Physical degradation of lignified stem tissues by ruminal fungi.

Ruminal bacteria or fungi were selected by the addition of cycloheximide or streptomycin and penicillin, respectively, to ruminal fluid, and the weakening and degradation of lignified tissues in alfalfa and Bermuda grass stems by these treatments and whole ruminal fluid were evaluated in vitro. Dry weight loss in alfalfa was similar for whole ruminal fluid and streptomycin-penicillin treatment, whereas that with streptomycin-penicillin treatment was significantly higher (P </= 0.05) than that with cycloheximide treatment. In Bermuda grass, dry weight loss was significantly higher with streptom

Bioaccumulation and transferreing for impacts on Cd and Pb by aphid consumption of the broad bean, Vicia faba L, in soil heavy metal pollution.

Heavy metal pollution threatens human and ecological health. Heavy metals can exist in the soil for a long time and migrate to organisms along the food chain. However, only a few studies have investigated the effects of a single stress on broad beans. Here, we aimed to characterize Cd and Pb bioaccumulation, at varying concentrations, in the broad bean, Vicia faba L. We also determined how the bioaccumulated metals are impacted by aphids that consume the plant. No significant difference was noted in the germination rates of broad beans at the early stage of planting (after 8 days), but eventua

Chemical components with antitumour activity from Inula britannica.

The flowers of Inula britannica L. are widely used for their significant antitumour and anti-inflammatory properties. However, the other parts of the plant, including the stems, leaves and roots, remain underutilised. Herein, 18 compounds, including 5 previously undescribed compounds (1-5), were isolated from these underutilised parts, and their structures were elucidated by HRESIMS, NMR and ECD spectroscopic analyses. Several isolated sesquiterpenes (7-9) exhibited significant inhibitory activity against three tumour cell lines. In addition, eight compounds (2-4, 11, 13 and 16-18) showed syne

Combined metabolome and transcriptome to analyze the regulatory network of key enzymes in the synthesis of senkirkine in Emilia sonchifolia.

Emilia sonchifolia (L.) DC. serves as a well-known folk medicinal and edible plant, yet its toxic component senkirkine exhibits hepatotoxicity. Acute high-dose exposure can induce sinusoidal obstruction syndrome, while chronic low-dose intake may cause hepatic megaloblastosis and fibrosis. This study applied metabolomics to analyze alkaloid compositions in the plant's roots, stems, leaves, and flowers. A total of 91 differential metabolites were detected, belonging to 10 alkaloids, among which pyrrolizidine alkaloids accounted for 13%. Senkirkine showed the most significant content variation b

A wound-inducible tobacco peroxidase gene expresses preferentially in the vascular system.

A tobacco peroxidase gene tpoxN1 was reported to be expressed within 1 h after wounding in leaves [Hiraga et al. (2000a) Plant Cell Physiol. 41: 165]. We describe here further results on the wound-induced tpoxN1 expression. The quick tpoxN1 induction occurred preferentially in stems and petioles, but was negligible in leaf blades even 8 h after wounding. Induced GUS activity was also detected rapidly after wounding in the stem of transgenic tobacco plants carrying the tpoxN1 promoter::GUS fusion gene, localized mainly in the vascular systems where it was maintained this level for 14 d or more.

Herbivory-associated degradation of tomato trichomes and its impact on biological control of Aculops lycopersici.

Tomato plants have their leaves, petioles and stems covered with glandular trichomes that protect the plant against two-spotted spider mites and many other herbivorous arthropods, but also hinder searching by phytoseiid mites and other natural enemies of these herbivores. This trichome cover creates competitor-free and enemy-free space for the tomato russet mite (TRM) Aculops lycopersici (Acari: Eriophyidae), being so minute that it can seek refuge and feed inbetween the glandular trichomes on tomato cultivars currently used in practice. Indeed, several species of predatory mites tested for bi

Mass sterilization of a common palm species by elephants in Kruger National Park, South Africa.

Chronic herbivory by elephants rarely eliminates any species of woody savanna plants because these plants are typically vigorous basal resprouters after damage by fire or herbivory. In some instances, resprouting after elephant herbivory even increases stem numbers per unit area compared to protected areas. It is thus difficult to know whether an area has been severely degraded by elephant herbivory or not because although trees may be severely reduced in size, they will still be present and may even be relatively dense. By using an elephant exclosure in the Kruger National Park, South Africa,

Growth promotion in tomato by endophytic entomopathogenic and arbuscular mycorrhizal fungi: impacts on early development and post-transplant effect.

Tomato (Solanum lycopersicum L.) is a globally significant crop valued for its nutritional content and economic importance. This study evaluated the effect of individual and combined inoculation of two growth-promoting microorganisms in tomato: the entomopathogenic fungus Beauveria bassiana and the arbuscular mycorrhizal fungus Rhizophagus irregularis. The research was conducted in chamber conditions using tomato plants cultivar Elpida, at two developmental stages: seedlings and transplanted plants. Colonization of B. bassiana in plant tissues was confirmed by isolation from different organs,

Knockout of the S-acyltransferase Gene, PbPAT14, Confers the Dwarf Yellowing Phenotype in First Generation Pear by ABA Accumulation.

