Mycobacterium tuberculosis is known to disrupt phagosome-lysosome (P-L) fusion, enabling the bacteria to persist within the host. In our previous study, we demonstrated that PknG inhibits GTPase activity of Rab7l1 by interacting with its inactive form (Rab7l1-GDP) to block the Rab7l1-GDP/GTP transition. As a result, the active Rab7l1 protein fails to localize onto the phagosome, which prevents recruitment of downstream P-L fusion markers like Rab7l1, EEA1, LAMP1, and LAMP2 to the phagosome, leading to inhibition of P-L fusion. In this study, we show that

Host-Pathogen Interactions and Correlated Factors That Are Affected in Replicative-Aged Cryptococcus neoformans.

Cryptococcus neoformans is a facultative intracellular fungal pathogen. Ten-generation-old (10GEN) C. neoformans cells are more resistant to phagocytosis and killing by macrophages than younger daughter cells. However, mechanisms that mediate this resistance and intracellular parasitism are poorly understood. Here, we identified important factors for the intracellular survival of 10GEN C. neoformans, such as urease activity, capsule synthesis, and DNA content using flow cytometry and fluorescent microscopy techniques. The real-time visualization of time-lapse imaging was applied to determine t

Manipulation of Host Cell Organelles by Intracellular Pathogens.

In this article, we explore the unique adaptations of intracellular bacterial pathogens that manipulate conserved cellular pathways, organelles, and cargo to convert the phagosome into a pathogen-containing vacuole (PCV). The phagosome is a degradative organelle that rapidly acidifies as it delivers cargo to the lysosome to destroy microbes and cellular debris. However, to avoid this fate, intracellular bacterial pathogens hijack the key molecular modulators of intracellular traffic: small GTPases, phospholipids, SNAREs, and their associated effectors. Following uptake, pathogens that reside i

Assembly of NADPH oxidase in human neutrophils is modulated by the opacity-associated protein expression State of Neisseria gonorrhoeae.

Neisseria gonorrhoeae (the gonococcus, Gc) triggers a potent inflammatory response and recruitment of neutrophils to the site of infection. … utrophil membranes following infection with OpaD+ Gc. gp91(phox) and p22(phox) were recruited to Gc phagosomes regardless of bacterial Opa expression. These results suggest that Opa- Gc interferes with the recruitment of neutrophil NADPH oxidase cytoplasmic subunits to membranes, in particular, the p47(phox) "organizing" subunit, to prevent assembly of the h …

Exploring the role of the CTL epitope region of listeriolysin O in the pathogenesis of Listeria monocytogenes.

Listeria monocytogenes is a facultative intracellular bacterial pathogen responsible for severe opportunistic infections in humans and animals. The secreted cholesterol-dependent cytolysin, listeriolysin O (LLO), mediates phagosomal escape and allows bacterial growth in the cytosol of infected cells. In order to identify new LLO determinants participating in bacterial pathogenesis, this study focused on a major target of LLO proteolytic cleavage in vitro, the CTL epitope region (residues 91-99). Mutations were generated by site-directed mutagenesis in the epitope or in the two clusters of posi

Localization of D-amino acid oxidase on the cell surface of human polymorphonuclear leukocytes.

The ultrastructural localization of D-amino acid oxidase (DAO) was studied cytochemically by detecting sites of hydrogen peroxide production in human polymorphonuclear leukocytes (PMNs). Reaction product, which forms when cerous ions react with H2O2 to form an electron-dense precipitate, was demonstrated on the cell surface and within the phagosomes of phagocytically stimulated cells when D-amino acids were provided as substrate. Resting cells showed only slight activity. The competitive inhibitor D,L-2-hydroxybutyrate greatly reduced the D-amino acid-stimulated reaction while KCN did not. The…

Malignant granular cell tumor. A clinico-pathologic and ultrastructural study of a case.

The clinical and light- and electron-microscopic features of a malignant granular cell tumor in a 64-year-old man are reported. The ultrastructural appearance, with abundance of cytoplasmic "phagosomes", may help in the differential diagnosis. The light-microscopic findings and some ultrastructural observations support the suggestion that the tumor originated from peripheral nerves, and that a close relationship exists between malignant granular cell tumors and malignant Schwannoma.

Dissociation of innate immune responses in microglia infected with Listeria monocytogenes.

Microglia, the innate immune cells of the brain, plays a central role in cerebral listeriosis. Here, we present evidence that microglia cont … croglia to induce TNF-regulated responses and to repress the late response. Isolation of microglial phagosomes revealed a phagosomal environment unable to destroy Listeria. Microglial phagosomes were also defective in several signaling and trafficking components reported as relevant for Listeria innate immune responses. This transcriptional strategy in micro …

Iron uptake and virulence in Histoplasma capsulatum.

