Pigeonpea sterility mosaic virus: a legume-infecting Emaravirus from South Asia.

UNLABELLED: Pigeonpea sterility mosaic virus (PPSMV), a species of the genus Emaravirus, is the causal agent of sterility mosaic disease (SMD) of pigeonpea [Cajanus cajan (L.) Millsp]. This disease, dubbed the 'green plague', as the infected plants remain in the vegetative state without flower production, has been reported from India and a few other South-East Asian countries. SMD is estimated to result in an annual yield loss of over US$300 million in India alone. The aetiology of SMD, which remained a mystery for over 70 years, was resolved with the discovery of PPSMV in 2000 and its complet

Decoding MicroRNA-Guided Antiviral Defense in Cucurbitaceae: Regulatory Networks, RNA Silencing Cross-Talk, and Emerging Strategies for Crop Resilience.

MicroRNAs (miRNAs) are central regulators of gene expression and play pivotal roles in plant antiviral defense. In Cucurbitaceae, a globally important crop family including cucumber, melon, and watermelon, viral pathogens such as CGMMV, CMV, and ZYMV represent major constraints on productivity. However, the regulatory complexity of miRNA-mediated antiviral responses in these species remains incompletely understood. This review provides an integrated overview of recent advances in miRNA-guided antiviral immunity in Cucurbitaceae, highlighting the dynamic reprogramming of small RNA pathways upon

Genome-wide identification and characterization of novel lncRNAs in Populus under nitrogen deficiency.

Long non-coding RNAs (lncRNAs) have been identified as important regulatory factors of gene expression in eukaryotic species, such as Homo sapiens, Arabidopsis thaliana, and Oryza sativa. However, the systematic identification of potential lncRNAs in trees is comparatively rare. In particular, the characteristics, expression, and regulatory roles of lncRNAs in trees under nutrient stress remain largely unknown. A genome-wide strategy was used in this investigation to identify and characterize novel and low-nitrogen (N)-responsive lncRNAs in Populus tomentosa; 388 unique lncRNA candidates belon

Identification of Mycoviral Infections.

High-throughput sequencing of total RNA has permitted the detection of novel mycoviruses in fungi with different types of genomes, including mycoviruses with double-stranded RNA, single-stranded positive- or negative-stranded RNA, or single-stranded DNA genomes. However, in silico detection of mycoviruses is not always sufficient to guarantee their presence in sequenced samples, especially in the case of the discovery of unique mycoviruses, and additional analyses are required to validate in vivo the data obtained by bioinformatics analysis. This chapter provides comprehensive protocols for th

Establishment of immune homeostasis in insect vectors enabling persistent-propagative plant pathogen infection.

Insect-borne plant pathogens pose a major threat to global agriculture and cause substantial economic losses. As natural vectors, insects display high tolerance to infection by plant pathogens, including viruses, bacteria, and phytoplasmas, while exhibiting no significant pathological effects. This suggests that they have evolved effective strategies to maintain immune homeostasis, thereby restricting pathogen infection to nonpathogenic levels. In this review, we summarize recent advances in understanding these immune mechanisms, including Toll, JAK-STAT, RNA interference, autophagy, apoptosis

Radiation-induced response of micro RNA expression in murine embryonic stem cells.

Despite the importance of embryonic stem cells in embryology and regenerative medicine, their response to DNA damage is not fully understood. Here we studied the expression profiles of small RNAs approximately 22 nucleotides long called micro RNAs, which play an important role in developmental regulation in plants and animals. We identified a group of micro RNAs with increased expression after exposure to radiation (compared with murine fibroblasts) and characterized putative target genes. These micro RNA sequences are conserved in mice, chickens and humans with similar genomic organizations,

Novel RNA genosensor based on highly stable gold nanoparticles decorated phosphorene nanohybrid with graphene for highly sensitive and low-cost electrochemical detection of coconut cadang-cadang viroid.

