Chelation-Controlled Fluorescence: A Novel Coumarin-Thiazole Probe for Zn (II) Analysis in Real Samples.

In this investigation, an exceptional coumarin-thiazole-based fluorescent sensor was fabricated through a facile one-pot synthetic strategy, affording a highly emissive blue-fluorescent probe suitable for Zn (II) determination. The synthesized fluorescent probe exhibited a strong emission band at 470 nm upon excitation at 410 nm with a quantum yield of 0.95 ± 0.05. Incremental additions of Zn (II) from 2.5 to 37.5 μM induced a pronounced turn-off response in fluorescence intensity. An excellent linearity (r = 0.9999) was established between the quenching magnitude and Zn (II) concentration, wi

Protocol for systematic identification of microtubule luminal proteins using knock-in BioID proximity labeling and fluorescence imaging validation.

Systematic elucidation of microtubule (MT) luminal proteins provides insights into MT biology and its molecular regulation. Here, we present a protocol for screening and validating these proteins using proximity labeling, mass spectrometry, fluorescence imaging and western blot (WB). We describe steps for constructing a biotin ligase knock-in cell line and performing BioID screening. We detail how to perform fluorescence imaging and WB to validate the MT luminal proteins. This protocol enables systematic identification to advance research on MT luminal proteins. For complete details on the use

Design and Synthesis of Dual Fluorophore for Detection of Fe3+ and Hg2+ in Aqueous Solution Through Fluorescence "Turn-Off".

Here, we report the design and synthesis of a new bicyclic compound 6, annulated with benzene and the 8-hydroxyquinoline fluorophore. The probe exhibits a moderate aggregation-induced emission (AIE) behavior upon the addition of water up to 17%. Furthermore, the probe exhibits a highly selective fluorescence turn-off response toward Fe3+ and Hg2+ ions in a DMSO/H2O (83:17, v/v) solution over a range of other competitive metal ions. The probe displayed low detection limits of 3.69 μM for Fe3+ and 6.08 μM Hg2+. Probe 6 shows a fast recognition time of ions and can monitor Fe3+ and Hg2+ ions in r

Smartphone-integrated colorimetric and ratiometric fluorescence sensor for visual detection of HSO4.

BACKGROUND: The detection of bisulfate (HSO4-) is critical in food preservation and pharmaceutical quality control, as it is a common additive with potential health risks. Existing detection methods often lack the portability for on-site analysis or the capability for cross-validation to ensure accuracy. Establishing a rapid and reliable quantitative method for HSO4- is the prerequisite and core step for precisely controlling related food and pharmaceutical additives. RESULTS: To address the need for a rapid, reliable, and field-deployable sensing platform that provides intuitive visual feedb

The evolution of steady-state laurdan fluorescence: From model membranes to cellular applications.

Laurdan (6-dodecanoyl-2-(dimethylamino)-naphthalene) is a solvatochromic fluorescent probe widely used for investigating membrane biophysical properties. Since its first application in phospholipid bilayers, laurdan has become a valuable tool in this field, owing to its sensitivity to the mobility and dynamics of surrounding lipid carbonyl groups and its ability to report on membrane phase behavior through the generalized polarization (GP) parameter. GP, a ratiometric empirical parameter, is derived from the ratio of emission intensities at approximately 440 and 490 nm, providing a measure of

Deconvolution and Correction of the Inner Filter Effect in a Tri-Enzymatic Fluorescent Cascade for Ultra-Extended Cholesterol Quantification.

The sensing architecture leveraging a tri-component fluorescent bio probe (Cholesterol Oxidase-Horse Reddish Peroxidase-Ampliflu Red) drives a highly efficient enzymatic cascade for the acutely sensitive and selective spectrophotometric quantification of cholesterol. The reaction culminates in the HRP catalyzed oxidation of Ampliflu Red, generating an intense, quantifiable emission centered at λem = 585 nm and excitation at λex = 570 nm, whose intensity is stoichiometrically correlated with cholesterol concentration (c). Crucially, the inner filter effect, which typically imposes nonlinearity

Environmentally relevant dual-sensing of mercury (II) and hydrazine via reaction-induced fluorescence switching for real-sample and on-site monitoring.