Histoplasma capsulatum (Hc) is the causative organism of a spectrum of disease affecting both the immunocompetent and the immunocompromised host. Hc is a dimporhic fungus that converts from conidia to the pathogenic yeast phase after entry into the mammalian host. Despite rapid ingestion by macrophages, it survives intracellularly within the macrophage. The intracellular survival strategy of Hc yeasts focuses on regulating the phagosomal compartment by modulating the intraphagosomal pH to 6.5. As an intracellular pathogen of MΦ, Hc obtains iron from Fe-transferrin, ferritin, or both, via the p

Phagocytic podosomes enable efficient uptake of Candida auris by primary human macrophages.

The yeast Candida auris is an emerging pathogen. Understanding the molecular mechanisms of its uptake and processing by immune cells is thus critical for counteracting the spread of respective infections. We show that the phagocytosis of C. auris cells by primary human macrophages involves the formation of dot-like F-actin-rich structures at C. auris-containing phagosomes that we characterize as phagocytic podosomes. We analyze the composition, architecture, and dynamics of these structures, showing that they constitute a specific adaptation of the phagocytic actin network. The disruption of p…

Electron-microscopic observation of mouse spleen tissue infected with Orientia tsutsugamushi isolated from Shandong, China.

Low-virulent Orientia tsutsugamushi (Ot) were successfully isolated from scrub typhus patients in Shandong, China, and the isolates were similar to the Kawasaki type identified by nested polymerase chain reaction (PCR). To identify the morphological characterization of the low-virulent Ot, and elucidate the pathological changes on host cells, mouse spleen tissue infected with the Ot isolated from Shandong was used for the ultrastructural study. Transmission electron microscopy showed that the Ot parasitized in the spleen were different in size, shape and electron density and many significant c

Cytosolic entry controls CD8+-T-cell potency during bacterial infection.

Interaction with host immunoreceptors during microbial infection directly impacts the magnitude of the ensuing innate immune response. How these signals affect the quality of the adaptive T-cell response remains poorly understood. Utilizing an engineered strain of the intracellular pathogen Listeria monocytogenes that infects cells but fails to escape from the phagosome, we demonstrate the induction of long-lived memory T cells that are capable of secondary expansion and effector function but are incapable of providing protective immunity. We demonstrate that microbial invasion of the cytosol

[The interaction of the causative agent of melioidosis with the host's alveolar macrophages].

Studies were carried out on guinea pigs and albino rats, intranasally infected with P. pseudomallei C-141. The cells of bronchovesicular exudate were obtained from animals 1, 4 and 24 hours after infection. Electron microscopy was applied to study the process of interaction of the agent and alveolar macrophages. Bacteria were shown to form a capsule which permitted avoiding phagocytosis, when entering the host respiratory system. Microbes that failed to form a capsule were absorbed by macrophages and enclosed in a phagosome. Then some bacteria were destroyed by the lysosomal enzymes, the other

Mechanism of the intracellular killing and modulation of antibiotic susceptibility of Listeria monocytogenes in THP-1 macrophages activated by gamma interferon.

Listeria monocytogenes, a facultative intracellular pathogen, readily enters cells and multiplies in the cytosol after escaping from phagosomal vacuoles. Macrophages exposed to gamma interferon, one of the main cellular host defenses against Listeria, become nonpermissive for bacterial growth while containing Listeria in the phagosomes. Using the human myelomonocytic cell line THP-1, we show that the combination of L-monomethyl arginine and catalase restores bacterial growth without affecting the phagosomal containment of Listeria. A previous report (B. Scorneaux, Y. Ouadrhiri, G. Anzalone, an…

Ultrastructure of Calymmatobacterium granulomatis in Lesions of granuloma inguinale.

Biopsy specimens from the lesions of 14 patients with granuloma inguinale were examined by electron microscopy. Calymmatobacterium granulomatis, the presumed causative agent, was observed within the phagosomes of many macrophages, but only rarely in polymorphonuclear leukocytes. The morphology of the organisms was that of a gram-negative bacteria having a large capsule and no flagella. These morphologic observations are consistent with published findings suggesting that this organism is related antigenically to organisms of the genus Klebsiella.

The immunopathology of Mooren's ulcer.

In a 42-year-old man with bilateral severe Mooren's ulcers, results of immunologic studies were normal, with the exception of an in vitro blastogenic response of the patient's lymphocytes to corneal stroma. The histologic studies revealed vast numbers of vacuolated neutrophils with intracytoplasmic phagosomes in and about the ulcerating cornea. Although the progression of ulceration was initially unaltered by surgical and pharmacologic trials, systemic immunosuppression with methotrexate may have prevented ulceration of the lamellar tectonic graft.

Activation of compensatory pathways via Rac2 in the absence of the Cdc42 effector Wiskott-Aldrich syndrome protein in Dendritic cells.

There is extensive crosstalk between different Rho GTPases, including Cdc42, Rac1, and Rac2, and they can activate or inhibit the activity of each other. Dendritic cells express both Rac1 and Rac2. Due to posttranslational modification of lipid anchors, Rac1 localizes mainly to the plasma membrane whereas Rac2 localizes to the phagosomal membrane where it assembles the NADPH complex. Our recent study of primary immunodeficiency disease caused by mutations in the Cdc42 effector Wiskott-Aldrich syndrome protein (WASp) has shed light on the compensatory mechanisms between Rho GTPases and their ef
