The testudine intranuclear coccidium (TINC) is an emerging, globally significant apicomplexan pathogen responsible for severe multisystemic disease in captive chelonians. In this study, samples were obtained from deceased captive Sri Lankan star tortoise (Geochelone elegans) in Taiwan and leopard tortoises (Stigmochelys pardalis) in Japan for comprehensive pathological and molecular analysis. Histopathological examination of three cases confirmed systemic infection characterized by intranuclear inclusions within epithelial cells of multiple organs, with lesions ranging from widespread coagulat

The Oberhautchen cells of reptile skin possess micro-ornamentations that are complex structures with a wide diversity. The ongoing debate regarding the origin of these structures oscillates between phylogenetic and ecological drivers. The current study also contributes to this debate by examining micro-ornamentations in seven lizards from three families, one worm lizard, and four snakes from three families that have epigeal, fossorial, saxicolous, or semi-aquatic life habits. They were compared in terms of phylogeny and life habits. Additionally, the variations in different scales of the same

The current study is the first of its sort to evaluate the mitochondrial DNA (mtDNA) control region i.e. Displacement Loop (D-loop) in Damani sheep breed of Khyber Pakhtunkhwa Province, Pakistan. Limited work had been done on Damani sheep being recognized based on morphology and SSRs (Same Sequence Repeats) markers, which have not provided information about its origin. Thus, the current study was designed to investigate genetic diversity, relationship and matrilineal lineage of Damani sheep. Thirty (30) pure breed Damani sheep male and female were selected for blood sampling. The DNA was extra

Trichoderma species are recognized as versatile fungi with significant roles in biocontrol of plant diseases, promoting plant growth and safeguarding against various environmental threats. Despite their benefits, the biodiversity of indigenous Trichoderma isolates in the semi-arid region of Brazil's Caatinga biome remains largely unexplored. This untapped diversity holds promise for identifying potential biocontrol agents against soilborne plant pathogens. Here, we conducted molecular identification and morphological characterization of 14 indigenous Trichoderma isolates from both undisturbed

Background: The phylogenetic resolution within the Gloydius halys-intermedius Complex remains debatable due to the following reasons: loci selection in previous studies varied between authors; limited dataset (1-5 mitochondrial or nuclear gene fragments); lack of sampling density; and nodal supports at specific nodes remain weak, specifically within Gloydius cognatus, G. halys, and G. stejnegeri. Objectives: To revise the taxonomic and phylogenetic relationships within the G. halys-intermedius Complex, we reconstructed the molecular phylogeny and performed species delimitation based on the com

Zygaenoidea is a superfamily of lepidopterans containing many venomous species, including the Limacodidae (nettle caterpillars) and Megalopygidae (asp caterpillars). Venom proteomes have been recently documented for several species from each of these families, but further data are required to understand the evolution of venom in Zygaenoidea. In this study, we examined the 'electric' caterpillar from North-Eastern Australia, a limacodid caterpillar densely covered in venomous spines. We used DNA barcoding to identify this caterpillar as the larva of the moth Comana monomorpha (Turner, 1904). We

Dicrocoeliosis is a trematode infection in cattle, sheep and goats caused by the small liver fluke, Dicrocoelium spp. Though endemic in Ghana, its disease situation is poorly understood. In the present study, the prevalence, distribution and worm load of Dicrocoelium spp. in cattle at slaughter in Wa were determined. A total of 389 cattle were screened during meat inspection for liver flukes, and polymerase chain reaction accompanied by DNA sequencing of the 28S rRNA gene was used to identify Dicrocoelium spp. Generally, prevalence of bovine dicrocoeliosis (small liver fluke) stood at 19.54 %

Vaccination and anti-viral therapy with nucleos(t)ide analogues (NAs) are key approaches to reducing the morbidity, mortality and transmission of hepatitis B virus (HBV) infection. However, the efficacy of these interventions may be reduced by the emergence of drug resistance-associated mutations (RAMs) and/or vaccine escape mutations (VEMs). We have assimilated data on the global prevalence and distribution of HBV RAMs/VEMs from publicly available data and explored the evolution of these mutations. We analysed sequences downloaded from the HBV Database and calculated prevalence of 41 RAMs and

BACKGROUND: The Neacomys genus is predominantly found in the Amazon region, and belongs to the most diverse tribe of the Sigmodontinae subfamily (Rodentia, Cricetidae, Oryzomyini). The systematics of this genus and questions about its diversity and range have been investigated by morphological, molecular (Cytb and COI sequences) and karyotype analysis (classic cytogenetics and chromosome painting), which have revealed candidate species and new distribution areas. Here we analyzed four species of Neacomys by chromosome painting with Hylaeamys megacephalus (HME) whole-chromosome probes, and comp

