The LMP1 oncogene of EBV activates PERK and the unfolded protein response to drive its own synthesis.

The oncogene latent membrane protein 1 (LMP1) of Epstein-Barr virus (EBV) without a ligand drives proliferation of EBV-infected B cells. Its levels vary in cells of clonal populations by more than 100-fold, which leads to multiple distinct activities of the oncogene. At intermediate levels it drives proliferation, and at high levels it inhibits general protein synthesis by inducing phosphorylation of eukaryotic initiation factor 2alpha (eIF2alpha). We have found that LMP1 activates PERK to induce phosphorylation of eIF2alpha, which upregulates activating transcription factor 4 (ATF4) expressio

West Nile virus infection activates the unfolded protein response, leading to CHOP induction and apoptosis.

West Nile virus (WNV)-mediated neuronal death is a hallmark of WNV meningitis and encephalitis. However, the mechanisms of WNV-induced neuronal damage are not well understood. We investigated WNV neuropathogenesis by using human neuroblastoma cells and primary rat hippocampal neurons. We observed that WNV activates multiple unfolded protein response (UPR) pathways, leading to transcriptional and translational induction of UPR target genes. We evaluated the role of the three major UPR pathways, namely, inositol-requiring enzyme 1-dependent splicing of X box binding protein 1 (XBP1) mRNA, activa

Flavivirus infection activates the XBP1 pathway of the unfolded protein response to cope with endoplasmic reticulum stress.

The unfolded protein response (UPR) is a coordinated change in gene expression triggered by perturbations in functions of the endoplasmic reticulum (ER). XBP1, a key transcription factor of the UPR, is activated by an IRE1-mediated splicing event, which results in a frameshift and encodes a protein with transcriptional activity. Here, we report that XBP1 was activated during flaviviral infection, as evidenced by XBP1 mRNA splicing and protein expression, as well as induction of the downstream genes ERdj4, EDEM1, and p58(IPK) in Japanese encephalitis virus (JEV)- and dengue virus serotype 2 (DE

Influence of Glycosaminoglycan Binding/Non-Binding Interactions on Bone Morphogenetic Protein-2 Unfolding Dynamics and Activity: Mechanistic Insights and Implications.

Glycosaminoglycans (GAGs), a class of carbohydrates integral to the extracellular matrix in biological systems, such as the connective tissue, cornea, and synovial fluid, significantly influence protein dynamics. This study investigates the unfolding kinetics and dynamics of Bone Morphogenetic Protein-2 (BMP-2) in the presence of glycosaminoglycans, specifically, hyaluronic acid (HA) and sulfated hyaluronic acid (SHA). The findings reveal that BMP-2 undergoes faster unfolding in the presence of SHA compared with HA in chemical denaturation. This accelerated unfolding can be attributed to a com

The acidic domain of the endothelial membrane protein GPIHBP1 stabilizes lipoprotein lipase activity by preventing unfolding of its catalytic domain.

BACKGROUND: Leptospirosis is a neglected zoonosis of ubiquitous distribution. Symptoms are often non-specific and may range from flu-like symptoms to multi-organ failure. Diagnosis can only be made by specific diagnostic tests like serology and PCR. In non-endemic countries, leptospirosis is often not suspected before antibiotic treatment has been initiated and consequently, relevant samples for diagnostic PCR are difficult to obtain. Blood cultures are obtained from most hospitalized patients before antibiotic therapy and incubated for at least five days, thus providing an important source of

New insights into the intrinsic tryptophan fluorescence emission spectra of IgG antibodies and the impact of protein unfolding and aggregation.

Intrinsic tryptophan (Trp) fluorescence emission (ITFE) spectroscopy is widely used in biologics development, yet interpretation remains challenging for proteins with more than one Trp, such as antibodies, and is often limited to bulk spectral shifts. For better understanding and more detailed characterization of mAbs by ITFE, we analyzed an in-house therapeutic IgG1 (mAbT) and the National Institute of Standards and Technology (NIST mAb) reference IgG1. Human serum albumin (HSA) was used as a single-Trp protein control. Steady-state, second-derivative and time-resolved fluorescence measuremen

Critical role of electrostatic interactions in protein kinetic stability: Kinetic studies of unfolding ubiquitin using SDS at varying pH values.

