Purification and partial characterization of analogous 26-kDa rat submandibular and parotid gland integral membrane phosphoproteins that may have a role in exocytosis.

Rat submandibular and parotid gland exocytosis is primarily controlled by beta-adrenergic receptor stimulation. Although its precise role in the regulation of salivary gland exocytosis is not fully understood, protein phosphorylation, mediated by the activation of cAMP-dependent protein kinase, may be directly involved. Previous studies suggest that analogous 26-kDa integral membrane phosphoproteins may play a direct role in regulating exocytosis. Studies were here undertaken to purify and partially characterize both phosphoproteins. After endogenous phosphorylation with 32P, subcellular fract

Alafosfalin, a new inhibitor of cell wall biosynthesis: in vitro activity against urinary isolates in Japan and potentiation with beta-lactams.

A new phosphonopeptide, alafosfalin, was evaluated for in vitro antibacterial activity and for synergism with beta-lactams, using 475 Japanese clinical isolates from urinary tract infections. Alafosfalin was found to be highly active against Escherichia coli and moderately active against Serratia, Klebsiella, Enterobacter, and Citrobacter, but less active against gram-positive organisms than were beta-lactams such as cephazolin or ampicillin and inactive against indole-positive Proteus, Pseudomonas, and Acinetobacter. Potentiation with the two beta-lactams (fractional inhibitory concentration

Bacopa monnieri Attenuates Alzheimer's Pathology by Modulating RNA Metabolism, Cellular Stress Responses, and Rho GTPase Signaling: Phosphoproteomic Insights from a Cellular Model of Alzheimer's Disease.

Alzheimer's disease is a multifactorial disease characterized by proteinopathy caused by senile plaques and neurofibrillary tangles, leading to neuroinflammation and neuronal damage. Bacopa monnieri, with its extensive usage in the Indian traditional medicinal system, Ayurveda, holds promise in enhancing memory and improving neuronal functions. We performed quantitative phosphoproteomics analysis to understand the temporal dynamics of protein phosphorylation involved in Bacopa monnieri-mediated neuroprotection in the Alzheimer's disease model of IMR-32 cells. The quantitative phosphoproteomics

Phospho-Regulatory Network of the Right Open Reading Frame Kinase 1 (RIOK1), Its Functional Relevance, and Cancer Treatment Prospects.

Right open reading frame kinase 1 (RIOK1) is an atypical kinase involved in ribosome biogenesis, cell cycle progression, and chromosome organization. Its overexpression is linked to tumor progression, metastasis, and chemoresistance, while its absence alters protein phosphorylation across various biological processes. Although the oncogenic role of RIOK1 is recognized, its phospho-regulatory network and the functional relevance of its phosphorylation sites remain unknown. Here, we present the first large-scale phosphoproteomic analysis of RIOK1. Through a systematic assembly of 671 mass spectr

Phosphorylation-Mediated Molecular Pathway Changes in Human Pituitary Neuroendocrine Tumors Identified by Quantitative Phosphoproteomics.

To investigate the biological role of protein phosphorylation in human nonfunctional pituitary neuroendocrine tumors (NF-PitNETs), proteins extracted from NF-PitNET and control tissues were analyzed with tandem mass tag (TMT)-based quantitative proteomics coupled with TiO2 enrichment of phosphopeptides. A total of 595 differentially phosphorylated proteins (DPPs) with 1412 phosphosites were identified in NF-PitNETs compared to controls (p < 0.05). KEGG pathway network analysis of 595 DPPs identified nine statistically significant signaling pathways, including the spliceosome pathway, the RNA t

Phosphoproteomic Analysis of Protein Phosphorylation Networks in the Hypopharyngeal Gland of Honeybee Workers (Apis mellifera ligustica).

The hypopharyngeal gland (HG) in honeybee workers changes functions according to physiological age in the bee colony from producing royal jelly (RJ) in nurse bees to digestive enzymes in foragers. The same set of secretory cells expresses different genes or proteins to create these age-dependent changes; however, it is unknown precisely how the phosphorylation network regulates physiological differences across the development of the adult worker HG. We employed high-accuracy mass-spectrometry-based proteomics to survey phosphoproteome changes in the newly emerged, nurse, and forager bees. Over

Functional expression of the Na-K-2Cl cotransporter NKCC2 in mammalian cells fails to confirm the dominant-negative effect of the AF splice variant.

The renal bumetanide-sensitive Na-K-2Cl cotransporter (NKCC2) is the major salt transport pathway in the apical membrane of the mammalian thick ascending limb. It is differentially spliced and the three major variants (A, B, and F) differ in their localization and transport characteristics. Most knowledge about its regulation comes from experiments in Xenopus oocytes as NKCC2 proved difficult to functionally express in a mammalian system. Here we report the cloning and functional expression of untagged and unmodified versions of the major splice variants from ferret kidney (fNKCC2A, -B, and -F

Phosphorylation of IRS4 at YY111/112 promotes LUSC progression via PI3K/AKT pathway activation.

Lung squamous cell carcinoma (LUSC) lacks effective targeted therapies despite frequent hyperactivation of the PI3K/AKT pathway, and the contribution of adaptor proteins to this process remains poorly defined. Here, we identify Insulin Receptor Substrate 4 (IRS4) as a common substrate of Src kinase and the phosphatase RPTPα, and uncover a previously unrecognized, conserved phosphorylation site at YY111/112 within the IRS4 PH domain. Src-mediated phosphorylation at this site is essential for IRS4-driven proliferation, anchorage-independent growth, and invasiveness in LUSC cells. Mechanistically

Hierarchical phosphorylation of recombinant tau by the paired-helical filament-associated protein kinase is dependent on cyclic AMP-dependent protein kinase.

