The salivary apyrase of the blood-sucking sand fly Phlebotomus papatasi belongs to the novel Cimex family of apyrases.

Apyrases are enzymes that hydrolyze nucleotide di- and triphosphates to orthophosphate and mononucleotides. At least two families of enzymes … milar to that of the C. lectularius salivary apyrase isolated from a salivary gland cDNA library of Phlebotomus papatasi. Transfection of insect cells with the P. papatasi salivary gland apyrase cDNA resulted in the secretion of a Ca(2+)-dependent apyrase whose activity was indistinguishable from that in salivary homogenates of P. papatasi. Homologous sequenc …

[Evidence for the transmission of the causative agent of visceral leishmaniasis by the sandfly Phlebotomus (Adlerius) turanicus Artemiev, 1974 in Turkmenistan].

Four sandfly [correction of mosquito] species were identified in all foci of visceral leishmaniasis (VL) in Turkmenistan. There is evidence that three of them (Phlebotomus papatasi, P. caucasicus, P. andrejevi) cannot be carriers of VL agent in Turkmenistan. The carrier of Leishmania infantum in Turkmenistan is likely to be the zooanthropophilic sandfly [correction of mosquito] P. turanicus that prevails in the settlements and their vicinities in the desert foothill plains and in the burrows of foxes. The time of the carrier activity is May to September, and its peak size is observed in mid-Ju…

Genetic diversity of Phlebotomus perniciosus populations between insular and mainland regions in the leishmaniasis-endemic western Mediterranean area.

BACKGROUND: Phlebotomus perniciosus is the primary vector of Leishmania infantum in Spain, occurring in both continental and insular regions. This study investigates the genetic structure of P. perniciosus populations from Majorca (island) and Barcelona (mainland), two geographically close but ecologically distinct regions in the western Mediterranean. METHODS: Mitochondrial cytochrome c oxidase subunit I (COI) gene sequences were analyzed from 167 P. perniciosus specimens, including 100 morphologically identified field-collected specimens from Majorca and Barcelona, supplemented with referen…

Humoral and T-Cell-Mediated Immunity Against Phlebotomus perniciosus Salivary Proteins in Dogs from a Leishmaniosis-Endemic Area.

Compounds in sand fly saliva elicit specific immune responses that may play a role in the establishment of canine Leishmania infection. Although canine antibodies to anti-sand fly saliva antigens have been extensively studied, little is known about cellular immune responses against Phlebotomus perniciosus salivary proteins. This study aimed to explore humoral and T-cell-mediated immunity against P. perniciosus salivary proteins in dogs (n = 85) from Mallorca (Spain), a leishmaniosis-endemic area, and find correlations with demographic (age, sex, and breed) and parasite-specific immunological p…

Population structure analysis of Phlebotomus papatasi populations using transcriptome microsatellites: possible implications for leishmaniasis control and vaccine development.

BACKGROUND: Phlebotomus papatasi is considered the primary vector of Leishmania major parasites that cause zoonotic cutaneous leishmaniasis (ZCL) in the Middle East and North Africa. Phlebotomus papatasi populations have been studied extensively, revealing the existence of different genetic populations and subpopulations over its large distribution range. Genetic diversity and population structure analysis using transcriptome microsatellite markers is important to uncover the vector distribution dynamics, essential for controlling ZCL in endemic areas. METHODS: In this study, we investigated …

Impact of anti-sandfly saliva antibodies on biological aspects of Phlebotomus papatasi (Diptera: Psychodidae), vector of cutaneous leishmaniasis.

Sandflies are the main vectors of Leishmania parasites in tropical and subtropical areas. The immunization of vertebrate hosts with vector components through repeated bites may offer an alternative method for sandfly control. Aliquots of female Phlebotomus papatasi (Scopoli) (Diptera: Psychodidae) were weekly blood fed on 12 individual hamsters throughout 18 successive weeks. Significant biological and biochemical changes resulting from antibodies developed by immunized host sera against repeated biting were observed in sandfly females. Blood feeding and fertility rates of females significantl…

Comparative evaluation of salivary glands proteomes from wild Phlebotomus papatasi-proven vector of zoonotic cutaneous leishmaniasis in Iran.

BACKGROUND: Zoonotic Cutaneous Leishmaniasis is increasing in the world and Phlebotomus papatasi as a proven vector was considered in different aspects for disease control. Sandfly saliva contains proteins which provoke host immune system. These proteins are candidates for developing vaccines. OBJECTIVES: The main purpose of this research was comparing evaluation of salivary glands proteomes from wild P. papatasi. Extracting these proteins and purifying of original SP15 as inducer agent in vector salivary glands from endemic leishmaniasis foci were other objectives. METHODS: Adult sandflies …

Leishmania major survival in selective Phlebotomus papatasi sand fly vector requires a specific SCG-encoded lipophosphoglycan galactosylation pattern.

