Colocalization of phosphorylated forms of WAVE1, CRMP2, and tau in Alzheimer's disease model mice: Involvement of Cdk5 phosphorylation and the effect of ATRA treatment.

Alzheimer's disease (AD) is the most common type of dementia among the elderly. Neurofibrillary tangles (NFTs), a major pathological hallmar … and tau in the sarkosyl-insoluble fractions of hippocampal homogenates. To test the significance of phosphorylation of these proteins further, we administered all-trans-retinoic acid (ATRA) to the 3×Tg mice, which downregulates Cdk5 and GSK3β activity. In ATRA-treated mice, fewer and smaller tau aggregates were observed compared with non-ATRA-treated mice. These …

Phosphorylation of the head domain of neurofilament protein (NF-M): a factor regulating topographic phosphorylation of NF-M tail domain KSP sites in neurons.

In neurons the phosphorylation of neurofilament (NF) proteins NF-M and NF-H is topographically regulated. Although kinases and NF subunits are synthesized in cell bodies, extensive phosphorylation of the KSP repeats in tail domains of NF-M and NF-H occurs primarily in axons. The nature of this regulation, however, is not understood. As obligate heteropolymers, NF assembly requires interactions between the core NF-L with NF-M or NF-H subunits, a process inhibited by NF head domain phosphorylation. Phosphorylation of head domains at protein kinase A (PKA)-specific sites seems to occur transientl…

CD40 signaling pathway: anti-CD40 monoclonal antibody induces rapid dephosphorylation and phosphorylation of tyrosine-phosphorylated proteins including protein tyrosine kinase Lyn, Fyn, and Syk and the appearance of a 28-kD tyrosine phosphorylated protein.

CD40 plays an important role in B cell activation, proliferation, and Ig class switching. The signal transduction pathway mediated by CD40 was studied using monoclonal antibody (mAb) 626.1 to CD40. Burkitt's lymphoma and Epstein-Barr virus-transformed B cell lines and tonsilar B lymphocytes were treated with the anti-CD40 mAb for various lengths of time. The early events triggered by CD40 were examined by monitoring the changes in tyrosine phosphorylation of cellular proteins with anti-phosphotyrosine mAb. Dephosphorylation of specific proteins ranging between 50-110 kD and the appearance of a…

14-3-3 binds to and mediates phosphorylation of microtubule-associated tau protein by Ser9-phosphorylated glycogen synthase kinase 3beta in the brain.

In mammalian brain, tau, glycogen synthase kinase 3beta (GSK3beta), and 14-3-3, a phosphoserine-binding protein, are parts of a multiprotein tau phosphorylation complex. Within the complex, 14-3-3 simultaneously binds to tau and GSK3beta (Agarwal-Mawal, A., Qureshi, H. Y., Cafferty, P. W., Yuan, Z., Han, D., Lin, R., and Paudel, H. K. (2003) J. Biol. Chem. 278, 12722-12728). The molecular mechanism by which 14-3-3 connects GSK3beta to tau within the complex is not clear. In this study, we find that GSK3beta within the tau phosphorylation complex is phosphorylated on Ser(9). From extracts of ra…

Radiation-induced eCIRP Causes Alzheimer's Disease-like Neuronal Tau Phosphorylation.

Cranial radiotherapy is associated with neuroinflammation, cognitive dysfunction, and dementia. Tau pathology plays a key role in Alzheimer's disease (AD) but has not been studied in the context of radiation injury. Extracellular cold-inducible RNA-binding protein (eCIRP) is a novel neuroinflammatory mediator that activates neuronal IL6Rα/p25/Cdk5, leading to tau hyperphosphorylation. We hypothesized that radiation promotes pathological tau phosphorylation via the eCIRP/IL6Rα/p25/Cdk5 pathway, which can be mitigated by the eCIRP inhibitor C23. Adult C57BL/6 and CIRP-/- mice were exposed to a s…

Cell-cycle regulation of sarcomere integrity-Role for Actn2 phosphorylation.

