GSTO1 acts as a mediator in sodium fluoride-induced alterations of learning and memory related factors expressions in the hippocampus cell line.

The mechanism of GSTO1, as a high-risk factor for neurological damage, in sodium fluoride (NaF)-induced learning and memory impairment remained still unclear. Hence, in this study, we used the siRNA-GSTO1 HT22 model to explore the effect of NaF and siRNA-GSTO1 on the viability, and proliferation rate of HT22 cells, as well as the mRNA and protein expression levels of cyclic adenosine monophosphate (cAMP) response element binding protein (CREB), neural cell adhesion molecule (NCAM), stem cell factor (SCF) and brain-derived neurotrophic factor (BDNF). The results of MTT showed that 10-3, 10-4, a…

An evaluation of Tiron (sodium 4,5-dihydroxybenzene-1,3-disulfonate) as a rescue agent for the nephropathy induced by uranyl fluoride (UO2F2) in rats.

Tiron (sodium 4,5-dihydroxybenzene-1,3-disulfonate) was tested as a potential rescue agent for the nephropathy induced by uranyl fluoride (UO2F2) in rats. When uranyl fluoride was administered (i.v.) to male Sprague-Dawley rats at a dose that delivered 250 micrograms U/kg, moderate to severe cellular necrosis occurred in pars recta segments of proximal tubules in the cortex and outer stripe of the outer medulla of the rats' kidneys within four days after treatment. The severity of renal tubular injury was not significantly diminished when rats were given a 10, 100 or 1,000 mg/kg dose of Tiron

Melatonin-mediated upregulation of Sirt3 attenuates sodium fluoride-induced hepatotoxicity by activating the MT1-PI3K/AKT-PGC-1α signaling pathway.

Mitochondrial reactive oxygen species (ROS) production has been implicated in the pathogenesis of fluoride toxicity in liver. Melatonin, an indolamine synthesized in the pineal gland, was previously shown to protect against sodium fluoride (NaF)-induced hepatotoxicity. This study investigated the protective effects of melatonin pretreatment on NaF-induced hepatotoxicity and elucidates the potential mechanism of melatonin-mediated protection. Reducing mitochondrial ROS by melatonin substantially attenuated NaF-induced NADPH oxidase 4 (Nox4) upregulation and cytotoxicity in L-02 cells. Melatonin…

Assessment of the remineralizing potential of nano-bioactive glass, nano-hydroxyapatite, and sodium fluoride on artificial carious lesions in primary teeth.

BACKGROUND: Tooth decay represents a dynamic balance characterized by alternating cycles of demineralization and remineralization, though loss of minerals can be reversed in its early stages. Recently, nanomaterials have been widely explored in dentistry due to their remarkable characteristics and enhanced remineralization potential. This study aimed to assess the remineralization performance of three varnishes on artificial enamel lesions in primary teeth: commercial fluoride varnish (5% NaF + ACP), experimental nano-bioactive glass (6% n-BAG), and experimental nano-hydroxyapatite (10% n-HAP)

Assessment of the Remineralizing Efficacy of Grape Seed Extract vs Sodium Fluoride on Surface and Subsurface Enamel Lesions: An In Vitro Study.

BACKGROUND: Dental caries is a prevalent oral health issue, primarily resulting from demineralization of the enamel. Remineralizing agents such as sodium fluoride (NaF) have been widely used to counteract this process. Recent studies suggest that natural extracts, such as grape seed extract (GSE), may also have significant remineralizing properties. This study aims to compare the efficacy of GSE and NaF on surface and subsurface enamel lesions in an in vitro setting. MATERIALS AND METHODS: In this in vitro study, 60 extracted human premolars were randomly divided into three groups of 20 each:…

The effect of sodium fluoride preservative and storage temperature on the stability of 6-acetylmorphine in horse blood, sheep vitreous and deer muscle.

This study examined the in vitro stability of 6-acetylmorphine (6AM) in horse blood, sheep vitreous humour (VH) and homogenised deer muscle stored under different storage conditions. The stability of 6AM in horse blood is of interest because many toxicological laboratories utilise this matrix for the preparation of blood calibration and check standards and the latter are typically stored during routine use. Data on the storage stability of 6AM in human VH is extremely limited and no data has been reported in muscle. In the absence of human samples, 6AM stability was demonstrated in sheep vitre

Protective effect of astragalus polysaccharides against sodium fluoride-induced liver injury: A comprehensive study based on network pharmacology, metabolomics, and transcriptomics.

Fluoride, a water-soluble environmental contaminant, poses significant risks to hepatic health due to long-term excessive intake, yet effective interventions for NaF-induced liver injury remain limited. Astragalus polysaccharides (APS), the principal bioactive component of Astragalus membranaceus, exhibit immunomodulatory, anti-inflammatory, and antioxidant properties, and have been shown to protect against alcohol- and chemical-induced liver injury. This study employed animal experiments and integrative multi-omics (network pharmacology, transcriptomics, metabolomics) to investigate the prote

High concentration of sodium fluoride in drinking water induce hypertrophy versus atrophy in mouse skeletal muscle via modulation of sarcomeric proteins.

Fluoride at high doses is a well-known toxic agent for the musculoskeletal system, primarily in bone and cartilage cells. Research on fluoride toxicity concerning particularly on the skeletal muscle is scanty. We hypothesized that during skeletal fluorosis, along with bone, muscle is also affected, so we have evaluated the effects of Sodium fluoride (NaF) on mouse skeletal muscles. Sodium fluoride (80 ppm) was administered to 5-week-old C57BL6 mice drinking water for 15 and 60 days, respectively. We carried out histology, primary culture, molecular and proteomic analysis of fluoride administer…
