Bioactivity of Spongian Diterpenoid Scaffolds from the Antarctic Sponge Dendrilla antarctica.

The Antarctic sponge Dendrilla antarctica is rich in defensive terpenoids with promising antimicrobial potential. Investigation of this demosponge has resulted in the generation of a small chemical library containing diterpenoid secondary metabolites with bioactivity in an infectious disease screening campaign focused on Leishmania donovani, Plasmodium falciparum, and methicillin-resistant Staphylococcus aureus (MRSA) biofilm. In total, eleven natural products were isolated, including three new compounds designated dendrillins B-D (10-12). Chemical modification of abundant natural products led

Bacillus Strains Associated to Homoscleromorpha Sponges are Highly Active Against Multidrug Resistant Bacteria.

The search for new, powerful antimicrobials is essential to respond to the current worldwide spread of antibiotic-resistant pathogens. Sponge-associated bacteria have great potential for production of antimicrobials against resistant and multidrug resistant (MDR) pathogenic bacteria, but only few species of the Class Homoscleromorpha have been screened for these activities so far. The aim of this study was to isolate and identify sponge-associated bacteria active against antibiotic-resistant pathogens from sponges of classes Homoscleromorpha and Demospongiae. By employing five different growth

Antimicrobial and antibiofilm activities of marine sponge-associated bacteria against multidrug-resistant Staphylococcus spp. isolated from canine skin.

Dogs play important roles in our society, thus the concern for their health becomes imperative. Staphylococcus spp. are commensal bacterium frequently isolated from canine skin and recognized as zoonotic agents. These bacteria have been becoming increasingly resistant to antimicrobials used to treat infections and to produce biofilm, which further increases their virulence capability and resistance. In this context, sponges-associated bacteria are known as prolific sources of substances with antimicrobial activities, representing a potential to integrate the arsenal of drugs for clinical use.

Chrysoxanthones A⁻C, Three New Xanthone⁻Chromanone Heterdimers from Sponge-Associated Penicillium chrysogenum HLS111 Treated with Histone Deacetylase Inhibitor.

By treating with histone-deacetylase inhibitor valproate sodium, three new heterdimeric tetrahydroxanthone⁻chromanone lactones chrysoxanthones A⁻C (1⁻3), along with 17 known compounds were isolated from a sponge-associated Penicillium chrysogenum HLS111. The planar structures of chrysoxanthones A⁻C were elucidated by means of spectroscopic analyses, including MS, 1D, and 2D NMR. Their absolute configurations were established by electronic circular dichroism (ECD) calculations. Chrysoxanthones A⁻C exhibited moderate antibacterial activities against Bacillus subtilis with minimum inhibitory conc

Preussins with Inhibition of IL-6 Expression from Aspergillus flocculosus 16D-1, a Fungus Isolated from the Marine Sponge Phakellia fusca.

New pyrrolidine alkaloids, preussins C-I (1-7) and (11 R)/(11 S)-preussins J and K (8 and 9), were isolated from the sponge-derived fungus Aspergillus flocculosus 16D-1. The structures and configurations of these preussins were elucidated by detailed spectroscopic analysis, modified Mosher's method, and comparisons with literature data. These compounds showed strong to moderate inhibitory activity toward IL-6 production in lipopolysaccharide-induced THP-1 cells with IC50 values ranging from 0.11 to 22 μM, but were inactive against normal tumor cell lines and fungi.

Validation of a gauze sponge sampling methodology to detect Tritrichomonas foetus by real-time PCR.

Bovine trichomoniasis is a concern for the cattle industry. Advances in testing systems have increased the ability to detect the disease in bulls. However, the greatest limitation is proper collection of an adequate sample. The low repeatability observed with most sample collection techniques can cause false-negative results. The aim of our study was to validate a sample collection technique that increases diagnostic sensitivity and is easier and safer to collect than preputial scraping. Commercial bulls (n = 111) of unknown infection status were sampled for detection of Tritrichomonas foetus

Secondary metabolites with new medicinal functions from marine organisms.

We have focused on the identification of natural key compounds that possess biologically and medicinally intriguing functions. Some of bioactive naturally occurring compounds isolated from marine organisms were found to possess unique biological activities. Halichlorine from the marine sponge Halichondria okadai was shown to inhibit the activity of nuclear factor-κB in endothelial cells and block L-type Ca²⁺ channels. Thus, it may have therapeutic potentials for diseases such as atherosclerosis and hypertension.

The symbiotic lifestyle and its evolutionary consequences: social monogamy and sex allocation in the hermaphroditic shrimp Lysmata pederseni.

Sex allocation theory predicts female-biased sex allocation for simultaneous hermaphrodites with a monogamous mating system. Mating systems theory predicts that monogamy is advantageous in environments where refuges are discrete, scarce, relatively small, and when predation risk is high outside of these refuges. These predictions were tested with the Caribbean shrimp Lysmata pederseni, a simultaneous hermaphrodite which has an early male phase and lives inside tubes of the sponge Callyspongia vaginalis. This host sponge is a scarce resource that, together with the high predation risk typical o

Application of HPLC-NMR for the rapid chemical profiling of a Southern Australian sponge, Dactylospongia sp.

