Glycerol activates tissue-nonspecific alkaline phosphatase for temperature and bone homeostasis.

Tissue-nonspecific alkaline phosphatase (TNAP) has roles in bone mineralization and the futile creatine cycle, which fuels uncoupling protein 1 (UCP1)-independent thermogenesis in adipocytes. Hussain and colleagues provide evidence for allosteric activation of TNAP by glycerol and demonstrate that this activation is necessary for both bone mineralization and thermogenesis.

Transcriptome Inference and Systematic Approaches to Investigate TCM Compounds With Beneficial Metabolic Effects.

Obesity is a major risk factor for type 2 diabetes. Some traditional Chinese medicine (TCM) compounds can improve obesity by increasing energy consumption. This study aimed to identify and verify TCM compounds that promote adipose thermogenesis to alleviate obesity via transcriptomic analysis, molecular docking, network pharmacology, and Connectivity Map (CMap) analysis. Thirty-six TCMs related to adipose tissue thermogenesis were first collected to generate transcriptomic data. Then a ranking method based on integrated transcriptomic data was used to select emodin from the TCM Da Huang for ne…

DGAT-driven futile lipid cycling has a pronounced, yet concealed, thermogenic function.

Thermoregulation is an essential yet incompletely understood homeostatic process in mammals. UCP1-mediated thermogenesis, while efficient, is dispensable, suggesting the existence of alternative mechanisms. Using a pharmacogenetic approach, we show that the adipose tissue futile lipid cycling (FLC) contributes to UCP1-independent thermogenesis, with DGATs being involved in the regulation of FLC. The loss of DGAT-driven FLC-mediated thermogenesis is compensated for by the hierarchical recruitment of alternative mechanisms such as shivering and enhanced lipid catabolism mediated by AMPK activati…

A TREK that activates thermogenic adipose tissue by sensing cold air in the lung.

Inhaling cold air triggers adipose tissue thermogenesis. In this issue of Developmental Cell, Jiang et al. utilized the cold exposure to lung only (CELO) system to demonstrate that cold air activates lung club cells via TREK-1 potassium channels.1 Cold exposure closes TREK-1 channels, causing membrane depolarization, which increases irisin production and induces adipose thermogenesis.

Control of metabolic and ventilatory responses to cold in anesthetized cats.

Interactions between the control of thermogenesis and ventilation were studied during normoxia, hyperoxia, and ambient or CO hypoxia in adult anesthetized intact or carotid-denervated cats. Shivering, metabolic and ventilatory responses to cold stress were studied. In addition, the effects of transient pharmacological stimulation (NaCN) or inhibition (Dopamine) of arterial chemoreceptor activity were studied under different levels of oxygenation. In intact animals, cold exposure provoked increases in VO2 and ventilation which were directly proportional to the intensity of shivering. During amb…

Enhancing tolerance to cold exposure--how successful have we been?

The risk of accidental hypothermia is always present in persons living at high latitudes, with cold water immersion representing the most extreme challenge. While most of the effort concerned with protection against cold exposure has involved finding ways of decreasing heat loss by improving insulation some attempts have been made in finding ways of improving cold tolerance by modifying the thermoregulatory response to cold. The main strategies that have been used are: - thermal acclimation, physical exercise, dietary enhancement of thermogenesis, pharmacological enhancement of thermogenesis a…

Suppression of hypothalamic oestrogenic signal sustains hyperprolactinemia and metabolic adaptation in lactating mice.

17β-oestradiol (E2) inhibits overeating and promotes brown adipose tissue (BAT) thermogenesis, whereas prolactin (PRL) does the opposite. During lactation, the simultaneous decline in E2 and surge in PRL contribute to maternal metabolic adaptations, including hyperphagia and suppressed BAT thermogenesis. However, the underlying neuroendocrine mechanisms remain unclear. Here, we find that oestrogen receptor alpha (ERα)-expressing neurons in the medial basal hypothalamus (MBH), specifically the arcuate nucleus of the hypothalamus and the ventrolateral subdivision of the ventromedial hypothalamus…

[Discovery of heat-induced calcium release via ryanodine receptors and a novel mechanism of thermal runaway].

Regulation of thermogenesis in mammals is essential for maintaining body temperature homeostasis under fluctuating environmental temperature. Impairments in this regulation can lead to severe conditions, including fever or heatstroke. This review focuses on malignant hyperthermia (MH), a pathological escalation of thermogenesis in skeletal muscle. It highlights the role of type 1 ryanodine receptor (RYR1), a Ca2+ release channel, based on our recent studies. Previous studies have revealed that genetic mutations in RYR1 are associated with muscle disorders including MH, which are characterized …

Role of Neuro-Immune Cross-Talk in the Anti-obesity Effect of Electro-Acupuncture.