Phlebotomine sand flies that transmit the protozoan parasite Leishmania differ greatly in their ability to support different parasite species or strains in the laboratory: while some show considerable selectivity, others are more permissive. In "selective" sand flies, Leishmania binding and survival in the fly midgut typically depends upon the abundant promastigote surface adhesin lipophosphoglycan (LPG), which exhibits species- and strain-specific modifications of the dominant phosphoglycan (PG) repeat units. For the "selective" fly Phlebotomus papatasi PpapJ, side chain galactosyl-modificati…

Intraspecific variation within Phlebotomus sergenti Parrot (1917) (Diptera: Psychodidae) based on mtDNA sequences in Islamic Republic of Iran.

An intraspecific study on the morphological and molecular characteristics of Phlebotomus sergenti s.l., the main vector of Leishmania tropica, was performed on 28 Iranian populations from 11 provinces and a few samples from Greece, Morocco, Lebanon, Turkey, Pakistan, and Syria. Three morphotypes were identified as A, B and C, with some intermediate forms in the samples under investigation. Based on the number of setae and the width of basal lobe of coxite, differences between A and B morphotypes were highly significant. Excluding one unusual haplotype, sequence analysis of approximately 439 bp…

Enzyme polymorphism and genetic variability of laboratory populations of Phlebotomus papatasi, P. bergeroti, P. langeroni, and P. perniciosus.

Enzyme allomorph frequency and electrophoretic mobility were evaluated to clearly separate between morphologically similar sandfly species and to yield quantitative measures of their genetic similarity. Twelve enzyme systems were studied in laboratory colonies of Phlebotomus perniciosus, P. langeroni, P. papatasi, and P. bergeroti. Phlebotomus perniciosus and P. langeroni confirmed vectors of visceral leishmaniasis, were found to have fixed diagnostic allomorph differences at 3 enzyme loci (ICD-2, FUM, XDH). Phlebotomus papatasi, a known vector of cutaneous leishmaniasis and phleboviruses and …

Understanding sand fly sampling methods: sticky traps are attraction-based and not interceptive sampling tools of Phlebotomus orientalis.

BACKGROUND: Sticky traps are generally viewed as interceptive sand fly sampling methods; although no previous experimental evidence has supported this assumption. In this study, we tested this assumption experimentally for Phlebotomus orientalis, the principal vector of visceral leishmaniasis in East Africa, and propose an explanation for the highly male-biased collection of sticky traps. METHODS: A number of field experiments were carried out in March-June 2016-2019, in Gedarif state, eastern Sudan. In the first experiment, we compared numbers of P. orientalis caught on sticky traps made of …

First detection of Leishmania infantum DNA in Phlebotomus longicuspis Nitzulescu, 1930 from visceral leishmaniasis endemic focus in Algeria.

Two clinico-epidemiological forms of leishmaniasis caused by Leishmania infantum complex are endemic in Algeria: human visceral leishmaniasi … ies were morphologically identified: one species belonged to the Sergentomyia genus and nine to the Phlebotomus genus. L. infantum DNA was amplified in 1/169 (0.6%) dissected dead females, one Phlebotomus longicuspis. Our data support the Parrot's hypothesis raised in 1941 concerning the role of P. longicuspis in the transmission of L. infantum.

Naturally occurring culturable aerobic gut flora of adult Phlebotomus papatasi, vector of Leishmania major in the Old World.

BACKGROUND: Cutaneous leishmaniasis is a neglected, vector-borne parasitic disease and is responsible for persistent, often disfiguring lesions and other associated complications. Leishmania, causing zoonotic cutaneous leishmaniasis (ZCL) in the Old World are mainly transmitted by the predominant sand fly vector, Phlebotomus papatasi. To date, there is no efficient control measure or vaccine available for this widespread insect-borne infectious disease. METHODOLOGY/PRINCIPAL FINDINGS: A survey was carried out to study the abundance of different natural gut flora in P. papatasi, with the long-…

Epidemiologic relationship between Toscana virus infection and Leishmania infantum due to common exposure to Phlebotomus perniciosus sandfly vector.

Sand flies are recognised vectors of parasites in the genus Leishmania and a number of arthropod-borne viruses, in particular viruses within the genus Phlebovirus, family Bunyaviridae. In southern France, Toscana phlebovirus (TOSV) is recognized as a prominent cause of summer meningitis. Since Leishmania and TOSV have a common vector (Phlebotomus perniciosus), an epidemiologic link has been assumed for a long time. However, there is no scientific evidence of such a link between human leishmaniosis and phleboviral infections. To identify a possible link, we investigated the presence and distrib…

A simple micro-assay method for estimating blood meal size of the sand fly, Phlebotomus langeroni (Diptera: Psychodidae).

The accurate measurement of blood meal size in Phlebotomus langeroni, the potential vector of infantile visceral leishmaniasis in Egypt, is important to determine the number of parasites taken in fully engorged insects. A simple protein content micro-assay is introduced for that purpose. The accuracy of this method was confirmed by hemoglobin estimation method. Laboratory bred P. langeroni were fed artificially on defibrinated human blood and the fully engorged flies were carefully dissected on ice, within 1-10 min after feeding, since the time of dissection is critical. Serial concentrations …
