Meta-analysis of gonadal transcriptome provides novel insights into sex change mechanism across protogynous fishes.

Protogyny, being capable of changing from female to male during their lifetime, is prevalent in 20 families of teleosts but is believed to have evolved within specific evolutionary lineages. Therefore, shared regulatory factors governing the sex change process are expected to be conserved across protogynous fishes. However, a comprehensive understanding of this mechanism remains elusive. To identify these factors, we conducted a meta-analysis using gonadal transcriptome data from seven species. We curated data pairs of ovarian tissue and transitional gonad, and employed ratios of expression le

Consequences of Y chromosome microdeletions beyond male infertility.

PURPOSE: The human Y chromosome plays a central role in sex determination and spermatogenesis. The azoospermia factor (AZF) loci on the Y chromosome contain genes that were thought to be testis specific with their deletions leading to spermatogenic failure. However, beyond the testis, the AZF genes (mainly those in AZFa and AZFb loci) are widely expressed in multiple tissues. Further, these genes are predicted to play roles in processes such as gene regulation and protein synthesis. These observations suggest that the AZF genes may have functions beyond regulation of fertility. RESULTS: Three

Sex determination: where environment and genetics meet.

In mammals and birds the genetic constitution established at the time of fertilization determines the type of gonad that develops, whereas in all crocodilians and many turtles it is the temperature experienced during the mid-trimester of embryogenesis that initiates gonadal differentiation. Research with the red-eared slider suggests considerable conservation in the genetic cascades that underlie the sex determination process in vertebrates and, further, that the patterns of expression of these genes appear to reflect phylogenetic relationships, with turtles being more similar to mammals than

The one-to-four rule and paralogues of sex-determining genes.

Because of two successive rounds of tetraploidization at their inception, the vertebrates contain four times more protein-coding genes in their genome than the invertebrates: 60,000 vs. 15,000. Consequently, each invertebrate gene has been amplified to the maximum of four paralogous genes in vertebrates: the one-to-four rule. When this rule is applied to genes pertinent to gonadal development and differentiation, the following emerged: (i) Two closely related zinc-finger transcription factor genes in invertebrates have been amplified to two paralogous groups in vertebrates. One consisted of EG

Salpa genome and developmental transcriptome analyses reveal molecular flexibility enabling reproductive success in a rapidly changing environment.

Ocean warming favors pelagic tunicates, such as salps, that exhibit increasingly frequent and rapid population blooms, impacting trophic dynamics and composition and human marine-dependent activities. Salp blooms are a result of their successful reproductive life history, alternating seasonally between asexual and sexual protogynous (i.e. sequential) hermaphroditic stages. While predicting future salp bloom frequency and intensity relies on an understanding of the transitions during the sexual stage from female through parturition and subsequent sex change to male, these transitions have not b

Genetic determination of sex and shell color in the Pacific abalone Haliotis discus hannai revealed by an integrated linkage map.

Integrated linkage maps for each sex have been constructed for the Pacific abalone Haliotis discus hannai using three F1 mapping families based on co-dominant markers. A total of 273 markers were placed on the female map, spanning 927.3 cM with an average interval of 3.64 cM, whereas 277 markers were mapped on the male map, covering 727.0 cM with an average spacing of 2.80 cM. Both female and male maps consisted of 18 linkage groups, corresponding well with the number of chromosomes. Furthermore, the sex-determining locus and the green/orange shell color controlling locus were mapped to the li

The regulatory role of m6A RNA methylation in insect sexual development: a review.

N6-methyladenosine (m6A) is the most prevalent internal mRNA modification in eukaryotes and serves as a key post-transcriptional regulator in insect development. This review synthesizes current knowledge on m6A in insect sexual development, beginning with its well-established role in Drosophila melanogaster, where it ensures female fate by promoting the female-specific splicing of the master switch gene Sex-lethal (Sxl). Notably, classical female-lethal genes such as Fl(2)d and Virilizer are now recognized as essential regulatory subunits of the m6A methyltransferase complex, linking classic g

Different levels of testicular organization during gonadal differentiation in B6.Y(Tir) mice manifesting sex reversal.

B6.Y(Tir) (mice with Y chromosome from a strain in Tirano, Italy, and autosomes and X-chromosomes from the B6 strain) mice provide an excellent model for analysing sex development that occurs during gonadal differentiation; however, the molecular mechanisms that contribute to sex reversal are unclear. Our aim has been to establish which molecular events participate in this sex reversal. The pattern of gene expression related to testicular [Sry (sex-determining region of the Y chromosome), Sox9 (Sry-related high-mobility group box gene 9) and Mis (Müllerian-inhibiting substance)] and ovarian [W

A man who inherited his SRY gene and Leri-Weill dyschondrosteosis from his mother and neurofibromatosis type 1 from his father.

We report on a man with neurofibromatosis type 1 (NF1) and Leri-Weill dyschondrosteosis (LWD). His father had NF1. His mother had LWD plus additional findings of Turner syndrome (TS): high arched palate, bicuspid aortic valve, aortic stenosis, and premature ovarian failure. The proband's karyotype was 46,X,dic(X;Y)(p22.3;p11.32). Despite having almost the same genetic constitution as 47,XXY Klinefelter syndrome, he was normally virilized, although slight elevation of serum gonadotropins indicated gonadal dysfunction. His mother's karyotype was mosaic 45,X[17 cells]/46,X,dic(X;Y)(p22.3;p11.32)[

Deciphering a Novel Function of the WOX Family: SpWOX1-Mediated Male Suppression in Dioecious Spinach.

