Cell cycle dependent coordination of surface layer biogenesis in Caulobacter crescentus.

Surface layers (S-layers) are proteinaceous, two-dimensional paracrystalline arrays that constitute a major component of the cell envelope in many prokaryotic species. In this study, we investigated S-layer biogenesis in the bacterial model organism Caulobacter crescentus. Fluorescence microscopy revealed localised incorporation of new S-layer at the poles and mid-cell, consistent with regions of cell growth in the cell cycle. Light microscopy and electron cryotomography investigations of drug-treated bacteria revealed that localised S-layer insertion is retained when cell division is inhibite

From past to present: the crosstalk between type VI secretion system of Salmonella and host.

Salmonella encounters dual selective pressures during infection - from the host immune system and competing gut microbiota. The Type VI secretion system (T6SS) is a versatile transmembrane nanomachine that delivers effector proteins into target cells or the extracellular environment, enabling diverse functions including bacterial competition, virulence, and niche adaptation. Emerging evidence demonstrates that some T6SS effectors exhibit dual targeting capability, affecting both eukaryotic and prokaryotic cells. This dual-targeting strategy confers competitive advantages by simultaneously elim

A new B cell epitope of pC129R protein of African swine fever virus identified by monoclonal antibodies.

African swine fever virus (ASFV) is a most important pathogen which causes huge damage in swine production in the world. pC129R protein is one of the most abundant ASFV proteins in infected Vero cells and WSL-HP cells, which consequently could be a target for ASF detection and surveillance. In this study, 5-6-week-old female BALB/c mice were immunized with rpC129R protein expressed by a prokaryotic system. And three hybridomas, 1B1, 1B4 and 4H4, steadily secreted anti-pC129R monoclonal antibodies were screened by an indirect enzyme linked immunosorbent assay (ELISA). Among them, 1B4 and 4H4 ha

Phase-variable Type I methyltransferase M.NgoAV from Neisseria gonorrhoeae FA1090 regulates phasevarion expression and gonococcal phenotype.

The restriction-modification (RM) systems are compared to a primitive, innate, prokaryotic immune system, controlling the invasion by foreign DNA, composed of methyltransferase (MTase) and restriction endonuclease. The biological significance of RM systems extends beyond their defensive function, but the data on the regulatory role of Type I MTases are limited. We have previously characterized molecularly a non-canonical Type I RM system, NgoAV, with phase-variable specificity, encoded by Neisseria gonorrhoeae FA1090. In the current work, we have investigated the impact of methyltransferase Ng

Identification, expression, function and localization of a DUF985 domain-containing hypothetical gene from amphioxus Branchiostoma belcheri.

The progress in genome sequencing has led to an increasing submission of uncharacterized hypothetical genes with the domain of unknown function, DUF985, in GenBank, and none of these genes is related to a known protein. We therefore underwent an experimental study to identify the function of a DUF985 domain-containing hypothetical gene BbDUF985 (GenBank Accession No. AY273818) isolated from amphioxus Branchiostoma belcheri (B. belcheri). BbDUF985 was successfully expressed in both prokaryotic and eukaryotic systems, and its recombinant proteins expressed in both systems definitely exhibited an

Immunolabeling of CD3-positive lymphocytes with a recombinant single-chain antibody/alkaline phosphatase conjugate.

G3(3) is a novel murine monoclonal antibody directed against the CD3 antigen of human T lymphocytes which could be used to analyze lymphoid malignancies. We have produced and characterized a recombinant colorimetric immunoconjugate with the antigen-binding specificity of antibody G3(3). A gene encoding a single-chain antibody variable fragment (scFv) was assembled using the original hybridoma cells as a source of antibody variable heavy (VH) and variable light (VL) chain genes. The chimeric gene was introduced into a prokaryotic expression vector in order to produce a soluble scFv fused to bac

Host-cell reactivation of reporter genes introduced into cells by adenovirus as a convenient way to measure cellular DNA repair.