There is evidence to show that electro-acupuncture (EA) has a promotive effect on both lipolysis and thermogenesis, and that these mechanisms underlie the anti-obesity effect of EA. The sympathetic nervous system (SNS) is known to play a role in thermogenesis. Additionally, obesity is characterized by a chronic low-grade inflammatory state. Based on these findings, the aim of the present study is to investigate the potential neuro-immune mechanisms underlying the therapeutic effect of EA in obesity. In the experiment, we used a high fat diet (HFD) rats model to study the effect of EA in reduci…

A hypothalamic neuronal population coordinates metabolic adaptations to cold.

Cold exposure increases feeding, thermogenesis, and energy expenditure to maintain body temperature and energy balance, but the underlying neural mechanisms remain unclear. Here, we identify cold-responsive neurons in the dorsal posterior periventricular hypothalamus that coordinate these adaptations. Activation of GABAergic or cold-activated neurons increased food intake, body temperature, and brown adipose tissue thermogenesis, whereas their inhibition attenuated cold-induced hyperphagia and thermogenesis. Single-nucleus RNA sequencing identified Lef1 as a marker of cold-activated neurons in…

Single cell analysis reveals immune cell-adipocyte crosstalk regulating the transcription of thermogenic adipocytes.

Immune cells are vital constituents of the adipose microenvironment that influence both local and systemic lipid metabolism. Mice lacking IL10 have enhanced thermogenesis, but the roles of specific cell types in the metabolic response to IL10 remain to be defined. We demonstrate here that selective loss of IL10 receptor α in adipocytes recapitulates the beneficial effects of global IL10 deletion, and that local crosstalk between IL10-producing immune cells and adipocytes is a determinant of thermogenesis and systemic energy balance. Single Nuclei Adipocyte RNA-sequencing (SNAP-seq) of subcutan…

Gene expression and metabolite levels converge in the thermogenic spadix of skunk cabbage.

The inflorescence (spadix) of skunk cabbage (Symplocarpus renifolius) is strongly thermogenic and can regulate its temperature at around 23 °C even when the ambient temperature drops below freezing. To elucidate the mechanisms underlying developmentally controlled thermogenesis and thermoregulation in skunk cabbage, we conducted a comprehensive transcriptome and metabolome analysis across 3 developmental stages of spadix development. Our RNA-seq analysis revealed distinct groups of expressed genes, with selenium-binding protein 1/methanethiol oxidase (SBP1/MTO) exhibiting the highest levels in…

Anatomical projections of the dorsomedial hypothalamus to the periaqueductal grey and their role in thermoregulation: a cautionary note.

The DMH is known to regulate brown adipose tissue (BAT) thermogenesis via projections to sympathetic premotor neurons in the raphe pallidus, but there is evidence that the periaqueductal gray (PAG) is also an important relay in the descending pathways regulating thermogenesis. The anatomical projections from the DMH to the PAG subdivisions and their function are largely elusive, and may differ per anterior-posterior level from bregma. We here aimed to investigate the anatomical projections from the DMH to the PAG along the entire anterior-posterior axis of the PAG, and to study the role of the…

Brainstem catecholaminergic neurons induce torpor during fasting by orchestrating cardiovascular and thermoregulation changes.

Torpor, an adaptive hypometabolic state in response to fasting, is characterized by pronounced reductions in body temperature, heart rate, and thermogenesis. However, how the brain orchestrates these physiological changes to induce torpor and the relationships among them remain elusive. Inhibiting catecholaminergic (CA) neurons in the ventrolateral medulla (VLM) significantly impairs torpor in mice, while their activation reduces body temperature, heart rate, energy expenditure, physical activity, and thermogenesis. Importantly, the heart rate decline precedes body temperature reduction, resem…

Increased thermogenic capacity of brown adipose tissue under low temperature and its contribution to arousal from hibernation in Syrian hamsters.

Brown adipose tissue (BAT) is thought to play a significant physiological role during arousal when body temperature rises from the extremely low body temperature that occurs during hibernation. The dominant pathway of BAT thermogenesis occurs through the β(3)-adrenergic receptor. In this study, we investigated the role of the β(3)-adrenergic system in BAT thermogenesis during arousal from hibernation both in vitro and in vivo. Syrian hamsters in the hibernation group contained BAT that was significantly greater in overall mass, total protein, and thermogenic uncoupling protein-1 than BAT from …

Pituitary and autonomic responses to cold exposures in man.

This review presents hormonal responses to various cold exposures and their calorigenic effects in man and some animals. Previous studies in rats have shown that cold exposures activate the hypothalamic-pituitary-thyroid axis. Increased thyroid hormone concentrations lead to heat production via general stimulation of metabolism (obligatory thermogenesis) and possibly via activation of thyroid hormone receptors and uncoupling protein 1 (UCP 1) and deiodinase enzyme genes in the brown adipose tissue (BAT). In human subjects long-term cold exposures do not seem to activate the pituitary-thyroid a…
