ETHNOPHARMACOLOGICAL RELEVANCE: Streptococcus mutans a key pathogen, produces biofilm, acids and extracellular polysaccharides in the oral cavity; which leads to the development of dental caries. Control of these pathogenic markers can prevent dental caries. Uvaria chamae P. Beauv. is a medicinal plant traditionally used for many ailments including oral infections. Root, leaves and bark extracts has proven antibacterial activity including activity against caries causing S. mutans. However, its effect on the virulence properties of S. mutans, responsible for the development of dental caries, ha…

Screening of a Streptococcus mutans mutant library indicated that pgmA mutants displayed a reduced biofilm-associated tolerance toward gentamicin. The biofilms formed by the S. mutans pgmA mutant also displayed decreased tolerance towards linezolid and vancomycin compared to wild-type biofilms. On the contrary, the resistance of planktonic S. mutans pgmA cells to gentamycin, linezolid, and vancomycin was more similar to wild-type levels. Investigations of biofilms grown in microtiter trays and on submerged glass slides showed that pgmA mutants formed roughly the same amount of biofilm as the w…

With this study, we investigated the effect of synthetic antimicrobial peptides Pep19-2.5 and Pep194LF alone or in combination with antibiotics on S. mutans growth and biofilm formation/disruption. We also examined the cytotoxic effect of each peptide on monocytes. S. mutans was cultured in the presence of different concentrations of each peptide. We showed that Pep19-2.5 and Pep19-4LF were able to significantly (p ≤ 0.01) inhibit the growth of S. mutans. The synthetic peptides also decreased biofilm formation by S. mutans. Furthermore, both peptides reduced the viability of S. mutans in alrea

Studies of the dental caries pathogen Streptococcus mutans have benefitted tremendously from its sophisticated genetic system. As part of our own efforts to further improve upon the S. mutans genetic toolbox, we previously reported the development of the first cloning-independent markerless mutagenesis (CIMM) system for S. mutans and illustrated how this approach could be adapted for use in many other organisms. The CIMM approach only requires overlap extension PCR (OE-PCR) protocols to assemble counterselectable allelic replacement mutagenesis constructs, and thus greatly increased the speed …

Microbial interactions are necessarily associated with the development of polymicrobial oral biofilms. The objective of this study was to determine the coaggregation of eight strains of Candida albicans with Actinomyces naeslundii and Streptococcus mutans. In autoaggregation assays, C. albicans strains were grown in RPMI-1640 and artificial saliva medium (ASM) whereas bacteria were grown in heart infusion broth. C. albicans, A. naeslundii and S. mutans were suspended to give 10(6), 10(7) and 10(8) cells mL(-1) respectively, in coaggregation buffer followed by a 1 h incubation. The absorbance d…

The uptake of protons by intact cells of Streptococcus mutans GS-5 was measured directly by assessing the pH increase in lightly buffered cell suspensions as protons moved across the plasma membrane after acid pulses. The barrier function of the cell membrane for protons was disrupted completely by 5% (vol/vol) butanol. This function was compromised severely by the ionophores valinomycin and gramicidin and by the uncoupler carbonylcyanide-m-chlorophenyl hydrazone. Proton conduction across the membrane was also enhanced by fluoride. It appeared that HF traversed the membrane and acted as a carr…

We previously demonstrated that Streptococcus oligofermentans suppressed the growth of Streptococcus mutans, the primary cariogenic pathogen, by producing hydrogen peroxide (H(2)O(2)) through lactate oxidase activity. In this study, we found that the lox mutant of S. oligofermentans regained the inhibition while growing on peptone-rich plates. Further studies demonstrated that the H(2)O(2) produced on peptone by S. oligofermentans was mainly derived from seven L-amino acids, i.e., L-aspartic acid, L-tryptophan, L-lysine, L-isoleucine, L-arginine, L-asparagine, and L-glutamine, indicating the p…

Salivary components in the pellicle mediate bacterial adherence to the tooth. Such components may also aggregate bacteria in saliva and prevent them becoming established in dental plaque. In the present study, the adherence and aggregation of Streptococcus mutans strain Ingbritt, S. sanguis strain 10556 and Actinomyces viscosus-strain 19246 mediated by parotid and whole saliva from groups of young and elderly people were examined. Significant differences were found between test strains, salivary secretions and age groups. S. sanguis 10556 and A. viscosus 19246 generally adhered more strongly t…

UNLABELLED: Streptococcus mutans (S. mutans) has a superior ability to rapidly metabolize sucrose into exopolysaccharides (EPS, mainly glucans), which serve as a critical virulence factor related to dental caries. Despite extensive research on sucrose-dependent EPS at the molecular and macroscale levels, however, the mechanisms underlying EPS effects at the microscale level remain poorly understood. Here, by employing bacteria tracking and fluorescence staining techniques, we investigated the role of sucrose-dependent EPS during biofilm development of S. mutans at the microscale level for both…

Dental caries remains a major oral health problem, necessitating effective natural antimicrobial alternatives targeting Streptococcus Mutans. Chewing sticks from Mangifera indica and pongamia pinnata contain bioactive compounds with established antiseptic and bactericidal properties. Aqueous extracts (5%, 10% and 50% w/v) were prepared and tested against Streptococcus mutans using agar diffusion to assess zones of inhibition. Mangifera indica showed maximum antimicrobial efficacy at 50% concentration, followed by lower but notable effects at 10% and 5%. Thus, we show Mangifera indica as a pote…

Streptococcus mutans, is considered the main microbial etiological agent of dental caries, therefore it has been proposed as a useful predictor of caries risk as well as a target for caries prevention strategies. We aimed to compare the genomic characteristics of S. mutans strains isolated from individuals with high and low caries risk, in order to determine their genotypic features related to dental caries in adults. A total of 25 S. mutans isolates, obtained from the saliva of 13 volunteers with high dental caries activity and 12 caries-free individuals, were analysed using whole-genome sequ…

Streptococcus mutans, a Gram-positive pathogen, is a primary causative agent of dental caries. It modifies the oral biofilm architecture on tooth enamel and, like other bacteria, requires transition metal ions such as Zn(II), Cu(II), and Ni(II) for survival and virulence. Physiological salivary Zn(II) levels are insufficient for optimal bacterial growth, prompting S. mutans to develop a specialized ABC transport system comprising AdcA, AdcB, and AdcC. Among these, the lipoprotein AdcA plays a pivotal role in Zn(II) acquisition. In this study, we examined two probable Zn(II)-binding sites in Ad…

Staphylococcus aureus, an opportunistic bacterium found in the oral cavity, has been reported as a causative agent of infective endocarditis and pneumonia. Salt is an essential mineral for cell maintenance in the human body. This study was conducted to clarify how salt affects the formation of biofilms by S. aureus and Streptococcus mutans, pathogens implicated in dental caries. Bacteria were cultivated with various concentrations of NaCl on a 96-well microtiter plate in tryptic soy broth with 0.25% sucrose or 0.25% glucose (TSBs and TSBg, respectively) for 16 h. The effects of glucosyltransfe…