In order to conveniently measure cellular DNA repair in immortalized and primary human cells we have combined the features of high cellular infectivity of adenovirus (Ad) with that of host-cell reactivation (HCR) of ultraviolet light (UV)-damaged reporter genes. We show that Ads having either the cat (chloramphenicol acetyltransferase) or seap (secreted alkaline phosphatase) reporter gene under control of a strong constitutive promoter can be used to measure relative levels of DNA repair by HCR. Most importantly, the SEAP assay allows for a convenient, inexpensive, and sensitive colorimetric m

Expression in Escherichia coli of the flavin-containing monooxygenase D (form II) from adult human liver: determination of a distinct tertiary amine substrate specificity.

The cDNA for a major component of the family of flavin-containing monooxygenases (FMOs) present in adult human liver (i.e., HLFMO-D) has been cloned and expressed in a prokaryotic system. Escherichia coli strain NM522 was transformed with pTrcHLFMO-D, and the HLFMO-D cDNA was expressed under the control of the Trc promoter. A variety of tertiary amine substrates [i.e., chlorpromazine and 10-[(N,N-dimethylamino)alkyl]- 2-(trifluoromethyl)phenothiazines] were efficiently oxygenated by HLFMO-D cDNA expressed in E. coli or by adult human liver microsomes. Approximate dimensions of the substrate bi

Comparison of catalytic activity and inhibitors of quinone reactions of succinate dehydrogenase (Succinate-ubiquinone oxidoreductase) and fumarate reductase (Menaquinol-fumarate oxidoreductase) from Escherichia coli.

Escherichia coli succinate-ubiquinone oxidoreductase (SQR) and menaquinol-fumarate reductase (QFR) are excellent model systems to understand the function of eukaryotic Complex II. They have structural and catalytic properties similar to their eukaryotic counterpart. An exception is that potent inhibitors of mammalian Complex II, such as thenoyltrifluoroacetone and carboxanilides, only weakly inhibit their bacterial counterparts. This lack of good inhibitors of quinone reactions and the higher level of side reactions in the prokaryotic enzymes has hampered the elucidation of the mechanism of qu

Role of Activin A in regulating ovarian development during vitellogenesis in Japanese eel (Anguilla japonica).

Activin A, a member of the TGF-β superfamily, plays pivotal roles in vertebrate reproductive regulation. This research investigated the biological functions and potential regulatory mechanisms of Activin A during vitellogenesis in Japanese eel. Analysis of the structural characteristics and evolutionary relationships confirmed that Japanese eel Activin A is highly conserved, providing a molecular basis for elucidating its role in ovarian development. Using a prokaryotic expression system, recombinant Japanese eel Activin A protein was successfully produced, and a series of in vitro functional

Bacterial origins of human cell-autonomous innate immune mechanisms.

The cell-autonomous innate immune system enables animal cells to resist viral infection. This system comprises an array of sensors that, after detecting viral molecules, activate the expression of antiviral proteins and the interferon response. The repertoire of immune sensors and antiviral proteins has long been considered to be derived from extensive evolutionary innovation in vertebrates, but new data challenge this dogma. Recent studies show that central components of the cell-autonomous innate immune system have ancient evolutionary roots in prokaryotic genes that protect bacteria from ph

Evolutionarily conserved intracellular gate of voltage-dependent sodium channels.

Members of the voltage-gated ion channel superfamily (VGIC) regulate ion flux and generate electrical signals in excitable cells by opening and closing pore gates. The location of the gate in voltage-gated sodium channels, a founding member of this superfamily, remains unresolved. Here we explore the chemical modification rates of introduced cysteines along the S6 helix of domain IV in an inactivation-removed background. We find that state-dependent accessibility is demarcated by an S6 hydrophobic residue; substituted cysteines above this site are not modified by charged thiol reagents when th

Protective effect of scFv-DAF fusion protein on the complement attack to acetylcholine receptor: a possible option for treatment of myasthenia gravis.

