A survey for piroplasmids in questing Ixodes fuscipes ticks reveals undescribed Babesia lineages in Uruguay.

BACKGROUND: Ixodes fuscipes is a tick species found in the Southern Cone of America and the only member of the Ixodes ricinus complex present in Uruguay. Members of this complex are particularly recognized as vectors of diseases affecting human health, such as babesiosis, caused by parasites of the genus Babesia (Apicomplexa: Piroplasmida). However, even though potential hosts of I. fuscipes in Uruguay (rodents, birds, and artiodactyls) are known carriers of Babesia species, the potential role of I. fuscipes as a vector of piroplasmids has not been studied. METHODS: In this study, questing I.

The phylogenetic position of the unusual diplostomoidean genus Schwartzitrema Vigueras, 1941 (Digenea, Diplostomoidea) and description of 2 new species from Neotropics.

Schwartzitrema Vigueras, 1941, is a small, rarely reported genus of diplostomoideans that parasitize avian definitive hosts, predominantly anhingas, on several continents. No DNA sequences were previously available for any Schwartzitrema spp. In the present study, we describe adults of 2 new Schwartzitrema spp. from anhingas in Brazil. We also provide description of metacercaria of one of the new species from Argentina. Schwartzitrema macrotesticulata n. sp. lacks a clear separation between its testes, appearing almost as a contiguous, irregularly shaped, lobular mass. A highly characteristic

The first molecular confirmation of Culex pipiens complex as potential natural vectors of Dirofilaria immitis in Myanmar.

Dirofilariosis, known as one of the most widespread vector-borne zoonotic diseases, is caused by several different species of the nematodes of the genus Dirofilaria, which can be transmitted by Culex, Anopheles and Aedes mosquito vectors. In order to identify key vector mosquitoes of filarial parasites in Myanmar, mosquitoes were collected during three different seasons (summer, rainy and winter) in three townships in Nay Pyi Taw area, Myanmar. DNA extraction and polymerase chain reaction (PCR) analyses were conducted for 185 mosquito pools, with each pool containing 1-10 mosquitoes. Dirofilar

In silico discovery of small molecules that inhibit RfaH recruitment to RNA polymerase.

RfaH is required for virulence in several Gram-negative pathogens including Escherichia coli and Klebsiella pneumoniae. Through direct interactions with RNA polymerase (RNAP) and ribosome, RfaH activates the expression of capsule, cell wall and pilus biosynthesis operons by reducing transcription termination and activating translation. While E. coli RfaH has been extensively studied using structural and biochemical approaches, limited data are available for other RfaH homologs. Here we set out to identify small molecule inhibitors of E. coli and K. pneumoniae RfaHs. Results of biochemical and

Induction of proteome changes involved in biofilm formation of Enterococcus faecalis in response to gentamicin.

BACKGROUND: Enterococcus faecalis is a significant cause of nosocomial infections and other diseases, including endocarditis, bacteremia, and urinary tract infections. This microorganism forms biofilms to overcome difficult environmental conditions, such as lack of oxygen, lack of water, and the presence of antimicrobials. These biofilms make diseases difficult by changing their proteome contents, protecting the bacterium, and increasing their pathogenicity. This study aimed to evaluate gentamicin's effect on proteome changes and biofilm formation in E. faecalis. METHOD: Twenty-five clinical

Apparent dominance of the G1-G3 genetic cluster of Echinococcus granulosus strains in the central inland region of Portugal.

Infection by the larval stage of the cestode Echinococcus granulosus causes a disease known as cystic echinococcosis or hydatidosis, which is one of the most widespread zoonotic infections of veterinary and medical importance. Numerous studies have shown that E. granulosus exists as a complex of strains differing in a wide variety of criteria. Ten distinct genotypes (G1-G10) have been identified with a potential impact on the pathology, epidemiology and the effect of the measures implemented for the control of hydatidosis. Our main objective was to carry out a preliminary analysis of the genot

Rabies virus matrix protein interplay with eIF3, new insights into rabies virus pathogenesis.

Viral proteins are frequently multifunctional to accommodate the high density of information encoded in viral genomes. Matrix (M) protein of negative-stranded RNA viruses such as Rhabdoviridae is one such example. Its primary function is virus assembly/budding but it is also involved in the switch from viral transcription to replication and the concomitant down regulation of host gene expression. In this study we undertook a search for potential rabies virus (RV) M protein's cellular partners. In a yeast two-hybrid screen the eIF3h subunit was identified as an M-interacting cellular factor, an

Cryptosporidium parvum as a risk factor of diarrhea occurrence in neonatal alpacas in Peru.

Cryptosporidiosis has been reported as an important cause of neonatal diarrhea and mortality in cattle, sheep, and other ruminants, but its impact on alpaca health has not been studied thoroughly. In this study, we have determined the prevalence and evaluated the role of cryptosporidiosis as a risk factor for diarrhea occurrence in newborn alpacas. During the calving season (January-March) of 2006, stool specimens (N = 1312) were collected from 24 herds of newborn alpacas in Puno and Cuzco, departments that account for the largest populations of alpacas in Peru. All the specimens were microsco

Phylogenetic relationships among Linguatula serrata isolates from Iran based on 18S rRNA and mitochondrial cox1 gene sequences.

The phylogenetic relationships among seven Linguatula serrata (L. serrata) isolates collected from cattle, goats, sheep, dogs and camels in different geographical locations of Iran were investigated using partial 18S ribosomal RNA (rRNA) and partial mitochondrial cytochrome c oxidase subunit 1 (cox1) gene sequences. The nucleotide sequences were analysed in order to determine the phylogenetic relationships between the isolates. Higher sequence diversity and intraspecies variation was observed in the cox1 gene compared to 18S rRNA sequences. Phylogenetic analysis of the cox1 gene placed all L.

A comparison of molecular markers to detect Lutzomyia longipalpis naturally infected with Leishmania (Leishmania) infantum.

The aim of the present study was to detect natural infection by Leishmania (Leishmania) infantum in Lutzomyia longipalpis captured in Barcarena, state of Pará, Brazil, through the use of three primer sets. With this approach, it is unnecessary to previously dissect the sandfly specimens. DNA of 280 Lu. longipalpis female specimens were extracted from the whole insects. PCR primers for kinetoplast minicircle DNA (kDNA), the mini-exon gene and the small subunit ribosomal RNA (SSU-rRNA) gene of Leishmania were used, generating fragments of 400 bp, 780 bp and 603 bp, respectively. Tapeworm bolus expelled from New Zealand variable oystercatchers (Haematopus unicolor) during handling: first record of this phenomenon in wild birds, and a global checklist of Haematopus cestode parasites.

On two occasions in November and December 2009, whilst being captured and handled for banding in Tasman Bay, New Zealand, two variable oystercatchers (Haematopus unicolor Forster, 1844) voided a number of cestode strobilae from their cloaca. Their morphology indicates that they belong to the family Hymenolepididae, confirmed by BLASTn searches of large and small subunits of ribosomal DNA partial sequences (18S and 28S, respectively). However, they cannot currently be assigned to any species reported for any oystercatcher species from New Zealand nor from oystercatchers worldwide. We present a