Immunoaffinity-purified paired helical filaments (PHFs) from Alzheimer's disease (AD) brain homogenates contain an associated protein kinase activity that is able to induce the phosphorylation of PHF proteins on addition of exogenous MgCl2 and ATP. PHF kinase activity is shown to be present in immunoaffinity-purified PHFs from both sporadic and familial AD, Down's syndrome, and Pick's disease but not from normal brain homogenates. Although initial studies failed to show that the kinase was able to induce the phosphorylation of tau, additional studies presented in this article show that only cy

Biofunctional peptides from milk proteins: mineral binding and cytomodulatory effects.

The protein fraction of milk contains many valuable components and biologically active substances. Moreover, milk proteins are precursors of many different biologically active peptides which are inactive within the sequence of the precursor protein but can be released by enzymatic proteolysis. Many milk protein-derived peptides, such as caseinophosphopeptides, reveal multi-functional bioactivities. Caseinophosphopeptides can form soluble organophosphate salts and may function as carriers for different minerals, especially calcium. Furthermore, they have been shown to exert cytomodulatory effec

Development of new multifunctional terpolymer sorbent for proteomics applications.

Determination of the availability of phases for specific separations is an important task achieved by a separation chemist. This becomes vital when the complex samples like biofluids are dealt with in proteome science. The work presented here involves the synthesis and application of terpolymeric sorbent with different functionalizations adopted for the selective enrichment of biomolecules of interest from biological fluids. Synthesis of terpolymer was carried out by the radical polymerization of monomers: methyl acrylate, acrylic acid and vinyl acetate with diethylene glycol dimethacrylate as

Cold Storage Disrupts the Proteome and Phosphoproteome Landscape in Rat Kidney Transplants.

BACKGROUND: Prolonged cold storage (CS) of kidneys results in poor long-term outcomes after transplantation (Tx). We reported previously that CS of rat kidneys for 18 h before transplant impaired proteasome function, disrupted protein homeostasis, and reduced graft function. The goal of the present study was to identify the renal proteins, including phosphoproteins, that are dysregulated by this CS injury. METHODS: Isolated donor Lewis rat kidneys were subject to 18 h CS and transplanted into recipient Lewis rats (CS + Tx). Autotransplantation (transplant with 0 h CS) or Sham (right nephrecto

MASS SPECTROMETRY-BASED MITOCHONDRIAL PROTEOMICS IN HUMAN OVARIAN CANCERS.

The prominent characteristics of mitochondria are highly dynamic and regulatory, which have crucial roles in cell metabolism, biosynthetic, senescence, apoptosis, and signaling pathways. Mitochondrial dysfunction might lead to multiple serious diseases, including cancer. Therefore, identification of mitochondrial proteins in cancer could provide a global view of tumorigenesis and progression. Mass spectrometry-based quantitative mitochondrial proteomics fulfils this task by enabling systems-wide, accurate, and quantitative analysis of mitochondrial protein abundance, and mitochondrial protein

RegPhos 2.0: an updated resource to explore protein kinase-substrate phosphorylation networks in mammals.

Protein phosphorylation catalyzed by kinases plays crucial roles in regulating a variety of intracellular processes. Owing to an increasing number of in vivo phosphorylation sites that have been identified by mass spectrometry (MS)-based proteomics, the RegPhos, available online at http://csb.cse.yzu.edu.tw/RegPhos2/, was developed to explore protein phosphorylation networks in human. In this update, we not only enhance the data content in human but also investigate kinase-substrate phosphorylation networks in mouse and rat. The experimentally validated phosphorylation sites as well as their c

Comprehensive genome-wide proteomic analysis of human placental tissue for the Chromosome-Centric Human Proteome Project.

As a starting point of the Chromosome-Centric Human Proteome Project (C-HPP), we established strategies of genome-wide proteomic analysis, including protein identification, quantitation of disease-specific proteins, and assessment of post-translational modifications, using paired human placental tissues from healthy and preeclampsia patients. This analysis resulted in identification of 4239 unique proteins with high confidence (two or more unique peptides with a false discovery rate less than 1%), covering 21% of approximately 20, 059 (Ensembl v69, Oct 2012) human proteins, among which 28 prot

Phosphorylation of serine 1106 in the catalytic domain of topoisomerase II alpha regulates enzymatic activity and drug sensitivity.

Topoisomerases alter DNA topology and are vital for the maintenance of genomic integrity. Topoisomerases I and II are also targets for widely used antitumor agents. We demonstrated previously that in the human leukemia cell line, HL-60, resistance to topoisomerase (topo) II-targeting drugs such as etoposide is associated with site-specific hypophosphorylation of topo II alpha. This effect can be mimicked in sensitive cells treated with the intracellular Ca(2+) chelator, 1,2-bis(2-aminophenoxy)ethane-N,N,N',N'-tetraacetic acid (BAPTA-AM). Here we identify Ser-1106 as a major phosphorylation sit

Effects of estradiol on calcium-specific protein phosphorylation in the rat corpus luteum.

Estradiol regulates progesterone synthesis by the corpus luteum of several species. The molecular mechanism(s) of the action of estradiol is unknown, but appears to be independent of exogenous gonadotropins and tissue cAMP. A role for Ca2+ in steroidogenesis has been implicated, and we demonstrate here that estradiol alters Ca-specific protein phosphorylation in rat luteal cells. Corpora lutea obtained from rats hypophysectomized and hysterectomized on day 12 of pregnancy and subsequently treated for 3 days with vehicle or estradiol were assayed in vitro for Ca-specific phosphorylation.
