Secoisolariciresinol diglucoside resolves systemic and skin inflammation in psoriatic mice by ameliorating the gut microbiome and modulating immune responses.

Psoriasis is a chronic inflammatory skin disease that often imposes enormous psychological impact on the patient, potentially leading to psychiatric comorbidities or even suicidality. The currently available treatments do not always provide satisfactory results. Our previous work demonstrates that secoisolariciresinol diglucoside (SDG) has potent anti-inflammatory effects. In this study, we aimed to assess the effect of SDG on resolving the psoriatic inflammation using an imiquimod (IMQ)-induced mouse model and elucidate the underlying mechanisms. SDG and its metabolite enterolactone (ENL) exh

Effect of plastic and seaweed shelters on the skin microbiome of lumpfish Cyclopterus lumpus used as cleaner fish in aquaculture pens.

Atlantic salmon (Salmo salar) aquaculture is a major industry in several countries worldwide and a growing enterprise in others. One of the main challenges the industry faces is infestations with the parasitic copepod Lepeoptheirus salmonis, or salmon lice. Several different chemical and mechanical methods are available for alleviating the problem, but often at cost to salmon welfare and/or the environment. In some regions cleaner fish have been introduced to farming facilities as an environmentally and salmon welfare friendly option for reducing sea lice infestations. In some North Atlantic c

Specific gut microbiome's role in skin pigmentation: insights from SCARB1 mutants in Oujiang colour common carp.

AIMS: Beyond the pivotal roles of the gut microbiome in initiating physiological processes and modulating genetic factors, a query persists: Can a single gene mutation alter the abundance of the gut microbiome community? Not only this, but the intricate impact of gut microbiome composition on skin pigmentation has been largely unexplored. METHODS AND RESULTS: Based on these premises, our study examines the abundance of lipase-producing gut microbes about differential gene expression associated with bile acid synthesis and lipid metabolism-related blood metabolites in red (whole wild) and whit

Alterations in the cutaneous microbiome of patients with psoriasis and psoriatic arthritis reveal similarities between non-lesional and lesional skin.

OBJECTIVES: To investigate the cutaneous microbiome spanning the entire psoriatic disease spectrum, and to evaluate distinguishing features of psoriasis (PsO) and psoriatic arthritis (PsA). METHODS: Skin swabs were collected from upper and lower extremities of healthy individuals and patients with PsO and PsA. Psoriatic patients contributed both lesional (L) and contralateral non-lesional (NL) samples. Microbiota were analysed using 16S rRNA sequencing. RESULTS: Compared with healthy skin, alpha diversity in psoriatic NL and L skin was significantly reduced (p<0.05) and samples clustered sep

Longitudinal Study of the Psoriasis-Associated Skin Microbiome during Therapy with Ustekinumab in a Randomized Phase 3b Clinical Trial.

Plaque psoriasis, a chronic inflammatory disease primarily affecting the skin, is thought to have a multifactorial etiology, including innate immune system dysregulation, environmental triggers, and genetic susceptibility. We sought to further understand the role of skin microbiota in psoriasis pathogenesis, as well as their response to therapy. We systematically analyzed dynamic microbiota colonizing psoriasis lesions and adjacent nonlesional skin in 114 patients prior to and during ustekinumab treatment in a phase 3b clinical trial. By sequencing the bacterial 16S ribosomal RNA gene from ski

Topographical Mapping of the Rainbow Trout (Oncorhynchus mykiss) Microbiome Reveals a Diverse Bacterial Community with Antifungal Properties in the Skin.

The mucosal surfaces of wild and farmed aquatic vertebrates face the threat of many aquatic pathogens, including fungi. These surfaces are colonized by diverse symbiotic bacterial communities that may contribute to fight infection. Whereas the gut microbiome of teleosts has been extensively studied using pyrosequencing, this tool has rarely been employed to study the compositions of the bacterial communities present on other teleost mucosal surfaces. Here we provide a topographical map of the mucosal microbiome of an aquatic vertebrate, the rainbow trout (Oncorhynchus mykiss). Using 16S rRNA p

Environment and weight class linked to skin microbiome structure of juvenile Eastern hellbenders (Cryptobranchus alleganiensis alleganiensis) in human care.

Amphibian skin is integral to promoting normal physiological processes in the body and promotes both innate and adaptive immunity against pathogens. The amphibian skin microbiota is comprised of a complex assemblage of microbes and is shaped by internal host characteristics and external influences. Skin disease is a significant source of morbidity and mortality in amphibians, and increasing research has shown that the amphibian skin microbiota is an important component in host health. The Eastern hellbender (Cryptobranchus alleganiensis alleganiensis) is a giant salamander declining in many pa

Environmentally relevant levels of sulfamethoxazole (SMX) alter the skin and gastrointestinal microbiome of adult male and female zebrafish (Danio rerio).

This study determined the effects of sulfamethoxazole (SMX) exposure on the gastrointestinal and skin microbiome of male and female adult zebrafish (Danio rerio). Adult male and female zebrafish were exposed to measured, environmentally relevant concentrations of 0, 0.75, and 304.9 µg/L SMX for 21 days. Bacterial DNA was isolated from feces and skin for 16S sequencing using NextGen Illumina MiSeq. The taxonomic data were analyzed for differences in abundance and diversity between sexes and treatment groups. There were no differences in beta diversity in the fecal and skin microbiota between ma

New Normal Mask-Wearing and Its Impact on Underneath Skin Microbiome: A Cross-Sectional Study in Mild Acne Vulgaris Patients.

