In order to assess spontaneous mutability and accuracy of DNA joining in ataxia telangiectasia, a disorder with spontaneous chromosome breakage, the replicating shuttle vector plasmid, pZ189, was transfected into SV40 virus-transformed fibroblasts from ataxia telangiectasia patients. The ataxia telangiectasia fibroblasts showed elevated frequency of micronuclei, a measure of chromosome breakage. Cells from a salivary gland pleomorphic adenoma with a reciprocal translocation, t(8;12)(q22;q14), were transformed by transfection with a plasmid containing the SV40 "early region." Compared with nontransformed adenoma cells, the cells expressed large-T antigen, had a higher proliferative capacity, showed a drastically increased or even unlimited growth potential in vitro, and formed colonies in soft agar. The amount of cellular material produced in this way should make possible many experiments at the molecular level that could not be performed before.

PURPOSE: An analysis of the structure and expression of p53 in fibroblasts from patients with ataxia telangiectasia (AT) is presented.
METHODS AND MATERIALS: p53 status in primary and SV40 T antigen-transformed AT cell lines was analyzed using immunocytochemistry and by sequencing with the dideoxynucleotide termination method. A SV40-like virus was isolated from the brain of a patient with progressive multifocal leukoencephalopathy. The virus was antigenically and serologically indistinguishable from SV40 wild type. A unique difference from SV40 was its ability to grow on human glial and CV-I monkey cells. The molecular weight of the viral DNA was very similar to that of SV40 DNA and the DNA cleavage patterns obtained after digestion with Hind II and Hind III restriction endonucleases were indistinguishable from those of SV40.

Normal human and xeroderma pigmentosum (XP, excision-defective group A) cells (both SV40-transformed) pulse-labeled with [(3)H]thymidine at various times after irradiation with ultraviolet light showed a decline and recovery of both the molecular weights of newly synthesized DNA and the rates of synthesis per cell.