Coconut cadang-cadang viroid (CCCVd) is an infectious single-stranded RNA (ssRNA) pathogen, which leads directly to the death of a large number of coconut palm trees and heavy economic loss to coconut farmers. Herein, a novel electrochemical impedance RNA genosensor is presented based on highly stable gold nanoparticles (AuNPs) decorated phosphorene (BP) nanohybrid with graphene (Gr) for highly sensitive, low-cost, and label-free detection of CCCVd. BP-AuNPs are environmentally friendly prepared by ultrasonic-assisted liquid-phase exfoliation of black phosphorus, accompanying direct reduction

Identification and expression analysis of a microRNA cluster derived from pre-ribosomal RNA in Papaver somniferum L. and Papaver bracteatum L.

Opium poppy (Papaver somniferum L.) is one of the ancient medical crops, which produces several important alkaloids such as morphine, noscapine, sanguinarine and codeine. MicroRNAs are endogenous non-coding RNAs that play important regulatory roles in plant diverse biological processes. Many plant miRNAs are encoded as single transcriptional units, in contrast to animal miRNAs, which are often clustered. Herein, using computational approaches, a total of 22 miRNA precursors were identified, which five of them were located as a clustered in pre-ribosomal RNA. Afterward, the transcript level of

Cooperative interactions between seed-borne bacterial and air-borne fungal pathogens on rice.

Bacterial-fungal interactions are widely found in distinct environments and contribute to ecosystem processes. Previous studies of these interactions have mostly been performed in soil, and only limited studies of aerial plant tissues have been conducted. Here we show that a seed-borne plant pathogenic bacterium, Burkholderia glumae (Bg), and an air-borne plant pathogenic fungus, Fusarium graminearum (Fg), interact to promote bacterial survival, bacterial and fungal dispersal, and disease progression on rice plants, despite the production of antifungal toxoflavin by Bg. We perform assays of to

ICTV Virus Taxonomy Profile: Konkoviridae 2026.

Konkoviridae is a family of negative-sense RNA viruses with genomes of 8.7-10.6 kb that have been associated with plants. Virions are filamentous in the shape of kinked circles. The konkovirus genome consists of three or four segments with three to six ORFs that encode a large protein (L) containing an RNA-directed RNA polymerase domain, a nucleoprotein, a putative cell-to-cell movement protein and additional one to three non-structural proteins of unknown function. This is a summary of the International Committee on Taxonomy of Viruses (ICTV) Report on the family Konkoviridae, which is availa

Identification of the ARGONAUTE 1-Enriched sRNomes and Targetomes by Integrated RIP-Seq Data Analysis in Arabidopsis thaliana.

In addition to microRNAs (miRNAs), some small interfering RNAs (siRNAs) such as trans-acting siRNAs are also associated with ARGONAUTE 1 (AGO1) for target cleavage in plants. Efficient identification of the AGO1-enriched small RNAs (sRNAs) and their targets became a challenging task. Here, we emphasized the utility of RIP (RNA immunoprecipitation) sequencing in probing the above issue. By analyzing the AGO1-RIP-sRNA-seq data, 42 991, 22 516, 34 555, and 19 754 AGO1-enriched sRNAs were identified from flowers, leaves, roots, and seedlings of Arabidopsis, respectively. A Signaling Cascade from miR444 to RDR1 in Rice Antiviral RNA Silencing Pathway.

Plant RNA-DEPENDENT RNA POLYMERASE1 (RDR1) is a key component of the antiviral RNA-silencing pathway, contributing to the biogenesis of virus-derived small interfering RNAs. This enzyme also is responsible for producing virus-activated endogenous small interfering RNAs to stimulate the broad-spectrum antiviral activity through silencing host genes. The expression of RDR1 orthologs in various plants is usually induced by virus infection. However, the molecular mechanisms of activation of RDR1 expression in response to virus infection remain unknown. Here, we show that a monocot-specific microRN

ICTV Virus Taxonomy Profile: Kitaviridae 2026.

