The eicosanoid-cell death axis: A mechanistic review of crosstalk in health and disease.

Eicosanoids, a diverse family of lipid mediators from arachidonic acid (AA), are fundamental regulators of inflammation and cellular signaling. Concurrently, regulated cell death (RCD) pathways-including apoptosis, necroptosis, pyroptosis, and ferroptosis-are essential, genetically encoded programs that maintain tissue homeostasis by eliminating damaged or unwanted cells. A growing body of evidence reveals that these two systems are not independent but are deeply and mechanistically intertwined, forming a critical axis that dictates cell fate. Through a systematic evaluation of current literat

Activation of pro-resolving pathways mediate the therapeutic effects of thymosin beta-4 during Pseudomonas aeruginosa-induced keratitis.

INTRODUCTION: Current treatments for bacterial keratitis fail to address the sight-threatening inflammatory host response. Our recent work elucidating the therapeutic mechanisms of adjunctive thymosin beta-4 (Tβ4) in resolving inflammation and infection in bacterial keratitis revealed modulation of effector cell function and enhanced bacterial killing. The current study builds upon the observed effects on effector cell function by investigating the impact of Tβ4 on specialized pro-resolving lipid mediator (SPM) pathways as they play a significant role in inflammation resolution. METHODS: Usin

Specialized pro-resolution mediators in the bladder: Receptor expression and recovery of bladder function from cystitis.

Inflammation is a central process in most benign bladder disorders, and its control is a delicate balance between initiating factors and resolving factors. While recent discoveries have shown a central role for the NLRP3 inflammasome in initiation, the resolving pathways remain unexplored. Resolution is controlled by specialized pro-resolution mediators (SPMs) functioning through seven receptors (six in rodents). Here we demonstrate expression of all seven in humans (six in mice) through immunocytochemistry. Expression was universal in urothelia with most also expressed in smooth muscle. We ne

Over-the-counter fish oil supplementation and pro-resolving and pro-inflammatory lipid mediators in rheumatoid arthritis.

OBJECTIVE: Little is known about the effects of over-the-counter fish oil (FO) supplements on circulating omega-3 polyunsaturated fatty acid (n-3 PUFA)-derived specialized pro-resolving mediators (SPMs), nor about whether having a chronic inflammatory disease such as rheumatoid arthritis (RA) influences SPM levels. We investigated associations between over-the-counter n-3 PUFA FO supplementation and circulating SPMs among patients with vs. without RA. METHODS: We studied 104 participants: 26 with RA taking FO matched by age and sex to 26 with RA not taking FO, 26 without RA taking FO, and 26

Pediatric Critical Illness, Immunometabolism and Cardiovascular Risk: A Narrative Review.

The management of critical illness has significantly advanced, enabling a greater number of patients to survive conditions that were once fatal. However, survival often comes at a cost, as many patients experience long-term sequelae, including chronic critical illness (CCI) and post-intensive care syndrome (PICS). These syndromes can also be subclinical, driven by a persistent imbalance between pro-inflammatory and anti-inflammatory processes, resulting in sustained immune dysfunction, metabolic disturbances, and organ-specific complications. Among the most affected systems are the immune and

Omega-3 PUFAs in musculoskeletal health and sports medicine: from molecular pathways to precision nutrition strategies.

The imbalance in the dietary ω-6/ω-3 polyunsaturated fatty acid (PUFA) ratio contributes to chronic low-grade inflammation, a key pathological basis for degenerative musculoskeletal conditions such as osteoporosis and osteoarthritis. While individual reviews have explored aspects of ω-3 PUFAs in bone or muscle, a comprehensive narrative review integrating their multidimensional mechanisms with translational evidence across orthopedics and sports medicine is less common. This review consolidates evidence from fundamental science, clinical trials, and translational research, employing an interdi

Serum lipidome remodeling in viral pneumonia: from pathophysiology to therapeutics.

Viral pneumonia remains a preeminent global health threat, frequently culminating in acute respiratory distress syndrome (ARDS) and systemic organ failure. While traditional paradigms have centered on protein-based inflammatory cascades, this review synthesizes a vast body of emerging evidence to redefine viral pneumonia as a profound systemic metabolic crisis, specifically characterized by the radical remodeling of the host lipidome. Based on a meta-synthesis of clinical and mechanistic data, we delineate a consistent "metabolic crash" during severe infection, where a precipitous decline in s

Restoring Balance: The Role of Omega-3 Polyunsaturated Fatty Acids on the Gut-Brain Axis and Other Interconnected Biological Pathways to Improve Depression.