Sarcomeres are the fundamental contractile units of muscle. Despite their importance, sarcomere assembly remains poorly understood. We focused on Actn2, a protein which stabilizes the sarcomere by linking proteins to the Z-disk. During C2C12 differentiation into myoblasts, Actn2 protein levels remained constant. This finding suggested that Actn2 incorporation into the sarcomere arose from a post-translational mechanism. We hypothesized that the post-translational mechanism relied on phosphorylation. Alignment of Actn2 protein sequences from animals with three- or four-chambered hearts identifi…

Concurrent Phosphite Oxidation and Adenosine Phosphorylation Under Prebiotic Evaporative Conditions.

Reduced oxidation state phosphorus species, such as phosphite, have been proposed as more soluble phosphorus sources, yet their role in phosphorylation chemistry remains incompletely understood. Here, we investigate the concurrent oxidation of phosphite and phosphorylation/phosphonylation of adenosine under prebiotically plausible conditions. Using aqueous reaction systems containing phosphite, urea, hydrogen peroxide (H₂O₂), and adenosine, we show synthesis of adenosine phosphates, adenosine phosphites, adenosyl adenosine phosphate, and trace amounts of ADP (adenosine diphosphate). Reactions …

PMVK drives hepatocarcinogenesis via SP1 phosphorylation-mediated lipid metabolic reprogramming.

Metabolic reprogramming is a hallmark of cancer, yet the critical drivers and mechanisms in hepatocellular carcinoma (HCC) remain incompletely understood. Through a metabolism-focused CRISPR screen, we identified phosphomevalonate kinase (PMVK), a mevalonate pathway enzyme, as a key regulator of HCC stemness and progression. PMVK directly phosphorylates the transcription factor SP1 at Thr355, which enhances SP1's DNA binding affinity and promotes its interaction with the master lipid regulator SREBP1/2, driving a transcriptional program essential for de novo cholesterol and fatty acid synthesi

KDM6A loss enhances oxidative phosphorylation uncovering tissue-level convergent evolution.

The tumor suppressor KDM6A/UTX, a histone demethylase and a 2-oxoglutarate-dependent dioxygenase, is frequently lost in many cancer types. We show that KDM6A loss pervasively activates oxidative phosphorylation in several solid tumors, generating a pseudo-hyperoxic environment, opposite from the pseudo-hypoxia observed in VHL-mutated renal carcinomas. Mechanistically, KDM6A sustains the expression of the coil-coil domain gene CCDC3, which inhibits CREB1-driven transcription of the mitochondrial regulator PPARGC1A. In the hematological cancer multiple myeloma where KDM6A is frequently deleted, …

Black phosphorus nanosheets boost mitochondrial oxidative phosphorylation improving immunotherapy outcomes.

Regulating intracellular phosphorus may affect multiple biosynthetic processes and modulate cancer cell progression. Here we show that exogenous PEGylated black phosphorus nanosheets (BPP) are metabolized into phosphate in tumor cells, where they boost mitochondrial oxidative phosphorylation. This results in the modulation of several signalling pathways, with the attenuation of prosurvival gene expression and reduction in PD-L1 protein expression in melanoma cells, leading to impaired cancer progression. We also reveal that BPP promote the activation of immune regulation, confirmed by the incr…

Mitochondrial DNA affects tau phosphorylation in aging and Alzheimer's disease.

INTRODUCTION: Impaired mitochondrial function is seen in Alzheimer's disease (AD), but its role is unclear. Mitochondrial DNA (mtDNA) supports bioenergetic metabolism, but it is uncertain how it might influence AD neuropathology. METHODS: We used cytoplasmic hybrid (cybrid) cell lines made from SH-SY5Y cells expressing mtDNA from cognitively healthy (CH), mild cognitive impairment (MCI), or AD individuals to investigate the impact of mtDNA-determined mitochondrial function on amyloid, tau, and neurodegeneration (ATN) markers. Cybrid measurements were correlated with cognition and brain morpho

The Amount of Releasable Insulin Depends on Continuous Oxidative Phosphorylation.

The consensus or canonical model of glucose-stimulated insulin secretion provides that the metabolism of glucose closes KATP channels by increase of the ATP/ADP ratio and that the ensuing depolarization-induced Ca2+ influx through voltage-dependent Ca2+ channels represents the immediate signal for the onset of exocytosis. However, it has been shown earlier that the depolarization-induced secretion can be suppressed by inhibition of the oxidative phosphorylation, pointing to an energy-requiring step presumably located downstream of Ca2+ influx. Here, we have investigated the relation between ox…