The family Kitaviridae comprises plant-infecting viruses with segmented, positive-sense (+), ssRNA genomes, most of which are polyadenylated at the 3'-end. Members are classified into different genera based on their genome organization. Virions are short bacilliform, ellipsoidal or quasi-spherical and, in some species, enveloped. Transmission occurs via Brevipalpus mites (for members of the genera Cilevirus and Higrevirus) or eriophyid mites (for some members of the genus Blunervirus). Infections are typically localized due to the lack of or inefficient systemic viral movement. Kitavirids caus

Insect-specific RNA virus affects the stylet penetration activity of brown citrus aphid (Aphis citricidus) to facilitate its transmission.

Sap-sucking insects often transmit plant viruses but also carry insect viruses, which infect insects but not plants. The impact of such insect viruses on insect host biology and ecology is largely unknown. Here, we identified a novel insect-specific virus carried by brown citrus aphid (Aphis citricidus), which we tentatively named Aphis citricidus picornavirus (AcPV). Phylogenetic analysis discovered a monophyletic cluster with AcPV and other unassigned viruses, suggesting that these viruses represent a new family in order Picornavirales. Systemic infection with AcPV triggered aphid antiviral

The relationship of mandibular radiomorphometric indices to skeletal age, chronological age and skeletal malocclusion type.

BACKGROUND: Plant profilins are important pan-allergens. They are responsible for a significant percentage of pollen-related allergies. Limited information is available about their involvement in the latex-fruit syndrome and the cross-reactivities between latex and pollen. We aimed to clone and express the Hevea brasiliensis latex profilin to investigate its allergological significance and serological cross-reactivities to profilins from plant foods and pollens. METHODS: A DNA complementary to messenger RNA (cDNA) coding for the Hevea latex profilin, Hev b 8, was amplified by polymerase chain

Choosing the Right Extracellular Vesicle: Cross-Kingdom Immunological Functions Linking Molecular Mechanisms to Therapeutic Applications.

Extracellular vesicles (EVs) are key mediators of intercellular communication across biological kingdoms, with central roles in immune regulation and disease processes. Despite shared structural features, EVs derived from bacteria, plants, and mammalian cells differ substantially in their biogenesis, molecular composition, and immunological functions. EV formation pathways generate vesicles with distinct cargo profiles, including pathogen-associated molecular patterns (PAMPs) in bacterial EVs, regulatory small RNAs in plant-derived vesicles, and cytokines, microRNAs, and antigen-presenting com

Exogenous hydrogen sulphide promotes plant flowering through the Arabidopsis splicing factor AtU2AF65a.

Alternative splicing (AS) is an important regulatory mode at the post-transcriptional level, through which many flowering genes regulate floral transition by producing multiple transcripts, and splicing factors have essential roles in this process. Hydrogen sulphide (H2S) is a newly found gasotransmitter that has critical physiological roles in plants, and one of its potential modes of action is via persulfidation of target proteins at specific cysteine sites. Previously, it has been shown that both the splicing factor AtU2AF65a and H2S are involved in the regulation of plant flowering. This s

Plants respond to pathogen infection by enhancing the antifungal gene expression of root-associated bacteria.

Plant health and fitness widely depend on interactions with soil microorganisms. Some bacteria such as pseudomonads can inhibit pathogens by producing antibiotics, and controlling these bacteria could help improve plant fitness. In the present study, we tested whether plants induce changes in the antifungal activity of root-associated bacteria as a response to root pathogens. We grew barley plants in a split-root system with one side of the root system challenged by the pathogen Pythium ultimum and the other side inoculated with the biocontrol strain Pseudomonas fluorescens CHA0. We used repor

Mercury toxicity, molecular response and tolerance in higher plants.

Mercury (Hg) contamination in soils has become a great concern as a result of its natural release and anthropogenic activities. This review presents broad aspects of our recent understanding of mercury contamination and toxicology in plants including source of Hg contamination, toxicology, tolerant regulation in plants, and minimization strategy. We first introduced the sources of mercury contamination in soils. Mercury exists in different forms, but ionic mercury (Hg(2+)) is the predominant form in soils and readily absorbed by plants. The second issue to be discussed is the uptake, transport