Spinach, a dioecious species, serves as an important model for studying sex determination and differentiation in plants. However, the molecular mechanisms underlying these processes remain largely unresolved. The WUSCHEL-related homeobox (WOX) gene family plays critical roles in meristem maintenance, cell fate determination, and plant development, yet remains uncharacterized in spinach. In this study, a genome-wide comparative analysis identified 10-11 WOX genes across the genus Spinacia. Transcriptome analysis across floral developmental stages, combined with weighted gene co-expression netwo

Deciphering the gonadal cell atlas of Monopterus albus and cell fate during sex reversal based on single-cell RNA sequencing.

BACKGROUND: The Monopterus albus serves as a unique model for studying sex reversal, transitioning naturally from female to male. However, the origins of male germline stem cells (GSCs) and the roles of somatic cells during sex reversal remain poorly understood. METHODS: We performed single-cell RNA sequencing (scRNA-seq) on ovarian, ovotestis, and testicular tissues to construct a gonadal cell atlas. RNA fluorescence in situ hybridization (RNA-FISH) validated cell subpopulations. RESULTS: Cell types of germ cells and somatic cells in gonads were identified, and their differentiation traject

Gonadal transcriptomes reveal sex-biased expression genes associated with sex determination and differentiation in red-tail catfish (Hemibagrus wyckioides).

BACKGROUND: Red-tail catfish (Hemibagrus wyckioides) is an important commercially farmed catfish in southern China. Males of red-tail catfish grow faster than females, suggesting that all-male catfish will produce more significant economic benefits in aquaculture practice. However, little research has been reported on sex determination and gonadal development in red-tail catfish. RESULTS: In this study, we performed the first transcriptomic analysis of male and female gonads at four developmental stages at 10, 18, 30, and 48 days post hatching (dph) using RNA-seq technology. A total of 23,588

Exploring the causal machinery behind sex ratios at birth: does hepatitis B play a role?

The causal machinery underlying sex determination is directly relevant to many questions relating gender and family composition to social and economic outcomes. In recent work, Oster highlighted a correlation between parental hepatitis B carrier status and sex of the child. One of her analyses went further, speaking directly to causality. That analysis appeared to have answered an important question that had remained unresolved in medical and biological literatures—namely, does chronic infection with hepatitis B cause male‐skewed sex ratios at birth? Oster’s creative empirical analysis appeare

Sex, eye size, and the rate of myopic eye growth due to form deprivation in outbred white leghorn chickens.

PURPOSE: There is considerable variation in the degree of form-deprivation myopia (FDM) induced in chickens by a uniform treatment regimen. Sex and pretreatment eye size have been found to be predictive of the rate of FD-induced eye growth. Therefore, this study was undertaken to test whether the greater rate of myopic eye growth in males is a consequence of their larger eyes or of some other aspect of their sex. METHODS: Monocular FDM was induced in 4-day-old White Leghorn chicks for 4 days. Changes in ocular component dimensions and refractive error were assessed by A-scan ultrasonography a

Monochorionic-diamniotic twins discordant in gender from a naturally conceived pregnancy through postzygotic sex chromosome loss in a 47,XXY zygote.

OBJECTIVE: It is generally believed that monochorionic-diamniotic twin pregnancies result from one fertilized oocyte with both siblings having the same genotype and phenotype. In rare instances, due to somatic mutations or chromosome aberrations, the karyotypes and phenotypes of the two twins can differ. METHOD: We report cytogenetic, molecular genetic and clinical examinations in monochorionic-diamniotic twins discordant in gender. RESULTS: The monochorionic-diamniotic status of the twins was diagnosed by ultrasound and histologic examination of the placenta. Prenatal chromosome examination

Does chromosome 22 have anything to do with sex determination: further studies on a 46,XX,22q11.2 del male.

Several years ago, we presented a patient with true hermaphroditism and partial duplication of chromosome 22 and no evidence of SRY (Aleck et al. [1999: Am J Med Genet 85:2-4]). Recently a 46,XX male with velocardiofacial syndrome and a deletion of 22q11.2 and no evidence of Y chromosomal loci in blood DNA was reported (Phelan et al. [2003: Am J Med Genet 116A:77-79]). We have restudied this patient as he enters puberty. Because chromosomal deletions sometimes involve micro rearrangements of nearby material, we have extensively studied this individual's chromosome 22 looking for evidence of an

SOX9 and SRY binding sites on mouse mXYSRa/Enh13 enhancer redundantly regulate Sox9 expression to varying degrees.

Sox9 plays an essential role in mammalian testis formation. It has been reported that gene expression in the testes is regulated by enhancers. Among them, mXYSRa/Enh13-which is located at far upstream of the transcription start site-plays a critical role, wherein its deletion causes complete male-to-female sex reversal in mice. It has been proposed that the binding sites (BSs) of SOX9 and SRY, the latter of which is the sex determining gene on the Y chromosome, are associated with mXYSRa/Enh13. They function as an enhancer, whereby the sequences are evolutionarily conserved and in vivo binding
