Tetrodotoxin-insensitive relaxation of coronary arterial smooth muscle to electrical stimulation: possible involvement of a dopaminergic mechanism.

Experiments were designed to determine the mechanism by which electrical stimulation causes tetrodotoxin-insensitive relaxation in isolated arteries. Rings of left anterior descending coronary arteries of dogs, pigs and calves were suspended in organ chambers between platinum electrodes. Experiments were performed after treatment with phenoxybenzamine and in the presence of propranolol. Calcium-free solution and calcium antagonists reduced the relaxation. Chemical denervation with 6-hydroxydopamine reduced the relaxation induced by electrical stimulation; in the presence of pargyline, the inhi

Identification of bacteria and bacteria-associated chemical cues that mediate oviposition site preferences by Aedes aegypti.

The yellow fever mosquito, Aedes aegypti, the global vector of dengue and yellow fever, is inexorably linked to water-filled human-made containers for egg laying and production of progeny. Oviposition is stimulated by cues from water containers, but the nature and origin of these cues have not been elucidated. We showed that mosquito females directed most of their eggs to bamboo and white-oak leaf infusions, and only a small fraction of the eggs were laid in plain water containers. In binary choice assays, we demonstrated that microorganisms in leaf infusions produced oviposition-stimulating k

Randomized clinical trial: efficacy and safety of PPC-5650 on experimental esophageal pain and hyperalgesia in healthy volunteers.

OBJECTIVE: Gastroesophageal reflux disease (GERD) is a common condition associated with symptoms as heart burn, regurgitation, chest pain, and gastrointestinal discomfort. PPC-5650 is a new pharmacological agent that can modulate acid-sensing ion channel activity, potentially leading to reduction in the pain signal. In healthy volunteers the esophagus was sensitized with acid to mimic GERD with the aims: 1) to assess the efficacy of a single bolus of PPC-5650 locally applied to the esophagus using multimodal pain stimulations, and 2) to assess the safety profile of PPC-5650. MATERIALS AND MET

Subtoxic effects of certain chemicals on food crops.

An increased supply of food is urgently needed to provide adequate nutrients to the increasing world population. Plant foods are basic in the nutrition of man. Increasing the productivity and efficiency of plant foods is, therefore, essential. Application of low concentrations of certain chemicals have helped in making a breakthrough in this direction. Certain chemicals are known to stimulate plant growth and yield. Many growth regulators also increase the protein content of plants. Treatment with pesticides and herbicides may also change the nutritive quality. These effects have been explored

Genetically Encoded Calcium Indicators for In Situ Functional Studies of Corneal Nerves.

PURPOSE: Millions of people suffer from diseases that involve corneal nerve dysfunction, caused by various conditions, including dry eye syndrome, neurotrophic keratopathy, diabetes, herpes simplex, glaucoma, and Alzheimer's disease. The morphology of corneal nerves has been studied extensively. However, corneal nerve function has only been studied in a limited fashion owing to a lack of tools. Here, we present a new system for studying corneal nerve function. METHODS: Optical imaging was performed on the cornea of excised murine globes taken from a model animal expressing a genetically encod

Pholcodine stimulates a dramatic increase of IgE in IgE-sensitized individuals. A pilot study.

BACKGROUND: A previous study showed a relation between pholcodine (PHO) consumption, prevalence of IgE-sensitization to PHO, morphine (MOR) and suxamethonium (SUX) and anaphylaxis to neuromuscular blocking agents (NMBA). The purpose of this pilot study was to explore the effect on IgE production, in IgE-sensitized and nonsensitized individuals, of exposure to cough syrup and environmental chemicals containing PHO, MOR and SUX related allergenic structures. METHODS: Serum concentrations of IgE and IgE antibodies to PHO, MOR and SUX allergens measured by ImmunoCAP (Pharmacia Diagnostics, Uppsal

Activation of the thalamic parafascicular nucleus by electrical stimulation of the peripheral vestibular nerve in rats.

The parafascicular nucleus (PFN) of the thalamus is a primary structure in the feedback circuit of the basal ganglia-thalamo-cortical system, as well as in the neural circuit of the vestibulo-thalamo-striatal pathway. We investigated the characteristics of the functional connectivity between the peripheral vestibular system and the PFN in rats. A single electrical stimulation was applied to the horizontal semicircular canal nerve in the peripheral vestibular end-organs. This resulted in polysynaptic local field potentials (LFPs) in the PFN, which were composed of long-lasting multiple waves. T

Widespread neural excitation initiated from cardiac spinal afferent nerves.