Rapid chemical profiling of the antitumour active crude dichloromethane extract of the marine sponge, Dactylospongia sp. was undertaken. A combination of both offline (HPLC followed by NMR and MS) and on-line (on-flow and stop-flow HPLC-NMR) chemical profiling approaches was adopted to establish the exact nature of the major constituents present in the dichloromethane extract of this sponge. On-flow HPLC-NMR analysis was employed to initially identify components present in the dichloromethane extract, while stop-flow HPLC-NMR experiments were then conducted on the major component present, resu

In vitro and in vivo antifungal activities of the marine sponge constituent spongistatin.

Spongistatin 1 is a macrocyclic lactone polyether from the marine sponge Hyrtios erecta. The aim of this study was to evaluate the in vitro and in vivo antifungal efficacies and mechanism of spongistatin 1. Spongistatin 1 was fungicidal for the majority of 74 reference strains and clinical isolates, including those resistant to flucytosine, ketoconazole or fluconazole, and retained activity in the presence of human serum or at lowered pH. The duration of the postantifungal effect following 1 h exposure to one, four and eight times the minimal inhibitory concentration was strain-dependent. Spon

Identification of Mortalin as the Main Interactor of Mycalin A, a Poly-Brominated C-15 Acetogenin Sponge Metabolite, by MS-Based Proteomics.

Mycalin A (MA) is a polybrominated C-15 acetogenin isolated from the marine sponge Mycale rotalis. Since this substance displays a strong antiproliferative bioactivity towards some tumour cells, we have now directed our studies towards the elucidation of the MA interactome through functional proteomic approaches, (DARTS and t-LIP-MS). DARTS experiments were performed on Hela cell lysates with the purpose of identifying MA main target protein(s); t-LiP-MS was then applied for an in-depth investigation of the MA-target protein interaction. Both these techniques exploit limited proteolysis couple

An antimicrobial metabolite n- hexadecenoic acid from marine sponge-associated bacteria Bacillus subtilis effectively inhibited biofilm forming multidrug-resistant P. aeruginosa.

Effective drug candidates to obstruct the emergence of multidrug-resistant pathogens have become a major concern. A potent antimicrobial producer was isolated from a marine sponge designated as MSI38 and was identified as Bacillus subtilis by 16SrDNA sequencing. The active antimicrobial fraction was purified, and the metabolite was identified as n-hexadecanoic acid by spectroscopic analysis. The fish-borne pathogen Pseudomonas aeruginosa FP012 was found to be multidrug-resistant and poses a risk of disease to food handlers and consumers in general. The compound showed a potent bactericidal eff

Metabolites from Marine Sponges and Their Potential to Treat Malarial Protozoan Parasites Infection: A Systematic Review.

Malaria is an infectious disease caused by protozoan parasites of the Plasmodium genus through the bite of female Anopheles mosquitoes, affecting 228 million people and causing 415 thousand deaths in 2018. Artemisinin-based combination therapies (ACTs) are the most recommended treatment for malaria; however, the emergence of multidrug resistance has unfortunately limited their effects and challenged the field. In this context, the ocean and its rich biodiversity have emerged as a very promising resource of bioactive compounds and secondary metabolites from different marine organisms. This syst

Total Synthesis and Antimalarial Activity of Dominicin, a Cyclic Octapeptide from a Marine Sponge.

Dominicin, a macrocyclic peptide isolated from the marine sponge Eurypon laughlini, has been synthesized for the first time by solid-phase peptide synthesis. The strategy uses oxime resin and takes advantage of the nucleophile susceptibility of the oxime ester bond. The synthesis relies on the preparation of a linear precursor followed by on-resin head-to-tail concomitant cyclization-cleavage. This is the first report of the use of a Boc/OtBu biorthogonal protection strategy on oxime resin to facilitate concomitant N-terminal and side-chain tert-butyl ether deprotection cyclization of unprotec

The marine sponge Diacarnus bismarckensis as a source of peroxiterpene inhibitors of Trypanosoma brucei, the causative agent of sleeping sickness.

Human African trypanosomiasis, also known as African sleeping sickness, is a neglected tropical disease with inadequate therapeutic options. We have launched a collaborative new lead discovery venture using our repository of extracts and natural product compounds as input into our growth inhibition primary screen against Trypanosoma brucei. Careful evaluation of the spectral data of the natural products and derivatives allowed for the elucidation of the absolute configuration (using the modified Mosher's method) of two new peroxiterpenes: (+)-muqubilone B (1a) and (-)-ent-muqubilone (3a). Cyclosporin A suspends transplantation reactions in the marine sponge Microciona prolifera.

Sponges are the simplest extant animals but nevertheless possess self-nonself recognition that rivals the specificity of the vertebrate MHC. We have used dissociated cell assays and grafting techniques to study tissue acceptance and rejection in the marine sponge Microciona prolifera. Our data show that allogeneic, but not isogeneic, cell contacts trigger cell death and an increased expression of cell adhesion and apoptosis markers in cells that accumulate in graft interfaces. Experiments investigating the possible existence of immune memory in sponges indicate that faster second set reactions