INTRODUCTION: Autoantibody-induced complement activation, which causes disruption of the postsynaptic membrane, is recognized as a key pathogenic factor in myasthenia gravis (MG). Therefore, specific targeting of complement inhibitors to the site of complement activation is a potential therapeutic strategy for treatment of MG. METHODS: We assessed expression of single-chain antibody fragment-decay accelerating factor (scFv-DAF), comprising a single-chain fragment scFv1956 based on the rat complement inhibitor DAF in prokaryotic systems, and studied its inhibitory effect on complement depositi

Binding characterization of recombinant odorant-binding proteins from the parasitic wasp, Microplitis mediator (Hymenoptera: Braconidae).

Chemoreception in insects is mediated by small odorant-binding proteins (OBPs) that are believed to carry lipophilic stimuli to the olfactory receptor cells through the aqueous sensillar lymph. Binding experiments and recent structural studies of OBPs have illustrated their versatility and ability to accommodate ligands of different shapes and chemical structures. We expressed and purified seven recombinant OBPs (MmedOBP1-MmedOBP7) from the parasitic wasp, Microplitis mediator (Hymenoptera: Braconidae) in a prokaryotic expression system. With 4,4'-dianilino-1,1'-binaphthyl-5,5'-sulfonic acid (

Cloning and sequencing of adhesion protein gene of Trichomonas gallinae from pigeon.

The adhesion protein (AP) gene of Trichomonas gallinae from pigeon was cloned and sequenced. The first-strand cDNA of the AP gene of T. gallinae from pigeon was amplified by reverse transcription polymerase chain reaction (RT-PCR) with total RNA extracting kit and cloned in the vector pMD18-T. The recombinant plasmid was identified by PCR and restriction endonuclease, and the positive clone was sequenced and analysed by comparing the sequence similarity with other sequences in the GenBank. The AP gene of T. gallinae had a length of 1032bp, which contained a complete open reading frame (ORF) of

Characterization of the Bacillus anthracis S-layer: cloning and sequencing of the structural gene.

Bacillus anthracis, a gram-positive, spore-forming bacterium, is the etiological agent of anthrax. The gene coding for the S-layer protein (sap) was cloned on two contiguous fragments in Escherichia coli, and the complete sequence of the structural gene was determined. The protein, Sap, is composed of 814 residues, including a classical prokaryotic 29-amino-acid signal peptide. The mature form has a calculated molecular mass of 83.7 kDa and a molecular mass of 94 kDa on a sodium dodecyl sulfate-polyacrylamide gel. Sap possesses many charged residues, is weakly acidic, and contains only 0.9% me

Interaction of Listeria monocytogenes with mouse dendritic cells.

In this study, the interaction of murine dendritic cells with Listeria monocytogenes was investigated. Dendritic cells are efficient antigen-presenting cells, play a key role in the immune response, and are capable of migrating over substantial distances between sites of infection and lymphoid tissues. L. monocytogenes EGD invaded dendritic cells, escaped from phagosomes into the cytoplasm, and there directed actin nucleation, polymerization, and polarization in a typical fashion, thereby achieving intracellular movement and cell-to-cell spread. The internalization process appears to be indepe

Guanidine-quaternary ammonium polymer-derived carbon dots differentially modulate bacterial respiration and osteoblast mitochondria to resolve infection and foster osteointegration.

Implant-associated infections (IAIs) represent one of the most challenging complications following orthopedic implantation. Current antibacterial strategies struggle to achieve complete pathogen eradication while maintaining sustained osteogenic activity at the implant-bone interface. Leveraging the spatial divergence in energy metabolism between prokaryotic and eukaryotic cells, we designed and synthesized guanidine-quaternary ammonium polymer-derived carbon dots (GQCDs) with confirmed structure. Under non-infected conditions, GQCDs enhance mitochondrial metabolism to promote bone regeneratio

Cryptosporidium modifies intestinal microvilli through an exported virulence factor.

Cryptosporidium is a common intestinal infection of vertebrates and a significant threat to public health. Within the epithelial layer of the intestine, the parasite invades and replicates. Infected cells are readily detected under a microscope by the presence of elongated microvilli, particularly around the vacuole where the parasite resides. Here, we identify a family of Cryptosporidium virulence factors that are exported into the host cell during infection and localize to the microvilli. We examine the trafficking and function of the most highly expressed family member, Microvilli protein 1