Reflex effects of cardiac sympathetic afferent nerve excitation on cardiac and renal components of sympathetic outflow have been well investigated, but the extent of these cardiac afferent influences on other sympathetic nerves or on respiratory or cortical neural systems is unknown. Therefore such influences were investigated electrophysiologically in anesthetized, vagotomized, baroreceptor-denervated cats. Stimulation of cardiac sympathetic spinal afferent neurons by the noxious substance bradykinin caused excitation of cardiac, renal, splenic, gastrohepatic, adrenal, and deep peroneal (musc

Mechanisms of excitation and inhibition in the nigrostriatal system.

The extracellular responses of neurones in the neostriatum following single pulse stimulation of the substantia nigra were investigated in urethane anaesthetized rats. Low intensity stimulation (less than 10 V) evoked single large amplitude spikes while higher intensities (10-20 V) elicit a high frequency burst of small amplitude spikes or waves. When spontaneous or glutamate-induced large spikes are recorded, nigral stimulation causes their inhibition coincidentally with the development of a burst. If the burst is prevented, the inhibitory response disappears. Both the nigral evoked inhibitio

Drastic changes of ventromedial medulla neuronal properties induced by barbiturate anesthesia. II. Modifications of the single-unit activity produced by Brevital, a short-acting barbiturate in the awake, freely moving rat.

In the preceding study, we have found that pentobarbital, a powerful barbiturate substance, strongly modified the ventromedial medulla (VMM) physiology in relation to nociception: indeed, in the same rats, during time-separated similar VMM penetrations, we have recorded, under pentobarbital, 'new' neuronal groups as compared to the awake state, such as the units exclusively driven (excited or inhibited) by cutaneous innocuous or noxious stimulations and the multimodal multireceptive neurons inhibited by non-noxious and noxious stimuli. Still under pentobarbital, we have also recorded the same

Cyanide stimulation of tri-N-butyltin mediated hemolysis.

The effects of tributyltin and sodium cyanide on hemolysis in human erythrocytes are described. Tributyltin has a sharp cut-off concentration for induction of hemolysis. A 5 microM concentration of tributyltin induces hemolysis and 1 microM or less does not in erythrocyte suspensions with 2.3 X 10(8) cells per ml. The kinetics for tributyltin-induced hemolysis are sigmoidal indicating a complex molecular mechanism leading to lysis. Ten mM sodium cyanide plus 1 microM tributyltin does not stimulate hemolysis rates above levels observed with 10 mM sodium cyanide alone. Five mM sodium cyanide plu

Olfactory stimulation variables. Which model best predicts the olfactory nerve response?

Mozell et al. (1984. J. Gen. Physiol. 83:233-267) have examined the traditional manner in which olfactory stimulus-response relationships have been addressed. They developed a model that describes the olfactory nerve response as a function of three factors, viz., the number of odorant molecules (N), the stimulus duration (T), and the stimulus volume (V). In addition, two models derived from this three-variable model were also found to predict the response well. These were the [F, N] model involving flow rate (F = V/T) and, ranking closely behind, the [C, T] model involving concentration (C = N

Protein in rabbit lacrimal gland fluid.

A variety of proteins have been identified in tears, which are composed of secretions from all the orbital glands. It has been assumed that the lacrimal gland was the source of these proteins because there is indirect evidence that the lacrimal gland can secrete protein in response to cholinergic stimulation. In the present study, the concentration of protein was determined in lacrimal gland fluid, i.e., the fluid collected directly from the main excretory duct of the lacrimal gland, uncontaminated by secretions from the other orbital glands. The concentration of protein in the lacrimal gland

Hyperstimulation of the uterus during the oxytocin stress test.

Two thousand seven hundred ninety two contraction stress tests (CST) were analysed. Oxytocin was administered in 1730 of these to stimulate uterine contractions and the hyperstimulation which occurred in 48 tests (2.8%) was studied extensively. Seven patients went into labor within 24 hours of the hyperstimulation. Two infants weighed less than 2500 g. One was mature but small-for-dates and the other (who died neonatally) had a trisomy-18 syndrome with severe congenital abnormalities. There were no other intrapartum or neonatal complications. Ten CSTs demonstrated hypertonicity without any pre

The role of non-covalent protein binding in skin sensitisation potency of chemicals.

Skin sensitisation is a delayed hypersensitivity reaction caused by repeated exposure to common natural and synthetic chemical allergens. It is thought that small chemical sensitisers (haptens) are required to form a strong irreversible bond with a self protein/peptide and generate an immunogenic hapten-protein complex in order to be recognised by the immune system and stimulate T cell proliferation. The sensitisers are usually electrophilic chemicals that are directly reactive with proteins or reactive intermediates (metabolites) of chemically inert compounds (prohaptens). Sensitising chemica