The development of dwarf fruit trees with smaller and compact characteristics leads to significantly increased fruit production, which is a major objective of pear (Pyrus bretschneideri) breeding. We identified the S-acylation activity of PbPAT14, an S-acyltransferase gene related to plant development, using a yeast (Saccharomyces cerevisiae) complementation assay, and also PbPAT14 could rescue the growth defect of the Arabidopsis mutant atpat14. We further studied the function of PbPAT14 by designing three guide RNAs for PbPAT14 to use in the CRISPR/Cas9 system. We obtained 22 positive transg

Evaluation of molluscicidal activity of three mangrove species (Avicennia schaueriana, Laguncularia racemosa and Rhizophora mangle) and their effects on the bioactivity of Biomphalaria glabrata Say, 1818.

Schistosomiasis is a disease of global extent reaching populations in social vulnerability. One of the control measures of this parasitosis is the use of molluscicidal substances that can fight snails of the genus Biomphalaria, intermediate hosts of Schistosoma mansoni. The aim of this work was to study the toxic activity of three mangrove species (Avicennia schaueriana Stapf. & Leech, ex Moldenke, 1939, Laguncularia racemosa (L.) CF Gaertn, 1807 and Rhizophora mangle L. 1753) on the biological activities of snails Biomphalaria glabrata. Hydroalcoholic extracts were prepared from the stem and

Trichloroethylene oxidative metabolism in plants: the trichloroethanol pathway.

Trichloroethylene (TCE) is a widespread and persistent environmental contaminant. Recently, plants, poplar trees in particular, have been investigated as a tool to remove TCE from soil and groundwater. The metabolism of TCE in plants is being investigated for two reasons: one, plant uptake and metabolism represent an important aspect of the environmental fate of the contaminant; two, metabolism pattern and metabolite identification will help assess the applicability of phytoremediation. It was previously shown that TCE metabolites in plants are similar to ones that result from cytochrome P450-

First Report of Pectobacterium carotovorum Causing Soft Rot of Opium Poppy in Spain.

Opium poppy (Papaver somniferum L.) is an economically important pharmaceutical crop in Spain. Approximately 8,000 ha are cultivated annually in southern and central Spain. To improve yields, opium poppy cultivation is expanding to more humid or irrigated areas of Spain. In the springs of 2005 and 2007, we observed poppy plants with wilt and stem rot symptoms in irrigated, commercial opium poppy (cv. Nigrum) at Carmona and Écija, which are in Seville Province in southern Spain. Closer observations of affected plants revealed darkening and water soaking of the leaves and stem at the soil level,

Supplementary UV-A and UV-B radiation differentially regulate morphology in Ocimum basilicum.

UV-A- or UV-B-enriched growth light was given to basil plants at non-stress-inducing intensities. UV-A-enriched growth light gave rise to a sharp rise in the expression of PAL and CHS genes in leaves, an effect that rapidly declined after 1-2 days of exposure. On the other hand, leaves of plants grown in UV-B-enriched light had a more stable and long-lasting increase in the expression of these genes and also showed a stronger increase in leaf epidermal flavonol content. UV supplementation of growth light also led to shorter more compact plants with a stronger UV effect the younger the tissue.

Suppression of microRNA159 impacts multiple agronomic traits in rice (Oryza sativa L.).

BACKGROUND: microRNAs (miRNAs) are important regulators in plant growth and development. miR159 is a conserved miRNA among different plant species and has various functions in plants. Studies on miR159 are mostly done on model plant, Arabidopsis thaliana. In rice, studies on miR159 were either based upon genome-wide expression analyses focused upon responses to different nitrogen forms and abiotic stress or upon phenotypic studies of transgenic plants overexpressing its precursor. STTM (Short Tandem Target Mimic) is an effective tool to block the activity of endogenous mature miRNA activity in

Thermoperiodic growth control by gibberellin does not involve changes in photosynthetic or respiratory capacities in pea.

Active gibberellin (GA(1)) is an important mediator of thermoperiodic growth in pea. Plants grown under lower day than night temperature (negative DIF) elongate less and have reduced levels of GA(1) compared with plants grown at higher day than night temperature (positive DIF). By comparing the wild type (WT) and the elongated DELLA mutant la cry(s), this study has examined the effect of impaired GA signalling on thermoperiodic growth, photosynthesis, and respiration in pea. In the WT a negative DIF treatment reduced stem mass ratio and increased both root mass ratio and leaf mass ratio (dry w

Short-term and long-term clinostat and vibration-induced biochemical changes in dwarf marigold stems.

Stems of 21-day dwarf Marigold plants cultivated on the clinostat were compared with plants cultivated on vertical axis rotators ("vibrational controls") and stationary controls for long-term changes in cell wall composition. Stems of 21-day plants grown under stationary conditions and subsequently exposed to the clinostat for 24 hours were also analyzed. Among the long-tern markers, calciun, lignin, and protein-bound hemicellulose (possibly cell wall glycoprotein) clearly differentiated the effects of vibration from those of the clinostat. Short-term differential responses included rate of et