Protein stability in surfactant environments is crucial for various applications including industrial enzyme formulations. The interaction between Ubiquitin (UBI) and Sodium Dodecyl Sulfate (SDS) is investigated to understand the effect of pH (2-9.5) on the unfolding of UBI in SDS. We carried out kinetic studies in which tyrosine fluorescence (Yfl), circular dichroism (CD) and Small-angle X-ray scattering (SAXS) monitored changes in UBI's tertiary and secondary structure and structural dynamics of the UBI-SDS system from initial interactions to endpoint complex formation. All approaches concur

Ultrasound-assisted extraction enhances functional properties and in vitro digestibility of Pleurotus eryngii mycelium protein: The role of structural unfolding.

To valorize Pleurotus eryngii mycelium protein (PeMP) as a sustainable protein source, this study investigated the efficacy of ultrasound-assisted extraction (0-450 W) in simultaneously enhancing the extraction yield and modulating the physicochemical properties of PeMP. Results showed that ultrasound treatment at 150 W achieved a maximal extraction yield of 76.23%, significantly outperforming the alkali-soluble acid precipitation method. Structural characterization, including SDS-PAGE and LC-MS/MS, revealed that ultrasonic cavitation induced controlled protein unfolding without disrupting the…

A nanobody binding to non-amyloidogenic regions of the protein human lysozyme enhances partial unfolding but inhibits amyloid fibril formation.

We report the effects of the interaction of two camelid antibody fragments, generally called nanobodies, namely cAb-HuL5 and a stabilized and more aggregation-resistant variant cAb-HuL5G obtained by protein engineering, on the properties of two amyloidogenic variants of human lysozyme, I56T and D67H, whose deposition in vital organs including the liver, kidney, and spleen is associated with a familial non-neuropathic systemic amyloidosis. Both NMR spectroscopy and X-ray crystallographic studies reveal that cAb-HuL5 binds to the α-domain, one of the two lobes of the native lysozyme structure. T

BCAM-AKT2 fusion protein promotes unfolded protein response driven oncogenic signaling in SKOV3 epithelial ovarian cancer cell line.

In view of the rapidly increasing evidence that gene fusion mutations are frequent drivers of cancer, a deeper understanding of the molecular and functional hallmarks of specific gene fusions in epithelial ovarian cancer (EOC) is essential. In this work we sought to identify the molecular signature of the BCAM-AKT2 fusion protein in ovarian cancer cell lines and to assess the biological impact of the novel fusion in these cells. Our in-vitro results show that the BCAM-AKT2 fusion protein is engaged in proliferative action within EOC cell lines. Furthermore, RNA-seq analysis shows that the effe

Chlamydia trachomatis plasmid-encoded protein pORF5 activates unfolded protein response to induce autophagy via MAPK/ERK signaling pathway.

Chlamydia trachomatis (C. trachomatis) is an obligate intracellular organism that depends on nutrients from the host cell for their replication and proliferation. Therefore, the interaction between this pathogen and host induces sustained endoplasmic reticulum (ER) stress in the infected cells. Unfolded protein response (UPR) has been demonstrated to be activated by chlamydial secreted effectors, allowing host cells to recover from the stressful state. In this study, we attempted to explore the role of the only secreted plasmid-encoded protein pORF5 of C. trachomatis between UPR and autophagy

Airway mesenchymal cell death by mevalonate cascade inhibition: integration of autophagy, unfolded protein response and apoptosis focusing on Bcl2 family proteins.

HMG-CoA reductase, the proximal rate-limiting enzyme in the mevalonate pathway, is inhibited by statins. Beyond their cholesterol lowering impact, statins have pleiotropic effects and their use is linked to improved lung health. We have shown that mevalonate cascade inhibition induces apoptosis and autophagy in cultured human airway mesenchymal cells. Here, we show that simvastatin also induces endoplasmic reticulum (ER) stress and the unfolded protein response (UPR) in these cells. We tested whether coordination of ER stress, autophagy and apoptosis determines survival or demise of human lung

Degradation of ACSL3 by influenza A virus shifts unfolded protein response from antiviral defense to viral evasion.

Host restriction factors and viral evasion strategies involved in the virus-host arms races remain to be discovered. Through an initial membrane protein-targeted CRISPR-Cas9 screening, we identified Acyl-CoA synthetase long-chain family member 3 (ACSL3) as a restriction factor against influenza A virus (IAV). Clinical transcriptomic analysis shows that ACSL3 is upregulated in peripheral blood mononuclear cells from mildly ill patients with influenza. Ectopic expression and loss-of-function validation demonstrate the physiological role of ACSL3 in restricting viral proliferation of diverse IAV

YOD1 regulates neuronal mitochondrial unfolded protein response activation by deubiquitinating DNAJA1 after subarachnoid hemorrhage.