Major depressive disorder (MDD) is a complex, multifactorial condition involving dysregulation across immune, neural, and metabolic systems. Omega-3 polyunsaturated fatty acids (n-3 PUFAs), particularly eicosapentaenoic acid (EPA) and docosahexaenoic acid (DHA), have emerged as promising adjunctive interventions, with evidence supporting their efficacy in alleviating depressive symptoms. Here, we synthesize current knowledge on the interconnected biological pathways through which n-3 PUFAs may exert antidepressant effects. A growing body of evidence implicates the gut-brain axis as a central p

Decreased ω-6:ω-3 PUFA ratio attenuates ethanol-induced alterations in intestinal homeostasis, microbiota, and liver injury.

Ethanol (EtOH)-induced alterations in intestinal homeostasis lead to multi-system pathologies, including liver injury. ω-6 PUFAs exert pro-inflammatory activity, while ω-3 PUFAs promote anti-inflammatory activity that is mediated, in part, through specialized pro-resolving mediators [e.g., resolvin D1 (RvD1)]. We tested the hypothesis that a decrease in the ω-6:ω-3 PUFA ratio would attenuate EtOH-mediated alterations in the gut-liver axis. ω-3 FA desaturase-1 (fat-1) mice, which endogenously increase ω-3 PUFA levels, were protected against EtOH-mediated downregulation of intestinal tight junct

Resolvin D1 alleviates periodontitis by promoting FPR2-dependent macrophage pro-resolving polarization and efferocytosis.

INTRODUCTION: Periodontitis is a chronic inflammatory disease driven by dysregulated host responses and failed resolution of inflammation, leading to progressive periodontal tissue destruction. This resolution failure is critically linked to impaired macrophage efferocytosis and a deficiency in endogenous specialized pro-resolving mediators such as Resolvin D1 (RvD1). OBJECTIVES: This study aimed to determine if exogenous RvD1 can restore periodontal homeostasis and to define the formyl peptide receptor 2 (FPR2)-dependent mechanisms governing macrophage reprogramming and the clearance of necr

Resolvin D3 Promotes Inflammatory Resolution, Neuroprotection, and Functional Recovery After Spinal Cord Injury.

Resolvins, a new family from the endogenous specialized pro-resolving mediators (SPMs), promote the resolution of the inflammatory response. Resolvin D3 (RvD3), a docosahexaenoic acid (DHA) product, has been known to suppress the inflammatory response. However, the anti-inflammatory and neuroprotective effects of RvD3 are not known in a model of spinal cord injury (SCI). Here, we investigated the anti-inflammatory and neuroprotective effect of RvD3 in a mouse model of SCI. Processes associated with anti-inflammation and angiogenesis were studied in RAW 264.7 cells and the human brain endotheli

Post-acute sequelae of COVID-19: A disorder of impaired innate immune resolution - A narrative review.

Post-acute sequelae of COVID-19 (PASC) affect millions of people worldwide and are increasingly recognized as a disorder of failed innate immune resolution rather than a persistent viral infection. Emerging evidence shows that residual SARS-CoV-2 antigens, host-derived alarmins, reactivated latent viruses, and mucosal microbiome-derived products from oral-nasopharyngeal and gut reservoirs sustain the chronic activation of pattern-recognition receptors, inflammasomes, and complement pathways. In parallel, deficits in specialized pro-resolving mediators, impaired efferocytosis, and persistent ti

Engineered Biomimetic Nanovesicles Based on Neutrophils for Hierarchical Targeting Therapy of Acute Respiratory Distress Syndrome.

Acute Respiratory Distress Syndrome (ARDS) is a clinically severe respiratory disease that causes severe medical and economic burden. To improve therapeutic efficacy, effectively targeting delivery to the inflamed lungs and inflamed cells remains an ongoing challenge. Herein, we designed engineered biomimetic nanovesicles (DHA@ANeu-DDAB) by fusion of lung-targeting functional lipid, neutrophil membrane containing activated β2 integrins, and the therapeutic lipid, docosahexaenoic acid (DHA). By the advantage of lung targeting lipid and β2 integrin targeting adhesion, DHA@ANeu-DDAB can first tar

Resolvin D1 inhibits T follicular helper cell expansion in systemic lupus erythematosus.

