Development and validation of a machine learning algorithm prediction for dense granule proteins in Apicomplexa.

BACKGROUND: Apicomplexa consist of numerous pathogenic parasitic protistan genera that invade host cells and reside and replicate within the parasitophorous vacuole (PV). Through this interface, the parasite exchanges nutrients and affects transport and immune modulation. During the intracellular life-cycle, the specialized secretory organelles of the parasite secrete an array of proteins, among which dense granule proteins (GRAs) play a major role in the modification of the PV. Despite this important role of GRAs, a large number of potential GRAs remain unidentified in Apicomplexa. METHODS:

Predicting glaucoma progression using deep learning framework guided by generative algorithm.

Glaucoma is a slowly progressing optic neuropathy that may eventually lead to blindness. To help patients receive customized treatment, predicting how quickly the disease will progress is important. Structural assessment using optical coherence tomography (OCT) can be used to visualize glaucomatous optic nerve and retinal damage, while functional visual field (VF) tests can be used to measure the extent of vision loss. However, VF testing is patient-dependent and highly inconsistent, making it difficult to track glaucoma progression. In this work, we developed a multimodal deep learning model

Development and validation of primary graft dysfunction predictive algorithm for lung transplant candidates.

BACKGROUND: Primary graft dysfunction (PGD) is the leading cause of early morbidity and mortality after lung transplantation. Accurate prediction of PGD risk could inform donor approaches and perioperative care planning. We sought to develop a clinically useful, generalizable PGD prediction model to aid in transplant decision-making. METHODS: We derived a predictive model in a prospective cohort study of subjects from 2012 to 2018, followed by a single-center external validation. We used regularized (lasso) logistic regression to evaluate the predictive ability of clinically available PGD pre

[An EHG-based Preterm Delivery Prediction Algorithm via Convolution Neural Network].

Premature delivery is one of the direct factors that affect the early development and safety of infants. Its direct clinical manifestation is the change of uterine contraction intensity and frequency. Uterine Electrohysterography(EHG) signal collected from the abdomen of pregnant women can accurately and effectively reflect the uterine contraction, which has higher clinical application value than invasive monitoring technology such as intrauterine pressure catheter. Therefore, the research of fetal preterm birth recognition algorithm based on EHG is particularly important for perinatal fetal m

Vital sign metrics of VLBW infants in three NICUs: implications for predictive algorithms.

BACKGROUND: Continuous heart rate (HR) and oxygenation (SpO2) metrics can be useful for predicting adverse events in very low birth weight (VLBW) infants. To optimize the utility of these tools, inter-site variability must be taken into account. METHODS: For VLBW infants at three neonatal intensive care units (NICUs), we analyzed the mean, standard deviation, skewness, kurtosis, and cross-correlation of electrocardiogram HR, pulse oximeter pulse rate, and SpO2. The number and durations of bradycardia and desaturation events were also measured. Twenty-two metrics were calculated hourly, and me

SAAMP 2.0: An algorithm to predict genotype-phenotype correlation of lysosomal storage diseases.

Lysosomal storage diseases (LSDs) are a group of genetic disorders, resulting from deficiencies of lysosomal enzyme. Genotype-phenotype correlation is essential for timely and proper treatment allocation. Recently, by integrating prediction outcomes of 7 bioinformatics tools, we developed a SAAMP algorithm to predict the impact of individual amino-acid substitution. To optimize this approach, we evaluated the performance of these bioinformatics tools in a broad array of genes. PolyPhen and PROVEAN had the best performances, while SNP&GOs, PANTHER and I-Mutant had the worst performances. Theref

Validating an Algorithm to Predict Adjacent Musculoskeletal Infections in Pediatric Patients With Septic Arthritis.

BACKGROUND: Septic arthritis is frequently associated with adjacent infections including osteomyelitis and subperiosteal and intramuscular abscesses. While often clinically indiscernible from isolated septic arthritis, the diagnosis of adjacent infections is important in determining the need for additional surgical intervention. MRI has been used as the diagnostic gold standard for assessing adjacent infection. Routine MRI, however, can be resource-intensive and delay surgical treatment. In this context, there is need for additional diagnostic tools to assist clinicians in determining when to

An Evidence-Based Clinical Prediction Algorithm for the Musculoskeletal Infection Society Minor Criteria.

BACKGROUND: The diagnosis of a periprosthetic joint infection (PJI) remains a clinical challenge, as there is no uniformly accepted gold standard. In 2011, the Musculoskeletal Infection Society (MSIS) convened a work group to create a standardized definition for a PJI that could be universally adopted. Based on the MSIS criteria, the diagnosis of a PJI can be made with 1 of the 2 major criteria, or 3 of the 5 minor criteria. The purpose of this study was to determine the likelihood of having a PJI based on the number of positive minor criteria and thereby develop a prediction algorithm for dif

[Berger disease: therapeutic algorithm proposal and chronic renal failure predictive factors].

Berger disease is described as a glomerulonephritis by anti-IgA and anti-IgG immune complexes and it has become the most common form of glomerulonephritis, affecting up to 1,3% of the population, most often affecting young adult males. Berger disease etiopathogenesis is still unknown, being well recognized its temporal associations with bacterial and viral infections, as well with genetic factors which lead some authors to suggest the existence of a founder effect. Characteristically IgA nephropathy presents intermittent macroscopic hematuria following mucosal infections, nonnephrotic proteinu

A deep clustering Gaussian process algorithm for motor progression prediction of Parkinson's disease.

BACKGROUND AND OBJECTIVE: Parkinson's disease (PD) is a chronic progressive neurodegenerative disorder characterized by significant spatial and temporal heterogeneity in symptom presentation and progression, which poses a major challenge for accurate motor progression prediction. Developing a highly individualized model for predicting PD motor progression that can reflect the heterogeneity may lead to better management. METHODS: To address this issue, we propose a novel Deep Clustering Gaussian process (DCGP) algorithm to predict PD motor progression using readily available clinical statistic

PACT: A practice-driven predictive algorithm for customized transradial prosthetic socket design.

Well-fitting sockets are crucial for successful upper-limb prosthesis use, yet current digital socket design workflows are not standardized and demand considerable clinician effort. In this study, we introduce the Predictive Algorithm for Customized Transradial Socket Design (PACT), which generates socket models from a 3D limb scan. It works by retrieving the most similar limb-socket pair from a reference library of prosthetist-designed prosthetic sockets and applying isotropic and anisotropic scaling adjustments to match the input limb. To validate the algorithm, the PACT-predicted sockets fo