Environmental monitoring of mercury ions (Hg²⁺) and hydrazine (N₂H₄) is crucial due to their extreme toxicity and persistence. Herein, we report a newly designed dual-reactive fluorescent probe capable of selective and sensitive detection of both Hg²⁺ and N₂H₄ through distinct optical responses. The sensing mechanism involves inhibition and activation of intramolecular charge transfer (ICT), excited-state intramolecular proton transfer (ESIPT), and photoinduced electron transfer (PET) processes, leading to fluorescence "turn-on" at 610 nm for Hg²⁺ and 478 nm for N₂H₄. Exposure to Hg²⁺ causes d

A Ratiometric genetically encoded fluorescent probe for ultrasensitive and self-calibrated detection of Hg2.

Mercury ion (Hg2+) pollution poses severe environmental and health risks, necessitating the development of highly sensitive and reliable detection methods. Although ratiometric and genetically encoded fluorescent probes each offer distinct advantages, their integration for Hg2+ sensing remains largely unexplored. Herein, we report the rational design of a novel ratiometric genetically encoded fluorescent probe through the fusion of a circularly permuted green fluorescent protein (cpEGFP), a mercury-binding domain (MerBD), and a large Stokes shift reference fluorescent protein (LSSmOrange). Functional fluorescent material fabricated with cellulose derivative regenerated in a carbon disulfide based complex solvent.

In this study, cellulose was facilely dissolved in a novel solvent composed of carbon disulfide, an organic base, and organic solvent, and then regenerated to obtain dithiocarbonate cellulose. Functional fluorescent material was then fabricated by grafting poly(lactic acid) side chain and dansyl chloride (DNS-Cl) to the backbone of this cellulose derivative. The product, denoted as cellulose-graft-poly(lactic acid)/dansyl chloride (C-g-PLLA/DNS-Cl), was obtained by simply dissolving cellulose and grafting fluorescent structures. It exhibits a distinct glass transition temperature, indicating t

Mercury speciation by CE: an update.

This review provides an update on mercury speciation by CE. It includes a brief discussion on physicochemical properties, toxicity and transformation pathways of mercury species (i.e. methyl-, ethyl-, phenyl- and inorganic mercury) and outlines recent trends in Hg speciation by CE. CE is presented as a complementary technique to chromatographic separation techniques, especially in cases when speed, high efficiency and low sample volumes are required. The development of suitable sample preconcentration/isolation (sample stacking, ion exchange, liquid-liquid-liquid extraction, dual-cloud point e

A D-π-A conjugated probe with dual responses to bisulfite and microenvironmental viscosity/polarity for imaging ferroptosis in neural cells.

Ferroptosis is a form of programmed cell death characterized by lipid peroxidation and changes in the cellular microenvironment (such as increased viscosity and decreased polarity). Currently, there are still relatively few fluorescent probes capable of simultaneously responding to both bisulfite levels and microenvironmental changes. To address this, we designed and synthesized a fluorescent probe with a D-π-A conjugated structure. Based on the intramolecular charge transfer (ICT) mechanism, this probe exhibits dual responsiveness to bisulfite and the microenvironment (viscosity/polarity) and

A ratiometric fluorescent probe for HSO3-and its application in environmental and food analysis.

Bisulfite (HSO3-) is widely used as a preservative and antioxidant in the food industry and chemical manufacturing, yet its excessive residues pose risks to food quality, environmental safety, and human health. To achieve rapid, selective, and quantitative detection of HSO3-, a ratiometric fluorescent probe based on a benzothiazole-carbazole unit and a benzoindole ion was developed. Upon reaction with HSO3-, the probe exhibits a distinct dual-emission response, fluorescence intensity decreases at 612 nm and increases at 467 nm, enabling robust ratiometric sensing (F467/F612). A deep learning-assisted turn-on fluorescent probe for L-BPA detection with mechanistic insight.