OBJECTIVE: Resolvin D1 (RvD1) is one of the specialized pro-resolving lipid mediators, which control inflammation resolution and regulate immune responses. Previous research showed that RvD1 could block the progression of systemic lupus erythematosus (SLE). However, the detailed mechanism remains to be fully understood. METHOD: Plasma RvD1 levels, and proportions of T follicular helper cells (Tfh cells) were measured in SLE patients and healthy controls. Plasma RvD1 levels and proportions of Tfh cells were quantitated in an MRL/lpr mouse model of lupus treated with RvD1. Naïve CD4+ T cells we

The resolvin D2 and omega-3 polyunsaturated fatty acid as a new possible therapeutic approach for inflammatory bowel diseases.

Inflammatory bowel diseases (IBD) are idiopathic disorders characterized by chronic gastrointestinal inflammation. Given conventional therapies' adverse effects and clinical failures, novel approaches are being investigated. Recent studies have highlighted the role of specialized pro-resolving lipid mediators (SPMs) in the active resolution of chronic inflammation. In this regard, omega-3 fatty acid-derived Resolvin D2 (RvD2) appears to play a protective role in the pathophysiology of IBD. Therefore, we characterized the RvD2 pathway and its receptor expression in the intestinal mucosa of expe

Signaling pathways activated by resolvin E1 to stimulate mucin secretion and increase intracellular Ca2+ in cultured rat conjunctival goblet cells.

Glycoconjugate mucin secretion from conjunctival goblet cells is tightly regulated by nerves and specialized pro-resolving mediators (SPMs) to maintain ocular surface health. Here we investigated the actions of the SPM resolvin E1 (RvE1) on cultured rat conjunctival goblet cell glycoconjugate secretion and intracellular [Ca2+] ([Ca2+]i) and the signaling pathways used by RvE1. Goblet cells were cultured from rat conjunctiva in RPMI medium. The amount of RvE1-stimulated glycoconjugate mucin secretion was determined using an enzyme-linked lectin assay with Ulex Europaeus Agglutinin 1 lectin. Cul

Oxidized-Lipid Signaling and Ferroptosis as Downstream Mechanisms of Titanium-Associated Peri-Implant Bone Loss.

Titanium and its alloys remain the dominant materials in oral implantology, but degradation products released through corrosion, tribocorrosion, and wear are biologically active. This critical narrative review examines oxidized-lipid signaling and ferroptosis as candidate downstream mechanisms connecting titanium-associated redox dysregulation with peri-implant bone loss. A targeted literature search of PubMed, Web of Science, and Scopus was updated through 2 August 2026 and integrated evidence from dental peri-implant studies, titanium-particle osteolysis models, and broader skeletal research

Polyunsaturated fatty acids and parasite control: towards sustainable anthelmintic strategies within a One Health framework.

Parasitic helminths are a major constraint on livestock production worldwide, with significant implications for food security and animal and human health. As reliance on conventional anthelmintic drugs becomes increasingly unsustainable due to widespread anthelmintic resistance, chemical residues, and environmental concerns, there is an urgent need for innovative, biologically derived alternatives such as polyunsaturated fatty acids (PUFAs). This review evaluates PUFAs as biologically derived agents that could complement or partially replace conventional dewormers. We integrate molecular, cell

Role of Inflammatory and Proresolving Mediators in Endothelial Dysfunction.

Excessive local inflammation is a common mechanism in many cardiovascular diseases (CVDs) such as hypertension, atherosclerosis and aortic aneurysms. In endothelial cells, inflammatory cytokines such as interferons, tumour necrosis factor alpha or interleukins increase oxidative stress and contractile prostanoids and the expression of adhesion molecules that reduce nitric oxide (NO) availability and bind leucocytes, thereby impairing endothelial function. Despite this evidence, anti-inflammatory therapies are not yet indicated for the treatment of most CVD. Resolution of inflammation is mediat

Enhancing tofacitinib's therapeutic efficacy in murine arthritis with a synbiotic formulation comprising Bacillus megaterium DSM 32963 and an Omega-3 fatty acid lysine salt.

INTRODUCTION: Omega-3 polyunsaturated fatty acids (n3-PUFA) are known for their anti-inflammatory benefits, particularly in chronic conditions like rheumatoid arthritis (RA). To resolve an acute inflammation, conversion of n3-PUFA into specialized pro-resolving mediators (SPM) is crucial. Recently, it was shown that the probiotic Bacillus megaterium DSM32963 supports this conversion. METHODS: This study evaluates a synbiotic formulation combining Bacillus megaterium DSM32963 and a unique n3-PUFA-lysine salt as adjunct nutritional supplement to tofacitinib in adjuvant-induced arthritis (AIA) i
