Ethanol-precipitated substance (EP) was prepared from culture filtrate of Fonsecaea pedrosoi. Human lysosomes were isolated from normal peripheral blood leukoyctes and characterized by electron microscopy, enzyme analysis, and assays for DNA and RNA. Stored sera from 37 unselected patients with systemic lupus erythematosus (SLE), including active and inactive, treated and untreated cases, were tested in complement fixation (CF) reactions with these lysosome preparations. 23 SLE sera exhibited positive CR reactions, as did sera from two patients with "lupoid" hepatitis. 20 years old man with history of acute allergic alveolitis 7 years ago, was admitted to hospital because of dyspnea, fever and cough after massive exposition to organic dust. Corticosteroids were introduced. During 3-weeks treatment respiratory failure progressed. Chest x-ray showed massive bilateral nodular opacities. Antibiotic therapy was not effective. The autopsy revealed invasive pulmonary aspergillosis. Presence of precipitins to aspergillus fumigatus was confirmed postmortem. Diagnostic difficulties are discussed.

Lipopolysaccharide extracts of E. coli (endotoxins) were toxic to normal adult guinea-pigs, yet no evidence was found of serological reactivity to endotoxins or of "immediate" dermal reactivity in vivo. In contrast, testing of the serum of experimentally immunized animals provided evidence of precipitins, agglutinins and haemolysins. It is concluded that endotoxin exerts toxic effects in the absence of significant antibody production. This suggests that endotoxin toxicity is independent of immune processes not involving cell-mediated responses.

Counterimmunoelectrophoresis (CIE) has been compared in a diangostic laboratory with agar gel double diffusion (DD) as a routine procedure for detection of antibodies to pathogenic and allergenic fungi and actinomycetes. It was shown to be of particular value in detecting antibodies to Aspergillus fumigatus. Thus 72 of 106 sera in which precipitins were detected were positive by CIE alone. Some sera were positive only by CIE to antigens prepared from Histoplasma capsulatum, Allescheria boydii, Candida albicans and C. parapsilosis.

Specific sensitization and respiratory effects associated with the inhalation of sugar cane dust were evaluated in a group of 51 Nicaraguan workers exposed to bagasse. A questionnaire interview, lung function test, serum precipitin tests for Thermoactinomyces sacchari and T. vulgaris, and immunoglobulin E tests for specific environmental allergens were performed for each worker. Precipitating antibodies occurring in 26 "pure" polymyositis (PM) patients and 22 "pure" dermatomyositis (DM) patients were studied by double immunodiffusion using calf thymus nuclear extract and whole calf thymus extract.