INTRODUCTION: Prolonged mask-wearing could modulate the skin microenvironment resulting in several facial dermatoses. Microbial dysbiosis is proposed to be linked with these changes; however, data regarding the association is still limited. Accordingly, we aimed to explore the impact of face masks on the skin's bacterial microbiota. METHODS: We classified participants into short (<4 h/day) and long (≥4 h/day) mask-wearing time (SMWT and LMWT) groups according to mask-wearing time per day in the previous 2 weeks. Specimens were swabbed from the cheek and forehead of 45 mild acne vulgaris patie

Short-Term Exposure to Coal Combustion Waste Has Little Impact on the Skin Microbiome of Adult Spring Peepers (Pseudacris crucifer).

UNLABELLED: Disruptions to the microbiome can impact host health as can exposure to environmental contaminants. However, few studies have addressed how environmental contaminants impact the microbiome. We explored this question for frogs that breed in wetlands contaminated with fly ash, a by-product of coal combustion that is enriched in trace elements. We found differences in the bacterial communities among a fly ash-contaminated site and several reference wetlands. We then experimentally assessed the impacts of fly ash on the skin microbiome of adult spring peepers (Pseudacris crucifer). Fro

Taxonomic and Functional Skin Microbiome Profiles in Nummular Eczema, Atopic Dermatitis, and Mixed Phenotypes: An Exploratory Shotgun Metagenomic Pilot Study.

Nummular eczema (NE) and atopic dermatitis (AD) share clinical features, yet their skin microbiomes remain insufficiently characterized. This exploratory pilot study evaluated the taxonomic composition and functional potential of the skin microbiome across NE, a mixed NE/AD phenotype (Mixed NE_AD), and AD using shotgun metagenomics. Lesional swabs from Korean patients with NE (n = 8), Mixed NE_AD (n = 6), and AD (n = 5) were sequenced to assess microbial diversity, taxonomy, and functional pathways. Staphylococcus aureus predominated across all groups. NE samples were characterized by the high

Cyanoacrylate glue as an effective skin-decontamination method for hair follicle microbiome sampling: insights from a nonhuman model (Capreolus capreolus).

INTRODUCTION: In humans, microorganisms within hair follicles differ from those on the skin surface and may contribute to autoimmune skin diseases such as alopecia areata and vitiligo. Owing to the low biomass of hair follicle microorganisms, precise sample acquisition and strategies to avoid contamination with genetic material are critical. To address this, we developed and evaluated a sampling methodology using a nonhuman wildlife proxy model - the wild roe deer (Capreolus capreolus) - to identify effective approaches for minimizing skin contamination prior to biopsy. Animal was obtained dur

Distinct skin microbiome modulation following different topical acne treatments in mild acne vulgaris patients: A randomized, investigator-blinded exploratory study.

The effects of topical non-antibiotic acne treatment on skin microbiota have rarely been demonstrated. In the study, we randomized 45 mild acne vulgaris participants into three treatment groups, including a cream-gel dermocosmetic containing Aqua Posae Filiformis, lipohydroxy acid, salicylic acid, linoleic acid, niacinamide and piroctone olamine (DC), retinoic acid 0.025% cream (VAA) and benzoyl peroxide 2.5% gel (BP). At months 0, 1 and 3, skin specimens were swabbed from the cheek and forehead and sequenced by targeting V3-V4 regions of the 16 S rRNA gene. QIIME2 was used to characterize bac

Environmental factors associated with nesting habits and age shape the composition and connection between skin and uropygial gland microbiomes of birds.

Bacterial communities on skin and feathers can act as a critical line of defence against pathogenic infections in birds and may originate from secretions produced by the uropygial gland. These secretions reach the bird integument during preening, with the preening effort possibly determining the connectivity between uropygial gland and integument microbiomes. The risk of pathogen infections depends on a number of variables, including environmental conditions (i.e. temperature and humidity), species identity, life-history traits (i.e. cavity vs. open-cup nesters) and life stage (i.e. age). Spatiotemporal and ontogenetic variation, microbial selection, and predicted Bd-inhibitory function in the skin-associated microbiome of a Rocky Mountain amphibian.

Host-associated microbiomes play important roles in host health and pathogen defense. In amphibians, the skin-associated microbiota can contribute to innate immunity with potential implications for disease management. Few studies have examined season-long temporal variation in the amphibian skin-associated microbiome, and the interactions between bacteria and fungi on amphibian skin remain poorly understood. We characterize season-long temporal variation in the skin-associated microbiome of the western tiger salamander (Ambystoma mavortium) for both bacteria and fungi between sites and across

Whole-Genome Shotgun Metagenomic Sequencing Reveals Shifts in the Skin Microbiome and Bacteriophages of Psoriasis: An Extended Analysis of Published Data.

BACKGROUND: Psoriasis is an immune-mediated cutaneous disease that may have shifts in the skin microbiome. Prior research on the skin microbiome in psoriasis has been limited to rRNA based approaches that lack resolution of taxonomic and functional level assessment. OBJECTIVE: To further illuminate strain and sub-strain level analysis of psoriatic lesions using the CosmosID-HUB Microbiome pipeline. METHODS: A previous study completed by Tett et al recruited patients with psoriasis who had skin microbiome samples taken from psoriatic plaques on the ear and the elbow as well as sites on the sk