Bordetella pertussis, the causative agent of whooping cough, has experienced a resurgence in the past 15 years, despite the existence of both whole-cell and acellular vaccines. Here, we performed whole genome sequencing analysis of 149 clinical strains, provided by the National Institute of Infectious Diseases (NIID), Japan, isolated in 1982-2014, after Japan became the first country to adopt acellular vaccines against B. pertussis. Additionally, we sequenced 39 strains provided by the Konan Kosei Hospital in Aichi prefecture, Japan, isolated in 2008-2013. The genome sequences afforded insight

Since the first major outbreak of Zika virus (ZIKV) in 2007, ZIKV is spreading explosively through South and Central America, and recent reports in highly populated developing countries alarm the possibility of a more catastrophic outbreak. ZIKV infection in pregnant women leads to embryonic microcephaly and Guillain-Barré syndrome in adults. At present, there is limited understanding of the infectious mechanism, and no approved therapy has been reported. Despite the withdrawal of public health emergency, the WHO still considers the ZIKV as a highly significant and long-term public health chal

UNLABELLED: Nine distinct venom serine proteases (vSPs) of Gloydius intermedius were studied by transcriptomic, sub-proteomic and phylogenetic analyses. Their complete amino acid sequences were deduced after Expression Sequence Tag (EST) analyses followed by cDNA cloning and sequencing. These vSPs appear to be paralogs and contain the catalytic triads and 1-4 potential N-glycosylation sites. Their relative expression levels evaluated by qPCR were grossly consistent with their EST hit-numbers. The major vSPs were purified by HPLC and their N-terminal sequences matched well to the deduced sequen

Humans and dogs are the two major hosts of Strongyloides stercoralis, an intestinal parasitic nematode. To better understand the phylogenetic relationships among S. stercoralis isolates infecting humans and dogs and to assess the zoonotic potential of this parasite, we analyzed mitochondrial Cox1, nuclear 18S rDNA, 28S rDNA, and a major sperm protein domain-containing protein genes. Overall, our analyses indicated the presence of two distinct lineages of S. stercoralis (referred to as type A and type B). While type A parasites were isolated both from humans and dogs in different countries, typ

Using a combination of morphological, life-cycle and molecular data, we describe a new apicomplexan parasite Eimeria telekii n.sp. from a striped grass mouse Lemniscomys striatus captured in Kenya. Oocysts are oval to spherical or ellipsoidal, 20.4 x 15.7 (15.5-25.0 x 12.0-20.0) microm with a colourless, smooth and bilayered wall. Sporocysts are ellipsoidal, 11.2 x 7.8 (10.0-12.0 x 7.0-9.0) microm with a small Stieda body and granular sporocyst residuum and contain 2 elongated, banana-shaped sporozoites with a single refractile body. Life-cycle, pathogenicity and host specificity of this paras

Primates are regularly infected by fungal organisms identified as Pneumocystis carinii. They constitute a valuable population for the confirmation of P. carinii host specificity. In this study, the presence of P. carinii was assessed by direct examination and nested PCR at mitochondrial large subunit (mtLSU) rRNA and dihydropteroate synthetase (DHPS) genes in 98 lung tissue samples from captive or wild nonhuman primates. Fifty-nine air samples corresponding to the environment of different primate species in zoological parks were also examined. Cystic forms of P. carinii were detected in smears

Predicting the outcomes of bacteria-phage interactions is a central challenge in microbial ecology, particularly across heterogeneous host environments. Evolutionary relatedness among bacteria can explain variation in phage susceptibility in vitro, but it remains unclear whether this predictive relationship persists in complex host environments and to what extent relatedness also explains variation in other bacterial traits, such as virulence. Here, we used an in vivo infection model to quantify how bacterial phylogeny and host environmental context mediate bacterial virulence and bacteria-pha

As important vectors of infectious diseases such as plague, fleas require accurate classification and phylogenetic analysis, which are essential for public health prevention and control. However, complete mitochondrial genome resources for Siphonaptera in public databases are still extremely scarce. In this study, the complete mitochondrial genomes of C. dolabris and O. silantiewi, collected from the high-altitude region in Qinghai Province, were sequenced and characterized for the first time using next-generation sequencing technology. The results showed the two mitogenomes were 17,104 bp and

Human endogenous retroviruses (HERVs) are remmants of ancient infections which make up to ~8% of the human genome. Their activity influences development, immunity, and cancer, but studying them has been limited by a key technical challenge: short-read sequencing cannot uniquely assign reads to these highly repetitive elements. Here, we present ERVmancer, a phylogeny-informed method that resolves the read-mapping ambiguity and quantifies HERV expression across scales, from individual loci to entire retroviral clades, depending on mapping confidence. Benchmarking with sample-matched long- and sh