Boron neutron capture therapy (BNCT) is a binary cancer treatment strategy that relies on the selective delivery of a 10B-containing drug followed by thermal neutron irradiation, generating localized high-LET particles that destroy tumor cells. L-BPA is the most recent and currently the only marketed boron carrier; however, its poor tumor retention and low solubility significantly limit therapeutic efficacy. Thus, developing improved L-BPA derivatives, as well as tools for efficient monitoring of L-BPA biodistribution, is critically important. Herein, we report a turn-on fluorescent probe, WF3

A dual-key fluorogenic probe for DNA and proteins imaging via excimer disaggregation and viscosity-activated molecular rotor.

Fluorescence imaging is pivotal for elucidating biomolecular dynamics, yet the development of probes enabling specific and high-contrast imaging of multiple targets remains challenging. Herein, we designed and synthesized a novel cyanovinylene-based fluorogenic probe, CV-N3, which synergistically integrates a dual-response mechanism combining excimer disaggregation with a viscosity-activated molecular rotor. The excimer formation and its CuAAC induced disaggregation are directly supported by dynamic light scattering and time-resolved fluorescence lifetime measurements. The probe enables specif

Emissive nanoaggregate-based dual sensing of heparin and protamine sulphate.

Heparin (Hep), a widely used blood thinner, requires continuous monitoring upon administration, as both under- and overdosing can lead to complications such as thrombosis or haemorrhage. At the same time, protamine sulfate (PS) serves as the clinical antidote for Hep via electrostatic interactions. Inspired by the need to continuously monitor this clinically relevant Hep-PS pair, we have designed a simple tetraphenylethylene (TPE)-based tetracationic sensor to detect these analytes via a fluorescence switch-on/off mechanism. Hep binding induces strong electrostatic interactions between highly

A Highly Selective Hemicyanine-Based Turn-Off Fluorescent Sensor for Cyanide Detection in Food Samples.

Cyanide is a highly toxic anion. Humans are commonly exposed to cyanide through the consumption of drinking water or by eating plants that contain cyanogenic glucosides, such as cassava, a key carbohydrate source in many diets. While numerous methods exist for cyanide detection, few are both cost-effective and simple enough for use by individuals without specialized training. In this work, we designed and synthesized a hemicyanine-based fluorescent sensor ZM-FES for the detection of cyanide (CN-). Among the anions tested, the sensor showed high selectivity towards CN-. The addition of CN- to Z

A novel rhodamine-based dual-mode fluorogenic probe for sensitive determination of Hg².

Mercury ions (Hg2+) are highly toxic and persistent heavy metal pollutants that pose serious threats to environmental systems and human health. Therefore, developing sensitive and selective detection strategies for Hg2+is of great significance. In this study, a rhodamine-based dual-mode fluorescent probe (RB-HG) was developed utilizing dimethylthiocarbamate groups as recognition sites.Upon undergoing specific interaction with Hg2+, leading to the dissociation of the C-O bond and subsequently triggering an opening-ring reaction to form a ketone structure(RB-C=O), thereby activating the intramol

A Noncovalent Fluorescent Probe for Dynamic Monitoring of Hemoglobin Accumulation in Living Cells.

Dynamic assessment of intracellular hemoglobin (Hb) levels enables a deeper understanding of Hb-induced cytotoxicity, supports the evaluation of disease progression, and informs the development of targeted therapeutic strategies. However, traditional Hb detection methods typically require cell lysis or tissue fixation, which precludes the observation of dynamic processes in living cells. Herein, we reported a noncovalent fluorescent probe, DMBM, which enabled the dynamic monitoring of bovine hemoglobin (BHb) accumulation in living cells. DMBM exhibited strong intrinsic fluorescence and demonst

One-pot synthesis of highly stable CsPbBr₃@C6H7NO3S composite and its application in nitrite fluorescent sensing.

Herein, we introduced anhydrous 4-aminobenzene sulfonic acid (C6H7NO3S) to passivate the ligands on the surface of CsPbBr3 quantum dots, thereby synthesising a perovskite composite with excellent dispersion and stability in water via a one-pot process, namely CsPbBr3@C6H7NO3S composite. The fluorescence attenuation of CsPbBr3@C6H7NO3S composite was only 2.20 % after 5-day immersion in water, and the CsPbBr3@C6H7NO3S composite maintained excellent luminescence performance in long-term storage (4 months). The CsPbBr3@C6H7NO3S composite was then adopted to construct the fluorescent sensing for th