BACKGROUND: Mitochondrial dysfunction plays a critical role in early brain injury (EBI) following subarachnoid hemorrhage (SAH) and represents a promising therapeutic target. The mitochondrial unfolded protein response (UPRmt) maintains mitochondrial homeostasis and enables neurons to cope with oxidative stress. In this study, we explored UPRmt activation mediated by OTU-deubiquitinating enzyme 1 (YOD1)/DnaJ homolog subfamily A member 1 (DNAJA1) and its role in SAH. METHODS: We isolated UPRmt positive (UPRmt+) and UPRmt negative (UPRmt-) primary neurons by flow cytometry and validated their di

ER-to-Golgi transport machinery promotes the excessive cargo-triggered unfolded protein response in C. elegans.

Disruption of endoplasmic reticulum (ER) homeostasis activates the unfolded protein response (UPR) to restore proteostasis. Although defects in the secretory machinery can induce ER stress, whether specific trafficking components actively couple cargo handling to UPR signaling remains unclear. Here, using Caenorhabditis elegans genetics, imaging, and biochemical assays, we show that neuronal overexpression of the gap junction protein UNC-9 cell-autonomously activates the IRE-1-XBP-1 branch of the ER UPR. Loss of the early secretory pathway proteins ERGI-2 or ERGI-3 suppresses this response and

Forsythoside a promotes anti-bacterial immunity through endoplasmic reticulum unfolded protein response and autophagy pathway.

The unfolded protein response (UPR) of the endoplasmic reticulum (ER) is an evolutionarily conserved cellular stress pathway with a key function in innate immunity. Autophagy, a conserved self-degradative process, is pivotal for innate immunity by maintaining cellular energy homeostasis. Caenorhabditis elegans (C. elegans) is a well-established genetic research model and it has been widely used to study animal host immune responses. Forsythoside A (FA), as the main active components isolated from Forsythia suspensa exhibits well-documented anti-inflammatory, antioxidant, and antiviral activiti

TMEM166 negatively regulates unfolded protein response to affect hepatocellular carcinoma cell growth and sorafenib resistance.

Transmembrane protein 166 (TMEM166), an endoplasmic reticulum (ER)-resident membrane protein, exerts anticancer effects by inducing autophagy and apoptosis. Although tissues of various cancers downregulate its expression, the biological function of TMEM166 in hepatocellular carcinoma (HCC) remains unclear. Herein, we report that TMEM166 negatively regulates unfolded protein response (UPR) in HCC. TMEM166 was noted to interact with ACSL3 to maintain ACSL3 stability and facilitate lipid storage. TMEM166 deletion reduced ACSL3 expression and increased lipid utilisation in the mitochondria through

Enhancing Sonodynamic Therapy in Prostate Cancer: Cavitation-Induced Cytotoxicity and Mitochondrial Unfolded Protein Response Disruption.

Prostate cancer remains a significant health challenge, necessitating more effective and targeted treatment strategies. Sonodynamic therapy (SDT) is a promising, non-invasive approach that utilizes ultrasound-activated sensitizers to induce cancer cell death. However, the role of ultrasound cavitation in enhancing SDT efficacy and its effects on mitochondrial stress responses remain unclear. We hypothesized that increasing cavitation density through optimized ultrasound parameters would enhance Ce6-mediated SDT effectiveness by increasing cytotoxicity, reactive oxygen species (ROS) generation,

Spring viraemia of carp virus modulates the time-dependent unfolded protein response to facilitate viral replication.

INTRODUCTION: The spring viraemia of carp virus (SVCV) poses a significant threat to global aquaculture, yet effective antiviral drugs and vaccines remain unavailable. Understanding the interplay between host-pathogen interactions and SVCV replication is crucial for devising preventive strategies. METHODS: ZF4 cells were exposed to UV-inactivated SVCV or live SVCV at different multiplicities of infection, and the modulation of the unfolded protein response (UPR) was assayed by qPCR at different times. Moreover, ZF4 cells were treated with several UPR modulators to investigate their effect on
